고지방식이 동물의 간 조직에서 크리신 투여 또는 중강도 운동이 Inflammasome과 열 발생 유전자발현에 미치는 효과 (The Effects of Either Chrysin or Moderate Exercise on Inflammasome and Thermogenic Markers in High Fat Fed Mice)

  • 본 연구 목적은 고지방식이 동물의 간 조직에서 크리신 투여 또는 중강도운동이 Inflammasome과 thermogenesis 유전자 발현의 차이를 규명하고자 시도되었다. 본 연구를 위해 정상식이군, 고지방식이군, 고지방식이+크리신 투여군, 고지방식이+중강도 운동군으로 분류한 후, 크리신 투여군은 16주간 50 mg/kg 농도로 투여하였으며, 운동군은 최대산소섭취량의 60-75%의 중강도 운동으로 실시되었다. 연구결과 크리신 그리고 중강도운동군은 지방조직, 간조직 무게 그리고 지방세포 크기가 고지방식이 군과 비교해 유의하게 감소하였다. Inflammasome 유전자 변화는 크리신 투여군 그리고 중강도 운동군에서 NLRP3. ASC, Casepase1 mRNA 발현이 고지방식이 군과 비교해 유의하게 감소하였다. 열발생마커로 알려진 PGC-1a, BMP7 mRNA 발현은 중강도 운동군에서만 고지방식 이군과 비교해 유의하게 증가했다. 결론적으로 중강도 운동은 고지방식이 동물에서 지방무게, Inflammasome, 그리고 열발생 유전자들의 발현을 비만을 억제하는데 긍정적인 영향을 미치는 것으로 보여진다. 하지만 크리신 투여는 열발생 유전자 발현에는 유의한 차이를 나타내지 못하였다. 향후 연구에서는 크리신의 비만억제 효과를 규명하기 위해 투여농도 기간을 고려한 다양한 연구가 진행되어야 할 것이다.

High-fat diet alters the thermogenic gene expression to β-agonists or 18-carbon fatty acids in adipocytes derived from the white and brown adipose tissue of mice

  • Purpose: Although activating thermogenic adipocytes is a promising strategy to reduce the risk of obesity and related metabolic disorders, emerging evidence suggests that it is difficult to induce adipocyte thermogenesis in obesity. Therefore, this study aimed to investigate the regulation of adipocyte thermogenesis in diet-induced obesity. Methods: Adipose progenitor cells were isolated from the white and brown adipose tissues of control diet (CD) or high-fat diet (HFD) fed mice, and fully differentiated white and brown adipocytes were treated with β-agonists or 18-carbon fatty acids for β-adrenergic activation or peroxisome proliferator-activated receptor (PPAR) activation. Results: Compared to the CD-fed mice, the expression of uncoupling protein 1 (Ucp1) was lower in the white adipose tissue of the HFD-fed mice; however, this was not observed in the brown adipose tissue. The expression of peroxisome proliferator-activated receptor gamma (Pparg) was lower in the brown adipose progenitor cells isolated from HFD-fed mice than in those isolated from the CD-fed mice. Norepinephrine (NE) treatment exerted lesser effect on peroxisome proliferator-activated receptor-γ coactivator (Pgc1a) upregulation in white adipocytes derived from HFD-fed mice than those derived from CD-fed mice. Regardless which 18-carbon fatty acids were treated, the expression levels of thermogenic genes including Ucp1, Pgc1a, and positive regulatory domain zinc finger region protein 16 (Prdm16) were higher in the white adipocytes derived from HFD-fed mice. Oleic acid (OLA) and γ-linolenic acid (GLA) upregulated Pgc1a expression in white adipocytes derived from HFD-fed mice. Brown adipocytes derived from HFD-fed mice had higher expression levels of Pgc1a and Prdm16 compared to their counterparts. Conclusion: These results indicate that diet-induced obesity may downregulate brown adipogenesis and NE-induced thermogenesis in white adipocytes. Also, HFD feeding may induce thermogenic gene expression in white and brown primary adipocytes, and OLA and GLA could augment the expression levels.

