오미자(Schizandra chinensis $B_{AILI}$.) 열매로부터 항경련 활성물질의 분리 (Isolation of Anticonvulsant Compounds from the Fruits of Schizandra chinensis$B_{AILI}$)

  • 항경련 활성을 보인 오미자의 열매로부터 활성물질을 분리하기 위하여 MeOH로 추출하고, 추출물을 EtOAc, n-BuOH 및 물로 분배, 추출하였다. 얻어진 각 분획에 대하여 중추 신경계에서 억제성 신경전달물질로 알려진 ${\gamma}-aminobutyric$ acid(GABA)의 대사조절효소들인 succinic semialdehyde reductase (SSAR) 및 succinic semi-aldehyde dehydrogenase(SSADH)의 활성을 측정하였다. 활성 억제효과를 나타낸 EtOAc 분획으로부터 silica gel column comatography를 반복하여 sesquiterpene 및 sterol 배당체 각 1종, lignan화합물 4종을 분리, 정제하였다. 각각의 화학구조는 NMR, MS 등의 스펙트럼 데이터를 해석하여, chamigrenal, daucosterol, gomisin A, gomisin H, gomisin N 및 schizandrin으로 동정하였다. 각 화합물에 대하여 활성을 측정한 결과, schizandrin을 SSADH와 1시간 전처리하였을 때 효소의 활성은 65% 억제되었으며, daucosterol의 경우에는 같은 조건에서 SSAR의 활성을 80% 억제하는 것을 관찰할 수 있었다.

가공방법을 달리한 오미자 분말차의 품질 특성 분석 (Evaluation of the Quality Characteristics in Omija (Schizandra chinensis Baillon) Powder Tea)

  • 오미자의 이용성 확대를 위한 건조 가공 제품의 가능성을 확인해보고자 생오미자 및 건조오미자를 이용하여 착즙, 추출, 동결건조, 과립성형 및 분무건조 등의 방법을 이용하여 다양한 오미자분말차를 제조하여 그 품질 특성을 비교 분석하였다. PG, EG 및 ES의 총가용성고형분 함량 및 탁도의 경우 다른 구에 비해 유의적으로 높았으며, pH 측정 결과 PG 및 EG가 다른 구보다 유의적으로 높았다. PG 및 EG의 색도 측정 결과 다른 구에 비해 높은 $L^*$값, 낮은 $a^*$값 및 $b^*$값으로 나타나 오미자의 가공 형태에 따른 유의적 차이가 뚜렷함을 알 수 있었다. PG 및 EG의 경우 다른 구에 비해 유의적으로 높은 환원당 함량, 낮은 총 페놀성 화합물의 함량, 낮은 항산화활성, 낮은 총 안토시아닌 함량 및 낮은 schizandrin 함량을 보여 오미자의 가공 형태에 따른 유의적 차이를 확인할 수 있었다. 또한 관능검사 결과 PG 및 EG는 낮은 붉은색 정도, 낮은 탁도 정도, 낮은 신맛 정도, 높은 쓴맛 정도 및 높은 단맛 정도를 보였으며, 종합적 기호도에서 PG 및 ES의 경우 가장 높은 점수를 받았다. 이와 같은 결과에서 붉은색과 항산화 활성, 안토시아닌 및 schizandrin 함량이 우수한 오미자 분말 제품을 위해 과립차보다 동결건조 및 분무건조분말차가 바람직하다고 판단된다.

HPLC에 의한 오미자 리그난 성분의 정량분석 (Quantitative Determination of Lignans from Schizandra chinensis by HPLC)

  • A reversed phase HPLC method was established for quantitative determination of lignans including schizandrin (1), gomisin A (2), deoxyschizandrin (3), gomisin N (4), and wuweizisu C (5) from Schizandra chinensis using a binary gradient of acetonitrile and water as a mobile phase with UV detection at 254nm. In result, contents of schizandrin (1), gomisin A (2), deoxyschizandrin (3), gomisin N (4), and wuweizisu C (5) in Schizandra chinensis from Moonkyung were 8.41${\pm}$0.30, 3.01${\pm}$0.13, 1.06${\pm}$0.05, 7.69${\pm}$0.30, and 1.68${\pm}$0.06 mg/g, respectively.

