전동차내 부유 미생물 저감방안에 관한 연구 (A Research to Decrease Airborne Microoganism the Train)

  • 서울메트로 지하철 1~4호선 이용객은 하루 평균 450만명이 지하철을 이용(2010년 상반기 수송인원 현황)하고 있으며 이것은 서울시 인구 57%에(2009년도 서울시 인구 총1,046만명) 가까운 사람들이 지하철을 이용하고 있는 것으로 나타남에 따라 전동차에 쾌적한 실내공기질 유지 및 관리는 무엇보다 중요하다. 특히 전동차내 실내공기질에 영향을 미치는 요인으로는 승객의 재채기, 호흡과 같은 생체 활동 및 피부각질 등에 의한 부유 미생물 작용에 있다(권순박 등, 2011). 바이러스와 같은 미생물 오염 물질이 승객의 인체의 호흡기, 점막부위, 피부 등에 접촉 할 경우 과민성 질환, 아토피 피부염, 전염성 질환, 알레르기성 질환, 호흡기 질환을 유발할 수 있는(환경부-국립환경과학원, 생활속 세균 이렇게 관리하세요) 산업 보건상의 문제로 대두되고 있어 적절한 환기 시스템 도입과 제습을 위한 냉방 등이 필요한 실정이다. 시민들의 관심과 보조를 맞추어 지하철 2호선 출 퇴근 및 평상시 전동차 객실내 부유 미생물을 측정한 결과 부유세균과 부유진균은 2011년도 85~385$cfu/m^3$, 67~98$cfu/m^3$로 기준치(별도의 법적 기준은 없으나, 부유세균은 국내 실내기준 800$cfu/m^3$, 부유진균은 세계보건기구의 가이드라인 150$cfu/m^3$) 보다 낮은 수준으로 나타났으나 유동인구가 많은 출 퇴근 시간대에는 미생물 오염 물질이 승객에게 쉽게 전염 될 가능성이 있을 것으로 사료됨에 따라 부유 미생물 오염방지를 위한 공기조화설비 성능개선 등 실질적인 개선방안을 찾는 것이 필요하다. 이에 본 연구에서는 2004년 이후 부유 미생물 제어장치에 대한 연구 논문을 분석하여 부유미생물이 승객들에게 미치는 영향, 환기량 증대방안 등의 다양한 인자에 대한 분석을 통하여 전동차내 공기질을 실질적으로 개선할 수 있는 방법을 연구 하였다.

Antiviral and Anti-Inflammatory Activities of Pochonin D, a Heat Shock Protein 90 Inhibitor, against Rhinovirus Infection

  • Human rhinoviruses (HRV) are one of the major causes of common cold in humans and are also associated with acute asthma and bronchial illness. Heat-shock protein 90 (Hsp90), a molecular chaperone, is an important host factor for the replication of single-strand RNA viruses. In the current study, we examined the effect of the Hsp90 inhibitor pochonin D, in vitro and in vivo, using a murine model of human rhinovirus type 1B (HRV1B) infection. Our data suggested that Hsp90 inhibition significantly reduced the inflammatory cytokine production and lung damage caused by HRV1B infection. The viral titer was significantly lowered in HRV1B-infected lungs and in Hela cells upon treatment with pochonin D. Infiltration of innate immune cells including granulocytes and monocytes was also reduced in the bronchoalveolar lavage (BAL) by pochonin D treatment after HRV1B infection. Histological analysis of the lung and respiratory tract showed that pochonin D protected the mice from HRV1B infection. Collectively, our results suggest that the Hsp90 inhibitor, pochonin D, could be an attractive antiviral therapeutic for treating HRV infection.

국내의 돼지 인플루엔자 바이러스(H1N1, H3N2)의 혈청학적 조사 (Sero-prevalence of swine influenza virus (H1N1, H3N2) In Korea)

  • Swine influenza is an acute, infectious respiratory disease caused by type A influenza viruses in pigs. In the previous studies, serological surveys have indicated the presence of H3N2 swine influenza virus (SIV) since 1995 in Korea. And the percentage of the antibody-positive rate was 39.12% in the survey determining the prevalence of H1N1 SIV antibodies in 2002. The purpose of this study was therefore to investigate the sero-prevalence of SIV regard to the age of the pig and the season between June 2004 and May 2005. In this study, a total of 932 sera were used. These sera were randomly selected from blood samples, which were submitted to Department of Veterinary Pathology, Kangwon National University and Department of Veterinary Virology, Seoul National University from June 2004 to May 2005. These sera have been tested by ELISA test kit (IDEXX Lab, USA) for the SIV H3N2, H1N1 respectively. SAS version 9.1 was used for the statistical analysis based on the age of the pig and the season. The overall sero-prevalence of the antibody against H3N2 SIV was 20.82% (194/932). The overall sero-prevalence of the antibody against H1N1 SIV was 37.23% (347/932). The overall dual sero-prevalence of the antibody against H3N2 and H1N1 SIV was 10.62% (99/932). H3N2 has significant difference in statistically regarding the age of the pig and the season (p<0.0001). H1N1 has significant difference in statistically regarding the age of the pig (p<0.0001) but has not significant difference in statistically regarding the season (p=0.5882).

