Modulation of Human Cardiac Progenitors via Hypoxia-ERK Circuit Improves their Functional Bioactivities

  • Recent accumulating studies have reported that hypoxic preconditioning during ex vivo expansion enhanced the self-renewal or differentiation of various stem cells and provide an important strategy for the adequate modulation of oxygen in culture conditions, which might increase the functional bioactivity of these cells for cardiac regeneration. In this study, we proposed a novel priming protocol to increase the functional bioactivity of cardiac progenitor cells (CPCs) for the treatment of cardiac regeneration. Firstly, patient-derived c-$kit^+$ CPCs isolated from the atrium of human hearts by enzymatic digestion and secondly, pivotal target molecules identified their differentiation into specific cell lineages. We observed that hCPCs, in response to hypoxia, strongly activated ERK phosphorylation in ex vivo culture conditioning. Interestingly, pre-treatment with an ERK inhibitor, U0126, significantly enhanced cellular proliferation and tubular formation capacities of CPCs. Furthermore, we observed that hCPCs efficiently maintained the expression of the c-kit, a typical stem cell marker of CPCs, under both hypoxic conditioning and ERK inhibition. We also show that hCPCs, after preconditioning of both hypoxic and ERK inhibition, are capable of differentiating into smooth muscle cells (SMCs) and cardiomyocytes (CMs), but not endothelial cells (ECs), as demonstrated by the strong expression of ${\alpha}$-SMA, Nkx2.5, and cTnT, respectively. From our results, we conclude that the functional bioactivity of patient-derived hCPCs and their ability to differentiate into SMCs and CMs can be efficiently increased under specifically defined culture conditions such as short-term hypoxic preconditioning and ERK inhibition.

The contamination prevention of diamond conditioner by anti-contamination film coating (오염방지막 코팅을 통한 Diamond Conditioner의 표면오염 방지)

  • 반도체 device의 성능을 향상시키기 위하여 패턴은 더욱 더 고 집적화 되고 배선 또한 다층배선 구조를 가지게 되었으며 요구되는 선폭 또한 더욱 미세화 되어 CMP 공정이 도입되게 되었다. 이러한 CMP 공정에 사용되는 소모품으로는 크게 세 가지의 중요한 부분으로 나눌 수 있다. 그것은 slurry와 pad, conditioner이다. 그중에 pad conditioning 공정은 CMP 공정시 pad의 마모에 따라 감소하는 removal rate(RR)값을 회복시키기 위한 공정으로 마모된 pad의 표면을 활성화 시켜주는 중요한 공정이다. 하지만 pad conditioning 공정을 장시간 진행하게 되면 conditioner 표면에 오염물이 발생하게 되며, 오염물로 인하여 wafer표면에 scratch 및 defect을 발생시키는 원인이 될 수 있다. 이러한 문제점을 보완하기 위하여 conditioner의 표면을 변화시켜 공정중의 오염이 발생하지 않도록 하는 것이 중요하다. 본 논문에서는 oxide CMP 실험을 통하여 conditioner표면에 오염물이 발생함을 확인하였으며 energy dispersive spectroscopy(EDS) 분석을 통하여 주오염물의 성분이 oxide slurry중 silica임을 확인하였다. Conditioner의 표면을 소수성으로 만들기 위하여 self assembled monolayer(SAM) 방법을 이용하여 표면에 코팅을 하였으며, 소수성 박막이 코팅된 conditioner와 코팅되지 않은 conditioner의 비교 실험을 통하여 오염 정도를 비교하였다.

The Relationship of Participant Motivation, Physical Self-concept, University Life Satisfaction of the Leisure Sport on the University Students (대학생 여가스포츠의 참여동기, 신체적자기개념, 대학생활만족과의 관계)

  • The purpose of this study is to analyze the relationship between students' motivation for participating in leisure sports, students' physical self-concept and the life satisfaction of university students. A total of 419 students were finally analyzed using the convenience sampling method of non-random sampling method after selecting those university students participating in leisure sports at a university located in J district. The results of analysis using SPSS version 21.0 program are as follows. First, there is a positive (+) correlation between the motives of participation of university students and their physical self-concept and satisfaction with university life. Second, the influence of the motive to participate in sports has a positive (+) effect on sports competence and this is affected by health fitness and social interaction, physical activity is affected by pleasure, health fitness, and conditioning, self-esteem, muscular strength is affected by pleasure, health fitness, condition, and society, flexibility is affected by pleasure, health fitness, and society. Third, the influence of participation motive had a positive (+) effect on school satisfaction is affected by pleasure, health fitness, physical condition, external display, socialization, and fellow satisfaction is effected by pleasure and condition.

