STUDIES ON THE CYSTINE COMPONENT IN THE SERICULTURAL PROTEINS OF BOMBYX MORI L. (가잠사단백질의 각과정에서의 Cystine 성분에 대한 연구)

  • The purpose of this treatise is to prove the presence of cystine in silk fiber through wide sampling throughout all the sericultural processes of Bombyx mori.; also to show that disulfide cross linkages exist in the silk fiber. The conclusions reached were as follows: 1. Crystalline cystine was obtained from silk fibroin using Folin's Method. 2. Analytical data showing the cystine content of silk fiber and its related materials were obtained using Sullvan's Method as follows: Material Percent Cystine A. Mulberry leaf protein 0.175 B. Silkworm egg 0.33 C. Silkworm Body, matured, fat extracted, without silk gland 0.41 D. Silk gland, matured 1.23 E. Silkworm feces none F. Silkworm pupa, fat extracted 0.30 G. Silkworm moth, fat extracted 0.60 H. Raw Silk 0.22 I. Fibroin 0.175 J. Sericin 0.30 3. The presence of cystine in the silkworm was substantiated the existence of 0.175 % methionine in mulberry leaves and 0.12% methionine in the silk gland. 4. Part of the sulfhydryl compounds in the silk gland is believed to transfer to serine and methionine, with the former being secreted into the liquid silk finally as silk fiber and the latter used for nutritive purposes in the growing of silk gland tissue. 5. The cystine content is variable by mulberry species, silkworm species, sex, breeding process, and other culturing environments. 6. Hybrid silkworms require more nutritive amino acids for effective growth than the original parents, and secrete less of them as silk fiber. 7. From such an observation, the amino acid composition of silk fiber is believed to be fairly flexible. Cystine if included in the amorphous part of the fiber, especially in sericin. 8. The result from enriching the silkworm diet with pure cystine or wool cystine did not result in any advantage, therefore it is believed that the natural cystine and methionine contents in the mulberry leafaregoodenoughforsilkwormnutrition. 9. The disulfide cross linkage in silk fiber was verified by using the Harris Method. Contraction took place following the treatment of the fiber with various salts and acids. Comparisons were made with wool fiber. 10. During these experiments, the fibrious structure of silk fiber and the net-globular liquid form were photographed microscopically. It is believed that the globules of liquid silk are net-formed by the inter attraction of the OH ion of the globular peptide and the H ion of water as shown by the hair cracking behavior of the film. The net-globular protein precipitation from the mulberry protein solution showed that mulberry is a proper diet for the formation of fibrous protein in the silk fiber. 11. The significance of the presence of cystine in silk fiber as emphasized in this paper should result in modification of the general conception that cystine is absent from this fiber. Influences of Several Chemicals on the Solubility of Cocoon Sericin in Water (제사용수(製絲用水)에 대(對)한 몇가지 화학약제(化學藥劑)의 처리(處理)가 견층(繭層) Sericin의 용해성(溶解性)에 미치는 영향(影響))

  • The purpose of this study is to find out influences of several chemicals on the solubility of cocoon sericin in water, and there are several results of use to control the solubility of cocoon sericin in water. The results obtained are summarized as follows : The chemical which shows the strongest accelerating power of the solubility of cocoon sericin in water is Sodium Hydroxide (NaOH), the second is Potassium Hydroxide (KOH), the third is Sodium Silicate ($Na_2SiO_3$), and the weakest is Sodium Bicarbonate ($NaHCO_3$) in order among noticed silk-reeling accelerators. The chemical which shows the strongest inhibiting power of the solubility of cocoon sericin in water is Form Aldehyde (HCHO), the second is Ammonium Alum ($Al_2(SO_4)_2{\cdot}(NH_4)_2SO_4{\cdot}24H_2O$), the third is Acetic Acid ($CH_3COOH$), the weakest is Hydrochloric Acid(HCl) in order among noticed silk-reeling inhibitors. Particulary Hydrochloric Acid (HCl), which is expected to show strong inhibiting power of the solubility of cocoon sericin in water, shows accelerating power of the solubility of cocoon sericin at high temperature over 80 degrees of Celsius thermometer in water.

Chromium(VI) Adsorption Behavior of Silk Sericin Beads

  • Silk sericin (SS) has been fabricated into beads using a 1 M LiCl/DMSO solvent and utilized as a heavy metal adsorbent. Among the various heavy metals, we targeted Cr(VI) for adsorption using SS beads and found that its adsorption depended on the coagulant used for the fabrication of the SS beads. When methanol was used as a coagulant, the beads had a better adsorption capacity than when ethanol was used except at pH 1. The adsorption behavior of Cr(VI) on the SS beads followed the BET isotherm. The maximum adsorption capacity was 33.76 mg/g at pH 2. The adsorption of Cr(VI) was confirmed by FT-IR and EDS analyses. Finally, the desorption was carried out using NaOH solution, and it was found that 73.19% of the adsorbed Cr(VI) could be detached.

  • Recently, B. mori proteins such as silk fibroin and silk sericin have been found to have a water-holding capacity, anti hydrogen peroxide toxicity, antioxidant activity and tyrosinase-inhibitory activity (Yeo 2006, Kurioka 1999 & 2004), implying its potential usefulness of the application for cosmetic and functional food(Yamazaki 1999 & Une 2000). We are tried to application for dietary resources of B. mori silk fibroin and sericin that were prepared to some of different molecular cutting these resources by preparative recycling HPLC system. In our studies with rats have demonstrated that consumption of these silk proteins are being prevents constipation effect and it is maybe enhances intestinal absorption of water and dietary effects. These some of useful results further suggest a usefulness of sericin as dietary resources for health.

