Anti-oxidant, Anti-inflammatory, and Wound Healing Activities of Selaginella tamariscina Leaf Extract (부처손 잎 추출물의 항산화, 항염 및 피부재생 효능)

  • The leaves of Selaginella tamariscina were used for the treatment of many diseases in traditional medicine. In the study, antioxidant, anti-inflammatory, and wound healing activities of the hot-water extract(STW) and 80%ethanol extract(STE) obtained from S. tamariscina were evaluated. As a result, the polyphenol content of STW and STE were 38.108±0.766 mg/g and 17.927±1.064 mg/g, respectively. The DPPH and ABTS radical scavenging activities with the IC50 values of the STW were over 2 times lower than that of the STE. In the MTT assay, RAW264.7 cell viability of two extracts was decreased by about 6% at 1 mg/mL, whereas for HaCaT cell viability increased by 18% at 50 ㎍/mL. In addition, STW and STE suppressed the production of nitric oxide(NO), Tumor-necrosis(TNF)-��. COX-2 and PGE2 in lipopolysaccharide(LPS) induced RAW264.7 cells. Furthermore, the STE showed wound healing effect through the promotion of skin cell migration in TNF-�� stimulated human keratinocytes. These results indicated that the STW and STE have the potential to be used as a new cosmetic active ingredients in skin care.

Evaluation of Antioxidant Fractions and Hair Loss Prevention Effects of Platycodon grandiflorum (도라지 분획물의 항산화 및 탈모예방 효과)

  • Free radicals are known to inhibit hair vitality by damaging the cell membranes of the hair follicles. The purpose of this study was to determine the antioxidant activities and the capacity for hair loss prevention of extracts from Platycodon grandiflorum. We prepared butanol (BF) and water (WF) fractions from P. grandiflorum. DPPH and ABTS radical scavenging activities were measured to investigate the antioxidant activities of the fractions. Both fractions exhibited dose-dependent antioxidant activities for DPPH radical production, and BF and WF almost completely suppressed ABTS radical production when supplied at 10 and 100 mg/ml, respectively. We confirmed a skin regeneration effect by treating human HaCaT skin cells with a range of BF and WF concentrations for 24 and 48 hr. The extract treatments accelerated cell proliferation. We also assayed the capacity of BF and WF to suppress inflammation using RAW264.7 cells. BF dose-dependently suppressed nitrous oxide (NO) production. Treatment of human hair follicle dermal papilla cells (HFDPC) with BF and WF promoted cell proliferation after 24, 48, and 72 hr of treatment when supplied at 10, 50, 100, and $200{\mu}g/ml$. Taken together, these results confirm the possibility of using BF and WF extracts from P. grandiflorum in formulating hair loss prevention products.

Anti-inflammatory Effect of the Extract of Gynostemma pentaphyllum cell from Ullengdo Island as Korean Endemic Plant (한국 고유 식물 울릉도 돌외 식물 세포 배양추출물의 항염증 효과)

  • The purpose of this study was to evaluate and verify the effectiveness of sustainable cosmetic raw materials developed from Gynostemma pentaphyllum, a plant native to Ulleungdo, in improving the skin barrier function and treating atopic dermatitis. Cells were derived from adult Gynostemma pentaphyllum plants, and suitable conditions for mass culture of the cells were established in a bioreactor. DNA components and amino acids extracted from this mass culture were identified from the HPLC fraction. In the in vitro efficacy evaluation results, changes in the expression levels of skin barrier-related proteins such as filaggrin (FLG) and Zonula occludens-1 (Zo-1) were insignificant. It was confirmed that the expression levels of the proteins thymic stromal lymphopoietin (TSLP) and interleukin-33 (IL-33) were significantly reduced. These results lead to the conclusion that Gynostemma pentaphyllum cell extracts have significant anti-inflammatory effects and that these extracts can be widely used as sustainable, nature-friendly active material in cosmetics with anti-inflammatory effects and targeted at improving atopic dermatitis.They may find use in anti-aging cosmetic products as well.

Glycosyltransformation of ginsenoside Rh2 into two novel ginsenosides using recombinant glycosyltransferase from Lactobacillus rhamnosus and its in vitro applications