수확시기에 따른 오미자의 항산화 활성과 리그난 성분 함량 변화 (Changes of Antioxidant Activity and Lignan Contents in Schisandra chinensis by Harvesting Times)

  • We investigated antioxidant activity and lignan contents by harvesting times to expand use of Schisandra chinensis (Turcz.) Baillon. Total polyphenol and flavonoid contents of seed was higher than those of flesh but there is not much difference in harvesting times. As $RC_{50}$ value, that was, the concentration of sample required for 50% reduction of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical, was 13.7~24.2 ${\mu}g/m{\ell}$ in seed thus it showed a high antioxidant activity. Among lignan components, schizandrin content was the highest and followed by gomisin N and gomisin A in all of flesh and seed. Also these components in seed were 4~9 times more contained than those of flesh. All of them were decreased by harvesting times in flesh. But the contents of schizandrin and gomisin N were high in August 3rd and September 15th in seed, respectively. As the results, the seed of S. chinensis had high antioxidant activity and lignan contents so it could be potentially developed as a resource.

오미자 수집종의 생육특성 및 생리활성 분석을 통한 우수자원 선발 (Selection of Superior Resources through Analysis of Growth Characteristics and Physiological Activity of Schisandra chinensis Collection)

  • Background: Various Schisandra chinensis (SC) varieties grow in diverse regions in Korea. However, there is no valid scientific evidence of these varieties. This study aimed to select the excellent resources in terms of the growth characteristics, antioxidant activities, and analysis of the active compounds of the SC collection. Method and Results: In total, 154 resources of SC were collected from various regions of Korea. The growth characteristics were measured by the number of fruit bunches, fruit number, and weight of 100 fruits. The antioxidant activities were investigated by analyzing the total flavonoid and total polyphenol contents and the radical scavenging activity of DPPH and ABTS. Schizandrin A, schizandrin c, gomisin a, and gomisin N were analyzed by HPLC. Each resource showed different growth characteristics. Among the antioxidative effects, the highest 20 resources showed high antioxidant activities in selected 29 resources. Analysis of the SC lignan index showed that all resources contained more than 1.16% of active compounds. Conclusions: All of the selected 29 SC resources were shown to have excellent growth characteristics, antioxidant activities, and bioactive compound richness. Especially, SC-004, SC-007, and SC-154 showed the best growth characteristics, and SC-22, SC-40, and SC-45 showed the best antioxidant activities and bioactive compound richness.

오미자 열매 부위별 이화학적 특성 (Chemical Components Composition on Different Parts of Fruit in Schisandra chinensis Baillon)

  • 오미자 열매를 과피, 과육 및 씨 등을 부위별로 분리한 후 일반성분을 분석한 결과 모든 부분에서 가용성 무질소물의 함량이 각각 20.72, 6.88 및 24.42%로 가장 많이 함유되어 있으며 열매에서 씨 부분이 차지하는 비율은 40% 이상이었다. 또한, 오미자의 유효성분인 schizandrin을 포함한 리그 난 화합물의 총량이 씨 부분에서 15.55 mg/g으로 가장 많이 함유되어 있고 schizandrin이 5.68 mg/g, gomisin N이 2.90 mg/g, gomisin A 1.55 mg/g, schizandrin C가 1.53 mg/g 함유되어 있었으며 열풍 건조한 오미자 열매 전과와 거의 비슷한 함량을 함유하고 있었다. 오미자의 유기산은 주로 과피 쪽에 있었으며 총유기산 함량은 12.99%를 함유하고 있었고 주된 유기산은 citric acid로 4.84%를 함유하고 있었다. 오미자의 지방 함량은 씨 부위 및 열풍 건조한 오미자 열매에서 각각 19.04%와 15.11%를 함유하고 있었고 지방을 구성하고 있는 주된 지방산은 linoleic acid로 각각 72.66%와 74.10%를 차지하고 있었으며, 그다음은 oleic acid로 각각 14.78%와 17.46%를 차지하고 있었다. 오미자의 주된 무기이온은 씨에서는 P로 725.35 mg/100 g을, 열풍 건조한 열매 전과에서는 K로 2,024.10 mg/100 g을 차지하고 있었다. 결론적으로 오미자를 활용하기 위해서는 리그 난 화합물이 많이 함유되어 있는 씨 부분을 충분히 활용할 수 있는 방법을 고려하여야 할 것으로 생각된다.

증숙 과정을 통한 소청룡탕의 뇌신경세포 보호 활성 변화에 관한 연구 (Neuroprotective Activity of So Cheong Ryong Tang with Steaming Process)

  • So Cheong Ryong Tang (SCRT) has been used as a traditional herbal medicine for the treatment of the bronchial asthma. In this study, the effect of steaming process on changes in 11 major compounds (homogentisic acid, ephedrine HCl, paeoniflorin, cinnamic acid, cinnam aldehyde, glycyrrhizin, 6-gingerol, schizandrin, methyleugenol, gomisin A and gomisin N) content and neuroprotective activity of SCRT were evaluated. Major compound content was slightly different with steaming produce. The contents of paeoniflorin, cinnamic acid, cinnam aldehyde, 6-gingerol and methyleugenol were decreased and homogentisic acid, ephedrine HCl, glycyrrhizin, schizandrin and gomisin A were increased by steaming process. The neuroprotective activity of steaming SCRT was determined in mouse hippocampal HT22 cells by MTT assay. As a result, neuroprotective activity of a steaming SCRT was higher than that of a non steaming SCRT. This study demonstrated that neuroprotective activity of SCRT can be improved through steaming process.

