상황버섯 균사체를 이용한 발효주의 추출물이 염증관련 단백질의 발현과 세포내 활성산소 생성에 미치는 영향 (Effect of the Evaporating Extract from Liquor of Fermentation Using Mycelium of Phellinus linteus on the Expression of Inflammatory Proteins and the Generation of Reactive Oxygen Species)

  • Phellinus linteus is known as a medicinal mushroom, which has the pharmaceutical activity on tumors and inflammatory diseases in traditional Oriental medicine. However, despite extensive pharmacological studies on P. linteus, the liquor of fermentation using mycelium of P. linteus(LFMP) has not been investigated. In the present study, it was examined the effect of the evaporating extract from LFMP(E-LFMP) on the expression of inflammatory proteins and the generation of reactive oxygen species in human hepatocarcinoma HepG2 cells. E-LFMP inhibited acetaldehyde-induced morphological change in HepG2 cells. Also, E-LFMP inhibits expression of inflammatory proteins including cyclooxygenase(COX)-1 and COX-2 through suppressing nuclear translocation of nuclear factor κB(NF-κB) and degradation of inhibitory κBα(IκBα). In addition, E-LFMP inhibits generation of reactive oxygen species(ROS) by hydrogen peroxide(H₂O₂) in HepG2 cells. These results suggest that LFMP has the pharmaceutical, especially anti-inflammatory, activity similar to P. linteus mushroom.

Dust particles-induced intracellular Ca2+ signaling and reactive oxygen species in lung fibroblast cell line MRC5

  • Epidemiologic interest in particulate matter (PM) is growing particularly because of its impact of respiratory health. It has been elucidated that PM evoked inflammatory signal in pulmonary epithelia. However, it has not been established $Ca^{2+}$ signaling mechanisms involved in acute PM-derived signaling in pulmonary fibroblasts. In the present study, we explored dust particles PM modulated intracellular $Ca^{2+}$ signaling and sought to provide a therapeutic strategy by antagonizing PM-induced intracellular $Ca^{2+}$ signaling in human lung fibroblasts MRC5 cells. We demonstrated that PM10, less than $10{\mu}m$, induced intracellular $Ca^{2+}$ signaling, which was mediated by extracellular $Ca^{2+}$. The PM10-mediated intracellular $Ca^{2+}$ signaling was attenuated by antioxidants, phospholipase blockers, polyADPR polymerase 1 inhibitor, and transient receptor potential melastatin 2 (TRPM2) inhibitors. In addition, PM-mediated increases in reactive oxygen species were attenuated by TRPM2 blockers, clotrimazole (CLZ) and N-(p-amylcinnamoyl) anthranilic acid (ACA). Our results showed that PM10 enhanced reactive oxygen species signal by measuring DCF fluorescence and the DCF signal attenuated by both TRPM2 blockers CLZ and ACA. Here, we suggest functional inhibition of TRPM2 channels as a potential therapeutic strategy for modulation of dust particle-mediated signaling and oxidative stress accompanying lung diseases.

The Impacts of Obesity on Psychological Well-being: A Cross-sectional Study about Depressive Mood and Quality of Life

  • Objectives : The aim of this study was to assess whether individuals who visit clinics to ask medical help for obesity treatment depict comparable levels of depression, body dissatisfaction, eating psychopathology and lower quality of life. Methods : This is a cross sectional study with 534 females who sought treatment for their obesity or overweight being recruited in seven clinical units in Seoul, Korea. The patients group was divided into two groups. The group 1 consisted of the patients with BMI >$25\;kg/m^2$. The women who showed BMI ${\leq}\;25\;kg/m^2$ among patients recruited for this study were classified as the group 2. The control group (group 3) was composed of 398 healthy females who have never tried to lose weight. Results : We found that group 1 had higher frequency of more than moderate level of depression than group 2 and group3 did. Both patients groups showed greater eating disordered attitudes and behaviors regardless of obese condition than the control group. Group1 showed relatively lower level of quality of life than group2 and group3 in terms of the quality of life related to physical well-being. In addition, the control group reported higher quality of life in psychological health than both patients groups did. Conclusions : In conclusion, it is necessary for clinicians to make a careful evaluation of depressive tendency and eating disorders when obese women seek for medical help. The combination of medical treatment and psychological approach for obese women would result in higher quality of life.

