Effects of Socheongryong-Tang on Inflammatory Cells in Asthmatic Mice (소청룡탕이 천식이 유발된 생쥐의 면역세포에 미치는 영향)

  • BACKGROUND : Socheongryong-Tang(小靑龍湯, SCRT), a herbal medicine, has been widely used for the control of respiratory disease in Eastern countries. Recent years SCRT was known as anti-allergic agent. However, its therapeutic mechanisms, especially an inhibitory effect on inflammatory cell infiltration and airway remodeling in allergic asthma are unclear. METHODS : We investigated the effects of SCRT on levels of IL-4, and IFN-g using flow cytometric analysis and infiltration of inflammatory cells in bronchoalveolar lavage fluid(BALF) using cytospin methods in allergen-induced asthma. RESULTS : SCRT decreased levels of IL-4, a Th2-driven cytokine. But INF-g, a Th1-driven cytokine, was not changed. With SCRT treatment, infiltration of inflammatory cells showed reduced tendency. CONCLUSION : In conclusion, we demonstrated that regulation of Th1/Th2 imbalance may be one of mechanism contributed to reduction of inflammatory cell infiltration.

Effects of Socheongryong-Tang on Immunoglobulin Production in Asthmatic Mice (소청용탕(小靑龍湯)이 천식(喘息)이 유발된 생쥐의 면역 글로불린 분비에 미치는 영향)

  • Objectives : Socheongryong-Tang(小靑龍湯, SCRT), a herbal remedy, has been widely used to treat respiratory disease such as cough and asthma in Oriental countries. Recent years SCRT was known as anti-allergic agent. However, its therapeutic mechanisms including immunoglobuline such as IgE, IgG1, IgG2a productions are unclear. Methods : We investigated the effects of SCRT on levels of antigen specific total antibody, IgE, IgG1, IgG2a using ELISA method in serum from allergen-induced asthma mice. Results : SCRT decreased level of antigen specific IgE significantly. And SCRT treated mice showed downward tendency of IgG1, a Th1 relative antibody, level. But, SCRT did not affect levels of antigen specific total antibody and IgG2a, a Th1 relative antibody. Conclusions : we demonstrated the strong possibility of SCRT as a complementary or alternative drug to western drug also demonstrated that regulation of Th1/Th2 imbalance may be one of mechanism contributed to treatment for respiratory disease by SCRT.

Effects of Socheongryong-tang and Kamichihyo-san on Mucin Secretion from Airway Goblet (소청용탕 및 가미치효산이 평흡기 배장세포로부터의 뮤신 분비에 미치는 영향)

  • In the present study, the author intended to investigate whether two oriental medical prescriptions named socheongryong-tang(SCRT) and Kamichihyo-san(KCHS) significantly affect mucin release from cultured hamster tracheal surface epithelial(HTSE) cells. Confluent HTSE cells were metabolically radiolabeled with ³H-glucosamine for 24 hrs and chased for 30 min in the presence of SCRT or KCHS to assess the effect of each agent on ³H-mucin release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase(LDH) release. Also, the effects of SCRT and KCHS on contractility of isolated tracheal smooth muscle were investigated. The results were as follows: (1) SCRT significantly inhibited mucin release from cultured HTSE cells, without cytotoxicity; (2) KCHS significantly increased mucin release without cytotoxicity; (3) SCRT and KCHS did not affect contractility of isolated tracheal smooth muscle. We suggest that the effects of SCRT and its components should be further investigated and it is of great value to find, from oriental medical prescriptions, novel agents which have the possible inhibitory effects on mucin release from the viewpoint of management of hypersecretion of airway mucus.

Effect of Socheongryong-tang on Punghan and Pungyeol Type Common Cold : A Double Blind, Placebo Controlled Study (풍한형 및 풍열형 감모에 대한 소청룡탕의 효과 - 이중맹검, 위약대조군연구)

  • Socheongryong-tang(SCRT) is widely used to treat the common cold. The purpose of this study was to evaluate the efficacy of SCRT on Punghan type(風寒型) and Pungyeol type(風熱型) common cold. 98 registered students with commom colds of recent onset were randomized to the double blind, placebo-controlled study. SCRT extract in capsule, $5.4g(1.8g{\times}3cap)$, orally dissolved 3 times a day. The severity of illness was assessed by the physician, using a 5-point scale on start and finish. In Pungyeol type common cold, no statistically significant differences were detected between the SCRT and placebo groups for any of the measured outcomes. In Punghan type common cold, SCRT significantly reduced rhinorrhea (p=0.034) and nasal stuffiness (p=0.048) compared with placebo. This study shows that SCRT is effective in treating cold symptoms in Punghan type common cold. If patients with cold are able to be administered SCRT according to common cold type, the benefit would be expected to increase.

