Purification of a New Elicitin from Phytopthora cambivora KACC40160 (Phytophthora cambivora KACC 40160으로부터 새로운 elicitin의 분리)

  • Elicitins, proteinaceous elicitors secreted from Oomycetes fungi (Phytophthora spp. and Pythium spp.), have been known as inducer of hypersensitive response (HR) in incompatible interactions between plant and pathogens. Five elicitins among many Korean Phytophthora species caused the reactions of distal HR in radish, chinese cabbage and some hot pepper cultivars, but not in cucumber and tomato. Because the isolation of elicitin from Phytophthora cambivora hasn't been reported yet, we have purified a cambivorein, a new member of the elicitin family, from the culture filtrate of Phytophtilora cambivora (KACC 40160) by using FPLC (Fast Protein Liquid Chromatography, AKTA) with sepharose S and Sephacryl HR columns. We confirmed that it induces necrosis activities in some hot pepper cultivars and its molecular weight is about 10 KDa by Tricine-SDS-PAGE. Comparison of amino acid sequences of its N-terminal ends also informed the identification of Iysine at the 13th position, which is characteristic of a kind of basic elicitin isoform $({\beta}-isoform)$. It Also showed that our elicitin is not identical with N-terminal sequences of many elicitins reported from Phytophthora spp..

Construction and Characterization of Vitreoscilla Hemoglobin (VHb) with Enhanced Peroxidase Activity for Efficient Degradation of Textile Dye

  • Pollution resulting from the discharge of textile dyes into water systems has become a major global concern. Because peroxidases are known for their ability to decolorize and detoxify textile dyes, the peroxidase activity of Vitreoscilla hemoglobin (VHb) has recently been studied. It is found that VHb and variants of this enzyme show great promise for enzymatic decolorization of dyes and may play a role in achieving their successful removal from industrial wastewater. The level of VHb peroxidase activity correlates with two amino acid residues present within the conserved distal pocket, at positions 53 and 54. In this work, sitedirected mutagenesis of these residues was performed and resulted in improved VHb peroxidase activity. The double mutant, Q53H/P54C, shows the highest dye decolorization and removal efficiency, with 70% removal efficiency within 5 min. UV spectral studies of Q53H/P54C reveals a more compact structure and an altered porphyrin environment (λSoret = 413 nm) relative to that of wild-type VHb (λSoret = 406), and differential scanning calorimetry data indicate that the VHb variant protein structure is more stable. In addition, circular dichroism spectroscopic studies indicate that this variant's increased protein structural stability is due to an increase in helical structure, as deduced from the melting temperature, which is higher than 90℃. Therefore, the VHb variant Q53H/P54C shows promise as an excellent peroxidase, with excellent dye decolorization activity and a more stable structure than wild-type VHb under high-temperature conditions.

Effect of different soybean meal type on ileal digestibility of amino acid in weaning pigs

  • An experiment was conducted to evaluate apparent (AID) and standardized (SID) ileal digestibilities of crude protein (CP) and amino acids (AA) with 6 soybean products in weaning pigs. A total of 14 weaning barrows with an initial body weight of $6.54{\pm}0.34kg$ were fitted with T-cannula at the distal ileum and allotted to 7 diets containing various soybean products. The soybean products used in the experiment were conventional soybean meal (CSBM), SBM fermented by Aspergillus oryzae GB-107 (FSBMA), SBM fermented by Bacillus subtilis PP6 (FSBMB), UV sterilized SBM fermented by Bacillus subtilis PP6 (UVFSBMB), SBM containing Bacillus subtilis PP6 (PSBM), and soy protein concentrate (SPC). Six corn-based diets were used and each of soybean products was added. All diets contained 5.0 g/kg of chromic oxide as an indigestible indicator and an N-free diet was used to measure basal endogenous losses of CP and AAs. Ileal CP digestibility did not differ by different soybean products. However, SIDs of Ile, Phe and Val were improved in pigs fed the FSBMB, UVFSBMB and SPC diets and the pigs fed the FSBMA diet showed higher SIDs of Phe and Val compared with those fed the CSBM diet (P < 0.05). The FSBMB diet had higher SIDs in most AAs compared with the FSBMA diet (P < 0.05), and higher SIDs of Lys, Ala, Pro, Ser, and Tyr compared with PSBM diet (P < 0.05). However, there was no response of UV-sterilization on the FSBMB in the SIDs of AAs. These results suggest that SIDs of AAs could be improved by the supplementation of fermented soybean products in the diet for weaning pigs but fermentation with Bacillus subtilis is more efficient in improving ileal AA digestibility than that with Aspergillus oryzae. Furthermore, probiotics supplementation in the CSBM and UV-sterilization of the FSBMB had no effects on chemical composition and ileal AA digestibility.