Ginsenoside Rg1 ameliorates chronic intermittent hypoxia-induced vascular endothelial dysfunction by suppressing the formation of mitochondrial reactive oxygen species through the calpain-1 pathway

  • Background: As the major pathophysiological feature of obstructive sleep apnea (OSA), chronic intermittent hypoxia (CIH) is vital for the occurrence of cardiovascular complications. The activation of calpain-1 mediates the production of endothelial reactive oxygen species (ROS) and impairs nitric oxide (NO) bioavailability, resulting in vascular endothelial dysfunction (VED). Ginsenoside Rg1 is thought to against endothelial cell dysfunction, but the potential mechanism of CIH-induced VED remains unclear. Methods: C57BL/6 mice and human coronary artery endothelial cells (HCAECs) were exposed to CIH following knockout or overexpression of calpain-1. The effect of ginsenoside Rg1 on VED, oxidative stress, mitochondrial dysfunction, and the expression levels of calpain-1, PP2A and p-eNOS were detected both in vivo and in vitro. Results: CIH promoted VED, oxidative stress and mitochondrial dysfunction accompanied by enhanced levels of calpain-1 and PP2A and reduced levels of p-eNOS in mice and cellular levels. Ginsenoside Rg1, calpain-1 knockout, OKA, NAC and TEMPOL treatment protected against CIH-induced VED, oxidative stress and mitochondrial dysfunction, which is likely concomitant with the downregulated protein expression of calpain-1 and PP2A and the upregulation of p-eNOS in mice and cellular levels. Calpain-1 overexpression increased the expression of PP2A, reduced the level of p-eNOS, and accelerated the occurrence and development of VED, oxidative stress and mitochondrial dysfunction in HCAECs exposed to CIH. Moreover, scavengers of O2·-, H2O2, complex I or mitoKATP abolished CIH-induced impairment in endothelial-dependent relaxation. Conclusion: Ginsenoside Rg1 may alleviate CIH-induced vascular endothelial dysfunction by suppressing the formation of mitochondrial reactive oxygen species through the calpain-1 pathway.

  • The history of studies in biology regarding reactive oxygen species (ROS) is approximately 40 years. During the initial 30 years, it appeared that these studies were mainly focused on the toxicity of ROS. However, recent studies have identified another action regarding oxidative signaling, other than toxicity of ROS. Basically, it is suggested that ROS are reactive, and degenerate to biomolecules such as DNA and proteins, leading to deterioration of cellular functions as an oxidative stress. On the other hand, recent studies have shown that ROS act as oxidative signaling in cells, resulting in various gene expressions. Recently ROS emerged as critical signaling molecules in cardiovascular research. Several studies over the past decade have shown that physiological effects of vasoactive factors are mediated by these reactive species and, conversely, that altered redox mechanisms are implicated in the occurrence of metabolic and cardiovascular diseases ROS is a collective term often used by scientist to include not only the oxygen radicals($O2^{-{\cdot}},\;{^{\cdot}}OH$), but also some non-radical derivatives of oxygen. These include hydrogen peroxide, hypochlorous acid (HOCl) and ozone (O3). The superoxide anion ($O2^{-{\cdot}}$) is formed by the univalent reduction of triplet-state molecular oxygen ($^3O_2$). Superoxide dismutase (SOD)s convert superoxide enzymically into hydrogen peroxide. In biological tissues superoxide can also be converted nonenzymically into the nonradical species hydrogen peroxide and singlet oxygen ($^1O_2$). In the presence of reduced transition metals (e.g., ferrous or cuprous ions), hydrogen peroxide can be converted into the highly reactive hydroxyl radical (${^{\cdot}}OH$). Alternatively, hydrogen peroxide may be converted into water by the enzymes catalase or glutathione peroxidase. In the glutathione peroxidase reaction glutathione is oxidized to glutathione disulfide, which can be converted back to glutathione by glutathione reductase in an NADPH-consuming process.

