Phylogeny of the Yeast Species Isolated from Wild Tiger Lily (Lilium lancifolium Thunb.) (야생 참나리(Lilium lancifolium Thunb.)로부터 분리한 효모의 분자계통학적 분석)

  • BACKGROUND: Yeast isolates associated with the leaves, stems, and flowers of the tiger lily needed to be identified using isolation methods that have previously been used effectively in yeast biotechnology. A culture-based approach was necessary for the isolation of many yeast strains associated with tiger lily. METHODS AND RESULTS: In this study, the homogenized leaves, stems, and flowers of tiger lily were spreaded onto GPY medium containing chloramphenicol, streptomycin, Triton X-100, and L-sorbose. A total of 82 yeast strains from the leaves, 94 and 97 yeast strains from the stems and flowers were isolated, respectively. Yeast isolates were identified by phylogenetic analysis based on internal transcribed spacer region sequencing. The yeast species isolated from the leaves comprised of 31 isolates of the genus Pseudozyma, 28 of Aureobasidium pullulans, and 11 of the genus Cryptococcus. Those isolated from the stems comprised of 40 of A. pullulans and 11 of Cryptococcus, and 95 of A. pullulans While, 1 isolate each of the genera Rhodotorula and Metschnikowia were isolated from the flowers. CONCLUSION: We identified site-specific yeast communities associated with tiger lily. These yeast isolates may have high potential for application in the field of biotechnology.

Isolation and Characteristics of ${\varepsilon}$-Caprolactam Utilizing Bacteria (${\varepsilon}$-Caprolactam 이용성(利用性) 세균(細菌)의 분리(分離) 및 그 성질(性質))

  • A bacterium which utilizes ${\varepsilon}$-caprolactam as a sole source of carbon and nitrogen was isolated from sludge of Shinchun river in Taegu and identified as Arthrobacter globiformis N-2-l. The growth medium for the optimum culture condition was composed of 0.4% ${\varepsilon}$-caprolactam, 0.02% $K_2HPO_4$, 0.05% $KH_2PO_4$, 0.02% $MgSO_4{\cdot}7H_2O$, 0.01% $FeCl_3{\cdot}6H_2O$ and 0.05% yeast extract. The optimum pH and temperature for growth were 7.0 and $30^{\circ}C$ respectively. The bacterial growth on the ${\varepsilon}$-caprolactam medium did not require any other organic nitrogen source such as yeast extract, although it was remarkably stimulated by the yeast extract. The bacteria utilized wide range of sugars and organic acids such as ${\alpha}$-ketoglutarate, adipate and P-hydroxybenzoate. The bacteria could use all kind of amino acids, ${\varepsilon}$-Caprolactam in the medium was consumed completely in the timecourse culture at $30^{\circ}C$ for 60 hr on the shaker by the bacteria. Decomposition product of ${\varepsilon}$-caprolactam by Arthrobacter globiformis N-2-1 was ${\varepsilon}$-aminocaproic acid.

The Removal of Toluene by a Granular Activated Carbon Bioreactor using Yeast (Yeast와 입상활성탄을 이용한 미생물반응기의 휘발성유기화합물 분해 특성)

  • A liquid culture of yeast "Candida tropicalis" was used in a fluidized bioreactor to achieve high removal efficiencies of volatile organic compounds (VOCs). In this study, granular activated carbon (GAC) was used as a fluidized material to improve adsorptive capacity as well as mass transfer of gaseous toluene, the model VOC. The GAC fluidized bioreactor demonstrated toluene removal efficiencies ranging from 50 to 80%, when inlet toluene loading varied in a range between 13.1 and 37.4 g/m$^3$-hr. The maximum elimination capacity determined in the GAC fluidized bioreactor was 172 g/m$^3$-hr at a toluene loading of 291 g/m$^3$-hr. Transient loading experiments revealed that the removal efficiency was remained unchanged during an increased loading period, and toluene introduced to the bioreactor was first absorbed to GAC and then slowly desorbed and became available to the yeast culture. Hence the fluidized GAC helped to achieve an improved mass transfer between the gas and liquid phases, resulting in high toluene removal capacity. Consequently, the GAC fluidized bioreactor using C. tropicalis can be successfully applied for the removal of VOCs, and is a feasible alternative over conventional processes such as packed-bed biofilters.

