Physiological activities of natural color powders and their mixtures (천연소재로부터 분리한 색소분말과 혼합물의 생리활성)

  • Seven kinds (acacia, cochineal, catechu, grape peel, persimmon, gallnut and clove) of color powders obtained from natural resources and their mixtures were evaluated for their biological activities, such as antioxidant (ABTS and DPPH radicals scavenging), cholesterol absorption, and COX-2 inhibitory activities. Catechu, gallnut and clove were selected for the further studies due to its the best activities. The cholesterol absorption, COX-2 inhibitory, ${\alpha}$-amylase inhibitory and ${\alpha}$-glucosidase inhibitory activities were measured using the mixtures of catechu, gallnut and clove. The ABTS radical scavenging activity of the seven types of natural color powders were higher than the DPPH radical scavenging activity. The cholesterol absorption activity was significantly higher in cloves. The COX-2 inhibitory activity was significantly lower in acacia. The ${\alpha}$-amylase inhibitory activity was higher in catechu and gallnut, indicating that there were no significant difference between two mixtures. The ${\alpha}$-glucosidase inhibitory activity was the highest in catechu, which was higher than that of the catechu-containing mixtures. From all these results, a synergistic effect could be obtained when utilizing a mixture of powders rather than using only individual type. Since the activity of each powder was different, further studies will be required for clarifying the interactions between mixtures.

Antioxidant and Nitrite Scavening Activity and α-Glucosidase Inhibitory Effect of Water Extract from Schizandra chinensis Baillon (오미자 열수 추출물의 항산화 활성과 아질산 소거능 및 α-Glucosidase 저해 효과)

  • The nutraceutical role of omija (Schizandra chinensis Baillon) water extract (OWE) was determined through the analysis of antioxidant activity, nitrite scavening activity, and xanthine oxdiase and $\alpha$-glucosidase inhibitory effects. Antioxidant activity of OWE was measured by using 2,2-diphenyl-1-picryl-hydrazyl (DPPH) free radical scavenging activity and superoxide dismutase-like activity (SODA). DPPH radical scavenging activity and SODA increased in a dose-dependent manner, and was about 49.0% at 2.5 mg/mL and 69.2% at 5 mg/mL, respectively. The xanthine oxidase and $\alpha$-glucosidase inhibitory activities of OWE were about 88.8% and 86.2% at 1 mg/mL, respectively. Nitrite scavenging activity of OWE was about 54.9%, 42.4%, and 34.2% on pH 1.2, 3.0, and 6.0 at 1 mg/mL, respectively. These results suggest that OWE has a strong antioxidant activity, and xanthine oxidase and $\alpha$-glucosidase inhibitory effects.

Antioxidant activity and inhibition activity against α-amylase and α-glucosidase of Smilax China L. (청미래덩굴(Smilax China L.) 추출물의 항산화 및 α-amylase와 α-glucosidase 저해활성)

  • This study was carried out to analyze the effects of water and 70% ethanol extract on the antioxidative and antidiabetic activities of Smilax china L., a vine shrub belonging to the lily family. The activities of the extracts were measured based on the total phenolic and flavonoid contents and through on the results of the antioxidant tests, such as the electron-donating ability, ABTs radical scavenging activities, SOD-like activity, xanthine oxidase inhibition effect, antioxidant protection factor (PF), TBARs content and ACE inhibition activity, and ${\alpha}$-glucosidase, and ${\alpha}$-amylase inhibition activity. The resulting total phenolic and flavonoid contents of the 70% ethanol extract from S. china L. were greater than those of the water extract from S. china L. With regard to the results of the antioxidant tests, such as the electron-donating ability, ABTs radical scavenging activity, SOD-like activity, xanthine oxidase inhibition effect, antioxidant protection factor (PF), and TBARs content, those from the 70% ethanol extract from S. china L. were greater than those from the water extract from S. china L. Also, with regard to the ACE inhibition effect and ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibition, those from the 70% ethanol extract from S. china L. were greater than those from the water extract from S. china L. All these findings show that the 70% ethanol extract from S. china L. has greater antioxidative and antidiabetic effects and can be used as a preventive agent for oxidation and diabetes.