THE EFFECT OF EXOGENOUS ELECTRIC CURRENTS ON CYCLIC NUCLEOTIDES IN FELINE ALVEOLAR BONE (외인성전류가 고양이 치조골의 cyclic nucleotides에 미치는 영향에 관한 연구)

  • There are evidences that exogenous electric currents are capable of enhancing bone formation and resolution, and that the conversion of the bioelectric response to biochemical activity provides the directional component of orthodontic tooth movement. In addition, evidence has implicated cyclic nucleotides in alveolar bone cellular activation mechanism during orthodontic tooth movement. In view of these evidences, this study was performed to investigate the effects of exogenous electric currents on cyclic nuclotide levels in feline alveolar bone and the possible clinical application of electric currents as an additional orthodontic tool. In the first study, three groups of three adult cats were subjected to application of a constant direct current of $10{\pm}2$ microamperes to gingival tissue near maxillary canine noninvasively for 1, 3, and 7 days respectively. In the second study, three groups of three adult cats each were treated by an electric-orthodontic procedure for 1, 3, and 7 days respectively. The left maxillary (control) canine received an orthodontic force of 80gm alone at time of initiation, while the right maxillary (experimental) canine received combined force-electric stimulation (80gm of force and $10{\pm}2$ microamperes of a constant D.C. currents). Alveola, bone samples were obtain from the mesial (tension and/or cathode) and the distal (compression and/or anode) sites surrounding maxillary canines as well as from contralateral control sites. The samples were extracted, boiled, homogenized, and the supernatants were assayed for cyclic nucleotides (cAMP, cGMP) by a radioimmunoassay method. And also the amount of tooth movement was measured in the second study. On the basis of this study, the following conclusions can be drawn: 1. The fluctuation pattern of cyclic nucleotide levels in alveolar bone treated by exogenous electric currents was similar to that treated by orthodontic force. 2. The cAMP levels in alveolar bone of electrically treated teeth significantly elevated above the control values. And of electrically treated teeth, the values of the anode sites were higher than those of the cathode sites. 9. The cGMP levels in alveolar bone of electrically treated teeth elevated above the control values at the initiation phase of treatment, but dropped below the control values at time of termination. And of electrically treated teeth, the values of the cathode sites were higher than those of the anode sites. 4. The rate of tooth movement in teeth . treated by force-electric combination increased with the length of treatment as compared to that treated by mechanical force alone.

Comparison on marginal fitness and mechanical properties of copings with zirconia block and CAM type (지르코니아 블록과 CAM 종류에 따른 코핑의 변연적합도와 기계적 특성 비교)

  • Purpose: This study provided the basic data for selection the zirconia block and CAM by means of marginal fitness observations, flexural strength test and hardness test. Methods: Three dental zirconia blocks(ABCera, NaturaZ, ST98) and two dental milling machines(CAD/CAM MS, DWX-50) were used in this study. Metal abutment(diameter 10 mm, height 5 mm, inclined angle $3^{\circ}$ taper, 1 mm chamfer margin) was fabricated by Ti customized abutment, and then zirconia copings were fabricated for each ten specimens. Silicone replica technique was used to observe the marginal fitness of cross-sections with a stereomicroscope at ${\times}50$ magnification. The dental zirconia blocks was cut into 10 pieces each having a size of $25mm{\times}5mm{\times}1mm$, and fabricated according to the manufacturer's instructions, and flexural strength was measured using a universal testing machine. For hardness test, a micro Vickers hardness tester was used as it was in the flexural strength test. Statistical analysis was performed by one way ANOVA and post-test was performed by Scheffe test. Results: For marginal fitness of bucco-lingual axial, ZU group($59.7{\pm}10.3{\mu}m$) was the lowest, followed by RA, ZA, ZD, RD, RU. For marginal fitness of mesio-distal axial, ZU group($59.3{\pm}10.2{\mu}m$) was the lowest, followed by RA, ZA, RD, ZD, RU. One-way ANOVA showed statistically significant difference between groups(p<0.05). For flexural strength, ABCera block($718.0{\pm}57.2MPa$) was the highest, followed by NaturaZ, ST98. For hardness, ABCera block($1550.3{\pm}19.8Hv$) was the highest, followed by ST98, NaturaZ. There was no significant difference in flexural strength and hardness between blocks(p>0.05). Conclusion: Based on the results of this study, the type of dental zirconia block did influence the marginal fitness, and all dental zirconia blocks are expected to be suitable for clinical application. The highest flexural strength and hardness were ABCera block, and no statistically significant difference was observed.

