Efficient expression of the Salmonella Typhimurium tdc ABCDEG operon involved in the degradation of L-serine and L-threonine requires TdcA, the transcriptional activator of the tdc operon. We found that the tdcA gene was transiently activated when the bacterial growth condition was changed from aerobic to anaerobic, but this was not observed if Salmonella was grown anaerobically from the beginning of the culture. Expression kinetics of six tdc genes after anaerobic shock demonstrated by a real-time PCR assay showed that the tdc CDEG genes were not induced in the tdcA mutant but tdcB maintained its inducibility by anaerobic shock even in the absence of tdcA, suggesting that an additional unknown transcriptional regulation may be working for the tdcB expression. We also investigated the effects of nucleoid-associated proteins by primer extension analysis and found that H-NS repressed tdcA under anaerobic shock conditions, and fis mutation delayed the peak expression time of the tdc operon. DNA microarray analysis of genes regulated by TdcA revealed that the genes involved in N-acetylmannosamine, maltose, and propanediol utilization were significantly induced in a tdcA mutant. These findings suggest that Tdc enzymes may playa pivotal role in energy metabolism under a sudden change of oxygen tension.
Thermal stress and elastic creeping stress analysis was conducted by finite element method to simulate start-up process of a boiler header of 500MW standard fossil power plant. Start-up temperature and operating pressure history were simplified from the real field data and they were used for the thermal stress analysis. Two kinds of thermal stress analysis were considered. In the first case only temperature increase was considered and in the second case both of temperature and operating pressure histories were considered. In the first analysis peak stress was occurred during the temperature increase from the room temperature. Hence cracking or fracture may occur at the temperature far below the operating maximum temperature. In the results of the second analysis von Mises stress appeared to be higher after the second temperature increase. This is due to internal pressure increase not due to the thermal stress. When the stress components of radial(r), hoop($\theta$) and longitudinal(z) stress were investigated, compression hoop stress was occurred at inner surface of the stub tube when the temperature increased from room temperature to elevated temperature. Then it was changed to tension hoop stress and increased because of the operating pressure. It was expected that frequent start-up and shut-down operations could cause thermal fatigue damage and cracking at the stub tube hole in the header. Elastic-creeping analysis was also carried out to investigate the stress relaxation due to creep and stabilized stress after considerable elapsed time. The results could be used for assessing the creep damage and the residual life of the boiler header during the long-tenn service.

In this study, anti-foaming agents of a fatty acid methyl esters was synthesized and confirmed by FT-IR and $^1H-NMR$ spectroscopy. Synthesized material of characteristic of -C=O group having a $1740cm^{-1}$ and -C-O group $1175cm^{-1}$ peak by using FT-IR, and it was confirmed that k. Surface tensions were measured by surface tensiometer CBVP-43. Their surface tension values was 17.7 to 21 dyne/cm. Anti-foaming abilities in the SLS solution was measured through the Ross-Miles method. Performance of the 4 type of anti-foaming agent was determined and the best anti-foaming agent was prepared using stearic acid methyl.

For a long time, evaluation of soundness and strength in concrete has been performed through ultrasonic velocity(UV), which is essential work in field assessment. Porosity in concrete is a major parameter indicating durability and strength, and UV passing concrete depends on porosity variation. In this paper, a modeling on UV through concrete is carried out considering porosity and the results are verified with those from test. Additionally UV in concrete under compression/tension loading condition is measured and UV modeling with loading condition is performed. Up to 50% of loading ratio, UV slightly increases and greatly drops at peak load in compression region, however it fluctuates in tensile region due to micro cracking in matrix. The proposed model shows a reasonable agreement with test results in control and compression region, and needs modification for tensile region considering micro cracks and local aggregate interlocking.
A marine microorganism producing biosurfactant was isolated from the oil polluted coast of Chung-Mu in Korea, and was identified as Pseudomonas sp.. It produced the biosurfactanl and its optimum culture conditions for pH and salt concentration were 8.0 and 3.0%, respectively. The productivity of biosurfactant from this strain was affected by the nitrogen source used. For the oil resolvability of the biosurfactant, the residual oil in the culture broth with 2% Kuwait crude oil at each time of 48, 96, and 132hr was investigated by gas chromatography. As result of this experiment, it was verified that the biosurfactant acted on C10-C14, of Kuwait crude oil and so the oil was decomposed. The biosurfactant isolated from the supernatant was purified by adsorption to Amberliter XAD-7 and followed by gel chromatography (Sephadex G-100) and HPLC. The purified biosurfactant showed a high value of emulsifying activity at $40^{\circ}C$ and the emulsifying stability was maintained at the temperature range of $30^{\circ}C$∼$60^{\circ}C$. The purified biosurfactant reduced the interfacial tension of Kuwait crude oil remarkably and showed improved dispersing ability compared to those of commercial surfactants such as Tween 80, Tween 60 and SDS.

The 1MW OTEC (Ocean Thermal Energy Conversion) platform was designed for application in equatorial seas. In this study, the OTEC platform was investigated using numerical and experimental methods. An octagon-shaped OTEC platform was investigated using the Ocean Engineering Basin of KRISO. These experiments included various tests of regular waves, irregular waves and irregular waves with current (wave+current). The responses of the platform in regular waves showed good agreement between the numerical and experimental results, including the motion RAO, wave run up, and mean drift force. The peak period of heave and pitch motions were observed around 0.5 rad/s, and the effect of the total reflection was found under short wave conditions. The standard deviation (STD) of the platform motion was checked in irregular waves of equatorial and Hawaiian seas. The STD of the pitch was less than $4^{\circ}$ different from the operability requirement under equatorial conditions and the surge STD of the wave frequency showed good agreement between the numerical and experimental results. The STD values of the surge and pitch were increased 66.6% and 92.8% by the current effects in irregular waves, but the pitch STD was less than $4^{\circ}$ under equatorial conditions. This study showed that the STD of the surge was affected by spring effects. Thus, the watch circle of the platform and tension of the mooring lines must be evaluated for a specific design in the future.