Comparison of Marker Components and Biological Activities of Socheongryong-tang by Different Extract Methods (소청룡탕 제조방법에 따른 성분 및 생리활성 비교)

  • Socheongryong-tang (SCRT) was one of the major traditional herbal medicines wildly used in the treatment of respiratory disease. SCRT is being commercially produced in the form of mix extracts powder and soft dry extract by different extract methods in the Korean Herbal Pharmacopeia (KHP). In this study, the contents of marker components and biological activities of the SCRT mix extract powder were compared with those of the SCRT decoction. To analyze the marker components of SCRT, nine marker from eight herbal preparations were chosen. And the method using high performance liquid chromatography (HPLC) with diode-array detector method was established for the simultaneous analysis. Method validation was accomplished by linearity, precision test, and recovery test. The contents of nine marker components in this extract was ascertained by ratio. The biological activities were examined the effect of SCRT on anti-oxidation and pro-inflammation mediated by LPS-stimulation. We confirmed that both of SCRT mix extrct powder and decoction have the similar contents on total polyphenol and flavonoid and inhibited the secretion of nitric oxide (NO), $IL-1{\beta}$, IL-6, tumor necrosis factor $(TNF)-{\alpha}$ and the expression of iNOS, COX-2, $IL-1{\beta}$, IL-6, $TNF-{\alpha}$. These results suggest that SCRT mix extract powder and decoction have a significant correlation.

Protective effect of Socheongryong-Tang on hydrogen peroxide-induced hepatotoxicity (소청룡탕(小靑龍湯)의 과산화수소로 유도된 간세포 독성에 대한 보호효과)

  • Objectives : Socheongryong-Tang (小靑龍湯, SCRT) has been widely used to treat respiratory disease. In this study, we investigated the protective effects of SCRT on hydrogen peroxide-induced hepatotoxicity. Methods : In the mouse primary liver cells, SCRT was pretreated for 1 h, and 1 mM $H_2O_2$ was treated to mouse primary liver cells. Cell viability was analyzed by using 3- 4,5-Dimethylthiazol-2-yl-2,5-diphenyltetrazolium bromide (MTT) assay. Also, the activity of AST, ALT and LDH were measured for the evaluation the protective effect of SCRT on $H_2O_2$-induced hepatotoxicity. Intracellular ROS level was analyzed by FACS. Results : SCRT pretreatment decreased $H_2O_2$-induced hepatotoxicity and intracellular ROS production. Pretreatment of SCRT significantly reduced the cytotoxic effect induced by $H_2O_2$, associated with reducing DNA fragmentation and AST, ALT, LDH activities. Conclusions : These results suggest that SCRT has protective effect against $H_2O_2$-induced hepatotoxicity.

Protective Effects of Socheongryong-tang on Elastase-Induced Lung Injury (Elastase로 유도된 폐손상에 대한 소청룡탕(小靑龍湯)의 보호효과)

  • Objective: This study aimed to evaluate the protective effects of Socheongryong-tang (SCRT) on elastase-induced lung injury. Materials and Methods: The extract of SCRT was treated to A549 cells and elastase-induced COPD mice model. Then, various parameters such as cell-based cytoprotective activity and histopathological findings were analyzed. Results: SCRT showed a protective effect on elastase-induced cytotoxicity in A549 cells. This effect was correlated with analysis for caspase 3 levels, collagen and elastin contents, and gene expression of TNF-${\alpha}$ and IL-$1{\beta}$ in A549 cells. SCRT treatment also revealed the protective effect on elastase-induced COPD mice model. This effect was evidenced via histopathological findings including immunofluoresence stains against elastin, collagen, and caspase 3, and protein level of Cdc2, cyclin B1, and phospho-Erk1/2 in lung tissue. Conclusion: These data suggest that SCRT has pharmaceutical properties on COPD. This study provides scientific evidence for the efficacy of SCRT for clinical application to patients with COPD.

