Evaluation of Sun Protection Factor (SPF) and Protection Factor of UVA (PFA) of the Sunscreen Containing Microalgal Extracts and MAAs (미세조류 추출물 및 MAAs 함유 자외선차단제의 자외선차단지수(SPF) 및 자외선 A 차단지수(PFA) 평가)

  • For the sunscreen development from natural resources, a possible usage of microalgal extracts or mycosporine-like amino acids (MAAs) was investigated. Sunscreens containing 7% of microalgal extracts or MAAs derived from microalgae, Chlamydomonas hedleyi, were prepared and they were applied to human research. Through this clinical research, the values of Sun Protection Factors, Sun Protector Factor (SPF) and Protection Factor of UVA (PFA), of sunscreen containing microalgal extracts or MAAs were determined: SPF values of microalgal extracts and MAAs indicated 9.07 and 9.42, respectively, while PFA ones did 2.43 and 2.41. Due to more than 2 of PFA value in both sunscreens, they can be classified into [PA+]. Taken together, although sunscreen containing microalgae-derived extracts or MAAs does not effectively protect UV irradiation, its capacity can be satisfied if inorganic UV-protecting compounds are added.

Role of Shinorine derived from Microalgae in skin protection (미세조류 유래 Shinorine의 피부세포에서의 효능)

  • In the present study, mycosporine-like amino acids (MAAs)were extracted from in Chlamydomonas hedleyi, and their function was investigated regarding to protective capacity against UV radiation and a possibility to be developed into functional suncream including MAAs by using natural compounds. In particulr, we assessed UV protective ability and anti-inflammation of shinorine in human skin cells. As a result, shinorine can protect the skin against damage by the absorption of energy from UV radiation and functions as an anti-wrinkling substrate through a increase of collagen synthesis. These data suggest that shinorine can be utilized not only as a substrate to protect UV radiation, but anti-aging material in cosmetic products.

Innate immune responses of common carp, Cyprinus carpio L. against antiviral activity inducers (항바이러스 활성 유도 물질에 대한 잉어의 선천성 면역 반응)

  • To investigate the innate immune response involved in early stage of anti-viral defence, carps were injected with UV-inactivated spring viraemia of carp virus (SVCV), poly inosinic:cytidylic acid (Poly I:C) and concanavalin A (Con A), respectively and examined lysozyme activity, serum complement activity and chemiluminescent (CL) response of leucocytes isolated from head kidney at 3 days post-injection. There was no significant difference in plasma lysozyme activities among all experimental groups. However, lysozyme activities of head kidney in the groups injected with antiviral activity inducers were significantly higher than those of the control injected with physiological saline. Bactericidal activities of serum of the groups injected with antiviral activity inducers were not significantly different from control group. However, the CL responses were significantly higher at lower dose of Poly I:C and Con A, whilst dose-dependent increase was shown in UV-inactivated SVCV-injected group. In the challenge test with 1×104 TCID50/fish of SVCV at 4 days post-injection, UV-inactivated SVCV- and Poly I:C-injected groups showed higher relative percent survival (RPS) than Con A-injected group. Furthermore, strong protection was observed in the group injected higher dose of Poly I:C although showed lower activities in lysozyme and CL response. These results suggested that Poly I:C might stimulate other factors belonging to non-specific immune system have induced protective immunity against the SVCV challenged.

The Skin Protecting Effects of Ethanolic Extracts of Eggplant Peels (가지 외피 에탄올 추출물의 피부보호효과)

  • We investigated the in vitro antioxidant and antimelanogenesis effects of ethanol extracts from eggplant (Solanum melongena L.) peels. The total phenolics and chlorogenic acid in ethanol extracts were 2,465 mg/100 g and 2.08 mg/100 g, respectively. ABTS radical scavenging activity, ferric reducing/antioxidant power assay, and malondialdehyde (MDA) inhibitory effect of the extracts increased in a dose-dependent manner. In addition, the extracts generally showed strong UV absorption in the range of UV-B (290-320 nm). The $IC_{50}$ of mushroom tyrosinase inhibitory activity of ethanol extracts from eggplant peels was 870 ${\mu}g/mL$. Importantly, the melanin syntheses of B16/F10 melanoma cells were decreased by extracts in a concentration-dependent manner. Overall, these results suggest that eggplant peels can be potentially applied as a anti-melanogenic agent as well as an antioxidant resource.

Photoprotective Potential of Anthocyanins Isolated from Acanthopanax divaricatus Var. albeofructus Fruits against UV Irradiation in Human Dermal Fibroblast Cells

  • Ultraviolet (UV) A penetrates deeply into the skin and induces the generation of reactive oxygen species (ROS) causing damage to fibroblasts, which leads to aging of the skin. However, the body has developed an antioxidant defence system against the harmful effects of ROS. Enzymes such as superoxide dismutase (SOD) and catalase (CAT) play critical roles on the removal of excess ROS in living organisms. In this study, the antioxidant activities of anthocyanins (cyanidin 3-galactoside and cyanidin 3-lathyroside) from Acanthopanax divaricatus var. albeofructus (ADA) fruits were investigated by xylenol orange, thiobarbituric acid reactive substances (TBARS), and antioxidant enzyme assay. As a result, generation of $H_2O_2$ and lipid peroxide induced by UVA-irradiation in human dermal fibroblast (HDF-N) cells was reduced by treatment of anthocyanins. Also, augmented enzyme (SOD and CAT) activities were observed in UVA-irradiated cells when treated with anthocyanin. In conclusion, the results obtained show that anthocyanins from ADA fruits are potential candidates for the protection of fibroblast against the damaging effects of UVA irradiation. Furthermore, anthocyanin may be a good candidate for antioxidant agent development.

