Multi-Residue Analysis of Fipronil and Its Metabolites in Eggs by SinChERS-Based UHPLC-MS/MS

  • A method for simultaneous detection of fipronil (F) and its metabolites fipronil desulfinyl (FD), fipronil sulfide (FS), fipronil sulfone (FSO) in chicken eggs was applied and validated. It includes single-step, cheap, effective, rugged, safe-based method (SinChERS) for sample preparation and ultra high performance liquid chromatography coupled with mass spectrometry (UHPLC-MS/MS) for chemical analysis. Results suggested that formic acid enhanced the recovery of 4 target residues and 1% supplementation to acetonitrile gained higher recoveries than that of 5%. SinChERS integrated extraction and clean-up steps into one, with shorter time (1.5 h) to operate and higher recoveries (97%-100%) than HLB, Envi-Carb-NH2 and quik-easy-cheap-effective-rugged-safe method (QuEChERS), and it consumed the smallest volume of extracting solvent (10 mL) as QuEChERS. Quantitative analyses using external standard method suggested the linear ranges of 4 target compounds were 1-20 ㎍/L with R2 >0.9947. The limit of detection (S/N>3) and quantification (S/N>10) were 0.3 ㎍/kg and 1 ㎍/kg. Recoveries ranged from 89.0% to 104.4%, and the relative standard deviations (n=6) at 1, 10, and 20 ㎍/kg were lower than 6.03%. Thirty batches of domestic eggs (500 g each) were detected by the established SinChERS-based UHPLC-MS/MS and no target residues were detected in all samples. The method developed in this study is a rapid, sensitive, accurate and economic way for multi-residue analysis of fipronil and its metabolites in eggs.

황화수소 가스 감지를 위한 고성능 변색성 섬유형 센서의 제작 및 개발 (Fabrication of High-Performance Colorimetric Fiber-Type Sensors for Hydrogen Sulfide Detection)

  • Hydrogen sulfide(H2S) gas is a high-risk gas that can cause suffocation or death in severe cases, depending on the concentration of exposure. Various studies to detect this gas are still in progress. In this study, we demonstrate a colorimetric sensor that can detect H2S gas using its direct color change. The proposed nanofiber sensor containing a dye material named Lead(II) acetate, which changes its color according to H2S gas reaction, is fabricated by electrospinning. The performance of this sensor is evaluated by measuring RGB changes, ΔE value, and gas selectivity. It has a ΔE value of 5.75 × 10-3 ΔE/s·ppm, showing improved sensitivity up to 1.4 times that of the existing H2S color change detection sensor, which is a result of the large surface area of the nanofibers. The selectivity for H2S gas is confirmed to be an excellent value of almost 70 %.

Automatical Cranial Suture Detection based on Thresholding Method

  • Purpose The head of infants under 24 months old who has Craniosynostosis grows extraordinarily that makes head shape unusual. To diagnose the Craniosynostosis, surgeon has to inspect computed tomography(CT) images of the patient in person. It's very time consuming process. Moreover, without a surgeon, it's difficult to diagnose the Craniosynostosis. Therefore, we developed technique which detects Craniosynostosis automatically from the CT volume. Materials and Methods At first, rotation correction is performed to the 3D CT volume for detection of the Craniosynostosis. Then, cranial area is extracted using the iterative thresholding method we proposed. Lastly, we diagnose Craniosynostosis by analyzing centroid relationships of clusters of cranial bone which was divided by cranial suture. Results Using this automatical cranial detection technique, we can diagnose Craniosynostosis correctly. The proposed method resulted in 100% sensitivity and 90% specificity. The method perfectly diagnosed abnormal patients. Conclusion By plugging-in the software on CT machine, it will be able to warn the possibility of Craniosynostosis. It is expected that early treatment of Craniosynostosis would be possible with our proposed algorithm.

