Micropropagation of Ulmus davidiana Planch was established via adventitious shoot formation from the segments of adventitious roots. Adventitious roots were produced directly from root segments of seedlings on a 1/2 SH medium plus various concentrations of IBA. The maximum growth of adventitious roots was observed in the presence of 2.0 mg/L IBA. After the segments of adventitious roots were cultured on various cytokinins (zeatin, 2-iP, BA, kinetin) and cytokinins plus auxin (IBA), formation of adventitious shoot was investigated. Among cytokinin treated, kinetin was the most effective on both adventitious shoot induction and number of shoots. Especially, 2.0 mg/L kinetin was the best to increase adventitious shoot induction (95.8%) and a number of shoots (8.4). Adventitious shoots were rooted on 1/2 WPM medium and the plantlets were acclimated 100% on composed soil (peatmoss : vermiculite 1 : 1).
Traditional culture technology of medicinal plants mainly depends on the field culture, which has many problems. With progress of modern culture technology, it has become possible to produce valuable secondary metabolites from medicinal plants. In this paper, we discuss about the pigment and saikosaponin production from too medicinal plants, Carthamus tinctorius and Bupleurum falcatum, through bioreactor culture system. A two-stage bioreactor culture system was established for the production of yellow and red pigments and saikosaponins by cell suspension cultures of Carthamus tinctorius and Bupleurum falcatum. In Carthamus tinctorius, balloon type airlift bioreactors and column type airlift bioreactors were employed for the tell culture and for the pigment production, respectively. The greatest pigment production was obtained on White medium supplemented with 4 mg/L kinetin, high levels of sucrose concentration and photosynthetic photon flux. In Bupleurum falcatum, adventitious roots were cultured in balloon type airlift bioreactors and the root growth was greatest on SH medium containing 5 mg/L IBA and 0.2 mg/L kinetin. HPLC analysis showed that the contents of main active saikosaponins a, c, and d in adventitious roots were almost the same as those in field cultured root.
Rhodiola sachalinensis is one of the most popular oriental medicines in East Asia. It is a perennial herb, belonging to the family Crassulaceae, which is mainly distributed in mountains at the altitudes of 1700-2500 m in Baek-Du mountain. Cultivation of this species for the production of medicine materials is not easy in nature, because of restrict habitats. In vitro production of roots can be applicable for the production of medicinal materials. Here, we investigated the optimal conditions for induction and proliferation of adventitious roots in in vitro culture system. Leaf, stem and root segments from R. sachalinensis were cultured on Murashige and Skoog(MS) medium supplemented with the various concentrations of IBA(Indole-3-butyric acid)(0.5, 1.0, 3.0, and 5.0 mg/L) and sucrose(10, 30, 50, and 100 g/L). The optimal explant for adventitious root induction was leaf segment. Induction of adventitious roots was highest on MS medium supplemented with 5.0 mg/L IBA and sucrose 30 g/L. In liquid culture, fresh weight of adventitious roots was highest(15.65 g) on 1/3 strength MS liquid medium supplemented with 5.0 mg/L IBA with 30 g/L sucrose. These results revealed the first attempt for the production of adventitious roots in R. sachalinensis.
The type of air lift bioreactor affected the root growth in ginseng adventitious root cultures. Among bioreactors used in this experiment, bulb type bubble bioreactor (BU) was the best to increase root growth (41.92 g dry weight). The kLa value representing the oxygen transfer capacity from medium to explants (6.98 h$^{-1}$ ) in BU with 5 cm bubble column was higher than other bioreactors. On the other hand, cylindric tube bioreactor (CT) without bubble column resulted in minimum root growth (38.55 g dry weight) and kLa value (5.25 h$^{-1}$ ). Furthermore, the root growth (50.30 g dry weight) in BU with 10 cm bubble column more increased than 5 cm bubble column. However, the kLa value do not affected the secondary metabolite such as ginsenosides. These results show that the bubble column in air lift bioreactor increase kLa value and increased kLa value stimulate the growth of ginseng adventitious roots.
This study was carried out to detennine the optimal submerged culture conditions for the growth increase of ginseng adventitious roots containing germanium by means of a fractional factorial design with four factors and three levels, using the response surface methodology (RSM). The ginseng (Panax ginseng CA. Meyer) adventitious roots were induced by plant growth regulators and cultured in a liquid SH medium. The effects of various $GeO_2$ and phosphoric acid ($H_3P0_4$) concentrations in the medium, $GeO_2$ addition time and the pH of the medium on the fresh weight of the ginseng adventitious roots were investigated. The optimum pH of the medium and the phosphoric acid concentration detennined by the partial differentiation of the model equation were 4.7 and 6.0 roM, respectively. The predicted optimal $GeO_2$ concentration was 10 ppm and the $GeO_2$ addition time did not affect the growth of ginseng adventitious roots. Under these conditions, the growth of the ginseng adventitious root containing germanium was predicted to be 2.47 g.