Preparation and Characterization of Planar-type Artificial Calamine Powder with a High Aspect Ratio for the Application to Ultraviolet and Blue Band Protection Cosmetics (자외선 및 블루영역 차단 화장품 응용을 위한 박막 판형 인공 칼라민 소재의 합성 및 특성 평가 연구)

  • In this study, we have prepared pure planar-type ZnO and calamine powder containing both ZnO and Fe2O3 components as a raw material for cosmetics with UV and blue band blocking functions. The planar-type ZnO ceramic powder having a high aspect ratio in the range of 20:1 to 50:1 was synthesized by precipitation method in a zinc acetate and sodium citrate mixed solution with the electrolyte obtained by power generation with a zinc-air battery. The content of Fe2O3 in the artificial calamine ceramic powder could be increased by increasing the amount of iron chloride solution added, and in this case, some of the blue region of visible light and ultraviolet light were remarkably absorbed. When potassium acetate was added, the decomposition of the Zn(OH)42- anion in the solution was promoted to facilitate the growth of ZnO crystal in the form of a barrier wall in the vertical direction on the (0001) plane, which could increase UV absorption by providing more opportunities. By controlling the amount of iron chloride solution and potassium acetate solution added, the composition and shape of the thin film plate-shaped artificial calamine ceramic powder can be optimized, and when applied to cosmetic formulations, the light transmittance of the blue region can be greatly reduced.

Characteristics of Non-nano Needle Type Zinc Oxide and Its Application in Sunscreen Cosmetics (Non-nano 막대형 산화아연의 특성 및 자외선 차단용 화장품에의 응용)

  • With increasing interest in the effectiveness and safety of sunscreen worldwide, research on the development of new inorganic sunscreen is also gaining momentum. In the present study, non-nano sized needle type zinc oxide, which can meet the regulation of nano-material as a recent problem, has been synthesized and surface-modified with cetyl alcohol to obtain needle type zinc oxide powder. Here, we also investigated their physical properties and evaluated their potential application as sunscreens. As a result of the experiment, the sunscreen with needle type zinc oxide powder, which was non-nano, showed similar UV-protection properties and transparency compare to that of 40nm size zinc oxide. It was further confirmed that the UV blocking effect was significantly increased when the needle type zinc oxide dispersion was applied to the sunscreen. Therefore, although the needle type zinc oxide is non-nano (200 nm) material, it has the potential to be applied to the product as an excellent transparency (improvement of whiteness), UV protection efficacy and smooth texture.

Direct Evidence for the Radioprotective Effect of Various Carbohydrates on Plasmid DNA and Escherichia coli Cells

  • Damage to cells exposed to radiation is primarily attributed to direct effects on the structure of cellular DNA. Radiation-induced damage of pBluescript SK plasmid DNA and Escherichia coli $DH5\alpha$ were examined in the presence of various branched oligosaccharides, polysaccharides, and/or 8-MOP (8-methoxypsoralen). Branched oligosaccharides efficiently protected DNA and cells exposed to ultrasoft X-ray and UV irradiation. In the presence of 0.2% (w/v) branched oligosaccharides and polysaccharides, DNA can be protected from damage due to W and ultrasoft X-ray by a factor of 1.3-2.1 fo1d and 3.2-8.3 fold, respectively. The protective effect of cells exposed to UV or ultrasoft X-ray was also observed by branched oligosaccharides. The combination of MOP, a photoreagent, with carbohydrates increased the protective effects for DNA and cells, compared with that of a single use of MOP or carbohydrate alone.

Preparation and Characterization of Cerium Oxide/Silica Composite Particles (세륨 옥사이드/실리카 복합입자 제조 및 특성분석)

  • Composite particles of porous silica and cerium oxide nanoparticles blocking UV/blue light were prepared through a dry coating process. Various composite particles were prepared by varying conditions such as the mixing ratio of cerium oxide and silica, and the chamber rotating speed of mechano fusion system. The surface morphology of the composite particles was observed with SEM and the composition was analyzed using X-ray fluorescence (XRF). When the cerium oxide/silica composite particles were dispersed in water, the transparency and dispersion stability of the colloidal solution were improved. In addition, the fluidity and spreadability of the particle powder were enhanced by making composite particles. These results show that cerium oxide/silica composite particles can be used as functional cosmetic ingredients for UV/blue light protection.

  • Hyperpigmentation on face is a highly anxiety-producing symptom, especially for women from the aspect of beauty. Pigmentation of the skin is related to the amount of melanin that provides protection against UV radiation. In vivo reflectance confocal microscopy is a non-invasive imaging tool allowing visualization of the skin without tissue alteration, by placing a microscopy directly on the living skin. The aim of this study was to develop the new evaluation method of whitening effects using in vivo reflectance confocal microscopy and to validate other instruments for measuring skin colors, and UV-induced hyperpigmentation was elicited on the inside skin of the forearm. It suggested that the new method for whitening effects using the confocal microscopy was useful to evaluate the de-pigmentation products and to easy for understanding to customers.