The study on the survival rates and crestal bone changes around the implants (임플란트 주위 골변화와 생존율에 대한 연구)

  • The success and failure of dental implants depends on various factors such as patient's systemic status, quantity and quality of surrounding bone, presence or absence of marginal infection and mechanical loading condition. The measurement of crestal bone changes around the implants is implemental to evaluate the success and long-term prognosis of the implant. This study was to evaluate the cumulative survival rate of the implants which had been placed in the Department of Periodontics, Chonnam National University Hospital between 1992 and 2003, and to observe the crestal bone loss around the implants which had at least 2 consecutive periapical radiographs after connecting the transmucosal abutment. The radiographs were scanned and digitalized, and the crestal bone levels on the mesial and distal surface of implants were measured using Image analyzer (Image Pro Plus, Media Cybernetics, USA), immediately after implant placement, at 2nd surgery, and 3 months, 6 months, 1 year, and every year thereafter. Any bone loss was not observed during the period between the 1stand 2nd surgery, and the bone loss was 0.86 ${\pm}$ 0.92 mm for the first year of loading after connecting the transmucosal abutment. After 1 year of loading, annual bone loss was 0.1 ${\pm}$ 0.27 mm, and total bone loss was 0.90 ${\pm}$ 0.80 mm (during the average follow-up periods of 22.5 ${\pm}$ 25.6 Mos), The implant, with smooth surface, in the mandible, and with the fixed bridge prosthesis showed greater bone loss, compared to those, with the rough surface, in the maxilla and with single crown. In systemically diseased patients (including DM or osteoporosis), the greater bone loss was observed. The cumulative survival rate among 432 implants was 94.10% for 7 years. Among 15 failed implants, 9 implants were removed due to mobility from disintegration of bone-implant interface. From this results, crestal bone loss around the implants were greatest during 1 year after transmucosal abutment connection, and various factors could affect peri-implant bone loss. To prevent and predict the bone loss around the implants and improve the prognosis, further comprehensive maintenance and follow-up schedules are required.

Reconstruction of Tibial Defects in Lower Extremity With Various Versions of Vascularized Fibula Transfer (다양한 형태의 생 비골 이식술을 이용한 경골의 재건)

  • Twelve cases in eleven patients with segmental bone defects were treated with contralateral fibula free flap and ipsilateral island fibula flap in an antegrade, retrograde or bidirectional flow fashion. Five cases were managed with free flaps and seven were with ipsilateral fibula island transfer. Among seven cases, antegrade fashion was three, retrograde was three, and bidirectional was one. All patients were related with open tibial fractures and its sequelae except one who had open foot bone fracture. According to Gustilo's classification, ten patients were type IIIb and one was type IIIc. Basically, antegrade-flow flaps based on the peroneal vessels as in the conventional free flap were used for the proximal or middle one-third tibial defects. On the contrary, retrograde-flow flaps based on the communicating branch between the peroneal and posterior tibial vessels were used for the middle or distal one-third of the tibia. Bidirection-flow flap based on intact peroneal vessels were used for the middle portion of the tibia. The patients who have undergone ipsilateral fibula island flap had one of the following problems: a previously failed free flap, below-knee amputation of the opposite leg because of open tibial fracture, refusal to use the contralateral sound leg, or poor general condition to stand a lengthy operation. Six of the patients who have got ipsilateral fibula island flap also had an associated fibula fracture on the same leg, which was ultimately used as one of the osteotomy sites. The follow-up period was from 1 to 10 years. Two cases of free flap were failed: one patient had below-knee amputation and the other patient had ipsilateral fibula transfer. Other cases were successful and excellent hypertophy of the transferred fibula was achieved. Time to bone union ranged from 4 to 11 months. Time to full weight bearing was from 5 to 13 months after surgery. All of the transferred fibulas showed hypertrophy after weight bearing. In one case, stress fracture was developed during ambulation, which was healed conservatively. Nonunion occurred in two cases, which were treated with a long leg cast and cancellous bone graft, respectively. Length discrepancy of the legs was noted. The limb was shorter by an average 0.5 cm in three cases, longer by 1.1 cm in one case. In the case of island fibula transfer, limited arc of rotation was not a problem. Other disabling complications were not seen. We believe that these diverse modalities using a vascularized fibula will make us more comfortable to handle major bone defects.

