Effect of Growth Regulators of Plant Regeneration from Rhodiola sachalinesis leaf segments (홍경천 (Rhodiola sachalinesis)의 엽육 절편으로부터 식물체 분화에 미치는 생장조절제의 영향)

  • Rhodiola sachalinensis has been used as a traditional medicine in Asia. We were germination in vitro seedling of grow naturally in Chang bai Moutain. And callus induction from leaf segments, treatmented plant regeneration in plant growth regulators (Auxins and cytokinins). We investigated optimal conditions for efficient plant regeneration through callus induction and shoots formation on medium with various kinds of growth regulators. Callus induction and adventitious shoots formation was achieved when cytokinin and auxin combinated to this experiment. Especially, there was the highest callus induction rates when we were used to 1 mg/L kinetin and 2 mg/L NAA $(98\%)$, Adventitious shoots formation wear obtained difference rate when cytokinin alone 1 mg/L BA $(96.6\%)$. And regenerated plantlet was acclimatized and transplanted to the soil, showed $100\%$ survival.

  • Iris dichotoma Pall. is an important endangered plant belonging to the family Iridaceae. A method was developed for the rapid micropropagation of I. dichotoma through plant regeneration from leaf, rhizome, and root explant-derived calli. Leaf, rhizome, and root segments were cultured on Murashige and Skoog (MS) medium supplemented with 2,4-dichlorophenoxy acetic acid (2,4-D; $0-3.0mg{\cdot}L^{-1}$) for callus induction. Callus production was highest at $1.0mg{\cdot}L^{-1}$ 2,4-D, where 73.8% and 45.5% of cultured rhizome and root cuttings, respectively, produced calli. The viable calli were maintained at an induced concentration of 2,4-D ($3.0mg{\cdot}L^{-1}$). They were then transferred to MS medium supplemented with various concentrations of 2,4-D ($0-3.0mg{\cdot}L^{-1}$) in combination with 6-benzyladenine (BA: 0, 1.0 and $3.0mg{\cdot}L^{-1}$) for adventitious shoot regeneration. The addition of a low concentration of 2,4-D into BA-containing medium significantly increased the frequency of shoot regeneration in leaf, rhizome, and root-derived calli. The highest number of adventitious shoots (26.4 per callus) formed at $0.5mg{\cdot}L^{-1}$ 2,4-D and 1.0 mg/l BA. For rooting of the shoots, half- strength MS medium supplemented with different concentrations of indole 3-butyric acid (IBA) $0-3.0mg{\cdot}L^{-1}$ was tested. The optimal results were observed using half-strength MS medium supplemented with $1.0mg{\cdot}L^{-1}$ IBA, on which 98% of the regenerated shoots developed roots with an average of 3.5 roots per shoot within 45 days. The plantlets raised in vitro were acclimatized and transferred to soil with 95% success. This in vitro propagation protocol will be useful for conservation and mass propagation of this endangered plant.

Medium Composition and Growth Regulator on Organogenesis Platycodon grandiflorum (Jacq.) A. DC. with Yellow Green Petals ('녹색 꽃잎 도라지'의 기관분화에 미치는 배지조성 및 생장조절제의 영향)

  • This study was carried out to determine the optimal medium composition and growth regulators for the micropropagation of Platycodon grandiflorum (Jacq.) A. DC. Nodes containing yellow green petals were used as plant materials to execute the study. The best performance of adventitious root development was found in 1/4 strength of MS basal salt and the growth was satisfactory in the concentration of 1/2 MS medium. The best condition for adventitious root development and growth was observed in the higher concentration (5%) of sucrose and activated charcoal free 1/4MS medium respectively. Adventitious roots were developed at the controlled culture medium at pH 4.8 with a tendency of suppression with higher levels of pH. However, it was prevailed that the development and growth depended on the concentration of agar. The lower concentration of agar (0.4%) was performed better than that of higher concentration (1.2%), whereas the agar concentration (0.4%) showed the best performance for the development and growth of adventitious roots. For the development of shoots containing node, BA combined with IAA was more effective than kinetin with IAA or NAA. The highest shoot development (3.9 shoots per explant) was performed on MS medium supplemented with 0.1 mg/L BA and 0.5 mg/L IAA.

