Emulsification of Crude Oil by Acinetobacter sp. SH-14

  • As basic study to evaluate the treatability of oil-contaminated environment with bacteria, isolation and characterization of crude oil-degrading bacterium were carried out. A bacterial strain SH-14 capable of degrading crude oil was isolated from contaminated soils by enrichment culture technique and identified as Acinetobacter sp. by morphological, cultural and biochemical characteristics, and so named Acinetobacter sp. SH-14. The optimal medium composition and cultural conditions for the growth and emulsification of crude oil by Acinetobacter sp. SH-14 used were crude oil of 2.0%, $KNO_3$ of 0.2%, $K_2HPO_4$ of 0.05%, and $MgSO_4\;{\cdot}\;7H_2O$ of 1.0%, along with initial pH 7.0 at $30^{\circ}C$. Acinetobacter sp. SH-14 showed to be resistant to chloramphenicol and utilized various hydrocarbons such as dodecane, hexadecane, isooctane, cyclo-hexane etc., as a sole carbon source. Acinetobacter sp. SH-14 harbored a single plasmid. By agarose gel electrophoresis and curing experiment it was found that the genes for crude oil components degradation were encoded on the plasmid.

Effects of Aerosol Colistin Treatment of Pneumonia Caused by Multi-drug Resistant Acinetobacter baumannii (다약제 내성 Acinetobacter baumannii 에 의한 폐렴에서 Colistin 분무치료의 효과)

  • Background: Acinetobacter infections are difficult to treat as they often exhibit multiple resistance to the antibiotics that are currently available for the treatment of pneumonia. Colistin is active against gram-negative bacteria, including the multiple drug resistant (MDR) Acinetobacter species. However, intravenous administration of colistin was abandoned because of its nephrotoxicity and neurotoxicity. The aims of this study were to examine the efficacy and safety of colistin administered by aerosol in the treatment of pneumonia caused by MDR Acinetobacter baumannii. Methods: We retrospectively reviewed the medical records of patients admitted to the intensive care unit (ICU) from Dec. 2006 to Aug. 2007 who had been diagnosed as suffering from pneumonia due to MDR Acinetobacter baumannii and had been treated with nebulized colistin. Results: 31 patients received aerosolized colistin. The average duration of the treatment was $14{\pm}7$ days and the daily dose of ranged from 225 mg to 300 mg. All patients received concomitant intravenous antimicrobial agents. The average length of the stay in the ICU was $34{\pm}21$ days and in the hospital $58{\pm}52$ days. The overall microbiological eradication was observed in 25 patients (80.6%). 14 of these (56%) were cured, and 11 (44%) were infected with other microorganisms. The overall crude mortality of the ICU was 48%. Nephrotoxicity and significant bronchial constriction did not occur in any patient during neublized colistin treatment. Conclusion: Nebulized colistin may be a safe and effective option in the treatment of pneumonia due to MDR Acinetobacter baumannii. Its role in therapy warrants further investigation in comparative studies.

Interactions between Indole-3-acetic Acid Producing Acinetobacter sp. SW5 and Growth of Tomato Plant (Indole-3-acetic acid를 생성하는 Acinetobacter sp. SW5와 토마토 식물 간의 상호작용)

  • Many rhizobacteria can promote plant growth through various direct or indirect mechanisms, and their production of phytohormones such as indole-3-acetic acid (IAA) may have pronounced effects on growth and development of plants. Rhizobacterial strain isolated from rhizosphere of foxtail (Setaria viridis), Acinetobacter sp. SW5 produced 118.1 mg/L of IAA and 4.5 mg/L of gibberellin ($GA_3$) in brain heart broth medium at 2 and 1 day of incubation, respectively. In a pot test the lengths of stem and root and fresh weight of the germinated tomato seedlings treated with Acinetobacter sp. SW5 significantly increased by 26.3, 33.3, and 105.3%, respectively compared to those of the uninoculated control in 12 weeks of cultivation. When the root exudate secreted from tomato seedlings was analyzed by HPLC, 3.75 ng mg tomato $root^{-1}$ of tryptophan which is an IAA precursor was detected. Acinetobacter sp. SW5 could produce $4.06{\mu}M$ of IAA from root exudate from 8 tomato seedlings. Together with the capability of growth of Acinetobacter sp. SW5 in the tomato root exudates, this IAA secreted by bacteria might contribute to enhance the growth of tomato plants.