Factors Effecting to the Stage of Change for Exercise on the Workers (일부 근로자의 운동행위 변화단계에 영향을 미치는 요인)

  • Objective: This study was performed to identify the related factors of change affecting the stage of change, on the assumption that there are various stages of change in the exercise behavior of workers. Method: The subjects of the study consisted of 138 workers of 10 workplaces located in Daejeon City, and data collected by using questionnaires were analyzed with SPSS/WIN 11.5 program. Results: The subjects were distributed in the stage of exercise behavior change as follows: precontemplation stage(10.9%), contemplation stage(22.5%), preparation stage(41.3%), action stage(8.7%), and maintenance stage(16.7%). Of the process of change depending on the stage of exercise behavior change, the consciousness raising(F=11.50, p=.00), dramatic relief(F=2.82, p=.02) showed significant difference in the cognitive process, and the counter conditioning(F=4.56, p=.00), reinforcement management(F=4.64, p=.00), self-liberation(F=9.46, p=.00), and stimulus control(F=13.28, p=.00) showed significant difference in the behavioral process. For decision-making depending on the stage of exercise behavior change, both the pros of decision-making(F=9.23, p=.00) and the cons of decision-making(F=2.45, p=.04) showed significant difference. Self-efficacy depending on the stage of exercise behavior change showed significant difference as F=11.50, p=.00. The related factors of change affecting the stage of exercise behavior change were the pros of decision-making, the cons of decision-making, and self-efficacy including 34.1% R-square. Conclusion: In order to change the exercise behavior of workers positively, they need to be stimulated to use the cognitive and behavioral process in the process of change properly, and to be induced to make a decision positively. And also exercise programs suitable to various characteristics of each worker as well as those of workplaces to enhance self-efficacy need to be applied after being devised. Through the further longitudinal research, it is necessary to analyze the various aspects, such as groups to select, to maintain, to stop or give up the exercise, and to avoid the change, and to study how the related factors of change affect diversity like the above.

A Study on Factors Influencing The State of Adaptation of The Hemiplegic Patients (편마비 환자의 퇴원후 적응상태와 관련요인에 대한 분석적 연구)