  • Background: Ginsenoside Rh2 is well known for many pharmacological activities, such as anticancer, antidiabetes, antiinflammatory, and antiobesity properties. Glycosyltransferases (GTs) are ubiquitous enzymes present in nature and are widely used for the synthesis of oligosaccharides, polysaccharides, glycoconjugates, and novel derivatives. We aimed to synthesize new ginsenosides from Rh2 using the recombinant GT enzyme and investigate its cytotoxicity with diverse cell lines. Methods: We have used a GT gene with 1,224-bp gene sequence cloned from Lactobacillus rhamnosus (LRGT) and then expressed in Escherichia coli BL21 (DE3). The recombinant GT protein was purified and demonstrated to transform Rh2 into two novel ginsenosides, and they were characterized by nuclear magnetic resonance (NMR) techniques and evaluated by 3-(4, 5-dimethylthiazol-2-yl)-2-5-diphenyltetrazolium bromide assay. Results: Two novel ginsenosides with an additional glucopyranosyl (6→1) and two additional glucopyranosyl (6→1) linked with the C-3 position of the substrate Rh2 were synthesized, respectively. Cell viability assay in the lung cancer (A549) cell line showed that glucosyl ginsenoside Rh2 inhibited cell viability more potently than ginsenoside Rg3 and Rh2 at a concentration of 10 μM. Furthermore, glucosyl ginsenoside Rh2 did not exhibit any cytotoxic effect in murine macrophage cells (RAW264.7), mouse embryo fibroblasts cells (3T3-L1), and skin cells (B16BL6) at a concentration of 10 μM compared with ginsenoside Rh2 and Rg3. Conclusion: This is the first report on the synthesis of two novel ginsenosides, namely, glucosyl ginsenoside Rh2 and diglucosyl ginsenoside Rh2 from Rh2 by using recombinant GT isolated from L. rhamnosus. Moreover, diglucosyl ginsenoside Rh2 might be a new candidate for treatment of inflammation, obesity, and skin whiting, and especially for anticancer.

Protective Effects of Novel Tripeptide Against Particulate Matter-induced Damage in HaCaT Keratinocytes (미세먼지에 의해 유발되는 인간각질형성세포 손상에 대한 신규 트리펩타이드의 보호 효과)

  • In this study, we investigated inhibitory effect of Tripeptide against particulate matter (PM)-induced damage in human keratinocytes. PM-induced cell death was inhibited by Tripeptide and the activity of aryl hydrocarbon receptor (AhR) also inhibited by Tripeptide resulting in reduced expression of its downstream targets, cytochrome P450 family 1 subfamily A member 1 (CYP1A1) and cyclooxygenase-2 (COX-2), which are responsible for toxic metabolites production and inflammation. Furthermore, PM-induced expressions of pro-inflammatory cytokines, matrix metalloproteinase-1 (MMP-1) and apoptosis-related factors were decreased by anti-oxidant activity of Tripeptide. From these results, it has been shown that the Tripeptide has protective effect against PM-induced skin damage not only through the inhibiting of keratinocyte death but also through the inhibiting the secretion of several damage-inducing factors to adjacent skin tissue. And the results suggested that Tripeptide with anti-pollution effect could be applied as a new functional cosmetic material.

Comparison of Antioxidant and Anti-inflammatory Activity of Korean Houttuynia cordata Thunb. Extracts (국내산 어성초(Houttuynia cordata Thunb.)추출물의 항산화 및 항염증 활성 비교)

  • In this study, Korean-cultivated Houttuynia cordata Thunb. (HC) was extracted anti-inflammatory effects of extracts were compared in order to confirm the possibility of natural cosmetics as a raw material. HPLC pattern analysis, cell viability assay, total phenolic contents, ABTS/DPPH radical scavenging analysis, measurement of ROS production, griess reagent assay, and luminex technique. The content of quercetin was higher in fresh Houttuynia cordata (HC-F) than in dried Houttuynia cordata (HC-D). Also, content of HC extracts shows a difference in the content of the two representative compounds (polyphenol and flavonoid) according to the stored differently. ROS production showed higher inhibitory rates at HC-F. The anti-inflammatory activity of HC-D was shown higher than that of HC-F. But, in the measurement of the reduction in inflammatory cytokines, IL-1β showed that HC-D had a relatively high reduction effect, and IL-6 and TNF-α showed a high reduction effect. And these results will be a basis for the development of cosmetology in skin care and improvement of skin problem.

  • Objectives : The aim of this study is to explore the potential of oriental medicine in managing rosacea through the regulation of the autonomic nervous system. Methods : We reviewed studies on the pathophysiology and medical treatment (both western and oriental medicine) of rosacea, as well as the relationship between rosacea and the autonomic nervous system, using four databases: PubMed, OASIS, RISS, and NDSL. Results : Rosacea is a chronic recurrent inflammatory disease characterized by symptoms such as facial flushing, inflammatory papules, and pustules. In Western medicine, symptomatic treatments like vasoconstrictors, doxycycline, and anti-inflammatory drugs are primarily used. According to the pathophysiological mechanisms of rosacea, the autonomic nervous system is closely related, particularly with sympathetic overactivity causing vasodilation and local inflammation in rosacea patients. Additionally, recent studies report that rosacea patients frequently exhibit neuropsychiatric symptoms such as anxiety, depression, and insomnia, which are closely linked to autonomic dysfunction and contribute to the worsening of skin symptoms. However, current studies on the use of oriental medicine for rosacea focus mainly on anti-inflammatory effects at the local level, similar to conventional treatments. Conclusions : Based on the close involvement of the autonomic nervous system in the pathophysiological mechanisms of rosacea and numerous studies showing that oriental medicine can effectively regulate autonomic function, applying such treatments to rosacea patients may improve not only skin symptoms but also the frequently associated neuropsychiatric symptoms like anxiety, depression and insomnia.

