Study on the Antioxidant and Anticancer Effects of Extract of Stamens of Nelumbo nucifera and Kaempferol (연수 추출물과 Kaempferol의 항산화 및 항암작용에 관한 연구)

  • Objectives : The present study was performed to clarify the antioxidant and anticancer effects of extract of stamens of Nelumbo nucifera and Kaempferol. Methods : Antioxidant effect was measured by DPPH-radical scavenging activity for electron donating ability (EDA), superoxide dismutase (SOD)-like activity for SOD and lipid peroxidation. Anticancer effect was assessed by MTT absorbance for cytotoxicity. Results : Vitamin E, Kaempferol and ethyl acetate extract of stamens of Nelumbo nucifera increased SOD-like activity and DPPH-radical scavenging activity dose-dependently. On the contrary, lipid peroxidation was time-dependently decreased. Furthermore, Kaempferol and ethyl acetate extract of stamens of Nelumbo nucifera significantly decreased the growth rate of C6 glioma cells. Conclusions : These results suggest that ethyl acetate extract of stamens of Nelumbo nucifera may be a putative antioxidant or anticancer substance.

DNA Cleavage Induced by the Reaction of Salsolinol with Cu,Zn-Superoxide Dismutase

  • Salsolinol, endogenous neurotoxin, is known to be involved in the pathogenesis of Parkinson's disease (PD). In the present study, we have investigated the oxidative damage of DNA induced by the reaction of salsolinol with Cu,Zn-SOD. When plasmid DNA incubated with salsolinol and Cu,Zn-SOD, DNA cleavage was proportional to the concentrations of salsolinol and Cu,Zn-SOD. The salsolinol/Cu,Zn-SOD system-mediated DNA cleavage was significantly inhibited by radical scavengers such as mannitol, ethanol and thiourea. These results indicated that free radicals might participate in DNA cleavage by the salsolinol/Cu,Zn-SOD system. Spectrophotometric study using a thiobarbituric acid showed that hydroxyl radical formation was proportional to the concentration of salsolinol and was inhibited by radical scavengers. These results indicated that hydroxyl radical generated in the reaction of salsolinol with Cu,Zn-SOD was implicated in the DNA cleavage. Catalase and copper chelators inhibited DNA cleavage and the production of hydroxyl radicals. These results suggest that DNA cleavage is mediated in the reaction of salsolinol with Cu,Zn-SOD via the generation of hydroxyl radical by a combination of the oxidation reaction of salsolinol and Fenton-like reaction of free copper ions released from oxidatively damaged SOD.

An Experimental Study on the Pond Sediment and Water Quality Purification using Oxygen Solubilization Device(OSD) System (산소용해수를 이용한 호소 저질 및 수질개선에 관한 실험적 연구)

  • The pollution in closing water can be caused by not only artificial factor like sewage but also natural factor like elution from sediment. In this study we analyzed Sediment Oxygen Demand (SOD) for verification of sediment purification effect and sediment elution experiment as well as general items like COD, TN, TP, SS to complement and assess the effect of sediment and water quality. The experiment result showed that the release rate of OSD system were 4 times and 3 times as large as control for P and Fe respectively. SOD for operated OSD system and control were $12.18gO_2{\cdot}m^{-2}{\cdot}d^{-1}$ and $47.95gO_2{\cdot}m^{-2}{\cdot}d^{-1}$. From water qualities analyzed by COD, TN, TP, SS, chlorophyll-a, the removal efficiency increase of TN, TP, chlorophyll-a and COD were about 10~20%, 40~50% and 10% respectively. In conclusion, OSD can contribute to improvement of both the waterbody and the sediment environment effectively.

Development of Resources for Functional Food and Biological Activity of Lentinus edoes mycelium (표고버섯 균사체의 생리활성과 기능성식품 소재 개발)

  • Objectives : This study was carried out to investigate an antioxidant ability, the change of antioxidant ability, antimicrobial activity and anticancer in functional meat ball and sausage prepared from Lentinus edoes mycelium and texturized soy protein. Methods : This experiments was carried out to investigate antioxidant ability(TBARS, DPPH, SOD-like ability), antibacterial activity and anticancer ability using sarcoma 180 extracts from Lentinus edoes mycelium. Result : Anticancer ability of Lentinus edoes mycelium showed a 28% survival rate and 63% of inhibition rate of tumor, which showed $1.30{\pm}0.4g$ of tumor weight. These results revealed an effective Lentinus edoes mycelium resources as anticancer sources. After heating peoducts prepared from Lentinus edoes mycelium, these products doesn't showed difference between after heating and before heating in measuring of SOD-like activity and DPPH. In DPPH experiment, ethanol extracts showed a high DPPH value as 90.85%, but hot water extracts showed 82.14% in DPPH value. Conclusion : In conclusion, it is very useful resources for preparing functional food on the basis of results from antioxidant(TBARS, DPPH, SOD-like ability), antibacterial activity and anticancer ability using sarcoma 180.

