Endophytes are microorganisms that live in the internal tissues of plants without harming the host plants. In this symbiotic relationship, the host plants provide nutrients and shelter to the endophytes, in turn, endophytes can promote the growth of host plants and act as a biological control agents against plant pathogens. Plant-microbe interactions like this are noted for natural methods for sustainable agriculture and environmental conservation. However, in spite of the infinite potential, there are only a few reports on the endophytes present in ginseng. In this study, we isolated and identified the endophytes from Panax ginseng seeds and evaluated the biological activities (IAA production ability, nitrogen fixation ability, phosphate solubilization capacity, siderophore production ability, and antifungal activities) of the endophyte isolates. Eight different endophytes were identified by 16S rRNA sequencing. Most of the endophytes have antibiotic and plant growth promoting (PGP) activities. Particularly, PgSEB5-37E have the highest antibiotic activity, both PgSEB5-37B and PgSEB5-37H have high PGP traits such as an abilities to produce IAA, solubilize phosphate and fix nitrogen. These results indicated that the endophytes from P. ginseng seeds may have applicable value to many industries. In order to use the isolated endophytes, quantitative analysis and field tests are needed to be performed.
Lichens are composite organisms consisting of a symbiotic association of a fungus with a photosynthetic partner (the photobiont or phycobiont), usually either a green alga or cyanobacterium. According to more recent studies, the biological activities of lichens and lichen substances include an antibiotic activity, antitumor and antimutagenic activity against human immunodeficiency virus (HIV), allergenic activity, plant growth inhibitory activity, and enzyme inhibitory activity. This study screened lichen extracts with a potent in vitro antifungal activity against plant diseases caused by phytopathogenic fungi. The compounds were isolated from Stereocaulon alpinum and Sphaerophorus globosus, and their chemical structures were identified as methyl hematommate, methyl β-orsellinate, 5-hydroxyferulic acid, sphaerophorin, and 2-heptyl-4,6-dimethoxybenzoic acid by electron ionization mass spectrometry (EI-MS) and nuclear magnetic resonance (NMR) spectral analyses. In vitro disease control against Alternaria mali, Cochliobolus miyabeanus, Colletotrium gloeosporioides, and Verticillum dahliae was evaluated. And among the five compounds, only methyl hematommate was effective against A. mali, C. miyabeanus, and C. gloeosporioides. The compounds were isolated from these lichens, which have a similar biosynthetic pathway, respectively. This is the first report of these compounds being isolated from these lichens.
To investigate the new Yoghurt starter, Lactobacillus helveticus YM-1, which was selected among 14 Lactobacillus strains, and Streptococcus thermophilus CH-1 were inoculated together in reconstituted non-fat skim milk medium and their growth characteristics and cultural conditions for the mixed culture were examined. The main results of this study were obtained as follows. The typical symbiotic growth was shown between the two strains and pH and temperature for optimal growth were 6.5 and 4$0^{\circ}C$, respectively. Heat treatment of milk was most effective at 10$0^{\circ}C$ for 30 min. The cell-free filtrate of Lactobacillus helveticus YM-1 had stimulatory effect on Streptococcus thermophilus CH-1 but the reverse case was slightly observed. Significant difference was observed in the proteolytic activities between Lactobacillus helveticus YM-1 and Streptococcus thermophilus CH-1. The former liberated 135$\mu\textrm{g}$ free amino acid per $m\ell$ of cultured milk, the latter 35$\mu\textrm{g}$ per $m\ell$.
In order to screen new allogeneic mixed lymphocyte culture (allo-MLR) inhibitor which is expected to be immunomodulating drug lead, we have investigated allo-MLR inhibitory activity on the marine-derived symbiotic microorganisms (1,895 strains) from the marine algae. The potent inhibitory activities (over 45% inhibition of proliferation at 10 and 2 ${\mu}g/ml$) without cytotoxicity were observed in the extracts of 46 strains. While, the significant stimulating activities (over 100% proliferation at 10 and 2 ${\mu}g/ml$) without cytotoxicity were observed in the extracts of 5 strains. In the second assay using 46 bioactive strains, 14 strains exhibited again significant allo-MLR inhibitory activity. Finally, 11 strains among the 14 strains inhibited proliferation and IFN-${\gamma}$ production of CD4+ T cells during the stimulation with specific antigen in the third assay. On the basis of above results, the marine algae is nice source for isolation of immunomodulating microorganism, and the marine algae-associated microorganism is also nice target for development of the new immunomodulating drug lead.