Effect of Medium Compositions on the Mycelial Growth of Inonotus obliquus (차가버섯의 균사체 성장에 대한 배지성분의 영향)

  • Effect of temperature($22-32^{\circ}C$), pH(5-7) and medium composition on the mycelial growth for the submerged culture of Inonotus obliquus. The concentrations of glucose, starch, peptone, yeast extract, $K_2HPO_4$, $MgSO_4{\cdot}7H_2O$ and $CaCl_2$ were examined in the ranges of 30-120 g/L, 0-10 g/L, 0-20 g/L, 0-15 g/L, 0-2 g/L, 0-1.5 g/L and 0-0.5g/L, respectively. The maximum mycelial growth of Inonotus obliquus was obtained for $26-27^{\circ}C$ and pH 6. The concentrations of glucose, yeast extract and $CaCl_2$, which gave the maximum mycelial growth of Inonotus obliquus, were 70 g/L, 5 g/L and 0.1 g/L, respectively. In the cases of starch, peptone and $K_2HPO_4$, the mycelial growth of Inonotus obliquus increased with increasing the concentrations. However, as the concentration of $MgSO_4{\cdot}7H_2O$ increased, the mycelial growth of Inonotus obliquus decreased. The medium for maximum mycelial growth of Inonotus obliquus consisted of (per 1 L): glucose, 70 g; peptone, 5-20 g; starch, 10 g; yeast extract, 5 g; $K_2HPO_4$, 2 g and $CaCl_2$, 0.1 g.

Characteristics of Culture and Isolating Lactic Acid Bacteria and Yeast from Sourdough (Sourdough로부터 젖산균과 효모의 분리 및 배양 특성)

  • This study was conducted to investigate the optimum conditions for the medium composition after isolating and identifying yeast and lactic acid bacteria from sourdough. It was found that the best quality lactic acid bacterium with acid product and flavor was identified as Leuconostoc species among isolated 115 lactic acid bacteria, the best Quality yeast with good fermentation and flavor was identified as Saccharomyces species among isolated 8 yeast. While the microbial growth with glucose or sucrose as sugar source was good, it was, selected that sucrose which is using commercially is better than glucose. The growth of lactic acid bacterium ; was good with 1% added sucrose whereas yeast needed more growth. Additionally, the medium for the optimum 1 growth of the yeast was composed of 0.5% wheat flour, 0.5% peptone and 3% sucrose, whereas lactic acid bacterium was composed of 0.5% wheat flour and 1% sucrose without peptone.

Purification and Characterization of ${\beta}-galactosidase$ from Nuruk Yeast (누룩 Yeast에서 유당분해효소의 분리 및 특성)

  • A strain of Nuruk yeast No. IS (NY-15) which produced high activity of ${\beta}-galactosidase$ was isolated from Nuruk, and the crude enzyme was prepared by whey permeate culture of the microorganism. The crude enzyme was purified 40-fold with a 7.7% yield by acetone and ammonium sulfate fractionational precipitation, and chromatography on DEAE-cellulose, DEAE-Sephadex A-50 and Agarose-PAPT. Purified ${\beta}-galactosidase$ from Nuruk yeast showed two types of subunit patterns; a slow moving band and a fast moving deeply stained band, both anode·migrating at pH 7.5. The molecular weight of the former was estimated to be about 130,000 and that of the latter was 96,000 by SDS-polyacrylamide gel electrophoresis. The optimum pH of the enzyme activity was 7.5 and maximum activity appeared at $40^{\circ}C$.

