Changes in the Growth and Biochemical Composition of Nannochloropsis sp. Cultures Using Light-Emitting Diodes (LED (Light Emitting Diode)를 이용한 Nannochloropsis sp.의 성장 및 생화학적 조성 변화)

  • The marine microalgae Nannochloropsis sp. was cultured under a metal halide lamp (MH) or light-emitting diodes (LEDs) of various wavelengths (blue, LB; red, LR; yellow, LY; green, LG; white, LW); changes in growth, total carotenoid, chlorophyll a, amino acid and fatty acid profiles were investigated. LB-exposed cultures exhibited the highest specific growth rate (SGR) (0.32), whereas LY-exposed cultures showed the lowest SGR (0.18). After cultivation for 9 days, the maximum dry cell weight (g/L) of LB-exposed cultures was significantly higher than that of those exposed to other light conditions (LB>MH>LW$${\geq_-}LG{\geq_-}$$LR>LY). The essential amino acid (EAA, %) contents of cultures exposed to LG, LB, LR and MH were higher than those exposed to LY and LW (P<0.05). Eicosapentaenoic acid and n-3 highly unsaturated fatty acid levels were significantly higher in MH-exposed cultures compared to those exposed to LY (P<0.05), with no marked difference compared to those exposed to LB, LG, LR and LW (P>0.05). The total carotenoid content was highest in LR-exposed cultures (18.0 mg/L), whereas MH showed the lowest (11.8 mg/L; P<0.05). Chlorophyll a content was highest in cultures exposed to LR compared to other light sources. These results suggest consistent differences in growth and biochemical composition after exposure to light of different wavelengths.

Influence of denitrified biofloc water on the survival rate and physiological characteristics of Pacific white shrimp juveniles, Litopenaeus vannamei (바이오플락 탈질수가 어린 흰다리새우, Litopenaeus vannamei의 생존율 및 생리특성에 미치는 영향)

  • This study investigates the effect of denitrified biofloc water on changes in the water quality parameters and the physiological characteristics of shrimps. Biofloc rearing water contains a large number of microorganisms and can rapidly stabilize the water quality and energy saving if reusable due to high water temperatures. Rearing water contain floating bacteria with both aerobic and anaerobic bacteria. Therefore, when the carbon source is added in limited air supply, the anaerobic state is activated and the denitrification process is possible. In this study, the denitrification water had the following properties: ammonia (6.9 mg L-1), nitrite (0.3 mg L-1), nitrate concentration (9.2 mg L-1), high pH (8.42) and alkalinity (590 mg L-1). The experimental group consisted of seawater (SW, control), a mixture of Seawater and denitrified biofloc water (DNW) in the ratio of 3:1, 1:1 and DNW only. All experiments were done in triplicate. As a result, the survival rate never changed even when 100% of the denitrification water was utilized. However, a body fluid analysis showed that creatine and BUN were increased due to index of stress and the tissue damage resulting from the high denitrified water content. Body fluid ions (Na+, K+, and Cl-) significantly decreased as the denitrified water content increased. It was recommended that the denitrification water be mixed with a certain ratio (less than 50%) in the future as it may affect the osmotic pressure control in shrimps.

Effect of Feeding on Postlarvae of Pacific White Shrimp, Litopenaeus vannamei during the Acclimation Process to Low Salinities in Seawater (해수 저염분 순치과정에서 먹이섭취가 흰다리새우, Litopenaeus vannamei 유생에 미치는 영향)