Rhizobium meliloti 102 F 51, the symbiotic partner of alfalfa, was mutagenized with N-methyl-N'-nitro-N-nitrosoguanidine (NTG) and UV-irradiation. Three group of mutants which form white, white-pink and red nodules were selected. The adetylene reduction activity, nodulation activity, amount of heme synthesis during the nodulation, and ${\delta}-aminolevulinic$ acid synthetase (ALAS) and ${\delta}-aminolevulinic$ acid dehydratase (ALAD) activities in free living rhizobia and bacteroid states of the each group of mutants were compared. The mutants forming white nodules showed lower acetylene reduction activity compared to those of red nodule forming mutants. The two key enzymes for the heme synthetic pathway, ALAS and ALAD activities of the mutants forming red nodules was much higher than those of the mutants forming white nodules in bacteroid state, however no significant difference was observed in free living state. In the nodules the ALAS was detected only in bacteroid fraction, while ALAD was detected both in bacteroid and plant fraction. ALAS was dramatically increased with the heme synthesis during the nodulation, while ALAD was decreased in plant fraction but slight increase was observed in bacteroid fraction.
Effects of environmental factors of N, P, pH, moisture, temperature and oxygen on growth and nitrogen fixation activity of kummerowia striate (Thunb.) Schindler seedling, bearing symbiotic root nodules, were quantitatively analyzed during the growing period. The specific nitrogenase activity (ARA) of nodules showed the maximum value of 187 μmol C₂H₄g fr wt-1 h-1 6 weeks after seeds were germinated. The total nitrogenase activities per plant attained as 1.56, 0.85, 0.09 and 4.0, 1.11, 0.04 μmol C₂H₄hr-1, respectively for the treatments of 1, 3 and 5 mM NO₃ ̄and NH₄+ on the 60th day. While the plant grown in N-free media for 20 days after treatments of 5 mM NH₄+for 40 days resulted in 30 mg fr wt of nodule formation and exhibited the relative activities of 152% and 162% for total and specific ARA in comparison with those of control plant grown with N-free for 60 days. Total biomass and ARA was by 70% and 86% lower in N and P deficiency, respectively. The N and P deficient plot showed 70% and 86% decreases of total biomass and ARA in comparison with those of control. The plant grown with N-free for 20 days after pretreatment with N and P free media for 40 days showed the relative values of 77%, 118% and 150%, respectively for nodule biomass, total and specific ARA in comparison with those of control. The treatment with acid or alkali gradients resulted in significant decreases of nodule biomass and ARA. The optimum temperature and pO₂for ARA were 30°C and 40 kPa, respectively. Two peaks of diurnal variation appeared at 11:00 and 23:00 o'clocks by the continuous light condition. The plants with water stress by temporary wilting point rsulted in 95~97% inhibition for nodule respiration, transpiration and specific ARA. Transpiration and ARA ware recovered to 88% and 38% of those of water unstressed plants, respectively, 6 hours after the plants were rewatered from water stressed condition.
Lichen-forming (LFF) or lichenicolous fungi (LCF) were isolated from the lichens collected at‘Backwoon’mountain area,‘Chiri’mountain area and‘Sorok’island in the southern regions of Korea and were screened for antagonistic efficacy against several phyto-pathogenic fungi. Symbiotic algae-free LFF and LCF were isolated by the following methods: I) discharged spores (ascospores), II) macerated thallus suspension and III) direct use of thallus fragments. Among 58 isolates obtained from 34 lichens, 8 isolates showed antifungal activity against Rhizoctonia solani. Antifungal activities of the strongest antagonistic isolate (LB9810) originated from the thallus of Parmelia quercina lichen were evaluated against 15 phyto-pathogenic fungi. When crude methanol extract of mycelia of the LB8910 isolate was employed at the rate of 0.5% (v/w), fungal growth of Magnaporthe grisea and Rhizoctonia solani was severly and Rhizoctonia solani was severly inhibited as much as approximately 60% compared to control. Growth of various food-borne same extract. The extract was successively partitioned with n-hexane, ethyl acetate and n-butanol. n-Hexane fraction displayed the strongest antifungal activities against R. solani. The LB9810 isolate was finally identified as Fusarium equiseti (Corda) Sacc., which has not been reported as LFF or LCF yet. Therefore, it is very likely that F. equiseti isolated it the study was originated from the contaminants associated with thallus fragments rather than from LFF or LCF.