GRASS와 Arc/Info를 이용한 DEM 데이터의 정확도와 에러 측정 (The Measurements of Data Accuracy and Error Detection in DEM using GRASS and Arc/Info)

  • GIS 데이타의 정확도 문제는 DEM과 같은 데이터의 유용성과 적용에 대한 서로 다른 견해를 불러 일으킨다. 데이터의 정확성은 좌표의 정확한 위치와 속성정보를 무작위적으로 검색하여 결정할 수 있다. DEM은 과거 보다는 손쉽게 취득할수 있고 이를 처리할수 있는 소프트웨어도 다양해 졌으나 GIS의 응용은 이미 만들어진 데이터에 따라 그 결과가 달라질수 있으므로 데이터의 정확도와 에러에 대한 주의를 기울일 필요가 있다. 본 연구의 목적은 1:24,000과 1:250,000 DEM 데이터를 이용하여 DEM의 정확도를 검색하고 데이터가 지닌 에러를 찾아내는 방법을 모색하는데 있다. GRASS와 Arc/Info를 이용하여 DEM을 레이어로 만들어내는 과정 또한 연구 되었다. 연구지역은 250 $km^2$의 면적을 지녔으며 연구 결과 1:250,000 DEM에서는 실제 등고값이 정상적으로 처리 되었으나 1:24,000 DEM에서는 실제의 등고값이 아닌 0으로 표현된 에러가 발견되었다.

  • SIMS는 뛰어난 원소검출감도와 깊이 분해능을 가지고 있어서 깊이에 따른 미량불 순물 분석에 필수적인 장비이지만, 시료와 불순물의 변화에 따라 이온화율과 깍이는 속도가 달라서 일어나는 matrix effect 때문에 표준 시료없이 정량분석을 할 수 없는 문제점이 있 다. 이런 SIMS의 단점을 보완하기 위한 방법으로 개발된 여러 가지 SNMS 기술 중 SIMS에 아무런 기계장치를 덧붙이지 않고도 정량화 개선효과를 가져오는 CsX+ SNMS에 대한 연구 를 진행하여, 지금까지 밝혀진 실리콘 산화막 등에서의 주성분원소 조성비분석을 통해 SNMS 기능을 확인하고 SIMS의 주 분석대상인 분순물 농도분석에의 적용가능성을 실험해 보았다. 이를 위해 실리콘에 BF2 이온 주입 후 붕소분포 분석시 강한 matrix effect를 나타 내는 불소의 효과를 SNMS와 SIMS로 비교하였으며, 검출한계와 dynamic range도 조사하였 다. 실험결과 CsX+ SNMS 기술은 matrix effect 때문에 실제분포와 다른 값으로 검출되는 불순물 시료분석에 적용할 수 있음을 알았다.

황산화미생물을 이용한 새로운 수(水)중 생태독성탐지 방법 (A New Methodology of Measuring Water Toxicity using Sulfur Oxidizing Bacteria)

  • 수중의 독성을 탐지하기 위하여 황산화미생물을 이용한 새로운 형태의 생태독성탐지 방법이 연구되었다. 황산화미생물이 산소 존재 하에서 황입자를 산화하여 황산을 만들게 되고 이는 EC의 증가 및 pH의 감소를 가져온다. 독성물질이 들어오면 미생물의 저해로 황산을 적게 만들게 되므로 EC의 증가 정도가 감소하게 된다. 인공하천수(EC= 0.12 mS/cm and pH=7.2)를 이용하여 황산화미생물을 배양하고 반응조를 HRT 30분으로 연속 운전한 결과 유출 인공하천수의 EC=0.5~1.2 mS/cm, pH= ~2.5이었으며 7일 동안 일정하게 유지되었다. 이러한 유입수와 유출수의 EC의 차이는 인공하천수가 독성이 없는 조건에서 일정 속도로 황산을 만들었기 때문이다. 아질산성질소 2 mg/L를 함유한 인공하천수를 넣은 결과 인공하천수 주입 후 1.5시간 경과 후 EC 값이 급격하게 감소하고 pH 값은 상승하여 독성을 쉽게 탐지 가능하였다. 최적의 체류시간은 30분이었으며 이는 황입자의 크기를 줄임으로써 더 짧은 체류시간에서 운전이 가능할 것으로 판단된다.