Screening of α-Amylase, α-Glucosidase and Lipase Inhibitory Activity with Gangwon-do Wild Plants Extracts (강원도 자생 산채 추출물의 α-Amylase, α-Glucosidase, Lipase 효소 저해활성 탐색)

  • We investigated ${\alpha}$-amylase, ${\alpha}$-glucosidase and lipase inhibitory activity of extracts collected from wild plants in Gangwon-do. 90 wild plants were collected and their water and ethanol extracts were obtained. Results of measuring ${\alpha}$-amylase inhibitory activity indicated more than 80% of activity inhibition in 10 mg/mL concentration for ethanol extracts of three plants and water extracts of two plants. For ${\alpha}$-glucosidase inhibitory activity, ethanol extracts of thirteen plants and water extracts of three plants showed more than 80% of activity inhibition in 10 mg/mL concentration. In the experiment of inhibiting lipase activity, ethanol extracts of seven plants and water extracts of one plants showed above 80% of activity inhibition in 10 mg/mL concentration. These results suggest that the selected extracts could be potentially used as a resource of bioactive materials for health functional foods.

Hypoglycemic Effect of the Methanol Extract of Soybean Sprout in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐에 있어서 콩나물 메탄올 추출물의 헐당강하효과)

  • To control blood glucose level as close to normal is the major goal of treatment of diabetes mellitus. $\alpha$-glucosidase is the enzyme to digest dietary carbohydrate and inhibition of $\alpha$-glucosidase could suppress postprandial hyperglycemia. The methanol extract of soybean sprout was tested for the inhibitory activities against $\alpha$-glucosidase in vitro. Soybean sprout extract inhibited yeast $\alpha$-glucosidase activity by 24.5% at the concentration of 5 mg/mL. The methanol extract of soybean sprout was subsequently subjected to sequential fractionation with hexane, ethyl acetate, butanol and water. Among the fractions tested ethyl acetate-soluble fraction showed relatively strong inhibition against $\alpha$-glucosidase by 36.3% at the concentration of 5 mg/mL. Acarbose, standard $\alpha$-glucosidase inhibitor, inhibited $\alpha$-glucosidase activity by 40.1%. The ability of soybean sprout extract to lower postprandial glucose was studied in streptozotocin-induced diabetic rats. Starch solution (1 g/kg) with and without the methanol extract of soybean sprout (500 mg/kg) was administered to diabetic rats after an overnight-fast by gastric intubation. A single oral dose of soybean sprout extract inhibited the increase in blood glucose levels significantly at 60, 90, 120, 180 min (p<0.05) and decreased incremental response areas under the glycemic response curve significantly (p<0.05). These results suggest that soybean sprout might exert hypoglycemic effect by inhibiting $\alpha$-glucosidase activity.

In vitro biological activities of lotus (Nelumbo nucifera) leaves extract (연잎(Nelumbo nucifera) 추출물의 in vitro 생리활성)

  • Antioxidative, anticoagulant, and α-glucosidase inhibitory effects of 60% (v/v) ethanolic extract from lotus (Nelumbo nucifera) leaves were investigated in vitro. The yield and polyphenol content of the lotus leaves extract were 35.3±1.1% and 186.2±5.2 ㎍ gallic acid equivalents/mg, respectively. The lotus leaves extract effectively scavenged free and cation radicals, and nitrite in a concentration-dependent manner, and possessed potent reducing power. The lotus leaves extract had remarkable anticoagulant and α-glucosidase inhibitory activities as a function of the extract concentration.

Screening of Natural Products for Anti-diabetic Activity and Analysis of Their Active Compounds (항당뇨 효능이 있는 천연물의 탐색 및 활성물질의 분석)

  • Modern people have an increased incidence of metabolic diseases due to changed eating habits, and diabetes is considered the most significant metabolic disease. Given that existing diabetes treatments are accompanied by side effects, the aim of this study was to identify traditional natural products that have anti-diabetic activity. The potential anti-diabetic and antioxidant activities of natural products were examined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay, α-glucosidase assay, and protein tyrosine phosphatase 1B (PTP1B) inhibition assay. Methanol extracts of Ulmus davidiana var. japonica, Acer tegmentosum branches, Nelumbo nucifera seeds, and Carthamus tinctorius seeds were found to have high anti-diabetic activity and further fractionated with solvents using ethyl acetate and butanol. Consequently, the ethyl acetate fraction of C. tinctorius seeds (MG-11-E) with high α-glucosidase and PTP1B inhibitory activity was selected. MG-11-E was subjected to preparative thin layer chromatography, and fraction #6 showed high α-glucosidase and PTP1B inhibitory activity. Fraction #6 was analyzed and fractionated via high performance liquid chromatography with 50% methanol as the mobile phase, and anti-diabetic activity was observed in the sample that eluted after 4 min as a single peak. The α-glucosidase inhibitory activity exhibited by this sample seemed to be greater than the PTP1B inhibitory activity; thus, it was concluded that a greater anti-diabetic therapeutic effect may be achieved by combining this agent with natural products that inhibit PTP1B activity.

