This study aimed to develop a regulation system for off-label drug use to secure the safe use of marketed drugs. We searched governmental documents for national and global regulating systems of off-label drug uses and a body of academic literature to explore current regulating trends. We included European Union, United Kingdom, United States of America, Australia and Japan, and critically reviewed the regulation of off-label drug use in four issues, which were a regulatory structure, safety control before and after off-label use, and information management. The findings of the present investigation called for several measures in off-label drug uses: enhancing prescribers' self-regulation, providing up-to-date information to prescribers for evidence-based practice and to patients for their informed consent, making evidence with scientific rigor, building an official registering process for off-label use in good quality and extending the role of pharmaceutical industry in pharmacovigilance. At last, we proposed a new system so as to regulate and evaluate off-label drug uses both at national and institutional level. In the new system, we suggested a clear-cut definition for clinical evidence that applicants would submit. We newly introduced an official 'Off-Label Drug Use Report' to evaluate the safety and clinical efficacy of a given off-label drug use. In addition, we developed an algorism of the regulation of off-label drug use within an institution to help set up the culture of evidence-based practices in off-label drug uses.
In this study, a cross-damped low vibration controller was developed to reduce vibration in ultra-precision dual stage driven by master/slave principle. In master-slave structure, the master stage leads the driving motion and the slave stage follows it so as to maintain a constant gap between two stages. In this structure, a small perturbation of master stage makes big oscillations in slave servoing stage, so a low damped master stage composed of voice coil motor makes a long vibration in settling area after driving motion profile. In this research, an effective feedback damping algorithm for increase the damping characteristics of the dual stage was developed. The designed velocity damping algorithm improves the system stability and reduces the settling time of the whole system. Simulation and experimental results show that the developed algorithm reduces settling time and improves the regulating performance.
Temperate mycobacteriophage L1 encodes an unusual repressor (CI) for regulating its lytic-lysogenic switching and, in contrast to the repressors of most temperate phages, it binds to multiple asymmetric operator DNAs. Here, ions like $Na^+$, $Cl^-$, and $acetate^-$ ions were demonstrated to facilitate the optimal binding of CI to cognate operator DNA, whereas $K^+$, $Li^+$, ${NH_4}^+$, $Mg^{2+}$, $carbonate^{2-}$, and $citrate^{3-}$ ions significantly affected its operator binding activity. Of these ions, $Mg^{2+}$ unfolded CI most severely at room temperature and, compared to $Mg^{2+}$, $Na^+$ provided improved thermal stability to CI. Furthermore, the intrinsic tryptophan fluorescence of CI was changed notably upon replacing $Na^+$ with $Mg^{2+}$ and these opposing effects of $Mg^{2+}$ and $Na^+$ were also noticed in their actions on the C-terminal fragment (CTD) of CI. Taken together, $Na^+$ appeared to be more appropriate than $Mg^{2+}$ for maintaining the biologically active conformation of CI needed for its optimal binding to operator DNA.
Various ZnO nanostructures were synthesized and ZnO nanostructure-based self-assembled transistors were fabricated. Compared to spindle and flower like nanostructure, the ZnO nanorod (NR) structure showed much stronger gate controllability, and greatly enhanced device performance, demonstrating that this structural variation leads to significant differences of the nanostructure network-based device performance. Also, patterned dry transfer-printing technique that can generate monolayer-like percolating networks of ZnO NRs has been developed. The method exploits the contact area difference between NR-NR and NR-substrate, rather than elaborate tailoring of surface chemistry or energetic. The devices prepared by the transferring method exhibited on/off current ratio, and mobility of ${\sim}2.7{\times}10^4$ and ${\sim}1.03\;cm^2/V{\cdot}s$, respectively. Also, they exhibited showing lower off-current and stronger gate controllability due to defined-channel between electrodes and monolayer-like network channel configuration. With multilayer stacks of nanostructures on stamp, the monolayer-like printing can be repeated many times, possibly on large area substrate, due to self-regulating printing characteristics. The method may enable high-performance macroelectronics with materials that have high aspect ratio.
This study proposes a new smart base isolation system that employs Magneto-Rheological Elastomers (MREs), a class of smart materials whose elastic modulus or stiffness can be varied depending on the magnitude of an applied magnetic field. It also evaluates the dynamic performance of the MRE-based isolation system in reducing vibrations in structures subject to various seismic excitations. As controllable stiffness elements, MREs can increase the dynamic control bandwidth of the isolation system, improving its vibration reduction capability. To study the effectiveness of the MRE-based isolation system, this paper compares its dynamic performance in reducing vibration responses of a base-isolated single-story structure (i.e., 2DOF) with that of a conventional base-isolation system. Moreover, two control algorithms (linear quadratic regulator (LQR)-based control and state-switched control) are considered for regulating the stiffness of MREs. The simulation results show that the MRE-based isolation system outperformed the conventional system in suppressing the maximum base drift, acceleration, and displacement of the structure.
