  • Background: IL-18 was originally cloned as a IFN-${\gamma}$ inducing factor in primed T cells. In synergy with IL-12, IL-18 has been shown to induce strikingly high levels of IFN-${\gamma}$ production by T cells and to enhance Th1 development. Also this cytokine exerts induction of Th2 development through IL-4 induction. Methods: Resting $CD4^+$ T cells were sorted by negative selection and activated by anti-CD3 plus anti-CD28 Ab. Expression of IL-12 binding sites, IL-18 binding sites, IL-18R ${\alpha}$, and GATA-3 mRNA were analysed by FACS and RT-PCR, respectively. Results: Resting $CD4^+$ T cells expressed IL-18R ${\alpha}$ chain but not IL-18 binding sites, suggesting a lack of IL-18R ${\beta}$ expression. IL-18R ${\alpha}$ was maintained on the Th1 and Th2 committed cells. IL-18 binding sites were induced on the Th1 but not Th2 cells. Exposure of these cells to IL-18 led to up-regulation of GATA-3 mRNA expression only in Th2 committed cells. To elucidate the relationship between IL-18R ${\alpha}$ expression and GATA-3 induction by IL-18, Th1 and Th2 committed cells were further cultured in medium with or without IL-12 for 2 days. IL-12 binding sites were maintained on the Th1 and Th2 cells regardless of IL-12 treatment, but IL-18R a expression was rapidly down-regulated on the IL12-untreated Th2 cells which did not induce GATA-3 mRNA expression followed by IL-18 stimulation. Conclusion: IL-12 supports expression of IL-18R ${\alpha}$ and GATA-3 mRNA expression was induced by IL-18 through IL-18R ${\alpha}$ without expression of IL-18 binding site in Th2 cells.

  • The ion exchange behaviour of the $Cs^{+1},\;Sr^{+2},\;and\;Th^{+4}$ in the system of $Cs^{+1},\;Sr^{+2},\;Th^{+4},\;and\;7Cl^{-}-H^{+}$ from Dowex HCR-W2, was examined in the loading and elution processes. $Th^{+4}$ was slowly adsorbed through the entire contact time between resin and solution and $Cs^{+1}\;and\;Sr^{2+}$ were adsorbed fast for the first few minutes of contact time. Because of the strong affinity of $Th^{+4}$, the longer contact time was allowed, the less amount of $Cs^{+1}\;and\;Sr^{2+}$ was adsorbed on the resin. The peak concentration of the resin phase $Cs^{+1}$ in the solution concentration of $Cs^{+1}:Sr^{+2}:Th^{+4}$ in the ratio of 2 : 2 : 1 in normality with total normality of 0.1N was produced at about 4 minutes of contact time and the peak time for $Sr^{+2}$ was 20 minutes. The loaded ions were eluted using hydrochloric acid. The loaded $Cs^{+1}$ was eluted at the low eluent concentration of less than 0.1N with less than 5% contamination of $Sr^{+2}$. The loaded $Th^{+4}$ was eluted at the high eluent concentration of greater than 1N. The best eluent concentration for eluting $Th^{+4}$ was 4N.

  • Background: Costimulation is a critical process in Ag-specific immune responses. Both B7.1 and CD28 molecules have been reported to stimulate T cell responses during antigen presentation. Therefore, we tested whether Ag-specific immune responses as well as protective immunity are influenced by coinjecting with B7.1 and CD28 cDNAs in a mouse HSV-2 challenge model system. Methods: ELISA was used to detect levels of antibodies, cytokines and chemokines while thymidine incorporation assay was used to evaluate T cell proliferation levels. Results: Ag-specific antibody responses were enhanced by CD28 coinjection but not by B7.1 coinjection. Furthermore, CD28 coinjection increased IgG1 production to a significant level, as compared to pgD+pcDNA3, suggesting that CD28 drives Th2 type responses. In contrast, B7.1 coinjection showed the opposite, suggesting a Th1 bias. B7.1 coinjection also enhanced Ag-specific Th cell proliferative responses as well as production of Th1 type cytokines and chemokines significantly higher than pgD+pcDNA3. However, CD28 coinjection decreased Ag-specific Th cell proliferative responses as well as production of Th1 types of cytokines and chemokine significantly lower than pgD+pcDNA3. Only MCP-1 production was enhanced by CD28. B7.1 coimmunized animals exhibited an enhanced survival rate as well as decreased herpetic lesion formation, as compared to pgD+pcDNA3. In contrast, CD28 vaccinated animals exhibited decreased survival from lethal challenge. Conclusion: This study shows that B7.1 enhances protective Th1 type cellular immunity against HSV-2 challenge while CD28 drives a more detrimental Th2 type immunity against HSV-2 challenge, supporting an opposite role of B7.1 and CD28 in Ag-specific immune responses to a Th1 vs Th2 type.