Nickel(II) Determination by Spectrophotometry Coupled with Preconcentration Technique in Water and Alloy Samples

  • A micro organism Agrobacterium tumifacient as an immobilized cell on a solid support was presented as a new biosorbent in a simple and sensitive spectrophotometry determination of Ni(II) in various samples using 4-hydroxy benzaldehyde-4-bromophenyl hydrazone as a color developing agent (λmax 497 nm) at pH 4.0 ± 0.2. Beer's law was obeyed over the range of 0.01-0.1 μg L-1. The molar absorptivity and Sandell's sensitivity were 1.285 × 105 L mol-1cm-1 and 0.007245 μg cm-2 respectively. Under these conditions, the preconcentration factor obtained was 82, and the detection limit achieved was 0.05 μg L-1. The detailed study of various interfering ions made the method more sensitive and selective. The recovery of Ni(II) from various samples range from 97.75 to 99.35%. The present method was successfully applied for the determination of Ni(II) in spiked, natural water and alloy samples. The proposed method was compared with reported methods in terms of Student's ‘t'-test and Variance ratio ‘f'-test which indicates that there is no significant difference between proposed and literature method at 95% confidence level.

선형시스템의 모델기반 고장감지와 분류 (Model-based fault detection and isolation of a linear system)

  • In this paper, we propose a model-based FDI(fault detetion and isolation) algorithm to detect and isolate fault in a linear system. The proposed algorithm is gased on an HFC(hydrid fault classifier) which consists of an FCART2(fault classifier by ART2 neural network) and an FCFM(fault classifier by fault models) which operate in parallel to isolate faults. The proposed algorithm is functionally composed of three main parts-parameter estimation, fault detection, and isolation. When a change in the system occurs, the estimated parameters go through a transition zone in which errors between the system output and the stimated output and the estimated output cross a predetermined thrseshold, and in this zone the estimated parameters are tranferred to the FCART2 for fault isolation. On the other hand, once a fault in the system is detected, the FCFM statistically isolates the fault by using the error between ach fault model out put and the system output. From the computer simulation resutls, it is verified that the proposed model-based FDI algorithm can be performed successfully to detect and isolate faults in a position control system of a DC motor.

PAX1 Methylation Analysis by MS-HRM is Useful in Triage of High-grade Squamous Intraepithelial Lesions

  • This study is aimed to investigate the role of paired boxed gene 1 (PAX1) methylation analysis by methylation-sensitive high-resolution melting (MS-HRM) in the detection of high grade lesions in atypical squamous cells cannot exclude high-grade squamous intraepithelial lesion (ASC-H) and compared its performance with the Hybrid Capture 2 (HC2) human papillomavirus (HPV) test. In our study, 130 cases with a diagnosis of ASC-H from the cervical cytological screening by Thinprep cytologic test (TCT) technique were selected for triage. Their cervical scrapings were collected and evaluated by using PAX1 methylation analysis (MS-HRM) and high-risk HPV DNA test (HC2), followed by colposcopy and cervical biopsy. Chi-square test were used to test the differences of PAX1 methylation or HPV infection between groups. In the detection of CIN2+, the sensitivity, specificity, the PPV, NPV and the accuracy of PAX1 MS-HRM assay and high-risk HPV (HR-HPV) tests were respectively 80.6% vs 67.7%, 94.9% vs 54.5%, 83.3%, vs 31.8%, 94.0% vs 84.4%, and 91.5% vs 57.7%. The PAX1 MS-HRM assay proved superior to HR-HPV testing in the detection of high grade lesions (CIN2+) in ASC-H. This approach could screen out the majority of high grade lesion cases of ASC-H, and thus could reduce the referral rate to colposcopy.

HPLC-UV검출방법으로 유도체화를 통한 비테인과 콜린의 동시분석 (Simultaneous determination of betaine and choline using derivatization by HPLC with UV detection)

  • Extraction of quaternary ammonium compounds (choline and betaine) from plant samples (spinach) using ion exchange resin (AG1, OH form) is a very simple and inexpensive approach. However, it is very hard to determine amounts of choline and betaine simultaneously using high-performance liquid chromatography-ultraviolet (HPLC-UV) detection. Unlike choline, betaine has low molar absorptivity in UV-visible (UV-Vis) region, which makes it difficult to carry out UV-Vis detection of betaine. The mixture of quaternary ammonium compounds (choline and betaine) was derivatized using 2-bromo acetophenone as a derivatizing agent. As a result, choline did not react with the derivatizing agent, whereas betaine formed a betaine derivative. This betaine derivative exhibited detectable UV absorption with baseline separation between choline and the betaine derivative. Thus, with this method, choline and betaine can be determined simultaneously by using the HPLCUV method through one-step derivatization, which is an easy, sensitive, and reliable method.