This paper proposes a novel high step-up non-isolated DC-DC converter, suitable for regulating dc bus in various inherent low voltage micro sources especially for photovoltaic (PV) and fuel cell sources. This novel high voltage Non-isolated Boost DC-DC converter topology is best replacement, where high voltage conversion ratio is required without the transformer and also need continuous input current. Since the proposed topology utilizes the stack-based structure, the voltage gain, and the efficiency are higher than other conventional non-isolated converters. Switches in this topology is easier to control since its control signal is grounding reference. Also, there is no need of extra gate driver and extra power supply for driver circuit, which reduces the cost and size of system. In order to show the feasibility and practicality of the proposed topology principle operation, steady state analysis and simulation result is presented and analyzed in detail. To verify the performance of proposed converter and theoretical analysis 360W laboratory prototype is implemented.
Bats influence overall ecosystem health by regulating species diversity and being a major source of zoonotic viruses. Hence, there is a need to elucidate their migration, population structure, and phylogenetic relationship. The complete mitochondrial genome is widely used for studying the genome-level characteristics and phylogenetic relationship of various animals due to its high mutation rate, simple structure, and maternal inheritance. In this study, we determined the complete mitogenome sequence of the bird-like noctule (Nyctalus aviator) by Illumina next-generation sequencing. The sequences obtained were used to reconstruct a phylogenic tree of Vespertilionidae to elucidate the phylogenetic relationship among its members. The mitogenome of N. aviator is 16,863-bp long with a typical vertebrate gene arrangement, consisting of 13 protein-coding genes (PCGs), 22 transfer RNA genes, 2 ribosomal RNA genes, and 1 putative control region. Overall, the nucleotide composition is as follows: 32.3% A, 24.2% C, 14.3% G, and 29.2% T, with a slight AT bias (61.5%). The base composition of the 13 PCGs is as follows: 30.3% A, 13.4% G, 31.0% T, and 25.2% C. The phylogenetic analysis, based on 13 concatenated PCG sequences, infers that N. aviator is closely related to N. noctula with a high bootstrap value (100%).
This paper reports the development of an oil flushing system combined with a microbubble generator. Oil flushing plays a crucial role in regulating the lubricant's performance during the lubricant replacement process. Moreover, harmful contaminants, such as sludge, wear particles, and rust, from piping systems or lubrication system can be removed by oil flushing. Oil flushing aims to increase the system's efficiency using a dedicated flushing oil, increasing of the supply pressure and generating a vortex. In addition, it helps the mechanical system or equipment achieve peak performance and reduces the potential for premature failure. However, the contaminant-removal applications of existing oil flushing system are limited. In this research, we aim to improve the performance of oil flushing system by incorporating a microbubble generator, which uses the venture effect to generate microbubbles and mixes them with lubricant. The microbubbles in the blended lubricant remove contaminants from the lubrication system more effectively. Structural mechanics and fluid dynamics are analyzed through fluid-structure interaction (FSI) analysis, and the numerical analysis results are used for the designing the system. The magnitude of the maximum stress is investigated based on the pressure results obtained by the CFD analysis; through the CFD analysis, the mixing ratio of air (bubble) and lubricant is evaluated using the volume of fluid (VOF) model according to the working conditions.

TJ은 다른 피부 장벽기능에도 영향을 미칠 수 있으며 화학, 미생물 또는 면역학적 피부장벽에게 영향을 받는다. 스트레스로 인한 피부 장벽 손상에 관한 보고는 있지만 사람피부에서 코티솔이 TJ을 조절한다는 보고는 없다. 스트레스 호르몬 코티솔이 TJ을 조절하는 기능을 확인하기 위해 각질형성세포에 코티솔을 처리하였다. 코티솔은 TJ 구성 성분의 유전자 발현과 구조를 조절하여 피부 장벽 기능을 손상시켰다. 또한 코티솔은 인공피부 모델에서 과립층 형성을 억제하였다. 이러한 실험결과를 통해 스트레스 호르몬 코티솔이 TJ를 조절함으로써 피부 장벽 기능에 손상을 일으키는 것을 확인할 수 있었다.
In this paper the modeling and control of a twin roll strip caster is investigated. Mathematical models for the strip casting process are obtained by analyzing five critical areas such that the molten steel level in the pool, solidification process, roll separating force and torque, roll dynamics including hydraulic actuators, and roll drive system. A two-level control strategy is proposed. At lower level, three local subsystems are independently feedback-controlled by suitable local controllers which perform well to the behaviors of each subsystem. They are a variable structure control of the molten steel level in the pool, an adaptive predictive control of the roll gap which is directly related to the strip thickness, and an $H^{\infty}$ control of the roll drive system. At higher level, all reference signals to the lower level subsystems are generated by an optimal controller in the perspective of regulating the strip thickness and roll separating force.
