Antimutagenic Effects of the Edible Mountain Herb Juices Against Trp-P-1 and 2-AF (Trp-P-1과 2-AF에 대한 산채류 생즙의 항돌연변이 효과)

  • On the mutagenicity induced by 3-amino-l,4-dimethyl-5-H-pyrido[4,3-b]indol (Trp-P-1) and 2-aminotluorene (2-AF), the antimutagenic effects of edible muntain herb juices were examined by the Ames assay using Salmonella typhimurium TA98 and TA100. Juices prepared from aralia bud, small water dropwort, mugwort, roots of belltlower and sedum didn't have mutagenicity. Most of sample juices showed the antimutagenicity. Especially, juices prepared from aralia bud, small water dropwort and mugwort were found to possess strong antimutagenic effects. Sedum was moderatly effective and root of belltlower had little effect on mutagenicity caused by Trp-P1 and 2-AF. The experimental results with TA98 were similar to those with TA100 in the antimutagenicity test of edible mountain herb juices. In this study, antimutagenicity on Trp-P-1 was more effective than that on 2-AF.

Effects of Hizikia fusiforme Fractions on Melanin Synthesis in B16F10 Melanoma Cells (톳 분획물이 B16F10 흑색종 세포에서의 멜라닌합성에 미치는 영향 연구)

  • The objective of this study was to evaluate the anti-melanogenic effects of Hizikia fusiforme (HF) fractions in ${\alpha}$-melanocyte stimulating hormone-induced B16F10 mouse melanoma cells. Ethanol extractions of Hizikia fusiforme (EEHF) were subjected to fraction by using dichloromethane (CFHF), ethyl acetate (EAFHF), butanol (BFHF), and water (WFHF). EEHF, CFHF, and EAFHF inhibited tyrosinase activity and melanin synthesis in B16F10 mouse melanoma cells in a dose-dependent manner. The melanin contents were inhibited by 40.5% and 33.2% in response to treatment with 50 ${\mu}g/ml$ of EEHF and CFHF, respectively. In addition, tyrosinase activities showed a 53.3% and 54.1% reduction in treatment with 50 ${\mu}g/ml$ of EEHF and CFHF. Western blotting analysis showed that EEHF, CFHF, and EAFHF inhibited tyrosinase, TRP-1, TRP-2, and MITF expression in a dose-dependent manner. In conclusion, these findings indicate that ethanol and dichloromethane fractions of Hizikia fusiforme, which inhibit melanin synthesis and tyrosinase activity, are effective skin-whitening agents.

  • Human skin is constantly exposed to ultraviolet (UV) radiation, polluted air, and chemical products. UV rays, in particular, will affect the skin in a variety of ways, including causing wrinkles, fine lines, rough skin, and xeroderma, thereby resulting in skin aging. This study aimed to investigate the whitening effects of Juniperus rigida Sieb., which is a cedar tree that is found throughout the world. The whitening efficacy that was measured by tyrosinase inhibition revealed 49.4% efficacy in water extract and 80.0% efficacy in ethanol extract. Among the B16F10 black cells, the effect of the ethanol extract was higher than the effect of the water extract in the restrain creation of melanin pigment, tyrosinase, microphthalmia-associated transcription factor (MITF), tyrosinase related protein-1 (TRP-1), and tyrosinase related protein-2 (TRP-2). Thus, the results of these studies demonstrated that the ethanol extract had greater efficacy than the water extract and Juniperus rigida Sieb. Ethanol extracts could be utilized as materials for functional cosmetics, such as whitening products.

Whitening Effect of Extracts and Fractions from Diospyros kaki calyx (감꼭지 추출물과 분획물의 미백효과)

  • This study was performed to determine the whitening effect of several solvent fractions of Diospyros kaki extracts. Fractions from ethanol extracts of D. kaki were prepared by a systematic fractionation procedure using hexane, ethyl acetate, butanol, and $H_2O$. The ethyl acetate fractions were used to evaluate the inhibitory effects on tyrosinase activity and melanin synthesis in B16F10 melanoma cells. Ethyl acetate fractions suppressed the expression of tyrosinase, TRP-1 and TRP-2 in B16F10 melanoma cells. These results showed that ethyl acetate fractions of D. kaki could be developed as a skin whitening material in cosmetics.

