The Protective Effects of Acupuncture on Oxidative Stress Caused by Cadmium in the Kidney (카드뮴으로 유발된 산화적 스트레스에 대한 침 자극의 신장 보호 효과)

  • Objectives : This study was performed to inquire into the protective effects of acupuncture on oxidative stress caused by cadmium accumulation in the kidney. Methods : Sprague-Dawley male($150{\pm}30g$) rats were stabilized for 1 week and divided into 5 groups: normal, control, $LR_3$ acupuncture, $BL_{23}$ acupuncture and sham acupuncture. For three days experimental groups received oral doses of cadmium 2 mg/kg twice a day. Acupuncture was applied bilaterally at each point 10 times for two weeks. The depth of stimulation was 1 mm at right angles and torsion of acupuncture was produced 2 times per second for 1 minute. The kidneys were extracted and weighed after two weeks, and renal function was confirmed through blood urea nitrogen(BUN). We measured reactive oxygen species of the serum and kidney, and compared expression levels of superoxide dismutase(SOD), catalase, glutathione peroxidase(Gpx), nuclear factor erythroid derived 2-related factor 2(Nrf-2), heme oxygenase-1(HO-1), nuclear factor-${\kappa}B$(NF-${\kappa}B)$, cyclooxygenase-2(COX-2), inducible nitric oxide synthase (iNOS), Bax and Cytochrome c. Results : The $LR_3$ acupuncture group and $BL_{23}$ acupuncture group experienced significantly increased kidney weight, and decreased BUN compared to control group. In terms of oxidative stress, the $LR_3$ acupuncture group and $BL_{23}$ acupuncture group experienced significantly reduced reactive oxygen species compared to the control group. Conclusions : The $LR_3$ acupuncture group and $BL_{23}$ acupuncture group experienced showed the effects of antioxidant, anti-inflammatory and apoptosis protection. The $BL_{23}$ acupuncture group was more effective than $LR_3$ acupuncture group.

Effect of Cnidium officinale Extract on Recovery Capability of Allergic Contact-Dermatitis in Rat (흰쥐의 알레르기성 접촉피부염의 회복에 미치는 천궁추출물의 영향)

  • The epidermal recovering-effects of the Cnidium officinale extract after inducing contact-dermatitis by 1-chloro-2,4-dinitrochlorobenzene (DNCB) showed by measuring thickness of skin, identifying IgE amounts in plasma, and activities of anti-oxidant enzymes using histochemical and biochemical methods. By three-time applications of the extract, morphological changes of dermatitis in impaired region were recovered dramatically and shape of skin surface and thickness of epidermis were restored to be normal. Also, lipid content was recovered to the level of normal state. We suggested that extract treatment lowered a hypersensitive reaction by decreasing of IgE level in blood and restored activities of superoxide dismutase and catalase. In conclusion, we proposed that C. officinale extract might be used as natural resources for treating effectively allergic contact-dermatitis.

Isolation of main component and antioxidant activities on the stem and root of Rosa rugosa (해당화 줄기 및 뿌리의 주성분 분리 및 항산화활성)

  • Total phenol content was obtained from the stem extract of 7.8 (g/100g, D.W.) and root extract of 10.5 (g/100g, D.W.) on Rosa rugosa. S-4 and R-2 compounds were isolated from the stem and root of R. rugosa. The structure of S-4 and R-2 compounds was assigned as catechin by $^{1}H,\;^{13}C$ CNMR signal and TOF-MS. The tannin content of stem extract was 752.5 (mg/100g, D.W.) as 6.3 of gallic acid, 61.5 of epicatechin and 684.8 of catechin. The tannin content of root extract was higher about 2 times (1676.0) as 6.8 of gallic acid, 160.3 of epicatechin and 1508.5 of catechin than that of stem. It was shown that catechin was main compound in the stem and root extracts of R. rugosa. Antioxidant activities were stronger low concentration, and were not different of stem and root extracts. As the results, stem and root of R. rugosa were evaluated, as natural antioxidant resource.