Protective Effect of Omega-3 of Polyunsaturated Fatty Acid Docosahexaenoic Acid on the Organic Mercury-Induced Cytotoxicity in Cultured NIH3T3 Fibroblasts

  • To clerify the protective effect of omega-3 of polyunsaturated fatty acid docosahexaenoic acid (DHA) on the cytotoxicity induced by organic mercury in cultured NIH3T3 fibroblasts. The measurement of cell viability on ogranic mercury wad done by XTT assay after NIH3T3 fibroblasts were cultured with various concentrations of methyl mercuric chloride (MMC). And also, the effect of DHA on the MMC-mediated cytotoxicity was examined by cell viability, and antioxidant effect of DHA was also assessed by superoxide dismutase (SOD)-like activity and the lipid peroxidation activity in cultured NIH3T3 fibroblasts. In this study, MMC decreased cell viability and $XTT_{50}$ value was determined at $50{\mu}M$ of MMC in these culture. In the effect of DHA against the cytotoxicity induced by MMC, DHA significantly increased the cell viability damaged by MMC in cultured NIH3T3 fibroblasts. And also, DHA showed the antioxidant effect by showing the increase of SOD-like activity and the decrease of lipid peroxidation activity. From these results, it is suggested that organic mercury such as MMC has highly toxic effect on cultured NIH3T3 fibroblasts, and also, omega-3 of polyunsaturated fatty acid, DHA showed the protection on MMC-induced cytotoxicity and antioxidant effect.

Antioxidant Activity and Anti-inflammatory Effects of Salix Koreensis Andersson Branches Extracts (버드나무(Salix Koreensis Andersson) 가지 추출물의 항산화 및 항염증 효과)

  • This study aims to compare and analyze a willow tree (Salix Koreensis andersson) extract's antioxidant and anti-inflammatory activity by investigating its: total polyphenol, flavonoid content, SOD-like activity, DPPH vitality. the willow tree was induced with LPS to determine its active anti-inflammatory effects. as a result, the willow methanol extract showed a higher total polyphenol and flavonoid content than those of willow distilled water extract, but the willow distilled water extract showed a higher SOD than that of willow methanol extract. in its DPPH scavenging ability, the willow methanol extract's antioxidant activity was higher than that of the willow distilled water extract. the willow extract's measurements such as the production of NO, inflammatory cytokine ($TNF-{\alpha}$, IL-6 measurement) were significantly reduced as its concentration level went down. according to the research outcomes, when induced, he will extract's macrophage produces mediator-like substances such as NO and inflammatory cytokine that can be used to alleviate the inflammatory response. therefore, the willow tree proved to be a useful raw plant material for the products designed to combat inflammatory activities due to its natural antioxidant and anti-inflammatory response substances such as NO and cytokine.

Antioxidant and Anticancer Effects of Agrimony (Agrimonia pilosa L.) and Chinese Lizardtail (Saururus chinensis Baill) (짚신나물, 삼백초의 항산화와 항암활성 효과)

  • The antioxidant activities of Agrimony (Agrimonia pilosa L.) and Chinese lizardtail (Saururus chinensis Baill) according to extraction methods were measured. SOD-like activity showed greater antioxidant effects with ethanol solvent than those with water. Ethanol extracts of Agrimony leaves showed the highest SOD-like activity of 94.4%. SOD-like activity differed according to the extraction solvents. The contents of polyphenolic compounds were higher in water extracts than those in ethanol extracts. The contents were 161.4 mg for Agrimony roots, 100.2 mg for Agrimony leaves, and 79.1 mg for Agrimony stalks in order. EDA in Agrimony leaves that were highest among medicinal plants were 83.4% in the water extract and 81.7% in the ethanol extract. The anticancer effects of the extracts by water and ethanol from Agrimony and Chinese lizardtail were experimented. The growth of stomach cancer cells, SNU-719 was inhibited 94.5% by the hexane fractions of Agrimony and also the growth of liver cancer cells, Hep3B was inhibited 83.2% by the hexane fractions of Agrimony, while the growth of normal cell, DC2.4 was not affected.