고려인삼(Panax ginseng)과 미국삼(Panax quinquefolium)의 승열작용에 대한 비교 고찰 (A comprehensive review of the comparative studies of Korean(Panax ginseng) and American(Panax quinquefolium) Ginsengs on the Thermogenic Response)

  • It has been extensively known in the high temperature regions such as southern China and south east Asia that Korean ginseng(Panax ginseng) intake raises body heat, while American ginseng(Panax quinquefolium) lowers it, based upon oriental medicine. This is likely due to the misunderstanding of the conception that Korean ginseng shows a warm property, resulting in enhanced blood circulation and stimulation of metabolism. It's further supported by the fact that current scientific studies have shown no notable side effects. Moreover, it has been demonstrated in animal experimental test that Korean ginseng shows a complementary effect in a high temperature environment, together with scientific evidence of a preventive effect of ginseng saponin against cold conditions, suggesting it to exhibit an adaptogenic effect normalizing the body under abnormal body conditions. However, there is a shortage of studies published concerning comparative clinical and animal studies of Korean and American ginsengs, and also, on natives of high temperature countries. Therefore, a further placebo-controlled clinical study has to be considered to elucidate scientifically clinical evaluation of Korean and American ginsengs on the body heat. In this article, we summarize the scientific publications reporting the clinical comparative study of Korean and American ginsengs on body heat response.

In vitro Anti-obesity Effect of 4-hydroxybenzyl Alcohol from Cudrania tricuspidata

  • The present study was investigated on in vitro anti-obesity effect of 4-hydroxybenzyl alcohol from Cudrania tricuspidata. We isolated various compounds from Cudrania tricuspidata. Among these compounds, anti-obesity effects of 4-hydroxybenzyl alcohol was examined by lipase activity assay, cyclic adenosine monophosphate (cAMP)-specific phosphodiesterase type IV (PDE4) activity assay, and citrate synthase activity assay. 4-hydroxybenzyl alcohol and Cudrania tricuspidata extracts inhibited the enzymatic activities of lipase, PDE4, and citrate synthase. Lipase is known to mediate the hydrolysis of triacylglycerol in adipose tissue and cholesterol esters in other tissue or cells. Also, PDE4 hydrolyses cAMP, a crucial secondary messenger for in metabolic pathways including glucose and lipid metabolism, lipolysis, and thermogenic function. 4-hydroxybenzyl alcohol and Cudrania tricuspidata extracts induced the inhibitory effect against each enzymatic activity on several specific substrates as observed by detection at 405 or 412 nm. These findings might be attributable to the inhibition of adipogenesis, and partial prevention of obesity. In conclusion, these results show that 4-hydroxybenzyl alcohol and Cudrania tricuspidata may be a critical candidate as a natural anti-obesity source.

Effect of Selenium on Cold Adapted Beef Cattle

  • Cattle in Alaska seemed to be tolerant to low blood selenium (Se) although arctic winter energy demands might exacerbate oxidative damage to tissues lacking protection by Se. The thermogenic properities of brown adipose tissue (BAT) and the long cold adaptation period for Alaskan cattle indicates that they might develop BAT. Eighteen mature beef cows with whole blood Se of< 19 ppb were used to examine possible relation-ships among Se deficiency, tissue lesions, and observable BAT. All cows were wintered on a diet low in Se and nine cows were given supplemental Se provided by intraruminal bolus. Blood Se was elevated (p < .05) from November through February by supplemental Se, but body weight and back fat thickness were unaffected (p < .10). Tissues were taken from two Se deficient cows, two cows supplemented with Se, and two cows given large doses of Se (300 mg by injection) four weeks before slaughter. Histopathological examination of 187 samples of fat (7 to 55 from each cow) showed no observable amount of BAT. Examination of other tissues showed no lesions attributable to Se deficiency.

Ginsenoside Rg1 promotes browning by inducing UCP1 expression and mitochondrial activity in 3T3-L1 and subcutaneous white adipocytes

  • Background: Panax ginseng Meyer is known as a conventional herbal medicine, and ginsenoside Rg1, a steroid glycoside, is one of its components. Although Rg1 has been proved to have an antiobesity effect, the mechanism of this effect and whether it involves adipose browning have not been elucidated. Methods: 3T3-L1 and subcutaneous white adipocytes from mice were used to access the thermogenic effect of Rg1. Adipose mitochondria and uncoupling protein 1 (UCP1) expression were analyzed by immunofluorescence. Protein level and mRNA of UCP1 were also evaluated by Western blotting and realtime polymerase chain reaction, respectively. Results: Rg1 dramatically enhanced expression of brown adipocyte-especific markers, such as UCP1 and fatty acid oxidation genes, including carnitine palmitoyltransferase 1. In addition, it modulated lipid metabolism, activated 5' adenosine monophosphate (AMP)-activated protein kinase, and promoted lipid droplet dispersion. Conclusions: Rg1 increases UCP1 expression and mitochondrial biogenesis in 3T3-L1 and subcutaneous white adipose cells isolated from C57BL/6 mice. We suggest that Rg1 exerts its antiobesity effects by promoting adipocyte browning through activation of the AMP-activated protein kinase pathway.