Soil Acclimatization of Calanthe discolor through Multiple Shoot Formation from Tissue Culture (새우난초(Calanthe discolor)의 조직배양으로부터 다신초형성을 통한 토양순화)

  • This experiment was conducted to establish the micropropagation of Calanthe discolor through multiple shoot formation from the culture of leaf, corm and root explants. Frequency of adventitious shoot formation from leaf explants was higher than those of corms and root explants. Frequency of adventitious shoot formation on medium with various concentrations of BA (0. 1.0, 3.0, and 5.0 mg/L) and NAA (0, 0.1, 0.5, and 1.0 mg/L) was tested. The maximun induction of adventitious shoot was obtained on half strength Murashige and Skoog (MS) medium supplemented with 3.0 mg/L BA and 1.0 mg/L NAA after 6 weeks of culture. Multiple shoots were transferred onto half strength MS medium with various concentrations of GA3 (0, 0.1, 0.5, 1.0, 3.0, and 5.0 mg/L). The number and length of multiple shoots on medium were highest on medium with 3.0 mg/L GA3. All the adventitious shoot grew well and rooted on half strength MS medium with 3.0 mg/L NAA. The plantlets were acclimatized up to 100% on sand with TKS-II or pearlite with TKS-II.

Factors Affecting Organogenesis from Mature Cotyledon Explants and Regeneration in Soybean

  • A successful, efficient system for multiple shoot induction and plant regeneration of soybean (Glycine max) was established. Four soybean genotypes were compared for organogenic responses on various media cultured under light conditions. The adventitious shoots (98%, 2.6 shoots/cotyledon) directly from one-day-old cotyledon after germination induced by the hormone treatment and its efficiency was higher than any other conditions. The optimal medium for the induction of multiple shoots from cotyledon in Pungsannamulkong(shoot formation rate, 98%), Lx 16 (83%) and IIpumgeomjeongkong(63%) was MS medium supplemented with 2 mg/L BAP, but for Alchankong(75%), MS medium supplemented with 1mg/L zeatin and 1mg/L IAA, 3% sucrose, 4% Phytagel. Higher root induction (88%) was observed from the shoots placed on rooting medium (hormone-free MS basal). Plantlets were transferred onto the same medium supplemented with 1% activated charcoal for further development. With this treatment, regenerated plantlets were obtained within 7-8 weeks (shoot induction for 4 weeks, rooting and shoot elongation for 3-4 weeks).

  • Holostemma annulare (Family Asclepiadaceae) is an invaluable vulnerable medicinal plant; the root tubers are used in Ayurveda medicine and by folk healers to treat various ailments. In this study, Schenk and Hildebrandt medium fortified with the cytokinins 6-benzyl adenine, kinetin, and auxins, including indole 3-butyric acid, indole 3-acetic acid, α-naphthaleneacetic acid, and 2,4-dichlorophenoxyacetic acid, were checked for their efficiency on root tuber induction from different explants. Adventitious root tubers were more successfully induced from in vitro leaf segments and shoots when cultured in Schenk and Hildebrandt medium supplemented with 0.5 mg/l of α-naphthaleneacetic acid. In addition, preliminary phytochemical analysis of in vitro root tubers and identification of different secondary metabolites were conducted. Thin layer chromatography and high performance thin layer chromatography analysis of the crude methanolic extracts of the in vitro root tuber identified the presence of lupeol, a bioactive triterpene. Adventitious root tuber induction offers a novel method for the in vitro production of bioactive metabolites that can be scaled up by bioreactors, thus ensuring the conservation and sustainable utilization of H. annulare. The study warrants further scale-up production and pharmacological investigation that can be extended for pharmaceutical needs.