HME (Hot-Melt Extrusion)를 이용한 오미자 씨 및 박의 수용성 및 항산화 효과 향상 (An Enhanced Water Solubility and Antioxidant Effects of Seed and Pamace of Schisandra chinensis (Turcz.) Baill Formulation by HME (Hot-Melt Extrusion))

  • Objectives : Schisandra chinensis (Turcz.) Baill contains many nutrients and exhibits high physiological functions. It has been shown that Schisandra seed and pamace contains more nutrients than fruits and thus have higher antioxidant efficacy. In this study, seed and pamace of Schisandra chinensis (Turcz.) Baill (SPSC) were treated with hot-melt extrudate (HME) extrusion to produce water-soluble nanoparticles. Methods : SPSC was treated with HME to prepare nanoparticles. In this process, excipients (hydroxypropyl methylcellulose, pullulan, 2-hydroxylpropyl-beta-cyclodextrin, lecithin) were added to prepare a hydrophilic polymer matrix. To compare and analyze the antioxidant effect and schizandrin content, total flavonoid content, total phenol content and ABTS assay were measured. To confirm the effect of increasing the water solubility of the particles, particle size and water solubility index measurements were performed. The molecular of the material was analyzed using Fourier transform infrared spectroscopy (FT-IR). Results : The particle size of HME extrudates decreased, while total phenols, flavonoids, schizandrin, antioxidant effect, and solubility increased. Through FT-IR, it was confirmed that the SPSC and the extrudate exhibit the same chemical properties. In addition, it was confirmed that when extracted with water, it exhibited a higher antioxidant effect than the ethanol extract. Conclusions : HME technology increased the solubility of SPSC, which are processing by-products, and improved their antioxidant effect to a higher degree. It was confirmed that SPSC could be used as an eco-friendly, high value-added material.

사염화탄소로 유발된 간독성에 대한 오미자 Schizandrin C 유도체 DDB 복합물 DWP-04의 예방효과 (Preventive effect of a Schizandrin C derivative DDB-mixed preparation (DWP-04) against hepatotoxicity induced by Carbon Tetrachloride)

  • The protective effects of the DWP-04 [DDB : selenium yeast: glutathione {31.1 : 6.8 : 62.1 (%, w/w)} against hepatotoxicity by carbon tetrachloride $(CCl_4)$ were studied in rats. The rats were intraperitoneally injected with $CCl_4$ (50% in com oil) at initial dose of 1 ml/kg followed by 0.5 ml/kg 3 times during 1 week. The DWP-04 (50, 100 or 200 mg/kg) or its vehicle was administered everyday before the start of $CCl_4$ injection for two weeks. $CCl_4$ induced hepatocelluar degeneration and necrosis, which led to a great increase in serum aminotransferase, alkaline phosphatase activity and serum lipid levels. It was found by biochemical analysis that $CCl_4$ treatment remarkably increased thiobarbituric acid reactive substances and physphatidylcholine hydroperoxide in hepatic tissues and induced antioxidant enzymes such as catalase and superoxide dismutase (SOD). Liver and serum lipids were significantly lower in rats fed on DWP-04 than in rats induced by $CCl_4$ only-treatment. These results suggested that the DWP-04 could be a promising candidate for the protection of liver injury based on the preventive effects against lipid peroxidation and serum biochemical parameters.

GC/MS에 의한 오미자 Lignan성분의 동정 (Identification of Lignan Compounds in Fruits of Schisandra chinensis BAILLON by Gas Chromatography/Mass Spectrometry)

  • GC/MS를 이용하여 오미자의 chloroform-methanol 추출물로부터 11종의 lignan 성분을 동정하였다. GC column은 SPB-1 fused silica capillary $(0.25mm\;ID{\times}30m,\;Supelco)$를 사용하였고 column oven의 온도는 $200^{\circ}C$부터 $300^{\circ}C$까지 분당 $4^{\circ}C$씩 승온하였으며 MS의 ionization voltage는 70eV (El mode)이었다. 동정된 lignan 성분은 gomisin J, deoxyschizandrin, gomisin N, schizandrin, wuweizisu C, gomisin A, angeloylgomisin H, tigloylgomisin H, angeloylgomisin Q, gomisin B 및 benzoylgomisin H이었으며, GC chromatogram 상에서 이들 화합물의 분리상태는 양호하였다.