Antioxidative Activity and Nitrite Scavenging Ability of Ethanol Extract from Phyllostachys bambusoides (대나무 에탄올추출물의 항산화 효과 및 아질산염 소거작용)

  • Efficacy of antioxidative activity and nitrite scavenging ability of ethanol extract of Phyllostachys bambusoides S. et Z. (P. bambusoides) was investigated. Electron-donating ability of ethanol extract at $RC_{50}$ was $116.75\;{\mu}g/mL$. After addition of 0.92 mg/mL ethanol extract, autooxidation of pyrogallol decreased to 44% by superoxide dismutase-like activity. In antioxidative activity of ethanol extract against linoleic acid during incubation times of 4 and 6 day at $40^{\circ}C$, TBA values decreased by 74.76 and 58.48% with addition of 50 mg/mL, respectively. Nitrite scavenging ability showed the most remarkable effect at pH 1.2, decreasing to 43% by addition of 0.2 mg/mL. These results suggest that ethanol extract of P. bambusoides can be used in bioactive and functional material.

Comparison of Biological Activities of Acer mono and A. okamotoanum Extracts by Water Extracton and Low Temperature High Pressure Extraction (고로쇠와 우산고로쇠 저온고압 수용성 추출물의 유용생리활성 비교)

  • Hepatoprotective and antioxidant activities of Acer mono and A. okamotoanum were compared after being extracted by low temperature and high pressure (LTHP) at 20 MPa and $60^{\circ}C$ for 15 minutes. Extraction yield of both A. mono and A. okamotoanum was increased about 40~43% by this extraction process. On scavenging activities, the bark of A. okamotoanum from this extraction process showed the highest activity as about 97%. This value was higher than that from conventional water extraction and A. mono extracts. Both of A. mono and A. okamotoanum showed high ability on nitrite scavenging, but decreasing tendency according to increasing of pH. On SOD-like test, A. okamotoanum had the highest activity as 46.28% at $1.0\;mg/m{\ell}$ concentration. A. okamotoanum extracted by LTHP also showed the highest activity as 197.38% in adding $1.0\;mg/m{\ell}$ concentration. Generally, the extracts from low temperature and high pressure extraction process are higher hepatoprotective and antioxidant activities than that from conventional water extraction. It can conclude that the bark of A. okamotoanum has better biological activities than other parts of A. mono.

Effects of Mulberry Leaf Extract on Oxygen Radicals and Their Scavenger Enzymes in Serum of Rats (뽕잎추출물이 혈청중의 활성산소 및 제거효소에 미치는 영향)

  • This study was designed to investigate the effects of mulberry leaf extract (MLE) on oxygen radicals and their scavenger enzymes in serum of rats. Sprague-Dawley (SD) male rats (160${\pm}$10g) were fed experimental diets (MLE-100 and MLE-300 groups) added 100 and 300mg/kg BW/day for 6weeks. Triglyceride (TG) levels were significantly inhibited (10% and 20%) in MLE-100and MLE-300 groups, but there were no significant differences in total, LDL-and HDL- cholesterol levels in both MLE-100 and MLE-300 groups. Hydroxyl radical ($.$OH) formations resulted in a marked decreases(20∼25%) in MLE-100 and MLE-300 groups compared with control group, while superoxide radical (O2.-)and hydrogen peroxide formations resulted in a considerable decreases(7∼10% and 5∼10%) in MLE-100 and MLE-300 groups compared with control group. Lipid peroxide (LPO)and oxidized protein(>C=O group) productions resulted in a significant decreases (10% and 6∼10%) in MLE-100 and MLE-300 groups compared with control group. Superoxide dismutase (SOD)and catalase (CAT) activities were remarkably increased (30% and 40∼55%) in MLE-100 and MLE-300 groups, but glutathione peroxidase (GSHPX) activities were significantly increased (10∼15%) in MLE-100 and MLE-300 groups compared with control group. These results suggest that anti-aging effect of mulberry leaf extract (MLE) may play a pivotal role in attenuating a various agerelated changes.