The present study presents a nonlinear finite element analysis (FEA) approach using the general program of Abaqus to evaluate the seismic response of unreinforced masonry walls strengthened with the steel bar truss system developed in the previous investigation. For finite element models of masonry walls, the concrete damaged plasticity (CDP) and meso-scale methods were considered on the basis of the stress-strain relationships under compression and tension and shear friction-slip relationship of masonry prisms proposed by Yang et al. in order to formulate the interface characteristics between brick elements and mortars. The predictions obtained from the FEA approach were compared with test results under different design parameters; as a result, a good agreement could be observed with respect to the crack propagation, failure mode, rocking strength, peak strength, and lateral load-displacement relationship of masonry walls. Thus, it can be stated that the proposed FEA approach shows a good potential for designing the seismic strengthening of masonry walls.
It is normal to observe the presence of numerous cracks in coal body. And it has significantly effective on the mechanical characteristics and realistic failure models of coal mass. Therefore, this paper is to investigate the influence of crack parameters on coal body by comprehensive using theoretical analysis, laboratory experiments and numerical simulation through prepared briquette specimens. Different from intact coal body possessing single peak in stress-strain curve, other specimens with crack angle can be illustrated to own double peaks. Moreover, the unconfined compressive strength (UCS) of specimens decreases and follow by increasing with the increase of crack angle. It seems to like a parabolic shape with an upward opening. And it can be demonstrated that the minimum UCS is obtained in crack angle $45^{\circ}$. In terms of failure types, it is interesting to note that there is a changing trend from tensile failure to tensile-shear mixing failure with tension dominant follow by shear dominant with the increase of crack angle. However, the changing characteristics of UCS and failure forms can be explained by elastic-plastic and fracture mechanics. Lastly, the results of numerical simulations are good consistent with the experimental results. It provides experimental and theoretical foundations to reveal fracture mechanism of coal body with non-penetrating single crack further.
A nonlinear finite element model (FEM) using ATENA-3D software to simulate the axially compressive behavior of circular steel tube confined concrete (CSTCC) columns infilled with ultra high performance concrete (UHPC) was presented in this paper. Some modifications to the material type "CC3DNonlinCementitious2User" of UHPC without and with the incorporation of steel fibers (UHPFRC) in compression and tension were adopted in FEM. The predictions of utimate strength and axial load versus axial strain curves obtained from FEM were in a good agreement with the test results of eighteen tested columns. Based on the results of FEM, the load distribution on the steel tube and the concrete core was derived for each modeled column. Furthermore, the effect of bonding between the steel tube and the concrete core was clarified by the change of friction coefficient in the material type "CC3DInterface" in FEM. The numerical results revealed that the increase in the friction coefficient leads to a greater contribution from the steel tube, a decrease in the ultimate load and an increase in the magnitude of the loss of load capacity. By comparing the results of FEM with experimental results, the appropriate friction coefficient between the steel tube and the concrete core was defined as 0.3 to 0.6. In addition to the numerical evaluation, eighteen analytical models for confined concrete in the literature were used to predict the peak confined strength to assess their suitability. To cope with CSTCC stub and intermediate columns, the equations for estimating the lateral confining stress and the equations for considering the slenderness in the selected models were proposed. It was found that all selected models except for EC2 (2004) gave a very good prediction. Among them, the model of Bing et al. (2001) was the best predictor.
The concrete damage plasticity (CDP) model is widely used to simulate concrete behaviour using either implicit or explicit analysis methods. To effectively execute the models and resolve convergence issues in implicit analysis, activating the viscosity parameter of this material model is a common practice. Despite the frequent application of implicit analysis to analyse concrete structures with the CDP model, the viscosity parameter significantly varies among available models and lacks consistency. The adjustment of the viscosity parameter at the element/structural level disregards its indirect impact on the material. Therefore, the accuracy of the numerical model is confined to the validated range and might not hold true for other values, often explored in parametric studies subsequent to validations. To address these challenges and develop a unified numerical model for varied conditions, a quasi-static analysis using the explicit solver was conducted in this study. Fifteen thick flat plates tested under load control with different geometries and different eccentric loads were considered to verify the accuracy of the model. The study first investigated various concrete material behaviours under compression and tension as well as the concrete tensile strength to identify the most reliable models from previous methodologies. The study compared the results using both implicit and explicit analysis. It was found that, in implicit analysis, the viscosity parameter should be as low as 0.0001 to avoid affecting material properties. However, at the structural level, the optimum value may need adjustment between 0.00001 to 0.0001 with changing geometries and loading type. This observation raises concerns about further parametric study if the specific value of the viscosity parameter is used. Additionally, activating the viscosity parameter in load control simulations confirmed its inability to capture the peak load. Conversely, the unified explicit model accurately simulated the behaviour of the test specimens under varying geometries, load eccentricities, and column sizes. This study recommends restricting implicit solutions to the viscosity values proposed in this research. Alternatively, for highly nonlinear problems under load control simulation, explicit analysis stands as an effective approach, ensuring unified parameters across a wide range of applications without convergence problems.