  • Ectopic eruption of the maxillary first permanent molar means that the molar erupts out of the normal position and is arrested in its eruption by the second primary molar. This local eruption disturbance results in a premature atypical resorption on the distal part of the second primary molar. In most irreversible cases, the second primary molar is lost prematurely, either by spontaneous exfoliation or by extraction, In cases of doubt as to whether the eruption is of the irreversible type or not, careful radiographic observation period for a few months would be valuable in evaluating the possibilities of the tooth's freeing itself. The purpose of this study was to determine the characteristics and occurrence of the ectopic eruption of the maxillary first permanent molar. A descriptive, observational, retrospective study was done using the radiographs of 25 conseutive patients, who were in the first phase of mixed dentition. A method was designed to evaluate the amount of pathologic resorption of the second maxillary primary molar and the mesial angulation of the first permanent molar. The study showed that the most important etiologic factor was the eruption path or mesial angulation of the first permanent molars relative the chosen reference lines.

Correlation of Preoperative Ki67 and Serum CA15.3 Levels with Outcome in Early Breast Cancers - a Multi Institutional Study

  • Background: To investigate the association between preoperative pathological Ki-67 labeling index and serum tumor marker cancer antigen 15-3 (CA 15-3) with clinic-pathological parameters and treatment outcomes in early breast cancer. Materials and Methods: A retrospective study at 4 cancer centers in Saudi Arabia and Egypt was performed. Data were collected for female patients diagnosed with unilateral early breast cancer between March 2010 and October 2013. Cases treated with neoadjuvant chemotherapy (NACT) followed by surgery and radiotherapy were included. NACT included 6-8 cycles of anthracycline and taxane based regimens. Trastuzumab and hormonal treatments were added according to HER2 and hormone receptor status. Baseline serum CA15.3 and pathological Ki67 levels were evaluated and correlated with disease free survival (DFS) and overall survival (OS). Results: A total of 280 pts was included. The median age was 49 years (38-66 y) and median overall survival was 35 (20-38) months (mo). Estrogen receptors (ER), progesterone receptors (PR) and HER 2 receptors were positive in 233 (83.2%), 198 (70%) and 65 cases (23.2%), respectively. High preoperative Ki67 and CA15.3 were noted in 177 (63.2%) and 131 (46.8%). A total of 45 (16%) patients had distal or local recurrence and 24 (8.6%) died of their disease. Most of the relapsed cases had high preoperative Ki-67 (n=41, 91%) and CA15.3 (n=28, 62%) values. All of the patients who died had a high Ki-67 but CA15.3 was high in 9 (37%) only. Mean DFS/OS in patients with high preoperative Ki-67 was 32 months /32 months as compared to 37 months/35 months in those with normal Ki-67 (p<0.001). Correlation of preoperative CA15.3 and survival was statistically not significant. Conclusions:Preoperative Ki-67 can be a predictive and prognostic marker. Higher levels are associated with poor DFS and OS in patients with early BC.

Energy and Nutrient Digestibility in Four Sources of Distillers Dried Grains with Solubles Produced from Corn Grown within a Narrow Geographical Area and Fed to Growing Pigs

  • Two experiments were conducted to determine energy and nutrient digestibility in four sources of distillers dried grains with solubles (DDGS) produced from corn and fed to growing pigs. The objective was to test the hypothesis that energy and nutrient digestibility in DDGS does not vary if samples are procured from ethanol plants that use similar production technologies and use corn that is grown within a narrow geographical area. The four sources of DDGS that were used were procured from ethanol plants that were less than 10 yr old and located within 250 km of each other. In Exp. 1, five growing barrows (initial BW = 71.4 kg) that were fitted with a T-canula in the distal ileum were allotted to a 5${\times}$5 Latin square design and used to measure apparent (AID) and standardized (SID) ileal digestibility of AA in the four sources of DDGS. Results of this experiment showed that the SID of CP and all AA except Cys and Pro were greater (p<0.05) in two of the DDGS sources than in the other two sources. Exp. 2 was conducted to measure the concentration of DE and ME and the apparent total tract digestibility (ATTD) of energy, N, P, ether extract, NDF, and ADF in corn and in the same four sources of DDGS as used in Exp. 1. Five pigs (initial BW = 29.7 kg) that were placed in metabolism cages and allotted to a 5${\times}$5 Latin square design were used. Results of Exp. 2 showed that the average DE and ME in DDGS were 4,072 and 3,750 kcal/kg DM, respectively, which was less (p<0.01) than the DE and ME in corn (4,181 and 4,103 kcal/kg DM, respectively). The average ATTD for P in DDGS was 56.1%, which was greater (p<0.01) than the ATTD for P in corn (31.9%). The ATTD for ADF in DDGS was also greater (p<0.05) than in corn, but the ATTD for ether extract and NDF were greater (p<0.05) in corn than in DDGS. It is concluded that energy and nutrient digestibility vary among sources of DDGS even when the DDGS is procured from ethanol plants that use corn grown within a narrow geographical region. Thus, factors other than corn growing region are responsible for the variability of energy and nutrient digestibility in DDGS.
