2018년 경남 양식어류에서 검출된 병원체 모니터링 (Monitoring of Pathogens Detected in Cultured Fishes of Gyeongnam in 2018)

  • The major cultured marine fishes in sea off the coast Gyeongsangnam-do Province, South Korea, were assessed and included 9.3% rockfish Sebastes schlegelii, 7.8% red seabream Pagrus major, and 2.1% rock bream Oplegnathus fasciatus. The number of insurance payments related to disease mortality in cultured fish in 2017 was fourfold that in 2016. Economic loss in aquaculture due to disease in cultured fish is high and represents an important inhibitory factor affecting marine fishery productivity. In 2018, diseases led to severe production losses in several aquaculture species: 40.0% in rockfish, 11.4% in olive flounder Paralichthys olivaceus, 10.0% in filefish Thamnaconus modestus, and 9.3% in red seabream. Fish-parasitic pathogens such as Microcotyle sebastis, Alella spp., and Dactylogyrus spp. enter mainly via the gills and skin surface. Among bacterial pathogens, Vibrio species were most common, with Vibrio harveyi being the dominant species causing infections in these fishes. The bacterium Lactococcus garvieae is thought to exhibit host specificity in fish. The fish species in the present study exhibited a higher tendency for infection by heterologous pathogens than by a single pathogen; therefore, it is necessary to devise new strategies for treating diseases in cultured fish.

어류 병원세균, Listonella anguillarum에 대한 Pseudomonas aeruginosa MB I-3의 항균 효과 (Antibacterial Effects of Pseudomonas aeruginosa MB I-3 against Listonella anguillarum)

  • 질병 치료를 위하여 사용되는 항균제는 효과적이고 사용이 편리한 장점이 있지만, 내성균 발생이나 항균물질 잔류와 같은 문제점들을 안고 있다. 따라서 어류 질병 치료 및 예방을 위해 어류와 인체에 안전한 생약제 개발과 함께 어류 질병 원인균의 과다 발생을 억제하며 어체의 건강을 유지시키기 위하여 유용 균주를 이용하고자 하는 연구가 다양하게 진행되고 있다. 본 연구는 이전 연구에서 분리되어 Listonella anguillarum에 대하여 생장 억제효과가 있는 것으로 밝혀진 Pseudomonas aeruginosa MB I-3 (MB I-3)를 이용하여 넙치의 비브리오병에 대한 생물학적 방제효과를 검토하였다. Double layered plate assay와 co-culture을 통하여 MB I-3의 L. anguillarum에 대한 생장 억제능력을 조사하였고, MB I-3 균주 배양액을 ethyl acetate로 추출하여 disk 확산법으로 추출물의 항균 효과를 확인하였다. 액체 및 고체 배양에서 생장이 억제된 L. anguillarum을 전자현미경으로 관찰하였으며, 넙치 치어 사육 수조에 L. anguillarum과 MB I-3 균주를 동시에 첨가하여 폐사율을 비교하였다. MB I-3는 8종의 병원성 비브리오균에 대하여 항균력을 나타내었으며, 96시간 동안 실시한 co-culture에서 L. anguillarum은 배양 후 9시간까지 생장 증가를 보였으나, 그 후 감소하는 경향을 보였다. 한편 MB I-3 배양액 추출물 또한 L. anguillarum에 항균활성을 보여 항균 물질이 ethyl acetate로 추출됨을 알 수 있었다. 전자현미경 관찰에서 L. anguillarum은 세포질의 밀도 감소 및 세포막의 swelling에 의한 세포 용해 현상을 보였다. 한편 MB I-3를 L. anguillarum과 함께 투여한 넙치 치어는 대조군에 비해 누적 폐사율이 약 20% 감소되는 결과를 보였으므로, MB I-3를 이용한 수산용 probiotics 개발 가능성을 시사하였다.