  • The purposes of this study are to delineate a profile of the state of a stroke patient's adaptation at 3 months after hospitalization and to explore the relationship between the level of adaptation and the variables which influence the adaptation of hemiplegic patients. To these ends, theoretical framework was derived basically from the stress adaptation model. The basic assumption underlying the level of adaptation is influenced by the presenting focal, contextual and residual stimuli. This group of stimuli is further operationalized and represented by a perception of stress. which is the perceived effect of the disability and by the mediating variables such as sociodemographic factors as an external conditioning variables and perceived social support and hardiness personality characteristics as an internal intervening variables. The dependent varibales in this study is the level of physical, psychological and social adaptation and is hypothesized to be a function of the interaction between 3 sets of variables namely, the perceived disability effect, external conditioning variables and internal intevening varibles. A total of fourty three subjects from 3 general hospitals in Seoul were observed and interviewed with the aid of 7 structured instruments. The data were collected twice on each subject : first at the pre-discharge period arid at 3 months post-discharge from hospital for the second time. The study was carried out for the period from February to August, 1988. The instruments used for the study include 4 existing scales and 3 scales developed by the researcher for this study. They are : 1) The ADL dependency scale and the scale of the clinical physical functions for the assessment of physical adaptation. 2) the SDS(self report of depression) to measure the level of psychological adaptation. 3) The scale for the amount of social activities for the measurement of the level of social adaptation. 4) The scale for the perceived effect of disability for the measurement of the focal stimuli. 5) The health related hardiness scale and the perceived interpersonal support self evaluation list(ISEL) for the measurement of the hardiness personality character and the perceived social support. The data obtained were analyzed using percentage, oneway ANOVA, Pearson coefficients correlation and stepwise multiple regression. The findings provide valuable information about the present level of physical adaptation at 3 months after discharge. The patient revealed a decreased ADL dependency and lowered limitation of physical function as compared with pre - discharge state. Psycholcgically, the average degree of depression at follow up was within normal range of depression. Socially, the amount of social activities was very low. The one way ANOVA and the correlational analysis revealed the relationship between the 3 sets of variables and the adaptation level as follows : 1) The perceived disability effect was related to the degree of the depression and the amount of social activities but was not related to the physical adaptation. 2) Among the sociodemographic variables, sex and education were related to the difference of ADL dependency and the change of physical function. These factors indicate that women more than men and educated more than the less educated were found more independent. The education was also related to the degree of depression suggesting that the higher the educational level, the more well adapted the patients were both physically and psychologically. Age, marital status and job state were not found to be related to the patient's adaptation level. 3) Among the internal intervening variables, the health related hardiness characteristic was related to the differences of ADL dependency, physical functions and the social activities, indicating that the higher the hardiness character the higher the level of physical and social adaptation. 4) The perceived social support, another internal intervening variable, was related to the degree of depression and the social activities. This data suggest that the higher the perception of social support, the better adapted the patients were psychogically and socially. In summarizing the results of the correlational analysis, the level of physical adaptation was influenced by sex, the years of education and the hardiness character. The level of psychological adaptation was influenced by the years of education, the perceived disability effect and the perceived social support. And the level of social adaptation was influenced by the perceived disability effect, the hardiness character and the perceived social support. The stepwise multiple regression analysis shows findings as follows : 1) The most important factor to explain the difference of ADL dependency was sex, indicating females were more independent than males. 2) The most important factor to explain the difference of physical function and the degree of depression was the patient's education level. 3) The strongest explaining factor for the amount of social activities was perceived self esteem(one of the subconcepts of perceived social support). Thus the most important factors influencing the level of adaptation were found to be sex, education, the hardiness character and self esteem. From the above findings, the significance of this study can be delineated as follows : 1) Corroboration of the assumed relationship between the various variables and the adaptation level as suggested in the conceptual model. 2) Support for the feasibility of the cognitive approach for nursing intervention such as hardness character training, counselling and teaching for self-care in the chronic patients.

Effect of Individualized Exercise Program for Preventing Metabolic Syndrome among IT Company Office Workers (IT 기업 사무직 근로자의 대사증후군 예방을 위한 맞춤형 운동프로그램의 효과)

  • Objectives: Interventions promoting physical exercise and healthy habits in workplaces have been shown to be effective in reducing risk factors for metabolic syndrome. This study was conducted to examine the effects of an individualized conditioning exercise program of IT company office workers with or at higher risk of metabolic syndrome. Methods: A total of 444 IT company office workers with or at higher risk of metabolic syndrome participated in a 3-month conditioning exercise program. Body composition data using bioelectrical impedance analysis and cardiopulmonary data using cardiopulmonary exercise testing from 53 individuals (mean age: 34.8 ± 7.1 years, sex : 21% female, height : 170.4 ± 6.8 cm, weight : 75.2±12.2 kg, body mass index : 25.8±3.3 kg/m2) who have successfully completed pre-test, intervention, and post-test were analyzed. The 12 weeks intervention encompassed: (1) health counseling (2) supervised exercise(endurance-based, aerobic exercise, or circuit training once a week for 50 minutes at heart rate reserve(HRR) of 77-95%) (3) self-directed exercise and biweekly health screening checks. Results: The results indicated a significant decrease in body weight, body fat mass and body mass index, respectively. Moreover, VO2peak, AT VO2 and AT Time significantly improved, respectively. Resting blood pressure(SBP/DBP) showed positive changes but were not statistically significant. We observed the correlation between characteristics of participants and rate of changes in cardiopulmonary outcomes of participants, there are no significant correlation. These results indicate positive changes in body composition and cardiorespiratory fitness parameters following individualized conditioning exercise program. Conclusions: Individualized workplace exercise program for preventing metabolic syndrome can lead to improvements in body composition and cardiorespiratory fitness.