This study aimed at examining the mediation effects of depressive symptoms on the association between social activity participation and marital satisfaction among couples in later life. The study included 1,196 married couples aged 65 or above who participated in the 7th Korean Longitudinal Study of Aging in 2018. The study variables were husbands' and wives' social activity participation, marital satisfaction, and depressive symptoms. Covariates were individual age, education level, subjective health, couples' household income, and number of children. The Actor-Partner Interdependence Mediation Model (APIMeM) and bootstrapping techniques were used to test the significance of the mediating impacts of depressive symptoms based on the dyadic data structure. The results showed that for both husbands and wives, active participation in social activities had significant effects on increasing marital satisfaction through lowering depressive symptoms. However, only the participation of husbands in social activities was positively associated with their own marital satisfaction by reducing their own and their partner's depressive symptoms. These findings suggest that the active social participation of individuals salient for promoting the quality of marital relationships in later life, particularly for preventing their own and their spouse's depressive symptoms.

The main purpose of this research is to evaluate the effect of light quality on the growth and physiological activities of Laurus nobilis plants growing indoors, the L. nobilis seedlings were grown under four types of lighting for 180 days. The seedlings were grown under 4 different treatments((n=9 seedlings per treatment): control, control supplemental UV-B irradiated(+UV), FR reduced(-FR) and simultaneously supplemental UV-B irradiated and FR reduced(+UV-FR)). It was found that UV-B irradiation(+UV) reduced dry weight and leaf area, and increased leaf thickness and the amount of UV-absorbing compounds per unit leaf area. In contrast, a reduction in far-red(FR) light did not affect any of these parameters. Interestingly, however, the elongation growth and net photosynthetic rate of the L. nobilis seedlings grown under simultaneous UV-B irradiation and FR reduction(+UV-FR) were significantly decreased than the control treatment. From these results, it is concluded that the light quality has a large effect on the indoor growth of L. nobilis. This study can suggest basic information for managed in the L. nobilis in indoor using light quality.
Polydnaviruses (PDVs) are a group of double-stranded DNA viruses symbiotic to some endoparasitoid wasps. Cotesia plutellae bracovirus (CpBV) is a PDV symbiotic to an endoparasitoid wasp, C. plutellae, parasitizing young larvae of Plutella xylostella. An early expressed gene, CpBV-ELP1, plays an important role in the parasitism by suppressing host cellular immunity by its cytotoxic activity against hemocytes. This study aimed to test its oral toxicity against insect pest by expressing it in a recombinant tobacco plant. A recombinant CpBV-ELP1 protein was produced using a baculovirus expression system and secreted to cell culture medium. The cell cultured media were used to purify CpBV-ELP1 by a sequential array of purification steps: ammonium sulfate fractionation, size exclusion chromatography, and ion exchange chromatography. Purified rCpBV-ELP1 exhibited a significant cytotoxicity against Spodoptera exigua hemocytes. CpBV-ELP1 was highly toxic to the fifth instar larvae of S. exigua by injection to hemocoel. It also showed a significant oral toxicity to fifth instar larvae of S. exigua by a leaf-dipping assay. CpBV-ELP1 was cloned into pBI121 vector under CaMV 35S promoter with opaline synthase terminator. Resulting recombinant vector (pBI121-ELP1) was used to transform Agrobacterium tumefaciens LBA4404. The recombinant bacteria were then used to induce callus of a tobacco (Nicotiana tabacum Xanthi) leaves and subsequent generation (T1) plants were selected. T1 generation tobacco plants expressing CpBV-ELP1 gave significant insecticidal activities against S. exigua larvae. These results suggest that CpBV-ELP1 gene can be used to control insect pests by constructing transgenic crops.