해양 유해세균에 대한 감태 추출물의 항균특성 (Antibacterial Property of Ecklonia cava Extract against Marine Bacterial Pathogens)

  • 사람과 어류에게 감염증을 유발하는 Edwardsiella sp., Streptococcus sp. 및 Vibrio sp. 의 해양 유해세균에 대한 해조류 추출물의 항균 활성을 조사하였다. 연구에 사용된 4종의 식용 해조류 중에서 감태 MeOH 추출물이 본 연구에서 사용 된 6종의 모든 해양 유해세균에 대해 넓은 범위의 항균 활성을 나타내었다. 감태의 MeOH 추출물의 유기용매 분획 추출물들 중에서, EtOAc 분획 추출물이 가장 높은 항균활성을 나타내었으며 6종의 해양 유해세균에 대하여 $128{\mu}g/mL$에서 $256{\mu}g/mL$의 MIC 값을 나타내었다. 또한, HPLC 분석에 의해 감태 EtOAc 분획 추출물에 phlorotannin 화합물인 dieckol이 다량 존재하고 있는 것이 확인 되었다. 결론적으로, 감태 추출물의 phlorotannin 화합물이 여러 유해세균에 대한 강한 항균활성을 나타내는 것처럼, 해양 유해세균에 대해서도 강한 항균활성을 나타내는 것으로 판단된다.

Identification and Functional Analysis of Vibrio vulnificus SmcR, a Novel Global Regulator

  • Recently, quorum sensing has been implicated as an important global regulator controlling the production of numerous virulence factors such as capsular polysaccharides in bacterial pathogens. The nucleotide and deduced amino acid sequences of smcR, a homolog of V. harveyi luxR identified from V. vulnificus ATCC29307, were analyzed. The amino acid sequence of SmcR from V. vulnificus was 72 to 92% similar to those of LuxR homologs from Vibrio spp. Functions of SmcR were assessed by the construction of an isogenic mutant, whose smcR gene was inactivated by allelic exchanges, and by evaluating its phenotype changes in vitro and in mice. The disruption of smcR resulted in a significant alteration in biofilm formation, in type of colony morphology, and in motility. When compared with the wild-type, the smcR mutant exhibited reduced survival under adverse conditions, such as acidic pH and hyperosmotic stress. The smcR mutant exhibited decreased cytotoxic activity toward INT 407 cells in vitro. Furthermore, the intraperitoneal $LD_{50}$ of the smcR mutant was approximately $10^2$ times higher than that of parental wild-type. Therefore, it appears that SmcR is a novel global regulator, controlling numerous genes contributing to the pathogenesis as well as survival of V. vulnificus.

참조기(Larimichthys polyactis)에서 분리된 Neobenedenia spp.에 대한 분자 생물 동정 및 계통수 분석 (Molecular identification and phylogenetic analysis of Neobenedenia spp. isolated from small yellow croaker (Larimichthys polyactis))

  • In this study, we determined the cause of a disease outbreak in small yellow croaker(Larimichthys polyactis) in Jeju island. The major external signs in the dead fish were hemorrhage of the skin. Vibrio harveyi were isolated from a few fishes and viruses were not detected from the diseased fish. However, flukes were confirmed on the skin and we conducted molecular identification and phylogenetic analysis of the isolated parasites. The obtained 28S rRNA sequence of our specimen(Accession No. OM333244) showed the highest homology with Neobenedenia girellae, while the COI sequence of our specimen showed the highest homology with N. melleni. Further sequence analysis with other genes and morphological observation are necessary for accurate identification.

Lipopolysaccharide 생합성에 관여하는 Vibrio anguillarum의 phosphomannomutase/phosphoglucomutase 유전자 cloning과 특성 (Cloning and Characterization of Phosphomannomutase/Phosphoglucomutase (pmm/pgm) Gene of Vibrio anguillarum Related to Synthesis of LPS)