Antihyperglycemic α-Glucosidase Inhibitory Activity of Ethanol Extract from Neolentinus lepideus (잣버섯(Neolentinus lepideus) 에탄올 추출물의 혈당상승 억제 효과)

  • For development a new anti-diabetic compound from edible mushroom, antihyperglycemic ${\alpha}$-glucosidase inhibitory activities of Pleurotus ostreatus, Pleurotus cornucopiae, Pleurotus salmoneostramineus, Pleurotus eryngii and Neolentinus lepideus were investigated on its water and ethanol extracts. ${\alpha}$-Glucosidase inhibitory activity of Neolentinus lepideus fruiting body showed the highest at 86.3% in the 95% ethanol extracts and water extract from Pleurotus cornucopiae was also higher at 48.5% among water extracts. Therefore, Neolentinus lepideus which showed very high ${\alpha}$-glucosidase inhibitory activity was selected as a new anti-diabetic agent-containing mushroom and the ${\alpha}$-glucosidase inhibitor was maximally extracted when treated with 95% ethanol at $30^{\circ}C$ for 48 hr. The ethanol extracts from Neolentinus lepideus fruiting body showed dosage-dependent hypoglycemic action after administration to 120 min in the SD-rat and streptozotocin-induced diabetic SD-rat.

Improvement of Cellobiose Dehydrogenase(CDH) and $\beta$-Glucosidase Activity by Phanerochaete chrysosporium Mutant (Phanerochaete chrysosporium 변이주에서의 Cellobiose Dehydrogenase(CDH)와 $\beta$-Glucosidase 활성 향상)

  • Cellobiose dehydrogenase(CDH) as a hemoflavoenzyme is secreted out of cell in the cellulose degradation. As CDH strongly bound to amorphous cellulose, it helps cellulose hydrolysis by cellulase. CDH may have an important role of saccharification process for bioethanol production. In this study, Phanerochaete chrysosporium ATCC 32629 was selected for the production of CDH among other strains tested. The optimal temperature and pH of CDH produced by P. chrysosporium ATCC 32629 were ${55^{\circ}C}$ and 4, respectively. To improve the activity of CDH, the mutation of P. chrysosporium was performed using proton beam that has high energy level partially. As a result, P. chrysosporium mutant with the high activity was selected at 1.2 kGy in a range of 99.9% lethal rate. The CDH and $\beta$-glucosidase activities of mutant were 1.4 fold and 20 fold higher than those of wild strain. Therefore, P. chrysosporium mutant with the high activities of CDH and $\beta$-glucosidase was obtained from mutation by proton beam irradiation.

Production and Anti-hyperglycemic Effects of ${\alpha}$-Glucosidase Inhibitor from Yeast, Pichia burtonii Y257-7 (효모 Pichia burtonii Y257-7에 의한 ${\alpha}$-Glucosidase 저해제의 생산 및 식후 혈당 상승 억제 효과)

  • In order to develop a new anti-diabetic ${\alpha}$-glucosidase inhibitor, we compared the ${\alpha}$-glucosidase inhibitory activity of the cell-free extracts of 48 strains of yeasts isolated from Korean fermented foods, and found that Pichia burtonii Y257-7 exhibited the highest ${\alpha}$-glucosidase inhibitory activity of 55.6%. The ${\alpha}$-glucosidase inhibitor was maximally produced when Pichia burtonii Y257-7 was cultured in LB broth (initial pH of 6.0) at $28^{\circ}C$ for 24 h. The ${\alpha}$-glucosidase inhibitor, partially purified by Sephadex G-50 gel permeation chromatography and systematic solvents extraction, revealed potent hypoglycemic effects in normal rats and streptozotocin-induced diabetic rats after the oral administration of starch.
