A Study on Nano-emulsion for Enhanced Transdermal Delivery of Hippophae rhamnoides Leaf Extract (비타민나무 잎 추출물의 피부 흡수 증진을 위한 나노에멀젼 연구)

  • In this study, nano-emulsions containing 0.01, 0.03, 0.05, and 0.10% ethyl acetate fraction of Hippophae rhamnoides (H. rhamnoides) leaf extracts were prepared. The particle size, particle size distribution and skin permeability of the nano-emulsions were evaluated for five weeks. Nano-emulsion was prepared by the sequential use of homogenizer and microfluidizer. Nano-emulsion containing the ethyl acetate fraction exhibited a monodispersed form. Nano-emulsion containing 0.03% ethyl acetate fraction was the most stable for five weeks. The in vitro skin permeation study of nano-emulsion containing 0.03% ethyl acetate fraction was carried out using Franz diffusion cell. The nano-emulsion showed a better skin permeability than that of O/W emulsion. These results indicate that the nano-emulsion containing the ethyl acetate fraction of H. rhamnoides leaf extract showed a remarkable stability and skin permeability than that of O/W emulsion.

Effects of HLB value on oil-in-water emulsions: Droplet size, rheological behavior, zeta-potential, and creaming index

  • Using mixed nonionic surfactants Span/Tween, we investigated the effects of HLB value on the O/W emulsion stability and rheological behaviors. In this study, MS-01 (Span 60 & Tween 60) and MS-02 (Span 80 & Tween 80) was used as mixed nonionic surfactants. We considered required HLB value 10.85 and selected corresponding HLB value range 8-13. The droplet size distributions, droplet morphology, rheological properties, zeta-potential and creaming index of the emulsion samples were obtained to understand the mechanism and interaction of droplets in O/W emulsion. The results indicated that optimal HLB number for O/W emulsions was 10.8 and 10.7, while using MS-01 surfactant and MS-02 surfactant respectively. MS-01 (HLB = 10.8) sample and MS-02 (HLB = 10.7) sample showed smallest droplet size and highest zeta-potential value. Rheological properties are measured to understand rheological behaviors of emulsion samples. All emulsion samples showed no phase separation until 30 days storage time at $25^{\circ}C$.

  • Long chain alcohols, the mixtures of 1-hexadecanol/1-octadecanol, were used as cosurfactants for O/W emulsion prepared with glycerol monostearate/POE(100) monostearate mixed nonionic surfactants, and the phase behavior and flow properties of O/W emulsions were observed. The transition temperature of long chain alcohol was varied with the composition of 1-hexadecanol/1-octadecanol and had the lowest value when the mixed ratio of 1-hexadecanol/1-octadecanol was 2/1. The liquid crystalline phase was formed as the addition of long chain alcohol and the secondry droplet, the flocculate of the emulsion particles, was made, and thus the viscosity of the emulsion was increased. When the temperature of emulsion system was under the transition temperature of long chain alcohol, the mobility of hydrocarbon group of long chain alcohol was restricted, and thus gel structure was formed and the viscosity of the the O/W emulsion was increased, but along with the time, the liquid crystalline phase was disappeared and the viscosity of emulsion was decreased. Long chain alcohol/nonionic surfactants/water formed the liquid crystalline phase when the long chain alcohol was added above the saturation point of solution(2 wt% in this experoment), and the secondry droplet didn't formed when the long chain alcohol was added more than a certain amount (10 wt% in this experiment).

  • An experimental research on the preparation of zinc oxide fine particles in w/o emulsions was conducted. Precipitation solutions were zinc nitrate aqueous solutions with hexamethylenetetramine(HMTA) as precipitant. The precipitation solutions formed stable w/o emulsions with kerosine in the presence of Span 80. Homogeneous precipitation reaction occurred in the w/o emulsion after the resultant w/o emulsion was heated above the decomposition temperature of HMTA and zinc oxide particles were precipitated. In some case, zinc oxide particles of bi-modal distribution were obtained. However, zinc oxide fine particles of narrow particle size distribution could be obtained, even when the initial zinc concentration of precipitation solution and the conversion to zinc oxide are both higher that those in bulk homogeneous precipitation.