Effects of Dyglomera® on leptin expression, pro-inflammatory cytokines, and adipocyte browning in 3T3-L1 cells

  • Dyglomera® is an aqueous ethanol extract derived from the fruit and pods of Dichrostachys glomerata. A previous study has revealed that Dyglomera regulates adipogenesis and lipolysis by modulating AMP-activated protein kinase (AMPK) phosphorylation and increased expression levels of lipolysis-related proteins in white adipose tissue of high fat diet-induced mice and 3T3-L1 adipocyte cells. To further investigate mechanisms of Dyglomera, additional studies were performed using 3T3-L1 cells. Results revealed that Dyglomera downregulated adipogenesis by inhibiting the protein kinase B/mammalian target of rapamycin signaling pathway and reconfirmed that it downregulated gene expression levels of proliferator-activated receptor (PPAR)-γ, CCAAT enhancer binding protein α, sterol-regulation element-binding protein-1c. Dyglomera also reduced adipokines such as tumor necrosis factor alpha, interleukin-1β, and interleukin 6 by regulating leptin expression. Moreover, Dyglomera promoted beige-and-brown adipocyte-related phenotypes and regulated metabolism by increasing mitochondrial number and expression levels of genes such as T-box protein 1, transmembrane protein 26, PR domain 16, and cluster of differentiation 40 as well as thermogenic factors such as uncoupling protein 1, proliferator-activated receptor-gamma co-activator-1α, Sirtuin 1, and PPARα through AMPK activation. Thus, Dyglomera not only can inhibit adipogenesis, but also can promote lipolysis and thermogenesis and regulate metabolism by affecting adipokine secretion from 3T3-L1 adipocytes.

비만치료를 목적으로 마테(Yerba Mate, Ilex paraguariensis)를 함유한 복합투여물의 효과 및 부작용에 대한 고찰 (A Review of Combination Effects and Adverse Effects of Yerba Mate (Ilex paraguariensis) on the Treatment of Obesity)

  • Objectives : The aim of this study is to review the combination effects and adverse effects of Yerba Mate (Ilex paraguariensis) on the treatment of obesity. Methods : Four databases were searched, including Pubmed, National assembly library, RISS4u, and NDSL, for studies published before March 31, 2016 using the keywords 'yerba mate', 'Ilex paraguariensis', and those related to 'obesity'. Clinical trials, and both in vivo and in vitro studies were classified according to the following categories: publication year, nationality, types and composition of test compounds, combination effects, adverse effects, and safety. Results : A total of 16 articles (14 clinical trials, one in vivo, one in vitro) were included in the analysis. Combination effects were reported from six YGD (Yerba Mate, Guarana, Damiana, Zotrim), five Meltdown, one Shred-matrix, one DBX (DymaBurn Xtreme), one XSIII research study, and adverse effects were reported from two YGD (Zotrim), four Meltdown, one Shred-matrix and one MetEF research study. YGD and Shred-matrix increased satiety while YGD and MetEF had adverse gastrointestinal effects potentially due to guarana. Meltdown and Shred-matrix increased energy expenditure and fat oxidation and had the adverse effects of increased heart rate and blood pressure due to the thermogenic effect of caffeine and yohimbine. DBX increased resting energy expenditure but no difference was observed in respiratory exchange ratio. XSIII reduced body weight gain and food intake more than each of the extracts did on their own. Conclusion : Taken together, the combination effects and adverse effects differed according to the composition of test compounds and the ratio of ingredients. Multicomponent approaches should be used for treating multifactorial diseases such as obesity. As a general conclusion, there is a place for a combination of herbal components including yerba mate, based on the principle of Kun-Shin-Choa-Sa for the treatment of obesity.