  • 수해양성 병원성 미생물로 알려진 Vibrio anguillarum으로부터 mannose-1-phosphate를 mannose-6-phosphate, glucose-1-phosphate를 glucose-6-phosphate로 가역적으로 변환시키는 phosphomannomutase/phosphoglucomutase (pmm/pgm)의 유전자를 sequencing하여 1338 bp의 open reading frame (ORF)을 밝혔다. 이는 446개의 아미노산을 포함하며 47,625 Da을 가지고 있다. 보고된 다른 Vibrio sp.의 pmm/pgm 유전자와 상동성을 비교하였을 때 V. mimicus V. vulnificus, V. splendidus, V. harveyi와 92.3%, 91.4%, 89.9%, 89.9%에 해당하는 상동성을 지니고 있었다. 증폭된 목적 유전자를 pET-28a(+) vector에 연결하여 대장균에서 단백질의 대량발현을 유도하였으며 이는 주로 soluble한 상태로 나왔다. Soluble fraction을 Ni-NTA column chromatography로 정제하여 약 50 kDa의 단백질을 얻었고 이는 주로 mannose-1-phosphate를 이용하는 효소로 확인되었으며 Mg2+ 이온이 존재할 때 효소의 활성이 나타나는 것을 확인할 수 있었다. 본 연구의 유전자는 낮은 온도의 stress하에서 발현이 증가됨을 Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR)을 통해 확인하였고, 상동성 재조합 (homologous recombination)에 의한 돌연변이 균주 제작을 통해 PMM/PGM protein과 lipopolysaccharide (LPS)의 생합성과의 관계를 규명하였다. V. anguillarum wild type과 mutant로부터 LPS를 분리하였고 sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE)후 silver staining을 통해 LPS의 high molecular weight (HMW) 부분인 O-antigen에서의 변화를 확인하였다. 또한 V. anguillarum wild type과 mutant의 growth와 viability를 확인한 결과 mutant가 wild type보다 정지기까지 더 낮은 생육을 보였으며 viability가 감소함을 확인하였다. 본 연구를 통하여 V. anguillarum의 pmm/pgm 유전자가 미생물의 생육과 LPS 생합성에 관여하고 있음을 알 수 있었다.

어류 병원성 세균에 대한 월계수(Laurus nobilis) 잎 유래 sesquiterpene lactone과 수산용 항생제의 병용효과 (Combinational effects of sesquiterpene lactones isolated from bay laurel (Laurus nobilis) leaves with antibiotics against fish pathogenic bacteria)

  • This study is a report on combinational effects between four sesquiterpene lactones (SLs) from bay laurel (Laurus nobilis) leaves, and oxytetracycline (OTC) or amoxicillin (AMX) against four fish pathogenic bacteria such as Vibrio anguillarum, V. harveyi, Edwardsiella tarda, and Streptococcus iniae. Individually, four SLs exerted little antibacterial activity against fish pathogenic bacteria. However, when combined with OTC or AMX, they showed synergistic interaction against pathogenic bacteria. Especailly, zaluzanin C (1) reduced the MIC of OTC (or AMX) eight-fold. Our results showed that combinations of SLs with antibiotics (ABTs) are more effective than ABTs alone to control pathogenic bacteria. The highest synergistic effect was observed when zaluzanin C (1) was combined with OTC or AMX against V. harvey or S. iniae, displaying significant reductions of MICs up to 8-fold (0.125 to 0.015 ㎍/mL and 0.0078 to 0.0009 ㎍/mL). In addition, zaluzanin C (1) improved the antibiotic potency of OTC against OTC resistant V. harveyi (250 ㎍/mL to 62.5 ㎍/mL). Synergism between ABTs and phytochemical such as SLs could be a therapeutically helpful concept to improve the efficacy of ABTs and prevent antibiotic resistance. These results suggest that SLs can be used as an alternative to reduce antibiotic resistance in aquaculture.

넙치에서 lincomycin 근육투여에 따른 휴약기간 도출 및 병원성 그람음성 세균에 대한 약효 평가 (Withdrawal time study of lincomycin administered intramuscularly to olive flounder Paralichthys olivaceus, and in vitro efficacy evaluation against Gram-negative bacterial pathogens)