Biochemical and cultural characteristics of mineral-solubilizing Acinetobacter sp. DDP346 (미네랄 가용화능을 갖는 Acinetobacter sp. DDP346의 생화학적 및 배양학적 특성)

  • In this study, to select strains suitable as microbial agent from among rhizosphere microorganisms present in rhizosphere soil and roots, the mineral solubilization ability, antifungal activity against 10 types of plant pathogenic fungi, and plant growth-promoting activity of rhizosphere microorganisms were evaluated. As a result, DDP346 was selected because it has solubilization ability of phosphoric acid, calcium carbonate, silicon, and zinc; nitrogen fixing ability; production ability of siderophore, indole-3-acetic acid, and aminocyclopropane-1-carboxylate deaminase; and antifungal activity against seven types of plant pathogenic fungi. DDP346 showed a 99.9% homology with Acinetobacter pittii DSM 21653 (NR_117621.1); phylogenetic analysis also revealed a close relationship with Acinetobacter pittii based on the 16S rRNA base sequence. The growth conditions of DDP346 were identified as temperatures in the range of 10-40 ℃, pH in the range of 5-11, and salt concentrations in the range of 0-5%. In addition, a negative correlation coefficient (r2 = -0.913, p <0.01) was shown between pH change and the solubilized phosphoric acid content of Acinetobacter sp. DDP346, and this is assumed to be due to the organic acid generated during culture. Consequently, through the evaluation of its mineral solubilization ability, antifungal activity against plant pathogenic fungi, and plant growth-promoting activity, the potential for the utilization of Acinetobacter sp. DDP346 as a multi-purpose microbial agent is presented.

  • Comparative biodegradation studies of liquid and solid alkanes and of two different solid alkanes were conducted by an isolated Acinetobacter sp., which degraded crude oil alkanes simultaneously. for the determination of degradation mechanism of hydrophobic crude oil constituents. Also a model oil experimental system composed of a solid alkane. heneicosane, as a substrate and a non-degradable non-aqueous phase liquid. pristane, as an oil matrix was established and studied. It was proposed that the Acinefobacter sp. utilized hydrophobic substrates directly on the surface of them with no difference in the degradation rates between the liquid and solid alkanes. On the basis of the results from the heneicosane/pristane system which imitates crude oil matrix containing solid constituents. the crude oil matrix was considered to reduce the bioavailability of contained substrates by reducing the specific surface area of substrates to contact with microorganisms.

Carbon Monoxide Dehydrogenase in Cell Extracts of an Acinetobacter Isolate (Acinetobacter sp.1의 일산화탄소 산화효소의 특성)

  • Extracts of CO-autotrophically grown cells of Acinetobacter sp. 1 were shown to use thionin, methylene blue, or 2,6-dichlorophenol-indophenol, but not NAD, NADP, FAD, or FMN, as electron acceptors for the oxidation of CO under strictly anaerobic conditions. The CO dehydrogenase (CO-DH) in the thes bacterium was found to be an inducible enzyme. The enzyme activity was determined by an assay based on the CO-dependent reduction of thionin. Maximal reaction rates were found at pH 7.5 and $60^{\circ}C$, and the Arrhenius plot revealed an activation energy of 6.1 kcal/mol(25.5kJ/mol). THe $K_m$ m/ for CO was $154{\mu}M$. Known metalchelating agents tested had no effects on the CO-DH activity. No divalent cations tested affect the enzyme activity significantly escept $Cu^{2+}$ which suppressed the activity completely. The enzyme was inhibited by glucose and succinate. The same extracts catalyzed oxidation of hydrogen gas and formate with thionin as electron acceptor. The CO-DH of Acinetobacter sp. 1 was to have no immunological relationship with that of Pseudomonas carboxydohydrogena.

Distribution of Antimicrobial Resistant Genes in Acinetobacter calcoaceticus-baumannii Complex Isolated from Clinical Specimens in Chungcheong, Korea (충청지역의 임상검체로부터 분리된 Acinetobacter calcoaceticus-baumannii Complex를 대상으로 항균제 내성 유전자 비교분석)