Literature Study on the Efficacy of Fel Sus Scrofa (저담의 효능에 관한 문헌연구)

  • Bile juice prevents deposition of cholesterol in the blood vessel, digests fat, and absorbs fatty acid and vitamins, and it plays a great role on metabolism. Recently, emotional stimulus and mentally over-depression cause a person to come to illness, and westernization of way of life makes more patients with cholelithiasis, resulting into without bile secretion after cholelithotomy. Ageing, and gastrectomy and kidney transplantation are also the causes of more cholelithiasis occurrences. To solve these medical problems, we studied how Fel Sus Scrofa, which is not different from human bile juice, was used in the traditional Korean medicine. We I researched flavor, property, efficacy of Fel Sus Scrofa and how it was used by folk medicine, and we studied the usage examples of Fel Sus Scrofa in Sanghanlon and Dongeuibogam. The property of Sus Scrofa is bitter and cold. Its efficacy is to deposit glycogen. So it is known that it has been widely used, with many edible forms, without any humoral loss, for the inflammatory disease from various fever, problems of urine and feces, cutaneous disease, pulmonary disease, opthalmopathy, fever, thirst from diabetes, hepatocystic duct disorder. Fel Sus Scrofa can be used internally and externally to prevent humoral loss, and to control cutaneous disease among various pediatric disorder full of fever. And as we have in mind that it also can be used to treat patients with cholestasis after cholecystectomy, it is expected that post study of it must be done.

Comparison of Topical Agents for Bactericidal and Wound Healing Effect in Pseudomonas aeruginosa-infected Wound (Pseudomonas aeruginosa 감염창상에 사용되는 국소제제들의 항균효과 및 창상치유 효과의 비교)

  • Purpose: Pseudomonas aeruginosa is an etiologic agent in serious wound infection. Pseudomonas aeruginosa infection is problematic because this organism is resistant to many antimicrobial drugs. The purpose of this study was to compare the bactericidal effect of commonly used topical agents and their effect on wound healing. Methods: Pseudomonas aeruginosa-infected full-thickness skin defect was developed on the mouse to compare 3 commonly used topical agents-Betadine, 2% Gentamicin solution and 0.3% Acetic acid with the control group. Wound size change, bacterial colony counts and histologic findings of each groups were analyzed. Results: The wound size decreased in all treated groups as compared with the control group. However, there was no statistical difference. Gentamicin solution group was showed the lowest bacterial colony count and statistically significant difference compared with the control group(p=0.032). Other treated groups were also effective against Pseudomonas aeruginosa, but not different statistically. Histologic findings revealed that epithelialization, granulation tissue formation and microvessel proliferation were increased and necrosis and inflammation were decreased in all treated groups compared to the control group, but not different statistically. Betadine group significantly increased granulation tissue formation compared to the control group (p= 0.041). Conclusion: There is no universal topical agent that enhances most aspects of wound healing while simultaneously decreasing the bacterial concentration. However, Gentamicin solution may be an optimal topical agent for Pseudomonas aeruginosa infected wound. Further study should experiment on human with Gentamicin solution to confirm a effect on Pseudomonas aeruginosa infected wound for clinical applications.

In vitro Anti-allergic Effects of Bangpungtongseong-san in Human Keratinocytes and Primary Mouse Splenocytes (간 각질 세포 및 마우스 비장 세포를 이용한 방풍통성산의 항알러지 효능 연구)

  • Bangpungtongseong-san(BPTSS, Fangfengtongsheng-san in Chineses) is a traditional herbal formula comprising 18 medicinal herbs. In the present study, we performed the simultaneous analysis for four compounds of BPTSS and examined anti-allergic effects in human keratinocytes and mouse splenocytes. The column for separation of four compounds was used Luna C18 column and maintained at 40℃. The mobile phase for gradient elution consisted of two solvent systems. The analysis was carried out at a flow rate of 1.0 mL/min with PDA detection at 254 and 280 nm. To evaluate production and expression of Th2 chemokines, ELISA and RT-PCR were conducted in tumor necrosis factor (TNF)-α and interferon (IFN)-γ-stimulated HaCaT cells with or without BPTSS or silymarin, a positive control for skin inflammation. To measure Th2 cytokines, primary mouse splenocytes were treated with BPTSS and performed ELISA for interleukin (IL)-4, 5, 13. Calibration curves were acquired with r2>0.9999. The contents of geniposide, liquiritin, baicalin, and glycyrrhizin in BPTSS were 5.06 ㎎/g, 7.33 ㎎/g, 27.56 ㎎/g, and 7.81 ㎎/g, respectively. BPTSS reduced TARC and RANTES production and mRNA expression in TNF-α and IFN-γ-treated HaCaT cells. BPTSS inhibited IL-4, 5, and 13 production in mouse splenocytes. Our data will be a helpful information to upgrade quality control and anti-allergic effects of BPTSS.