Plant Regeneration Via Organogenesis on Petiole of Centella asiatica (L.) Urban

  • An efficient plant regeneration of C. asiatica was achieved from organogenesis using petiole explants of in vitro plantlet on MS basal medium controled with different plant growth regulators (NAA,2,4-D, IAA kinetin, and BA). Best results that 50%, efficiency of regeneration per explant for regeneration were obtained with IAA $17.13\;{\mu}M$ and BA $8.9\;{\mu}M$. Formation of adventitious shoots via organogenesis from the petiole explant was verified by histological sectioning of plantlets. Regenerated plants were transplanted into soil.

Shoot Proliferation and Plant Regeneration from Suspension-Cultured Cells of Dianthus gratianopol (패랭이꽃속 Dianthus gratianopol의 현탁배양세포로부터 Shoot 증식과 식물체 재분화)

  • Conditions for efficient organogenesis and plant regeneration from Dianthus gratianopol suspension cultured cells were established. Shoot-forming calli of glossy surface, pale green and knobby type were selected from leaf explant-derived calli and were suspension-subcultured every week in CP liquid medium with 1.0 mg/L 2,4-D and 0.5 mg/L BAP. Combinations of 1.0 mg/L 2,4-D and 0.5 mg/L BAP, and 1.5 mg/L 2,4-D and 0.5 mg/L BAP were effective for the induction of regenerative callus from the suspension cultured cell clusters. Multiple shoot primordia were initiated from the green spots of these regenerative callus and formed shoots on MS medium with 1.0 mg/L TDZ and 0.5 mg/L PAA. Shoot regeneration frequency (calli regenerating at least one shoot) was about 87%. For plant regeneration, proliferated shoots were excised and transferred to MS medium with 0.1 mg/L NAA for root initiation after 9 weeks of culture. The regenerants were potted in soil and formed the flowering buds and petals. Also, adventitious shoots were formed from the excised green shoot primordia of regenerative callus and these shoots proliferated successfully and regenerated to whole plants.

Multiple Shoot Formation by Apical Meristem Culture in Ipomoea batatas Poir. (고구마 정단분열 조직배양에 의한 多芽體 형성)

  • In sweet potato cultivars 'Mokpo #29' and 'Sanchunza', shoots from extplants were formed 100% on the MS medium with 0.1 ㎎/L NAA and 2.0 ㎎/L BA after 30 days of culture and roots produced from the base of stem at frequencies of 66.7% ('Mokpo #29') and 69.2% ('Sanchunza'), respectively, The media with 0.5∼4.0 ㎎/L BA were produced the greatest frequency of multiple shoot and the most of shoots developed rapidly into normal plantlets with rooting within 60 days of culture. Whereas the cultivar 'Keumsi' failed to produce normal shoot multiplication on the medium with cytokinins alone because of callusing of adventitious shoots. When single shoots with 1 to 2 nodes were excised from the multiple shoot or shoots covered with callus devoid of root and transferred to MS medium with 4.0 ㎎/L BA or kinetin. Host divided shoots showed the callus induction at the stem base and it was enable to obtain regenerated plantlets with shoot and root normally.

Plant Regeneration Depending on Explant Type in Chrysanthemum coronarium L.

  • Plant regeneration depending on explant type was inves-tigated with cotyledon, hypocotyl, and leaf explants of garland chrysanthemum (Chrysanthemum coronarium L.) cultured on MS basal medium supplemented with various concentrations of SAP and NAA combination. Among the three different types of explants, hypocotyl explants grown on MS medium containing $1.0{\mu}M\;NAA,\;1.0{\mu}M\;BA\;and\;6{\mu}M\;AgNO_3$ produced the highest adventitious shoots (4.67 per explant). Hypocotyl explants not only produced more vigorous shoots, which regenerated aster than the cotyledon and leaf explants. An efficient root formation was observed in MS medium containing $3\%$sucrose. The concentration of NAA did not show significant effects on root formation. Results from this experiment suggested that hypocotyl explants were efficient for the regeneration of garland chrysanthemum.