  • Arabidopsis trp1 mutant plants, deficient in phosphoribosyI anthranilate transferase (PAT) activity, accumulate anthranilate compounds, which render them blue fluorescence. The visible phenotype of trp1 makes the PAT gene an excellent reporter gene in the mutant. In order to develop a system for the homologous recombination using the phenotypic characteristic of trp1-100, we established optimum conditions for the isolation and regenera tion of protoplast from auxin-conditioned, trp1-100 root cultures. Trvptophan had to be supplemented in the germination medium for the efficient cell division and subsequent plant regeneration. When 10 uM tryptophan was added to the germination medium, we obtained the highest yield of protoplasts ($3{\times}10^6 cells/g$) and the best viability (92%). Thirty percent of root protoplast derived from meristematic cells underwent cell division within 5 days in callus-induction medium. Regenerated rosette leaves (2-3 mm) were transferred to rooting medium and finally acclimated to the soil for flowering.

Inhibitory Effects of Herb and Seeds of Oenothera odorata on the Melanogenesis (월견초 및 월견자의 멜라닌 생성 억제효과)

  • 목적 : 월견초는 다량의 불포화 지방산인 리놀렌산과 감마 리놀렌산을 함유하고 있으며, 천식성 기침이나 아토피피부염에 효능이 있는 것으로 알려져 있다. 본 연구는 월견초의 전초와 종자 추출물의 피부 멜라닌 합성에 대한 억제 효과를 조사하였다. 방법 : B16F10 멜라닌세포주를 이용하여 멜라닌, tyrosinase 활성 및 세포생존율을 측정하였다. 또한 멜라닌 합성- 관련효소인 tyrosinase, TRP-1, TRP-2의 단백질발현과 $\alpha$-MSH를 처리하여 색소침착을 유도 한 뒤 단백질 발현을 조사하였다. 결과 : 월견자는 B16F10 세포의 멜라닌 합성을 $5\;{\mu}g/ml$$10\;{\mu}g/ml$ 농도에서 각각 대조군의 81.3%, 68.3%로 억제하였고 tyrosinase의 활성도 이와 유사하게 억제하였다. 멜라닌 합성-관련효소들의 단백질발현을 관찰한 결과 월견초와 월견자는 tyrosinase 발현을 억제하였으며 TRP-1과 TRP-2의 발현에는 영향을 주지 않았다. 특히 $\alpha$-MSH에 의한 과색소 유도 시 tyrosinase 발현이 현저하게 감소되었으며, 월견자의 멜라닌 합성 억제 효과가 월견초 보다 높게 나타났다. 결론 : 이상의 연구 결과 월견자는 멜라닌세포의 tyrosinase 단백질 발현과 tyrosinase 활성을 억제하여 멜라닌 생성을 감소시키는 것으로 사료된다.

Inhibitory effects of Kirengeshoma koreana Nakai on Melanogenesis in B16F10 melanoma cells

  • Kirengeshoma koreana Nakai (K. koreana)was Saxifragaceae and rare plants in Korea, which is classified as an Critically Endangered (CR) species in Korea. Therefore, most of the studies on it were ecological and taxonomic, and there are no studies on biological activity. In this study, we evaluated the whitening activity of K. koreana extract (KKE). Melanogenesis Inhibitory effects were demonstrated by western-bot and RT-PCR for the effects of KKE on MITF, tyrosinase, TRP-1 and TRP-2 in IBMX-treated B16F10 melanoma cells. IBMX were reported as melanin synthesis enhancers. It could increase intracellular melanin synthesis by activation of the microphthalmia-associated transcription factor (MITF) signaling pathway. KKE showed no cytotoxicity at B16F10. In addition, KKE effectively inhibited the protein and mRNA levels of MITF, tyrosinase, TRP-1 and TRP-2. In conclusion, KKE inhibited melanin synthesis by inhibiting the expression of MITF and its downstream pathways tyrosinase, TRP-1 and TRP-2. Therefore, it was confirmed that K. koreana is a valuable resource for functional cosmetic and biomaterials.