Protective Role of Fucoidan in Cerebral Ischemia-Reperfusion Injury through Inhibition of MAPK Signaling Pathway

  • Fucoidan has been reported to exhibit various beneficial activities ranging from to antivirus and anticancer properties. However, little information is available about the effects of fucoidan on cerebral ischemia-reperfusion injury (IRI). Our study aimed to explore the effects of fucoidan on cerebral IRI, as well as the underlying mechanisms. Sprague-Dawley (SD) rats were randomly subjected to four groups: Sham, IRI+saline (IRI+S), IRI+80 mg/kg fucoidan (IRI+F80), and IRI+160 mg/kg fucoidan (IRI+F160). Fucoidan (80 mg/kg or 160 mg/kg) was intraperitoneally injected from 7 days before the rats were induced to cerebral IRI model with middle cerebral artery occlusion (MCAO) method. At 24 h after reperfusion, neurological deficits and the total infarct volume were determined. The levels of inflammation-associated cytokines (interleukin (IL)-$1{\beta}$, IL-6, myeloperoxidase (MPO), and tumor necrosis factor (TNF)-${\alpha}$), oxidative stress-related proteins (malondialdehyde (MDA) and superoxide dismutase (SOD)) in the ischemic brain were measured by enzyme-linked immunosorbent assay (ELISA). Besides, the levels of apoptosis-related proteins (p-53, Bax, and B-cell lymphoma (Bcl)-2) and mitogen-activated protein kinase (MAPK) pathway (phosphorylation-extracellular signal-regulated kinase (p-ERK), p-c-Jun N-terminal kinase (JNK), and p-p38) were measured. Results showed that administration of fucoidan significantly reduced the neurological deficits and infarct volume compared to the IRI+S group in a dose-dependent manner. Also, fucoidan statistically decreased the levels of inflammation-associated cytokines, and oxidative stress-related proteins, inhibited apoptosis, and suppressed the MAPK pathway. So, Fucoidan plays a protective role in cerebral IRI might be by inhibition of MAPK pathway.

Characterization of an Iron- and Manganese-containing Superoxide Dismutase from Methylobacillus Sp. Strain SK1 DSM 8269

  • A superoxide dismutase was purified 62-fold in seven steps to homogeneity from Methylobacillus sp. strain SK1, an obligate methanol-oxidizing bacterium, with a yield of 9.6%. The final specific activity was 4,831 units per milligram protein as determined by an assay based on a 50% decrease in the rate of cytochrome c reduction. The molecular weight of the native enzyme was estimated to be 44,000. Sodium dodecyl sulfate gel electrophoresis revealed two identical subunits of molecular weight 23,100. The isoelectric point of the purified enzyme was found to be 4.4. Maximum activity of the enzyme was measured at pH 8. The enzyme was stable at pH range from 6 to 8 and at high temperature. The enzyme showed an absorption peak at 280 nm with a shoulder at 292 nm. Hydrogen peroxide and sodium azide, but not sodium cyanide, was found to inhibit the purified enzyme. The enzyme activity in cell-free extracts prepared from cells grown in manganese-rich medium, however, was not inhibited by hydrogen peroxide but inhibited by sodium azide. The activity in cell extracts from cells grown in iron-rich medium was found to be highly sensitive to hydrogen peroxide and sodium azide. One mol of native enzyme was found to contain 1.1 g-atom of iron and 0.7 g-atom of manganese. The N-terminal amino acid sequence of the purified enzyme was Ala-Tyr-Thr-Leu-Pro-Pro-Leu-Asn-Tyr-Ala-Tyr. The superoxide dismutase of Methylobacillus sp. strain SK1 was found to have antigenic sites identical to those of Methylobacillus glycogenes enzyme. The enzyme, however, shared no antigenic sites with Mycobacterium sp. strain JC1, Methylovorus sp. strain SS1, Methylobacterium sp. strain SY1, and Methylosinus trichosproium enzymes.