Effects of Ginseng Saponins in Energy Metabolism, Memory, and Anti-neurotoxicity

  • Ginseng has been used as a key constituent in traditional medicine prescriptions for centuries. Other than its well-known anti-stress and adaptogenic properties, ginseng has also been shown to be very effective in treating age-related deterioration in metabolic and memory functions. Although it is generally believed that the saponin (GS) fraction of the ginseng root accounts for the bioactivity of ginseng, a direct demonstration on which ginsenoside does what is still generally lacking. In the past decade, our laboratory has endeavored to identify the active GS components involved in energy metabolism, memory, and anti-neurotoxicity. To examine the ergogenic effects of GS in enhancing aerobic capacity, rats were subjected to either severe cold ($40^{\circ}C$ under helium-oxygen, two hours) or exercise workload $(70\%\;VO_{2}max,$ to exhaustion). Acute systemic injection (i.p.) of ginseng GS (5-20 mg/kg) significantly elevated both the total and maximum heat production in rats and improved their cold tolerance. However, pretreating the animal with the optimal dose (10 mg/kg) of GS devoid of $Rg_1\;and\;Rb_1$ failed to elicit any beneficial effects in improving cold tolerance. This indicates that either $Rb_1\;and/or\;Rg_1$ may be essential in exemplifying the thermogenic effect of GS. Further studies showed that only pretreating the animals with $Rb_1(2.5-5\;mg/kg),\;but\;not\;Rg_l,$ resulted in an increase in thermogenesis and cold tolerance. In contrast to the acute effect of GS on cold tolerance, enhancement of exercise performance in rats was only observed after chronic treatment (4 days). Further, we were able to demonstrate that both $Rb_1\;and\;Rg_1$ are effective in enhancing aerobic endurance by exercise. To illustrate the beneficial effects of GS in learning and memory, a passive avoidance paradigm (shock prod) was used. Our results indicated that the scopolamineinduced amnesia can be significantly reversed by chronically treating (4 days) the rats with either $Rb_1\;or\;Rg_1$ (1.25 - 2.5 mg/kg). To further examine its underlying mechanisms, the effects of various GS on ${\beta}-amyloid-modulated$ acetylcholine (ACh) release from the hippocampal slices were examined. It was found that inclusion of $Rb_1$ (0.1 ${\mu}M$), but not $Rg_1$, can attenuate ${\beta}-amyloid-suppressed$ ACh release from the hippocampal slices. Our results demonstrated that $Rb_1\;and\;Rg_1$ are the key components involved in various beneficial effects of GS but they may elicit their effects through different mechanisms.

한국인의 에너지 섭취권장량 변화 (Change of Recommended Energy Intake for Korea)

  • 1962년 한국인 영양권장량의 제정부터 7차 개정을 거쳐 한국인 영양섭취기준으로 전환되고, 한국인 영양섭취기준이 개정되기까지 에너지를 중심으로 섭취권장량의 양적 변화 및 산출 방법 변화를 살펴보았다. 이에 근거하여 한국인의 에너지 섭취권장량의 개정 방향 및 설정 방법 등에 대하여 검토하고, 설정의 새로운 기법 마련을 위한 기초자료로 활용될 수 있기를 기대한다. 에너지 섭취권장량은 기초대사량(또는 휴식대사량), 활동대사량, 식이성 발열효과의 3가지 요인을 합한 양이다. 1962년 체중을 대입하여 산출하는 공식으로 시작하여 95년에는 휴식대사량(체중 대입 산출)에 평균활동계수를 적용하는 공식으로 전환되었다. 그리고 2005년 연령, 체격(신장, 체중)과 함께 4단계 활동단계를 구분, 대입하여 산출하는 에너지필요추정량이라는 새로운 에너지 섭취권장량이 소개되었다. 1962년 제정 이후 50년이 조금 안 되는 기간 동안 다소의 차이는 있으나 모든 연령대에서 에너지 섭취권장량이 감소하였으며, 이는 생활환경의 변화 즉 활동(노동)량의 감소가 원인으로 생각된다. 비만 인구의 증가와 이에 따른 만성 질환 이환율이 증가함에 따라 에너지 섭취권장량만은 다른 영양소와 달리 개인별 산출 및 적용이 권고되고 있으며, 현재 개인별 산출이 가능하다. 에너지 섭취권장량의 보다 정확한 추정을 위하여 우리 국민 대상의 에너지 소비량 연구 결과를 적용한 우리 국민의 공식이 마련될 수 있기를 기대한다.