Determination of Emulsion Stability Index in W/O Emulsion (유중수적형 유화계내에서의 유화안정지수 산출법의 확립)

  • To evaluate the emulsion stability indices of W/O emulsion system, we developed the simple and sensitive "VOLUMETRIC METHOD". This technique involved the first step of homogenizing the milk fat-water system with Ultra-turrax T25, then the volume of the added water phase was measured immediately. After quiescent incubation in test tubes at room temperature for a desired storage time, the bottom volume of the separated water layer was measured. And then "emulsion stability index(ESI)" was calculated by the following equation : $ESI=(1-V_s/V_a){\times}100$, where $V_a$ means the volume of the added water in the W/O emulsion and $V_s$ represents the volume of the separated water in the W/O emulsion for a desired storage time. The emulsion stability indices of W/O emulsion system at sorbitan trioleate, span 60, and tween 20 were $95.4{\pm}1.8$, $56.1{\pm}2.8$, and $41.6{\pm}2.2$ respectively. Furthermore, the differences between "VOLUMETRIC METHOD" and "Titus et al method" were less than 5.0 of ESI Value.

Moisturizing Effect and Durability of Sun Protection Factor (UVA/B) Activity with Multiple Emulsion (W/O/W) System (멀티플 에멀전(W/O/W) 시스템을 이용한 자외선차단성능(UVA/B)의 내수성과 보습효과)

  • This study is to evaluate moisturizing effect and durability of UV A/B blocking activity with multiple (W/O/W) emulsion system. Most of the sun protective products come to be hot issue having both high SPF and long-lasting activity as using special products when is going out, mountain climbing and sports. Also, many consumers prefer the products which have the excellent waterproofing activity of sun care cosmetics as well as the non-sticky feeling that carried out the study of the sensorial science and texture preference. Therefore, development of the specific formulation using this multiple (W/O/W) emulsion technology, it has O/W type hydro skin feel having soft and moist texture when it is treated on the skin. Finally, this formulation is instantly changed to W/O type feel after adsorbed into the skin. The purpose of this study is to get high SPF lasting effect having high water resistance tactivity with high functional multiple (W/O/W) emulsion cream. We used major ingredients, UV-B absorbers were selected with ethylhexyl methoxycinnamate, isoamyl-p-methoxycinnamate, ethylhexylsalicylate, and octocrylene, UV-A absorbers were selected with butylmethoxydibenzoylmethane, bis-ethylhexyloxyphenol methoxy phenyltriazine. SPF effect of O/W type cream was 34.1. SPF effect of W/O/W type cream was 40.6 (increased about 19%). Water resistance effect after 4 hours, SPF effect of O/W type cream was 3.6 (quickly drop down). SPF effect of W/O/W type cream having 81.0 % waterproofing effect was 32.7 (decreased about SPF 7.9). Moisturizing effect of O/W cream at first was superior comparing multiple emulsion. But after 3 hours quickly was drop-down. Moisturizing effect of multiple emulsion was high comparing O/W type and other sun block creams after 4 hours was constantly maintaining water-content.

Effect of Surfactant Micelle on Lipid Oxidation in Corn Oil-in-Water Emulsion with Phenol Compounds (Phenol성 물질이 첨가된 Corn Oil-in-Water Emulsion의 산화에 미치는 Surfactant Micelle의 영향)