Comparison of Physiological Activities of Radish Bud (Raphanus sativus L.) according to Extraction Solvent and Sprouting Period (추출용매 및 발아시기에 따른 무순 추출물의 생리활성 비교)

  • This study extracted radish bud (Raphanus sativus L.) and investigated its nitrite scavenging activity, superoxide dismutases (SOD)-like activity, tyrosinase inhibition activity, xanthine oxidase inhibition activity, and angiotensin-converting-enzyme (ACE) inhibition activity according to extraction solvent and sprouting period. For nitrite scavenging activity, each extract recorded its highest level of 81.44~89.71% at pH 1.2. Radish bud extracts on sprouting days 4 and 8 showed greater scavenging activities than those on sprouting day 12 at pH 1.2 and pH 4.0. There were differences in scavenging activity according to extraction solvent based on water extract exhibiting improved scavenging activity. Ethanol extract recorded scavenging activity of 16.12% at pH 6.0, which was similar to those of ethanol and methanol radish bud extracts on sprouting day 12. SOD-like activity of radish bud extracts was in the range of 4.57~27.05%. For comparison purposes, SOD-like activity of L-ascorbic acid was 52.15%, which was higher than that of radish bud extracts. Acetone and methanol extracts showed high SOD-like activities on sprouting day 8. SOD-like activity of radish bud extracts on sprouting day 12 significantly decreased to 4.57~15.59%. Radish bud extracts recorded good tyrosinase inhibitory activities on sprouting 8 and 12, whereas methanol extracts recorded the greatest tyrosinase inhibitory activity at 62.65~84.89%. Radish bud extracts recorded xanthine oxidase inhibition activity of 21.26~29.52% on sprouting day 4, and acetone extracts showed the highest level of xanthine oxidase inhibition activity. Xanthine oxidase inhibitory activity tended to decrease with sprouting period compared early on. ACE inhibitory activity was in the range of 12.48~51.78% according to sprouting period and extraction solvent. Ethanol extracts on sprouting day 8 showed the highest ACE inhibitory activity of 51.78%. These results will hopefully contribute to research into the identification of materials and development of products for natural functional foods.

Biological Activities of Maca (Lepidium meyenii) Extracts (마카 추출액의 생리활성 효과)

  • This study was conducted to determine the optimal extraction conditions for maca by comparing the yields, total polyphenol contents, superoxide dismutase (SOD)-like activity and the nitrite scavenging ability. The proximate composition analysis showed 6.57% moisture, 12.83% crude protein, 1.05% crude fat, 4.80% ash and 74.75% carbohydrate. Maca was extracted with 7 different solvents (water, methanol, ethanol, acetone, ethyl acetate, chloroform and hexane) and the extracts were tested for biological activities. The extraction yields of water, methanol and ethanol extracts were 46.2%, 21.4% and 16.8%, respectively. Acetone, ethyl acetate, chloroform and hexane exhibited very low extraction yield, ranging from 0.2 to 1.0%. Total polyphenol contents and the nitrite scavenging ability were the highest in water extract. Electron donating ability and the SOD-like activity were the highest in methanol extract. When water extract was drawn out at different extraction temperatures (30, 70, $100^{\circ}C$) and time (1, 3, 5 hr), the improved biological activities (total polyphenol contents, electron donating ability, SOD-like activity and nitrite scavenging ability) were found in extracts treated at $100^{\circ}C$ for 3 or 5 hrs.

Physiological Activities of Ethanol Extracts from Different Parts of Ailanthus altissima (가죽나무(A. altissima) 부위별 에탄을 추출물의 생리활성)

  • The study was carried out to optimize the electron donating ability (EDA), superoxide dismutase (SOD)-like activity, nitrite scavenging ability, and the inhibitory activities of xanthine oxidase and tyrosinase of Ailanthus altissima ethanol extracts from roots, stems and leaves. The EDA of the roots and stem extracts were 64.04% and 63.27% at $1,000{\mu}g/mL$, respectively. The SOD-like activity of the leaves extract was the highest (50.00%) at $1,000{\mu}g/mL$. The nitrite scavenging ability of the leaves extracts was over 98% at pH 1.2 and 3.0. The xanthine oxidase inhibitory rates of the extracts were $93.62%{\sim}95.40%$ and the tyrosinase inhibitory rate of the roots was the highest (62.01%) at the concentration of $1,000{\mu}g/mL$. These results indicated that the roots extract showed the highest EDA and tyrosinase inhibition, while the leaves extract had tile highest SOD-like activity and nitrate scavenging ability.