  • Background: Bronchial asthma is characterized by chronic eosinophilic inflammatory airway disease associated with bronchial hyperresponsiveness and reversible airway obstruction. Bronchial inflammation in asthma may depend in part on the activation of T helper lymphocytes that elaborate proinflammatory cytokines. T helper (Th) lymphocytes can be divided into two categories; Th1 lymphocytes, which secrete IL-2, IL-12 and IFN-$\gamma$, and Th2 lymphocytes, which secrete IL-4, IL-5, IL-6 and IL-10. Th2 lymphocytes appear to induce allergic responses, whereas Th1 lymphocytes induce delayed-type hypersensitivity response. Some infections, such as tuberculosis, cultivate a Th1 immunological environment and inhibit Th2 lymphocytes function. The presence of such infections might inhibit Th2 immune responses and thus protect development of atopic diseases. Method: 15 patients with allergic bronchial asthma, 10 patients with intrinsic bronchial asthma, and 10 healthy volunteers were studied. The serum concentrations of IFN-$\gamma$, IL-12, IL-4, IL-5, and IL-10 were measured by ELISA method and tuberculin skin test was estimated in different groups. Results: The positive response rates of tuberculin test were 46.7% in patients with allergic asthma, 100% in patients with intrinsic asthma and 60% in normal controls. The positive response rates were significantly lower in patients with allergic asthma than those of in patients with intrinsic asthma (p<0.05). Degree of responses to tuberculin test were $12.0{\pm}9.6mm$ in patients with allergic asthma, $18.4{\pm}4.5mm$ in patients with intrinsic asthma and $10.9{\pm}8.8mm$ in normal controls. The degree of responses were significantly reduced in patients with allergic asthma than those of patients with intrinsic asthma (p<0.05). The serum levels of IL-5 in patients with allergic asthma were significantly higher than in patients with intrinsic asthma and normal controls (p<0.05), although it was insignificant. the serum levels of IL-4 and IL-10 in patients with allergic asthma were higher than that of intrinsic asthma and normal controls. The serum levels of IL-12 and IFN-$\gamma$ in patients with allergic asthma and intrinsic asthma were significantly lower than those in normal controls(p<0.05). The serum levels of total immunoglobulin E (IgE) and peripheral blood eosinophile counts in patients with allergic asthma were significantly higher than those in normal controls. Peripheral blood esinophil counts had a significant correlation with the serum levels of total IgE, IL-5 and IL-10 in patients with allergic asthma (p<0.05). Conclusion: These results have showed that Th1 lymphocyte functions were lowered and Th2 lymphocyte functions were elevated in patients with allergic asthma than those in normal controls. Suppression of Th1 lymphocyte functions by activation of Th2 lymphocyte might be one of the important aspects of pathogenesis in allergic bronchial asthma.

  • To secure instream flow at the Yudeung urban stream reach of Daejeon city in South Korea, Yudeung upstream diversion was designed with total water storage of $59{\times}10^4m^3$, and the upstream Seongol reservoir was planned to raise the bank with various sizes. Downstream streamflows were simulated by considering outflows from upstream diversion and reservoir, and after then flow durations were analyzed and compared with flows of no reservoir condition. In case of no diversion or reservoir upstream, flow durations were $1^{st}$ flow of $84.72m^3/s$, $95^{th}$ flow of $2.10m^3/s$, $185^{th}$ flow of $0.92m^3/s$, $275^{th}$ flow of $0.42m^3/s$, and $355^{th}$ flow of $0.31m^3/s$. In case of upstream diversion, flow durations were $1^{st}$ flow of $94.38m^3/s$, $95^{th}$ flow of $2.96m^3/s$, $185^{th}$ flow of $1.22m^3/s$, $275^{th}$ flow of $0.50m^3/s$, and $355^{th}$ flow of $0.35m^3/s$. The increase flow rates were $0.04m^3/s$ in $355^{th}$ flow, $0.08m^3/s$ in $275^{th}$, and $0.30m^3/s$ in 185th. In case of Seongol reservoir with effective storage capacities of $365{\times}10^4m^3$, $544{\times}10^4m^3$, $750{\times}10^4m^3$, and $992{\times}10^4m^3$, flow durations were $85.5{\sim}83.9m^3/s$ on $1^{st}$ flow, $2.85{\sim}2.57m^3/s$ on $95^{th}$ flow, $1.16{\sim}1.27m^3/s$ on $185^{th}$ flow, $0.64{\sim}0.99m^3/s$ on $275^{th}$ flow, and $0.56{\sim}0.94m^3/s$ on $355^{th}$ flow. The increase flow rates were $0.25{\sim}0.63m^3/s$ in $355^{th}$ flow, $0.22{\sim}0.57m^3/s$ in $275^{th}$, and $0.24{\sim}0.35m^3/s$ in $185^{th}$. The more the sizes of upstream reservoirs increased, the $1^{st}$ and $95^{th}$ flows decreased in which coefficients of determination were 0.92, 0.99, respectively and the $185^{th}$, $275^{th}$, and $355^{th}$ flows increased in which coefficients of determination were 0.93 to 0.99.