Effects of Curcuma longa Rhizoma on MIA-induced Osteoarthritis in Rat Model (강황(薑黃)이 MIA 유도 골관절염 모델에 미치는 영향)

  • Objectives: The aim of this study was to investigate the anti-inflammatory effects of Curcuma longa rhizoma extract in an experimental rat model of osteoarthritis. Methods: Osteoarthritis was induced in rats by injecting monosodium iodoacetate (MIA) into the knee joint cavity of rats. The rats were divided into 5 groups (Normal, Control, positive comparison, low (CL) and high (CH) concentration groups). Rats in the low concentration (CL) group had MIA-induced osteoarthritis; they were treated with Curcuma longa rhizoma extract at a dose of 50mg/kg body weight. Rats in the high concentration (CH) group had MIA-induced osteoarthritis; they were treated with Curcuma longa rhizoma extract at a dose of 100mg/kg body weight. Hind paw weight distribution and ROS levels were measured. At the end of all treatments, changes in alanine aminotransferase (ALT), aspartate aminotransferase (AST), blood urea nitrogen (BUN), and creatinine levels were analyzed. In addition, inflammatory protein levels were evaluated by western blot analysis. Results: In this study, hind paw weight distribution significantly improved in the CL and CH groups, while. Reactive oxygen species (ROS) production significantly decreased in both. The levels of ALT, AST, BUN, and creatinine did not significantly change in either group. The production of nicotinamide adenine dinucleotide phosphate oxidase 4 (NOX4), $p47^{phox}$, and Ras-related C3 botulinum toxin substrate 1 (RAC1) decreased in both. Catalase, heme oxygenase-1 (HO-1) and superoxide dismutase (SOD) significantly increased in the CL and CH groups, respectively. Nuclear factor erythroid 2 (Nrf2) increased, but there were no significant differences between the experimental and control groups. Inflammatory cytokines, including nuclear factor-kappa Bp65 (NF-${\kappa}Bp65$), interleukin-1beta (IL-$1{\beta}$), and tumor necrosis factor-alpha (TNF-${\alpha}$), decreased significantly in both the CL and CH groups. Conclusions: Our results showed that Curcuma longa rhizoma extract has anti-inflammatory effects. Anti-inflammatory activity is regulated by the inhibition of inflammatory cytokines and mediators, such as NF-${\kappa}B$, therefore, it suppresses cartilage damage as well.

Anti-amnesic and Antioxidant Effect of Bunsimgieum (Fenxinqiyin) on Scopolamine-Induced Memory Impairment in Mice (Scopolamine 유발 기억력 손상 마우스 모델에서 분심기음의 항산화 및 기억력 감퇴 억제 효과)

  • Objectives: The purpose of this study was to confirm the anti-amnesic effects of Bunsimgieum (BSGE) through its favorable acetylcholine (ACh) and, acetylcholinesterase (AChE) activity, choline acetyltransferase (ChAT) mRNA expressions, and antioxidant effect on scopolamine (Sco)-induced memory impairment in C57BL/6 mice. Methods: Six groups, a total of 20 intact or 100 Sco-induced mice were used in this study, based on their body weight. Half of each group underwent passive avoidance tests and the measurement of hippocampus AChE activity, ACh content, and ChAT mRNA expression, The remaining half of each group underwent a Morris water-maze test and antioxidant defense system measurement as well. Results: Significant reductions in the step-through latency times from the passive avoidance test and reductions in the escape latency times from the Morris water-maze test were observed with increases of hippocampal AChE activities and, reductions in ACh contents and ChAT mRNA expression in hippocampus, as a result of Sco intraperitoneal treatment, in this study. Additionally, the increases in cerebral cortical MDA levels and, reductions in GSH contents, SOD activities, and CAT activities were demonstrated in the Sco control mice compared with the intact vehicle control mice, respectively. However, 28 days of consecutive oral pre-treatment of BSGE hot water extracts of 400, 200, and 100 mg/kg, respectively, markedly and dose-dependently inhibited Sco treatment-related amnesia. Conclusions: The results demonstrate that the oral administration of BSGE hot water extracts reduces Sco-induced memory impairment, through preserving ACh, related to ChAT mRNA expressions, causes AChE inhibition, and enhances the cerebral antioxidant defense system.