  • Sericin 정착은 친수성 단백질인 세리신을 화학처리에 의하여 소수성으로 변화시켜 fibroin 에 밀착, 응고시켜 생사를 양모와 유사한 섬유로 변화시켜 제품의 다양화를 기할 목적으로 많이 행하여졌으며 이에 따른 증량효과도 크게 기대되어진다. 종래의 세리신 정착방법들은 생사의 제리신 정착외에 공업적으로 그 가치가 크게 인정되었던 견사, 생사 층 등에도 광범위하게 응용되어 많은 특허가 있는데 이들 정착방법을 대별하면 (1) Aldehyde 처리에 의한 세리신 정착 (2) Cr 감류처리에 의한 세리신 정착 (3) Epoxy 화합물의 처리에 의한 세리신 정착 (4) 합성수지 처리에 의한 세리신 정착등을 들 수 있겠으며 그 중에서도 특히 Aldehyde 에 의한 정착 (주로 formaldehyde 사용) 과 Cr 감류에 의한 정착이 가장 많이 행해진 경향을 보인다.   • Silk protein has been investigated by many researchers to apply to biomedical field. We reviewed biomedical applications of silk protein such as matrix of wound dressing and drug delivery system. Since silk fibroin/ poly (ethylene glycol) (PEG) semi-interpenetrating polymer networks showed good mechanical properties and wound healing phenomena, it can be used as wound dressing materials. Sericin nanoparticles pre- pared by conjugation with PEG and silk protein/ poloxamer mixture gel are expected to become a deliv- ery as matrix for hydrophobic drug.

  • Silk organza can make the silhouette of clothes bulky and rich, owing to its stiffness. And silk organza in the gum has better dyeability so can be dyed more bright and deeper color than scoured silk because silk organza has sericin gum that has better dyeability than fibroin. So, silk organza came into the most broad use as the material for Hanbok, especially in summer. But the silk organza has poor color fastness. Sericin is removed from organza when silk organza go through scouring. The purpose of this research is to find out the optimum conditions that the scoured silk fabrics can get stiffness by persimmon juice treatment. For experiments, non scoured, partially scoured and fully scoured silk fabrics were prepared by alkali treatments. Then 3 kinds of silk fabrics were treated with the concentration of 1, 5, 10, 50 and 100% of persimmon Juice. The change of fiber surface of the various silk fabrics were observed By SEM. The hand values of those fabric samples also were measured hand values by Kawabata Evaluation System and stiffness by Cantilaver method. By scouring, the silk organza got more flexibility and less cohesiveness between their fibers. And it can be a little more bulky by removing gum. Namely, silk organza lost its unique stiff and crisp handle. By the persimmon juice treatment, tannin component could be coated on the fiber surface. so that the stiffness of the fabrics were improved.

Activity and Stability of Immobilized Enzyme on Silk Sericin Bead

  • In present preliminary report, we showed the possibility of silk sericin (SS) in enzyme immobilization. SS beads were prepared and enzymes were immobilized on it. The specific activity of immobilized a-chymotrypsin retained more than 87% compared to the free enzyme. The immobilized a-chymotrypsin has better stability against ethanol especially those immobilized on SS beads coagulated in methanol. Immobilized trypsin and lipase had also comparable apparent activity compared to free enzyme. Our result indicates that SS could be a good candidate for enzyme immobilization support due to its hydrophilicity.

Improvements Caused by Silk Sericin Extract Derived from Silkworm in Blood Glucose and Lipid Concentration in Diabetic Rats (누에 유래 실크 세리신이 당뇨성 흰쥐의 혈당 및 지질 농도 개선에 미치는 영향)

  • The objective of this study was to assess improvements caused by silk sericin extract in blood glucose and lipid concentrations in the sera of streptozotocin(STZ, 55 mg/kg B.W., I.P. injection)-induced diabetic rats fed on experimental diets for 5 weeks. Serum concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, triglyceride(TG) and phospholipids(PL) were remarkably higher in the diabetic group(group BSW) and STZ(I.P.)-plus-silk sericin extract group(group ESS) than in the control group(group CG, basal diet-plus-water). However, the serum concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, TG and PL were lower in the ESS group than in the BSW group, whereas the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration in the ESS group were higher than in the BSW group. The activities of alkaline phosphatase(ALP) and aminotransferase(AST, ALT) in serum were lower in the ESS group than in the diabetic BSW group. The results shown above suggested that silk sericin extract effectively improves blood glucose and lipid concentrations in the sera of STZ-induced diabetic rats.

Dietary Effect of Silk Protein on Colon Cancer of Animal (실크 단백질 식이공급이 동물대장암에 미치는 영향)

  • This study was done for the dietary effect of silk protein, molecular controlled silk sericin and silk fibroin, on colon cancer of animal. The ICR mice were given weekly injections of 1,2-dimethylhydrazine (DMZ) for the initial 10 mg/kg with four weeks, and fed a diets supplemented with 3% (v/v) molecular controlled sericin and fibroin for 6 weeks, respectively. Then, the body weight, efficiency of dietary, typical histologic appearance of animal colon tissue and COX-2 effect were studied, too. Molecular controlled sericin protein are shown more higher dietary effect on animal colon cancer than fibroin supplemental case. The results suggest a potential usefulness of sericin as a chemopreventive for colon carcinogenesis and the development functional food.