  • This study was to evaluate the effect of Samjayangchin-tang (STYCT) on mouse Th1 and Th2 cells' differentiation and ovalbumin (OVA)-induced allergic inflammation. The proliferation of mouse CD4 T cells and the secretion of Th1/Th2 cytokines under the influence of STYCT extract were measured as well as the amount of ${\beta}-hexosaminidase$ in RBL-2H3 cells and the levels of $TNF-{\alpha}$ and IL-6 secretion in Raw264.7 cells. BALB/c mice were orally administered with STYCT extract and simultaneously inoculated with OVA to induce allergic reaction and measure the level of total IgE, OVA-specific IgE and the production of IFN- g, IL-4, IL-5 by the spleen cells. When mouse CD4 T cell were stimulated with anti-CO3 and anti-CD28 for 48 hours in various concentrations of STYCT extract, it decreased proliferation of CD4 cells. CD4 T cells under Th1/Th2 polarizing conditions for 3 days with STYCT resulted in mild decrease of IFN- g in Th1 cells and significant decrease of IL-4 in Th2 cells. STYCT extract had a dose-dependent inhibitory effect on antigen-induced release of ${\beta}-hexosaminidase$ in RBL-2H3 cells. Treatment of STYCT extract on LPS stimulated Raw 264.7 cells showed dose-dependent decrease in IL-6 production. Oral administration of STYCT extract on OVA-induced allergic mice showed an inhibitory effect on the levels of total serum IgE and OVA-specific IgE by 53% and 44%, respectively. Culture of spleen cells with OVA resulted in significant increase of IFN- g by 54% and significant decrease of IL-4 and IL-5 by 42%, and 29%, respectively. The results show that STYCT does not strongly induce mouse T cells to transform into Th1 or Th2 but it has an anti-allergic effect in vitro, and that it also corrects the unbalance between the reactions of Th cells in allergic diseases.

  • Monazite is a phosphate mineral that contains thorium (Th) and rare earth elements. The Th concentration in monazite can be as high as 500 ppm, and it has the potential to be used as fuel in the nuclear power system. Therefore, this study aimed to conduct the techno-economic analysis (TEA) of Th extraction in the form of thorium oxide (ThO2) from monazite. Th can be extracted from monazite through an alkaline fusion method. The TEA of ThO2 production studied parameters, including raw materials, equipment costs, total plant direct and indirect costs, and direct fixed capital cost. These parameters were calculated for the production of 0.5, 1, and 10 ton ThO2 per batch. The TEA study revealed that the highest production cost was ascribed to installed equipment. Furthermore, the highest return on investment (ROI) of 21.92% was achieved for extraction of 1 ton/batch of ThO2, with a payback time of 4.56 years. With further increase in ThO2 production to 10 ton/batch, the ROI was decreased to 5.37%. This is mainly due to a significant increase in the total capital investment with increasing ThO2 production scale. The minimum unit production cost was achieved for 1 ton ThO2/batch equal to 335.79 $/Kg ThO2.

  • This study was undertaken to investigate the effect of indomethacin on prostaglandins in 4-Nitroquinoline-N-Oxide (4-NQO) induced palatal carcinoma of albino rats. 128 Sprague-Dawley strain albino rats-about 100g in body weight-were used in this study, divided into as belows; 1. Normal group (16-albino rats) with no treatment, 2. Control group (16-albino rats) treated with prophylene application onto palatal mucosa 3 times a week. 3. Experimental group I (48-albino rats) treated with 0.5% 4-NQO in prophylene application onto palatal mucosa 3 times a week. 4. Experimental group II (48-albino rats) treated with 0.5% 4-NQO in prophylene application with administered $20{\mu}g/ml$ of indomethacin in drinking water ad. lib. Four animals were sacrificed 7th, 13th, 19th, and 25th week respectively in normal and control group, and 7th, 9th, 11th, 13th, 15th, 17th, 19th, 21st, 23rd, 25th, 27th and 29th week respectively in experimental group I and II at each time. The palatal and lingual tissues were excised and kept frozen at $-70^{\circ}C$. Densitometer scan and Beta-counting counter were used for the thin layer chromatography of the arachidonic acid metabolites. The obtained results were as belows; 1. In experimental group I, prostaglandin $D_2$, and prostaglandin $E_2$ were increased, while 6-keto-prostaglandin $F_{1{\alpha}}$ and thromboxane $B_2$ were decreased in relative abundances of arachidonic acid metabolites. And there was little change in prostaglandin $F_{1{\alpha}}$ 3. In experimental group II, prostaglandin $D_2$, and prostaglandin $E_2$ were increased, while 6-keto-prostaglandin $F_{1{\alpha}}$ and thromboxane $B_2$ were decreased in relative abundances of arachidonic acid metabolites. And there was little change in prostaglandin $F_{2{\alpha}}$ also. 4. In the range of increase in prostaglandin $D_2$, and prostaglandin $E_2$, and that of decrease in 6-keto-prostaglandin $F_{1{\alpha}}$ and thromboxane $B_2$, in relative abundances, there was wider in experimental group I than in group II. 5. In the range of increase in prostaglandin $D_2$, and prostaglandin $E_2$, and that of decrease in 6-keto-prostaglandin $F_{1{\alpha}}$ and thromboxane $B_2$, in relative abundances, there was wider in palatal mucosa than in lingual mucosa in experimental group I and II.