  • This study focused on the effects of feeding on postlarvae of shrimp, Litopenaeus vannamei, during the identified acclimation time to low salinity. A total of 5 different salinity groups with or without feeding (32, 24, 16, 8, and 2 psu, 1 liter, triplicates) were prepared, and 30 shrimp were settled at PL21 (postlarvae) and placed in each group. After 24 hours of the experimentation process, the survival rate of the fed and starved groups was observed to be lower in the 2 psu group compared to other salinity groups, with the rate of 86.6% and 81.1%, respectively. The condition index of glucose and triglyceride, which are important factors for osmoregulation and as energy sources, was 4.2-7.6 times and 2.7-3.4 times higher in the fed groups than the starved groups at all the levels of salinities. The creatine level increased by 1.1-1.5 times in the starved groups as compared to the fed groups. Likewise, the activity of all the digestive enzymes like, lipase, ${\alpha}$-amylase, trypsin, and alkaline protease were clearly higher in the fed groups (ANOVA, p<0.05). Apparently, it was observed that feeding is effective for the postlarvae of shrimp, which shows a characteristic fast metabolism and larval development, during the acclimation period to low salinity.

  • Diffusion of SO4-- ion in hardened cement paste with slag and siliceous powders such as silica fume and white carbon was investigated. Ca(OH)2 from hardenend cement paste was dissolved by sea-water and then gypsum was formed from the reaction of Ca++ in hardenend cement paste and SO4-- ions in MgSO4 solution. A part of the gypsum by reaction with calcium aluminate hydrates formed ettringite. Amounts of SO4-- ions passed through hardened cement paste was less than that of Cl- ions(Dcl-) in hardened cement paste were 0.1∼0.6${\times}$10-11$\textrm{cm}^2$/sec and 1∼4${\times}$10-8$\textrm{cm}^2$/sec respectively.

Occurrence of Pontoniine Shrimp, Periclimenes brevicarpalis (Decapoda: Caridea: Palaemonidae) in Korean Waters

  • Two specimens of pontoniine shrimp, Periclimenes brevicarpalis (Schenkel, 1902), having a symbiotic relationship with sea anemones are reported for the first time in Korea. The specimens were collected by SCUBA diving in Jejudo Island. It has a transparent body with a few large white spots on the carapace, abdomen, telson and uropods. Blue bands are on the cheliped and pereiopods. Five brown eyespots with orange centers are on the telson and uropods. The morphology is described and illustrated with a color image of the living specimens. Two species of Korean Periclimenes, P. ornatus, and P. brevicarpalis, can be distinguished by the position of the anterior dorsolateral spine of the telson. This study extends its previously known range from Japan to Korea. Korean pontoniine now includes six species belonging to five genera of Conchodytes, Cuapetes, Onycocaris, Periclimenaeus, and Periclimenes.

  • A single specimen of Epinephelus lanceolatus, belonging to Epinephelinae of the family Serranidae, was newly collected from Jeju Island, South Korea, in December 2009. Epinephelus lanceolatus is characterized by anterior lateral-line scales with branched tubules, cycloid scales, irregular white spots, and fins with numerous small black spots. We propose a Korean name, "Dae-wang-ba-ri' for this species.

  • Breadmaking is a complex system in which many variables govern the production of acceptable bread. Lipids, a minor component of wheat flour, function importantly in bread-making. Shortening, or fat, is one of the essential ingredients in commercial baking. Beneficial shortening effect depends on type and quantity of lipids present in wheat flour and also on wheat flour quality. Surfactants have been used in baking industry during last decade because certrain surfactants can replace shortening and/or natural flour lipids. A proper combination of lipidshortening-surfactant is more useful in the production of specialty breads such as whole wheat breads, high protein breads, high fiber breads or even non-wheat composite breads rather than in the production of regular white breads. This presentation is a review of recent studies on the contribution of flour lipids, shortening, and surfactants, alone or in combination in the production of breads; illustrations are mainly from data obtained in the author's laboratory.

  • In this study of statistical characteristics of roll response of ships to narrow band exciting moment generated by passing white noise through a linear filter is investigated. The parameters of linear filler are determined by comparing the results of exciting moment generated through filler equation tilth those evaluated from JONSWAP spectrum. The statistical results of the roll response of shops are presented.