Microbiological Characteristics of Alcoholfermenting Wild Yeast, Aureobasidium pullulans P-1 and Its Makgeolli Fermentation Characteristics (알코올 발효성 야생 효모, Aureobasidium pullulans P-1의 균학적 특성과 막걸리 발효 특성)

  • The goal of this study was to investigate the microbiological characteristics of the ethanol-producing wild yeast, Aureobasidium pullulans P-1, isolated from flowers growing near the Yedang reservoir, Chungnam province, Korea, and in addition, to optimize its fermentation ability for the production of Makgeolli. A. pullulans P-1 was oval-shaped, and formed ascospores and pseudomycelium. The P-1 strain was a halophilic and sugar tolerant yeast which grew in 15% NaCl and 50% glucose-containing yeast extract-peptone-dextrose media. The P-1 strain was also resistant to 20% ethanol. Changes of the physicochemical properties during Makgeolli fermentation by A. pullulans P-1 were investigated. A maximum of 8.45% ethanol was obtained when a mixture of cooked rice, 150% water, and 35% ipguk per cooked rice was fermented by 5% A. pullulans P-1 culture broth at $25^{\circ}C$ for 10 days. Antihypertensive angiotensin I-converting enzyme inhibitory activity in the Makgeolli ferment produced by A. pullulans P-1 reached a high of 71.1% after 10 days.

Production and Characterization of Selenium Peptide from Saccharomyces Cerevisiae (효모를 이용한 selenium peptide 생산 및 특성 연구)

  • Selenium containing peptide was produced by culturing yeast with selenium, Selenium was broadly incorporated in the various size of proteins based on the GPC analysis of the total yeast protein. The ratio of selenium to protein increased with the concentration of added selenium in the culture medium. Antioxidant activity (glutathione peroxidase-like activity) was proportional to the concentration of selenium concentration in the peptide. Different size of proteins were obtained by hydrolyzing the total yeast protein by protease XIV. Average molecular weight of selenium peptide was analyzed by GPC. Glutathione peroxidase (GPx) activity of the selenium peptide increased as the size of peptide decreased. Sodium selenite had strong inhibition on the yeast growth than sodium selenate. The ratio of selenium to protein was higher with sodium selenate than with sodium selenite. These results showed the potentials of selenium peptide production by yeast cultivation.

Efficiency of Various Microbial Foods for Tigriopus japonicus Mori (Tigriopus japonicus Mori에 대한 수종 미생물의 이료효과)

  • Microbial organisms including yeast, Acinetobacter sp. AG-3, Chlorococcum sp., Chlorella sp. and some of their combinations were tested to evaluate growth efficiency of Tigriopus japonicus. Body length and weight of the copepoda were measured during four days experiment. Acinetobacter sp. AG-3, dried yeast(produced Wago), Chlorella sp., Chlorococcum sp. and mixed culture were used as food sources. Yeast(Y.) was the most effective food for the growth during nauplius stage and efficiencies of bacteria(Bact.)+chlorococcum sp.(RA), Chlorococcum sp.(RA), Bact.+chlorella sp.(Ch.), Bact. and Ch. decreased in odor, while for the growth of copepodite and adult, Bact.+RA was the most effective food with decreased efficiency of Y., RA, Bact. + Ch., Bact., Ch. in order. The ratio of weight gain to the food uptaken, after the weight and food units were converted to carbon, was between 21.6 and $68.7\%$, This result suggests that some kinds of bacteria, algae and mixed cultural microorganisms could be good food sources for the growth of copepoda.

  • A potent lytic strain was selected by an extensive screening test of microorganisms isolated from soils and sewages on the medium containing baker's yeast as a carbon source. This strain (M-10) was identified to a strain of Humicola sp. by the Genera of Fungi (Clements, 1964). The strain was cultured on the basal medium composed of 2% of baker's yeast, 0.3% of $K_2HPO_4$, 0.01% of $MgSO_4{\cdot}7H_2O$, 0.1% of yeast extract in a shaking incubator. Cultural conditions for lytic enzyme production has been studied, and the results obtained were as follows: 1. The Optimal conditions for lytic enzyme production were: initial pH 5.5 to 6.0, temperature $33^{\circ}C$ in shaking culture. 2. Among the various carbon sources, baker's yeast (4%) was the best for lytic enzyme production, increasing the level of activity eight, times higher than when grown on glucose (1%). 3. The most effective concentration of $K_2HPO_4\;and\;MgSO_4{\cdot}7H_2O$ in the basal medium for lytic enzyme production was 0.1% and 0.01% respectively. 4. When the strain was cultured under the optimal conditions, the production of lytic enzyme was maximized in 72 hours.