  • 본 연구는 lincomycin (LM)의 근육투여에 따른 저수온(13℃)에서의 잔류 특성 및 넙치에서 병원성 세균으로 발견되는 그람음성 세균에서의 시험관 내 in vitro 약효를 평가하기 위하여 수행하였다. LM을 10, 20 및 40 mg/kg의 용량으로 단회 투여하였을 때, 근육내 농도가 잔류허용기준인 0.1 mg/kg 이하로 감소하는 기간은 각각 32일, 33일 및 55일로 평가되었다. 또한, 현장 분리 균주 그람음성 세균인 Edwardsiella piscida 및 Vibrio harveyi에 대하여 LM의 minimal inhibitory concentration (MIC)로 평가된 in vitro 약효는 각각 >256 ㎍/mL 및 32 ㎍/mL을 나타냈다. 연구 결과, 넙치 양식장에서 사용되는 LM의 임상용량인 10 mg/kg을 고려하였을 때의 휴약기간을 30일 이상으로 설정하면 합리적일 것으로 판단된다. 또한 LM이 넙치 연쇄상구균증 치료 목적으로 투여될 때 그 동시에 감염된 그람음성 세균에는 약효가 발휘되지 않을 것으로 추정한다.

Photoinactivation of major bacterial pathogens in aquaculture

  • Background: Significant increases in the bacterial resistance to various antibiotics have been found in fish farms. Non-antibiotic therapies for infectious diseases in aquaculture are needed. In recent years, light-emitting diode technology has been applied to the inactivation of pathogens, especially those affecting humans. The purpose of this study was to assess the effect of blue light (wavelengths 405 and 465 nm) on seven major bacterial pathogens that affect fish and shellfish important in aquaculture. Results: We successfully demonstrate inactivation activity of a 405/465-nm LED on selected bacterial pathogens. Although some bacteria were not fully inactivated by the 465-nm light, the 405-nm light had a bactericidal effect against all seven pathogens, indicating that blue light can be effective without the addition of a photosensitizer. Photobacterium damselae, Vibrio anguillarum, and Edwardsiella tarda were the most susceptible to the 405-nm light (36.1, 41.2, and $68.4J\;cm^{-2}$, respectively, produced one log reduction in the bacterial populations), whereas Streptococcus parauberis was the least susceptible ($153.8J\;cm^{-2}$ per one log reduction). In general, optical density (OD) values indicated that higher bacterial densities were associated with lower inactivating efficacy, with the exception of P. damselae and Vibrio harveyi. In conclusion, growth of the bacterial fish and shellfish pathogens evaluated in this study was inactivated by exposure to either the 405- or 465-nm light. In addition, inactivation was dependent on exposure time. Conclusions: This study presents that blue LED has potentially alternative therapy for treating fish and shellfish bacterial pathogens. It has great advantages in aspect of eco-friendly treating methods differed from antimicrobial methods.

Expression and DNA Sequence of the Gene Coding for the lux-specific Fatty Acyl-CoA Reductase from photobacterium phosphoreum

  • The nucleotide sequence of the luxC gene coding for lux-specific fatty acyl-CoA reductase and the upstream DNA (325bp)of the structural gene from bioluminescent bacterium, Photobacterium phosphoreum, has been deternubed. An open reading frame extending for more than 20 codons in 325 bp DNA upstream of luxC was not present in both directions. The lux gene can be translated into a polypeptide of 54 kDa and the amino acid sequences of lux specific reductases of P. phosphoreum shares 80, 65, 58, and 62% identity with those of the Photobacterium leiognathi, Vibrio fischeri, Vibrio harveyi, and Xehnorhabdus luminescenens reductases, respectively. Analyses of codon usage, showing that a high frequency (2.3%) of the isoleucine codon, AUA, in the luxC gene compared to that found in Escherichia coli genes (0.2%) and its absence in the luxA and B genes, suggested that the AUA codon may play a modulator role in the expression of lux gene in E. coli. The structural genes (luxC, D, A, B, E) of the P. phosphoreum coding for luciferase (${\alpha}$,${\beta}$) and fatty acid reductase (r, s, t) polypeptides can be expressed exclusively in E. coli under the T7 phage RNA polymerase/promoter system and identificationof the [35S]methionine labelled polypeptide products. The degree of expression of lux genes in analyses of codon usage. High expression of the luxC gene could only be accomplished in a mutant E. coli 43R. Even in crude extracts, the acylated acyl-CoA reductase intermediate as well as acyl-CoA reductrase activities could be readily detected.