Production of Healthy Shrimp(Penaeus chinensis, Penaeus japonicus) Seedling Free from White Spot Baculovirus(WSBV) (White Spot Baculovirus(WSBV) 미감염 새우(Penaeus chinensis, Penaeus japonicus) 종묘 생산)

  • For the production of healthy shrimp(Penaeus chinensis, Penaeus japonicus) seedlings free from the white spot baculovirus(WSBV), we tried to disinfect shrimp eggs with iodine. A relative hatching rate of over 50% has resulted from the treatments of fertilized eggs of fresh shrimps(P. chinensis) with an effective povidon-iodine concentration of 20-200 ppm for 30 seconds and 60 seconds, respectively. The 60 sec treatment group with a 20 ppm effective povidon-iodine concentration showed nearly a 50% relative hatching rate. The over 50 ppm treatment group, however, did not result in hatching show a relative hatching rate. These hatching rates were similar to those in the fertilized eggs of kuruma prawns treated with povidon-iodine solutions. In case of washing fertilized eggs with UV-irradiated seawater 33.3% relative hatching rate in fresh shrimps was much lower than 55.2% in kuruma shrimps. Treatment fertilized eggs of 2 species of shrimp with 200 ppm iodine for 30 sec did not affect the survival rate of the larvae until the early stage of the post larvae even though the hatching rates were low. In order to produce healthy shrimp seedlings non-infected by WSBV, the shrimp eggs should be washed with UV-irradated seawater and sterilized with a low povidon-iodine concentration for a short time.

Effect of Soluble-silicate or Chitosan Foliar Spray on Ginseng Cultivated in Blue-white Plastic Film House

  • The experiments were performed in the Jinan (elevation: 300 meters above sea level), Jeollabuk-do. Seedlings (n = 63 per $3.3m^2$) of ginseng cultivar (Cheonpung, Yeonpung) were planted on April 10, 2015. Shading material of plastic film house was blue-white film. Before the Planting seedling, silicate (3 kg/10 a) or chitosan (40 kg/10 a) was fertilized and foliar sprayed on the leaves 1000 times dilution solution once a month from May to September every year. The growth results of 5-year old ginseng surveyed in 2018 are as follows. The average air temperature in the plastic film house was the highest at $26.6^{\circ}C$ and $26.5^{\circ}C$ in July and August, respectively, and the highest temperature was $40.5^{\circ}C$ in July. The maximum daily temperature of $35^{\circ}C$ or more was 30 days, with the average soil temperature being $24.9^{\circ}C$ in August. The chemical properties of the test soil are as follows. pH was 6.4~6.9 level and EC was 0.35~0.46 dS/m. The organic matter content was 33.5~41.4 g/kg, and available-P content was 251.9~306.8 mg/kg. Exchangeable cations contents, such as K, Ca and Mg were all the appropriate ranges. The soil microbial density surveyed by the dilution plate method was 10~50 times higher than that of control (Non-treatment) and actinomycete density was 3~6 times higher. Pathogens of the genus Fusarium by Metagenome analysis decreased 91.3% and 68.2% respectively in the foliar sprayed of chitosan and soluble-silicate. The light intensity (PAR) in the blue-white film plastic film house gradually increased until July and then decereased, with the average of light intensity in March-October was $120.3umol/m^2/s$. The growth of aerial parts such as plant height and stem length was better than non-sprayed group in silicate or chitosan treatments and Yeonpung cultivar was superior to the Cheonpung cultivar. The SPAD value was higher in Yeonpung cultivar foliar sprayed with soluble-silicate. The growth of underground parts such as root length and taproot length were better in chitosan and soluble-silicate treatment than control, especially in Yeonpung cultivar foliar sprayed with chitosan was good in taproot length and taproot diameter, and fresh weight of root was 60.1 g. Ginsenoside contents were 24.9 mg/g and 22.4 mg/g, in the Cheonpung cultivar foliar sprayed with soluble-silicate or chitosan respectively, 28% and 15% higher than control (19.5 mg/g). The incidence of disease by Alteraria panax and Botrytis cinerea was 3~9% and 4~9%, respectively. High temperature damage rate was 3~5%.