Antiviral Activity of Fritillaria thunbergii Extract against Human Influenza Virus H1N1 (PR8) In Vitro, In Ovo and In Vivo

  • Influenza viruses cause respiratory diseases in humans and animals with high morbidity and mortality rates. Conventional anti-influenza drugs are reported to exert side effects and newly emerging viral strains tend to develop resistance to these commonly used agents. Fritillaria thunbergii (FT) is traditionally used as an expectorant for controlling airway inflammatory disorders. Here, we evaluated the therapeutic effects of FT extracts against influenza virus type A (H1N1) infection in vitro, in ovo, and in vivo. In the post-treatment assay, FT extracts showed high CC50 (7,500 ㎍/ml), indicating low toxicity, and exerted moderate antiviral effects compared to oseltamivir (SI 50.6 vs. 222) in vitro. Antiviral activity tests in ovo revealed strong inhibitory effects of both FT extract and oseltamivir against H1N1 replication in embryonated eggs. Notably, at a treatment concentration of 150 mg/kg, only half the group administered oseltamivir survived whereas the FT group showed 100% survival, clearly demonstrating the low toxicity of FT extracts. Consistent with these findings, FT-administered mice showed a higher survival rate with lower body weight reduction relative to the oseltamivir group upon treatment 24 h after viral infection. Our collective results suggest that FT extracts exert antiviral effects against influenza H1N1 virus without inducing toxicity in vitro, in ovo or in vivo, thereby supporting the potential utility of FT extract as a novel candidate therapeutic drug or supplement against influenza.

공기 중 바이러스 포집 장비의 효율성 평가 (Assessment of Instrument Efficiency in Detecting Airborne Virus)

  • In livestock industry, damage caused by the epidemic diseases such as Foot-and-Mouth Disease (FMD), Highly-Pathogenic-Avian-Influenza (HPAI) and Porcine-Reproductive-and-Respiratory-Syndrome (PRRS) was very serious. The financial loss incurred from FMD alone which occurred on Nov. 2011 in Korea was estimated at 3 billion won, 23 % of annual livestock industry production. The livestock industry in Korea has greater risk of disease infection because of high density production, etc. Investigating the spread of livestock diseases should consider both direct and indirect contact as well as other various factors including airborne. Airborne infection of livestock disease was first hypothesised in the early 1900s, however, field experimental studies are still limited. Furthermore, no protocol is available in detecting airborne viruses in the field. In this study, effective virus samplers were investigated by comparative analysis of the type of samplers used detect to airborne virus. Laboratory experiments were conducted to compare virus samplers such as Bio-sampler, Dust-sampler, Compact-Cascade-Impactor (CCI) and Microflow in detecting PRRSV. Samples were analyzed by Reverse-Transcription PCR to assess the efficiency of the instrument in detecting the airborne virus. First, samples were classified into five levels according to light intensity of gel images and then the classified results were normalized. In every case, Bio-sampler and Dust-sampler were comparable with each other and have shown to be more effective than CCI and Microflow samplers.

Survival of Microorganisms on Antimicrobial Filters and the Removal Efficiency of Bioaerosols in an Environmental Chamber

  • Exposure to bioaerosols causes various adverse health effects including infectious and respiratory diseases, and hypersensitivity. Controlling exposure to bioaerosols is important for disease control and prevention. In this study, we evaluated the efficacies of various functional filters coated with antimicrobial chemicals in deactivating representative microorganisms on filters or as bioaerosols. Tested functional filters were coated with different chemicals that included (i) Ginkgo and sumac, (ii) Ag-apatite and guanidine phosphate, (iii) $SiO_2$, ZnO, and $Al_2O_3$, and (iv) zeolite. To evaluate the filters, we used a model ventilation system (1) to evaluate the removal efficiency of bacteria (Escherichia coli and Legionella pneumophila), bacterial spores (Bacillus subtilis spore), and viruses (MS2 bacteriophage) on various functional filters, and (2) to characterize the removal efficiency of these bioaerosols. All experiments were performed at a constant temperature of $25^{\circ}C$ and humidity of 50%. Most bacteria (excluding B. subtilis) rapidly decreased on the functional filter. Therefore, we confirmed that functional filters have antimicrobial effects. Additionally, we evaluated the removal efficiency of various bioaerosols by these filters. We used a six-jet collision nebulizer to generate microbial aerosols and introduced it into the environmental chamber. We then measured the removal efficiency of functional filters with and without a medium-efficiency filter. Most bioaerosol concentrations did not significantly decrease by the functional filter only but decreased by a combination of functional and medium-efficiency filter. In conclusion, functional filters could facilitate biological removal of various bioaerosols, but physical removal of these by functional was minimal. Proper use of chemical-coated filter materials could reduce exposure to these agents.