Anti-allergic effects of So-Cheong-Ryong-Tang in ovalbumin-induced allergic rhinitis model (난알부민 유도 알레르기 비염 마우스에 대한 소청룡탕(小靑龍湯)의 효능 실험연구)

  • Objective : Allergic rhinitis (AR) is an allergic inflammation of the nasal airways. Socheongryong-Tang traditionally has been used in Korea for treatment of allergy diseases. In the present study, we investigated whether SCRT suppresses the progression of AR in vivo and in vitro. Method : In this study, we investigated the effect of oral administration of SCRT on the Ovalbumin(OVA)-induced allergic rhinitis mouse models. Additionally, to find a possible explanation for the anti-inflammation effects of SCRT, we evaluated the inflammatory cytokine levels in LPS-stimulated mouse inflammation cell (RAW264.7) and mouse splenocyte. Results : The finding of this study demonstrated that SCRT reduced mast cells and inflammation cells infiltration in OVA-induced nasal cavity. Additionally, SCRT inhibited the production of inflammatory cytokines in splenocyte. However SCRT don't inhibited the production of inflammatory cytokines in RAW264.7 cells. Conclusion : Taken together, our results showed that oral administration of SCRT beneficial effects in allergic rhinitis, suggesting that SCRT might be a useful candidate for the treatment of allergic rhinitis.

Effects of Inhalable Microparticles of Socheongryong-tang on Chronic Obstructive Pulmonary Disease in a Mouse Model (COPD 동물 모델에서 소청룡탕 흡입제형의 효과)

  • Objectives: This study aimed to evaluate the effects of microparticles of Socheongryong-tang (SCRT) on chronic obstructive pulmonary disease (COPD) in a mouse model. Methods: The inhalable microparticles containing SCRT were produced by spray-drying with leucine as an excipient, and evaluated with respect to the aerodynamic properties of the powder by Andersen cascade impactor (ACI). Its equivalence to SCRT extract was evaluated using lipopolysaccharide (LPS) and a cigarette-smoking (CS)-induced murine COPD model. Results: SCRT microparticles provided desirable aerodynamic properties (fine particle fraction of $49.6{\pm}5.5%$ and mass median aerodynamic diameter of $4.8{\pm}0.3{\mu}m$). SCRT microparticles did not show mortality or clinical signs over 14 days. Also there were no significant differences in body weight, organ weights or serum chemical parameters between SCRT microparticle-treated and non-treated groups. After 14 days the platelet count significantly increased compared with the non-treated group, but the values were within the normal range. Inhalation of SCRT microparticles decreased the rate of neutrophils in blood, granulocytes in peripheral blood mononuclear cells (PBMC) and bronchoalveolar lavage fluid (BALF) and level of TNF-${\alpha}$ and IL-6 in BALF on COPD mouse model induced by LPS plus CS. This effect was verified by histological findings including immunofluorescence staining of elastin, collagen, and caspase 3 protein in lung tissue. Conclusions: These data demonstrate that SCRT microparticles are equivalent to SCRT extract in pharmaceutical properties for COPD. This study suggests that SCRT microparticles would be a potential agent of inhalation therapy for the treatment of COPD.

A Study on Effectiveness of Socheongryong-tang for Allergic Rhinitis : A Systematic Review and Meta-analysis (알레르기 비염에 대한 소청용탕(小靑龍湯)의 효과 평가 연구 : 체계적 문헌고찰과 메타분석)

  • Objectives : The purpose of this study is to assess the effectiveness and safety of Socheongryong-tang(SCRT) for allergic rhinitis(AR). Methods : We searched randomized controlled trials(RCTs) that used SCRT for AR in 8 databases(PubMed, Cochrane Library, CNKI, CiNii Articles, OASIS, NDSL, KISS, KMbase) from their inception until August 2019. The primary outcome was effective rate and scores evaluating the improvement of AR symptoms. The secondary outcome was quality of life scale, adverse events, recurrence rate, and laboratory indicators. Two researchers assessed the risk of bias in the included trials through the Cochrane Risk of Bias tool independently. The study synthesized outcomes using RevMan 5.3. Results : Eighteen RCTs involving 1686 participants were included in this review. The risk of bias was unclear for the majority of the included studies. Meta-analysis of 12 RCTs showed that there was no statistically significant difference between the SCRT group and usual care group in the effective rate(RR 1.18, 95% CI(0.98, 1.41), p=0.09, I2=46%). Meta-analysis of 5 RCTs showed that the combination treatment group of SCRT and usual care was significantly higher than the usual care group in the effective rate(RR 1.24, 95% CI(1.12, 1.38), p<0.0001, I2=0%). The SCRT group was more effective in improving nasal symptoms and quality of life than the placebo group according to one RCT. Mild adverse events such as dry mouth were identified in 5 RCTs, but no serious adverse events were reported. Conclusion : This review showed that in terms of the effective rate for AR, there was no statistically significant difference between SCRT and usual care and the combination treatment of SCRT and usual care was more effective than usual care. There were no serious adverse events. However, it is difficult to make a definite conclusion because of few included studies and heterogeneity between studies, and the quality of included studies was mostly insufficient. Further well-designed randomized controlled trials are needed.