  • The purpose of this research was to determine the effect of phenol compounds from green tea leaves and surfactant micelles on lipid oxidation in com oil-in-water emulsion (O/W). The concentration of phenol and surfactant in continuous phase of the O/W with exceed Brij 700 and phenol compounds was measured. The particle size of O/W with phenol (100 ppm) increased with increasing added exceed surfactant $(0{\sim}2.0%)$ and the concentration of surfactant and phenols in the continuous phase higher than these of control. Lipid oxidation rates, as determined by the formation of lipid hydroperoxides and headspace hexanal, in the O/W emulsions containing phenol compounds (100 ppm) and exceed surfactant $(0{\sim}2.0%)$ decreased with increasing concentration of exceed surfactant. The ability of the phenol compounds and exceed surfactant to inhibit hydroperoxide and headspace hexanal producing as lipid oxidation in O/W was BHT>procyanidin B3-3-O-gallate> (+)-gallocatechin > (+)-catechin and 2% > 1 % > 0% of exceed surfactant. These results indicate that phenol compounds and exceed surfactant could alter the physical location of hydroperoxide in O/W.

Removal study of As (V), Pb (II), and Cd (II) metal ions from aqueous solution by emulsion liquid membrane

  • Emulsion Liquid Membrane (ELM) is a prominent technique for the separation of heavy metal ions from wastewater due to the fast extraction and is a single-stage operation of stripping-extraction. The selection of the components (Surfactant and Carrier) of ELM is a very significant step for its preparation. In the ELM technique, the primary water- in-oil (W/O) emulsion is emulsified in water to produce water-in-oil-in-water (W/O/W) emulsion. The water in oil emulsion was prepared by mixing the membrane phase and internal phase. To prepare the membrane phase, the extractant D2EHPA (di-2-ethylhexylphosphoric acid) was used as a mobile carrier, Span-80 as a surfactant, and Paraffin as a diluent. Moreover, the internal (receiving) phase was prepared by dissolving sulphuric acid in water. Di-(2- ethylhexyl) phosphoric acid such as surfactant concentration, carrier concentration, sulphuric acid concentration in the receiving (internal) phase, agitation time (emulsion phase and feed phase), the volume ratio of the membrane phase to the receiving phase, the volume ratio of the external feed phase to the primary water-in-oil emulsion and pH of feed were studied on the percentage extraction of metal ions at 20℃. The results show that it is possible to remove 78% for As(V), 98% for Cd(II), and 99% for Pb(II). Morphology and swelling property of chitosan microapsules and microbeads prepared by W/O emulsion (W/O 에멀젼에 의한 chitosan microcapsule 및 microbead의 morphology와 팽윤성)

  • Chitosan microcapsules and microbeads were prepared by W/O emulsion method, and their morphologies were observed through SEM. The microcapsules have skin layer of 8 Um and 250 Um of mean diameter, The swelling test showed higher s welling ability in protic solvents than in aphotic solvents. After containing moth-yl violet in the microcapsules, the release patterns were investigated. The results sho wed that the addition of Iysozyme in pH 5.1 acetate buffer accelerated the re-lease rate. In case of the microbeads, the mean diameter was about 70 Um. The surface of the microbeads showed porous structures. The swelling ability of the beads revealed two times higher than the one of the microcapsules.

Development of Water Soluble Tocopherol Emulsion Using Surfactants (계면활성제를 이용한 수분산 Tocopherol 유화물의 개발)

  • This study was carried out to make water-soluble tocopherol emulsion which can be applicable directly in water. The molecular weight of tocopherol was 340 to 360 and tocopherol emulsion model was decided as O/O/W/W type. In correlation between stability of emulsion and surface tension, the stability in surface tension of emulsion was from 40 to 46 dyne/cm. In the case of lower than 40 dyne/cm of surface tension, the stability of the emulsion was lower. Lipophilic surfactants, especially for a polyglycerine polyricinoleate in 20%, 30% and 40% tocopherol emulsion, was the most effective in emulsion stability. A higher stability of the emulsion among hydrophilic surfactants in the tocopherol emulsion was obtained in the following order; polyglycerine monostearate> polyglycerine monooleate> polyoxyethylene (20) sorbitan monooleate$\geq$ polyoxyethylene (20) sorbitan monolaurate.