  • Defensins are antimicrobial peptides that participate in the innate immunity of hosts. Humans constitutively and/or inducibly express ${\alpha}$- and ${\beta}$-defensins, which are known for their antiviral and antibacterial activities. This review describes the application of human defensins. We discuss the extant experimental results, limited though they are, to consider the potential applicability of human defensins as antiviral agents. Given their antiviral effects, we propose that basic research be conducted on human defensins that focuses on RNA viruses, such as human immunodeficiency virus (HIV), influenza A virus (IAV), respiratory syncytial virus (RSV), and dengue virus (DENV), which are considered serious human pathogens but have posed huge challenges for vaccine development for different reasons. Concerning the prophylactic and therapeutic applications of defensins, we then discuss the applicability of human defensins as antivirals that has been demonstrated in reports using animal models. Finally, we discuss the potential adjuvant-like activity of human defensins and propose an exploration of the 'defensin vaccine' concept to prime the body with a controlled supply of human defensins. In sum, we suggest a conceptual framework to achieve the practical application of human defensins to combat viral infections.

울산지역의 개 인플루엔자 바이러스의 항체보유 실태 조사 (Prevalence on protective serum antibodies of canine influenzae virus in Ulsan area)

  • Canine influenza virus (CIV) is an emerging pathogen that causes severe and acute respiratory disease in dogs. In 2006, the H3N2 CIV was first identified in dogs from Guangdong province in China. The nine isolates were grouped together with the canine H3N2 viruses isolated from dogs and cats in Korea. The possible interspecies transmission of influenza A virus is very important. We carried out a serological retrospective study using invited canine serum. The hospital invited 123 dogs, first vaccination group were revealed with CIV antibody positive rate of 81.8%. the second vaccination group were detected a positive rate of 91.2%. Antibody generation rate was higher in 3~10 years dogs. Protective antibody titers were detected from 2 weeks to 12 months. thereafter below the protective antibody. The results indicate that H3N2 CIV may have been consistently circulating in dog populations. Recently. These findings showed that H3N2 CIV has the capacity to replicate in and transmit among cohoused dogs and underscore the need for continued public health surveillance. Considering the result continuous management and prevention system against CIV is required at the concentrated animal care centers. The importance of CIV surveillance in this region for understanding the genesis of this virus, and it is important to remain aware of the potential of H3N2 CIV to be transmitted from dogs to the human population.

A Docking Study of Newly Found Natural Neuraminidase Inhibitor: Erystagallin A

  • It's a threat for the public health that H1N1 (Influenza virus A) causes disease and transmits among humans. WHO (world health organization) declared that the infections caused by the new strain had reached pandemic proportions. The approved neuraminidase inhibitors (Zanamivir and Oseltamivir) and related investigative drug (BCX-1812) are potent, specific inhibitors of influenza A and B viruses. These drugs are highly effective to prevent influenza A and B infections. Early therapeutic use reduces illness duration and respiratory complications. Recently, we found one of the potent inhibitor of erystagallin A ($IC_{50}$ of 2.04 ${\mu}M$) for neuraminidase target, this inhibitor shows most similar structure to its natural substrate, sialic acid. Therefore, we chose 1l7f to get the receptor structure for docking study among many crystal structures. A docking study has been performed in Surflex-Dock module in SYBYL 8.1. In the present study, we attempt to compare the docking studies of pterocarpin and erystagallin A with neuraminidase receptor structure. In the previous report, the methoxy group of pterocarpin had H-bonding with Arg residues. The present docking results for erystagallin A showed the backbone of hydroxyl group shows significant H-bonding interactions with Arg152 and Arg292. The results showed that erystagallin A interacts more favorably with distinctive binding site rather than original active site. Therefore, we tried to reveal plausible binding mode and important amino acid for this inhibitor using docking and site id search calculations of Sybyl. The results obtained from this work may be utilized to design novel inhibitors for neuraminidase.

국내 돼지 유산태아에서 원인 바이러스 검출과 유병률 조사 (Detection and prevalence of viral pathogens from aborted fetuses and stillborn piglets in Korea)

  • This experiment was conducted to investigate the presence of recognized abortifacient viruses from aborted fetuses and stillborn piglets in cases of reproductive failure in sows by PCR. A total of 219 samples of aborted fetuses or stillborn piglets, submitted to the School of Veterinary Medicine of Kangwon National University between 2006 and 2009 May, were collected from 5 provinces in Korea. Abortifacient virus infections were detected in 82 (37.4%) out of 219 aborted fetuses or stillborn piglets as well as on 39 (40.2%) out of 97 pig farms. The major viral infections were porcine circovirus type 2 (PCV2), porcine reproductive and respiratory syndrome virus (PRRSV) and Aujesky's disease virus (ADV) for which 46 (21%), 19 (8.6%) and 16 (7.3%) were positive, respectively, with 9 fetuses had complicated infection of PCV2+PRRSV or ADV or both. And 8 (3.6%) for SIV, 3 (1.3%) for PPV and 1 (0.4%) for JEV were positive as minor viral infection. The results suggest that PCV2, PRRSV, ADV is apparently the most important viral infectious agents associated with fetal infection leading to abortion or stillbirth in Korea. SIV, PPV and JEV might have a minor impact on reproductive disease.