In order to study the effect of cyclohexylamine (CHA) and polyamines during adventitious root formation, the correlation of adventitious root formation with endogenous polyamine content was investigated in inoculated soybean cotyledons at the rooting medium supplemented with CHA, spermidine and spermine. Adventitious root formation was inhibited in medium containing 10$^{-3}$ -10$^{-2}$ M CHA. Adventitious root formation was not formed in treatment with 10$^{-2}$ M CHA plus polyamine. It was inhibited in 2.5 $\times$ 10$^{-3}$ M CHA alone and reverved in 2.5 $\times$ 10$^{-3}$ M CHA plus 10$^{-4}$ M spermine. Putrescine content was lower in 10$^{-2}$ M CHA and 10$^{-2}$ M CHAplus 10$^{-4}$ M spermine treatment than in control after 3 days of incubation. However, it was higher in 2.5 $\times$ 10$^{-3}$ M CHA and 2.5 $\times$ 10$^{-3}$ M CHA plus 10$^{-4}$ M spermine treatment than in control. Spermine content was higher in all CHA treatments than in control, while spermidine content was lower than in control.
Nitric oxide (NO) is a potent antihypertensive and vasodilator which plays an important role in regulating vascular tones. In this study, we investigated the effects of adventitious root extracts of wild ginseng on NO production and NO linked physiological activities. When human endothelial cell line (ECV304) was incubated with either water extracts of wild ginseng adventitious root (WE) or aqueous fraction of butanol extracts of wild ginseng adventitious root (ABE), considerable amounts of NO were released by the cells. The level of endothelial nitric oxide synthase (eNOS) expression was unchanged and about 6% of the angiotensin converting enzyme (ACE) was inhibited with treatment of ABE. The vasodilating activities of pulmonary artery rings in response to different doses of extracts were shown as 44.8% and 91.3% in 2.5 mg/ml WE and 0.1 mg/ml ABE, respectively. The blood pressure lowering effect was observed from the oral administered spontaneously hypertensive rat (SHR) with the lowest blood pressure (154.5${\pm}$8.6 mmHg) after 8 h. The blood pressure was recovered to the initial level after 24 h.
This study was carried out to investigate the dose-dependent effect of methyl jasmonate on both the adventitious root growth and the accumulation of various eleutherosides in the bioreactor culture of Eleutherococcus senticosus adventitious roots. The highest biomass production (5.4 g DW/L) was observed in the absence of methyl jasmonate and the root growth was significantly decreased by increasing the methyl jasmonate concentration. However, methyl jasmonate stimulated the production of both eleutheroside B, E and $E_1$. The highest level of eleutheroside B (359.9 ${\mu}g$/g DW) was obtained at 40 ${\mu}M$ of methyl jasmonate, while eleutheroside E and $E_1$ was accumulated at the highest level by the addition of 10 ${\mu}M$ of methyl jasmonate. Total eleutheroside was increased up to 3818.1 ${\mu}g$ per liter when 10 ${\mu}M$ of methyl jasmonate was applied. In addition, when the adventitious roots were cultured with 20 ${\mu}M$ of methyl jasmonate, the highest levels of eleutheroside B, E and $E_1$ were observed at the 12th, 3th and 9th days of culture, respectively.
This study describes culture conditions for a plant regeneration system via a combined pathway of somatic embryogenesis and organogenesis in root explant cultures of the commercial rose cultivar 'Charming'. Root explants formed white calluses at a frequency of 30% after 6 weeks of culture on Schenk and Hildebrandt (SH) medium supplemented with $11mg\;1^{-1}$ 2,4-dichlorophenoxyacetic acid. After 6 weeks of transfer to SH medium without growth regulators, initial white calluses gave rise to globular somatic embryos at a frequency of 2.8%, which were subsequently dedifferentiated to embryonic tissues. Somatic embryos or embryonic tissues initially derived from root explants did not undergo development beyond cotyledonary stage. To produce adventitious shoots, embryonic tissues were sliced and cultured on SH medium with $0.5mg\;1^{-1}$ 6-benzyladenine. After 4 weeks of culture, 28% of embryonic tissue explants formed adventitious shoots. Regenerated shoots were rooted on half strength SH medium with $0.1mg\;1^{-1}$${\alpha}-naphthalaneacetic$ acid and subsequently grown to maturity. Root-derived embryonic tissues were proliferated by subculture, while retaining the capacity for shoot production for a few years.