Development of Protocol for Standardized Emotion Induction in Children (아동 정서 유발 프로토콜 개발)

  • The aim of this study was to develop a research protocol that provides standardized conditioning stimuli in effectively evoking emotion-specific responses in children. The protocol targets five specific emotions(i.e., happiness, sadness, anger, distress, and boredom) by introducing study participants to a combination of music, color, stories, and dolls. This research protocol also includes a self-report emotion assessment scale specially developed for this study to verify the types of emotion induced. Evaluation of the method entailed triangulation of the results from the subjects' self-reported emotional state corresponding to each conditioned stimulus as well as behavioral observations conducted by researchers. Findings suggest that this new protocol effectively evokes five emotions in correspondence to the way it was intended for appropriate emotion induction. Additionally, results showed that female children than male children are more likely to feel boredom; and school-aged children were more likely to react to sadness than pre-school children.

The Level of Body Perception, Obesity, and Factors associated with Stages of Exercise and Dietary Habits Change in University Students: Application of the Transtheoretical Model (대학생의 체형 인식과 비만 정도 및 운동과 식습관 변화단계 영향요인: 범이론적 모형 적용)

  • Purpose: The purpose of this study was to identify the level of body perception, obesity, and factors associated with stages of exercise and dietary habits change in university students. Methods: A descriptive design was used. Data were collected from 279 university students using structured questionnaires. Results: The body perception was significantly associated with self rated health (F=27.89. p<.001), weight satisfaction (F=137.74. p<.001) and weight control interest (F=39.19. p<.001). The BMI was significantly associated with weight satisfaction (F=34.08. p<.001) and weight control interest (F=19.59. p=.003). But the body perception and BMI were not significantly associated with depression (F=2.17, p=.073, F=0.86. p=.462), the change of exercise (F=25.91. p=.055, F=20.34. p=.061), and dietary habits (F=12.96. p=.677, F=11.29. p=.505). The related factors affecting the change of exercise behavior were counter conditioning, self efficacy, eating behavior, and helping relationship, including 30.4% R-square. The related factor affecting the change of dietary habit was only eating behavior, including 20.0% R-square. Conclusion: This study can provide useful information to develop effective programs for the change of exercise and dietary habits considering university students' stages of change according to the transtheoretical model.

Analysis of The Stages of Change in Exercise Behavior of Women in Nursing College based on Transtheoretical Model (범이론적 모형을 적용한 간호여대생의 운동행동 변화단계 분석)

  • The study was performed to identify the process of change, decisional balance, self-efficacy and social support corresponding to the stage of exercise behavior change and the applicability of social support of women in nursing college based on a Transtheoretical Model. The subjects consisted of 223 women in nursing college by convenience sampling and the data were analyzed by IBM SPSS 21.0 program. The subjects were distributed in each stage of change of behaviors: 17.9% in the precontemplation stage, 54.3% in the contemplation stage, 16.6% in the preparation stage, 4.5% in the action stage and 6.7% in the maintenance stage. Analysis of variance showed that consciousness raising, dramatic relief, self reevaluation, social liberation, counter conditioning, helping relationship, reinforcement management, self liberation, stimulus control, pros, cons and self-efficacy were significantly associated with the stages of exercise behaviors. But there was no significant difference in social support. The transtheoretical model would be applicable to explain the exercise behavior of women in nursing college. This study will be useful information for developing effective exercise programs considering nursing female students' stages of change in exercise behavior.

Self-Cleaning of Mortar Mixed with Photocatalyst by Using Methylene Blue Solution (메틸렌블루 용액을 이용한 광촉매 혼입 모르타르의 방오성능 평가)

  • An experimental study to investigate the effect of self-cleaning of mortar mixed with photocatalyst was performed out in this study. Test parameters included the photocatalyst content and surface roughness of the specimens. The experimental mortar specimens were manufactured by mixing a photocatalyst by cement weight of 2.5%, 5.0%, 7.5% and 10.0%. In addition, the surface roughness was categorized into three cases. They included flat surface condition, little surface roughness(medium roughness), and high surface roughness. After mortar specimens were cured for 28 days, they were illuminated by an ultraviolet lamp for 24 hours and immersed in a methylene blue conditioning solution for 12 hours. Thereafter, an ultraviolet(UV) lamp was illuminated on the specimens for 48 hours in an experimental chamber and then the color change of methylene blue solution was measured by using a spectrophotometer over illuminating time of UV lamp. The color change of methylene blue tended to increase as photocatalyst contents increased. Test results meant that photocatalyst was effective for self-cleaning in mortar. However, the color change of the methylene blue solution did not show a noticeable tendency at different surface roughness conditions. It might be due to the uneven photocatalyst distribution on the surface of mortar specimens.