  • The antimutagenic activity of resveratrol on the mutagenicity induced by Trp P-1 (3-amino-1,4-dime-thyl-5H-pyrido{4,3-b}indole) was studied using the Ames test with Salmonella typhimurium TA98 and 100. Trp P-1 showed strong mutagenecity in S. typhimurium TA98, but was higly decreased mutagenecity in S. typhimurium TA100. This result suggests that the mutagenecity of Trp P-1 can be mainly induced by the DNA lesions causing frame shift. Resveratrol itself did not show antibacterial effect upon 300 $\mu\textrm{g}$/assay. Resveratrol showed the strongest inhibitory effect with dose dependent manner on the mutagenicity induced by Trp P-1. The inhibition rates of resveratrol at concentration of 2, 10, 25, 50, 100, 300 $\mu\textrm{g}$/assay were 13%, 37%, 52%, 65%, 81%, 89%, respectively.

The Effects of Soybean Protopectinase on Melanin Biosynthesis (효소(Protopectinase) 처리한 대두가 세포내 멜라닌 생성에 미치는 영향)

  • This study was performed to assess the antioxidant activities and whitening effects of protopectinase enzymes and mechanical maceration from soybeans on melanin synthesis. The whitening effects of enzyme treatment and mechanical maceration were examined by an in vitro mushroom tyrosinase assay and by assessing markers in B16BL6 melanoma cells. We assessed inhibitory effects on the expression of melanogenic enzymes, including tyrosinase, tyrosinase-related protein 1 (TRP-1), and tyrosinase-related protein 2 (TRP-2) in B16BL6 cells. Inhibitory effects on free radical generation were determined by measuring DPPH and hydroxyl radical scavenging activities. In DPPH radical scavenging activity, enzyme treatment and mechanical maceration had a potent anti-oxidant activity in a dose-dependent manner and significantly inhibited tyrosinase activity in vitro and in B16BL6 melanoma cells. There was also an inhibition in the expression of tyrosinase, TRP-1, and TRP-2 in B16BL6 melanoma cells. Our results show that soybean protopectinase treatment inhibits melanogenesis, with the underlying mechanism possibly due to the inhibition of tyrosinase activity and tyrosinase, TRP-1, and TRP-2 expression. We suggest that soybean protopectinase should be contained as natural active ingredients for antioxidant and whitening cosmetics.

Antioxidant and Whitening Effects of Sorbus commixta HEDL Cortex Extract (정공피 추출물의 항산화 활성 및 미백효과에 관한 연구)

  • This study was performed to assess the antioxidant activities and whitening effects of Sorbus commixta HEDL cortex on melanin synthesis. The whitening effects of Sorbus commixta HEDL cortex water extracts were examined by in vitro mushroom tyrosinase assay and B16BL6 melanoma cells. We assessed inhibitory effects of Sorbus commixta HEDL cortex water extracts on expression of melanogenic enzyme proteins including tyrosinase, tyrosinase-related protein 1 (TRP-1) and tyrosinase-related protein 2 (TRP-2) in B16BL6 cells. Inhibitory effects of Sorbus commixta HEDL cortex onto free radical generation were determined by measuring DPPH and hydroxyl radical scavenging activities. Our results indicated that Sorbus commixta HEDL cortex water extracts effectively inhibited free radical generation. These results show that Sorbus commixta HEDL cortex inhibited melanin production on the melanogenesis. The underlying mechanism of Sorbus commixta HEDL cortex on whitening activity may be due to the inhibition of tyrosinase activity and tyrosinase, TRP-1, TRP-2 expression. We suggest that Sorbus commixta HEDL cortex may be contain new natural active ingredients for antioxidant and whitening cosmetics.