  • Species that belong to the Acinetobacter calcoaceticus-baumannii (Acb) complex are major causes of hospital-acquired infections. They are important opportunistic pathogens. These species are usually multidrug resistant (MDR), and the therapeutic options to treat the infections caused by these species are limited. In the present study, we investigated fluoroquinolone resistance mechanisms in 53 ciprofloxacin resistant Acinetobacter species isolates in Chungcheong, Korea. Antimicrobial susceptibilities were determined using the disk-diffusion method. Detections of genes and identification of mutations associated with fluoroquinolone resistance were carried out using PCR and DNA sequencing. In our study, 47 out of 53 ciprofloxacin resistant Acinetobacter isolates harbored sense mutations at the 83rd residue (serine to leucine) in the gyrA gene as well as at the 80th residue (serine to leucine) in the parC gene. Among the 47 isolates harboring sense mutations in gyrA and parC gene, 44 isolates were A. baumannii and 3 isolates were A. pittii. Plasmid-mediated quinolone resistance (PMQR) determinants were detected in isolates in our study. Among the 46 ciprofloxacin resistant A. baumannii isolates, 41 showed type A, B, or F banding patterns on their REP-PCR profiles. This result suggests that clonal relation and horizontal spreading of the bacterial isolates have been around hospitals in Chungcheong area. To prevent colonization and disseminations of fluoroquinolone resistance Acb complex isolates, continuous investigation and monitoring of antimicrobial resistant determinants of MDR isolates are needed.

Effect of Phosphate Solubilizing Bacteria Application on Soil Chemical Properties and Chrysanthemum Growth in Greenhouse Cultivation Area with High Salt Accumulation (염류집적 국화 시설재배지 토양에서 인산분해미생물 시용이 토양화학성 및 국화생육에 미치는 영향)

  • This study was carried out to develop the soil amendment practice by phosphate solubilizing bacteria application in greenhouse chrysanthemum cultivation area with high salt accumulation. The experimental site (ShinWoo Flower, GwangJu) has been cultivated chrysanthemum for 15 years and showed significant salt accumulation. The phosphate solubilizing bacteria, Pseudomonas putida (KSJ11), Acinetobacter calcoaceticus (KSJ3) and Acinetobacter calcoaceticus (WP20) formulated on vermiculite for easy use, were applicated. Each 250L of phosphate solubilizing bacteria was applied for $82m^2$ before planting. Acinetobacter calcoaceticus (KSJ3; WP20) increased the amount of soluble phosphorus in an effective level. Particularly, Acinetobacter calcoaceticus (WP20) increased not only the level of soluble phosphorus but also potassium, calcium and magnesium resulting in the increase of EC in the soil. The level of nematode was also decreased with the non-treated increased. As a result, we suggest that selected phosphate solubilizing bacteria (WP20) could be a useful practice for soil amendment in chrysanthemum plantation soil and provided an opportunity to reduce the use of the fertilizer during the cultivation period.

Enzymological Localization of Carbon Monoxide Dehydrogenases in Pseudomonas carboxydovorans and Acinetobacter sp.1 (Pseudomonas carboxydovorans와 Acinetobacter sp.1의 일산화탄소 산화효소의 세포내 분포에 대한 효소학적 연구)

  • The localization of carbon monoxide dehydrogenases (CO-DHs) in Pseudomonas carvoxydovorans and Acinetobacter sp.1 was examined by comparison of the distribution of CO-oxidizing activity between soluble and particulate fractions obtained after disruption of CO-grown cells by sonic oscillation and of spheroplasts by osmotic shock. When the cells were broken by sonic oscillation, most of the CO-DH activity was recovered from soluble fractions. However, disryption by osmotic lysis of spheroplasts revealed that the enzyme activity is present in the cell membrane. The results indicated the CO-DHs in these cells are loosely attached to the cytoplasmic membrane.

Isolation and Characterization of Acinetobacter sp. WC-17 Producing Chitinase

  • The bacterial strain WC-17 able to produce chitinase was isolated from soil using an enrichment technique. The isolated strain was identified as Acinetobacter sp. judging by their morphological and physiological characterisitics. The optimal culture conditions for the production of chitinase of Acinetobacter sp. WC -17 are 1.5% colloidal chitin and 1 % tryptone at $30^{\circ}C$ with pH 6.5. Since the enzyme was rapidly produced in a culture supplied with chitin, glucose, or N-acetylglucosamine but not with other polymers and monosaccharide, the enzyme was considered to be an inducible enzyme. Notably N- acetylglucosamine and glucose were found to be effective inducers at low concentrations but repressors at excessive concentrations. The cultural supernatant of Acinetobacter sp. WC-17 inhibited the growth of phytopathogenic fungi such as P.oryzae, R.solani, and F.solani. Among the phytopathogenic fungi tested, P.oryzae was the most sensitive. The conventional agar plate (PDA containing 1 % colloidal chitin) method also produced the same result.