The Effect of UV-A and Reactive Oxygen Species on Glycosylation and Fragmentation of Calf Skin Collagen

  • Non-enzymatic glycosylation and fragmentation of collagen molecule were investigated by irradiating Ultraviolet A(UV-A) with or without scavengers of reactive oxygen species (ROS) in the presence of glucose. Non-enzymatic glycosylation was increased by UV-A at high concentration of glucose. It was reduced in the presence of the scavengers of superoxide radical and singlet oxygen, but not reduced in the presence of hydroxy radical scavenger. Fragmentation of collagen was increased by UV-A, but it was decreased in the presence of all ROS scavengers tested. Superoxide radical and singlet oxygen produced by autoxidation of glucose without UV-A may encounter the initial phase of glycosylation. Data presented here suggest that UV-A affects only on the fragmentation process, but all ROS except hydroxy radical act on both processes. It appears that hydroxy radical does not act on the glycosylation process.

Detection of Oxygen Species Generated from Ag2Se-Graphene Heterojunction Photocatalysts with Excellent Visible Light Driven Photocatalytic Performance

  • Reactive oxygen species (ROS) can be produced by interactions between sunlight and light-absorbing substances in natural water environments and can completely destroy various organic pollutants in waste water. In this study, we used graphene oxide modified $Ag_2Se$ nanoparticles to enhance photochemically generated oxygen (PGO) species activity. Surface area and pore volumes of the $Ag_2Se-graphene$ ($Ag_2Se-G$) samples showed catastrophic decrease due to deposition of $Ag_2Se$. The generation of reactive oxygen species was detected through the oxidation reaction of DPCI to DPCO. The photocurrent density and the PGO effect increase in the case of the use of modified graphene. The PGO effect of the graphene modified with $Ag_2Se$ composites increased significantly due to a synergetic effect between graphene and the $Ag_2Se$ nanoparticles. The photocatalytic activity of sample was evaluated by measuring the degradation of organic pollutants such as methylene blue (MB) and industrial dyes such as Texbrite BA-L (TBA) under visible light.

Effects of Streptozotocin, Bisphenol A and Diethylstilbestrol on Production of Reactive Oxygen Species and Lipid Peroxidation in the Boar Sperm

  • Streptozotocin (STZ), bisphenol A (BPA), and diethylstilbestrol (DES) are known as endocrine disruptors, occurs oxidative stress in animal cells. Generally, oxidative stress induces reactive oxygen species (ROS) and lipid peroxidation of sperm and lead to decreased viability and fertility in pigs. Therefore, we investigated the influence of STZ, BPA and DES on ROS production and lipid peroxidation on boar sperm. Collected sperm were incubated with semen extender containing $10{\mu}M\;STZ$, $10{\mu}M\;BPA$ and $20{\mu}M\;DES$ for 3, 6 and 9 hours. Intracellular ROS and lipid peroxidation of sperm were analyzed by 2', 7'-dichlorofluorescein diacetate and malondialdehyde methods. The results show that, intracellular ROS was not significantly different among the all treatments, but lipid peroxidation was significantly increased in STZ group at 3 hour after incubation with boar sperm (P<0.05). These results suggest that STZ stimulates lipid peroxidation more than ROS production and may exert a negative effect on sperm fertility.

  • Purpose: Interleukin (IL)-8 is one of pro-inflammatory cytokines. Reactive oxygen species (ROS) are reduced metabolites of $O_2$. Aggregatibacter actinomycetemcomitans is one of representative periodontopathogens. To investigate the role of A. actinomycetemcomitans in IL-8 expression of periodontal ligament (PDL) cells, we estimated the production of IL-8 and ROS in A. actinomycetemcomitans treated PDL cells. Methods: The IL-8 production was determined by enzyme-linked immunosorbent assay. The ROS production was estimated using H2DCFDA and FACS. Results: A. actinomycetemcomitans increased the production of IL-8 and ROS at 10, 100, and 500 multiplicity of infection. N-acetylcysteine, an antioxidant of ROS, down-regulated the production of IL-8 induced by A. actinomycetemcomitans. Conclusions: These results suggest that A. actinomycetemcomitans induces IL-8 production and ROS may act as a mediator in this process.

누에의 에탄올 추출물이 체내 활성산소 생성에 미치는 영향 (Effect of Ethanolic Extract of Silkworm on Reactive Oxygen Species Formation in vivo)

  • The purpose of this study is to investigate the effects of silkworm extracts (SWE) on reactive oxygen species formation in mice (C57BL/6). Mice were administrated intraperitoneally with SWE (20 mg/kg/day) for 14 days. All animals were sacrificed 24 hours after the last SWE treatment and then extracted the blood and brain tissue in mouse. The researcher measured several parameters related to reactive oxygen species formation, malondialdehyde (MDA) and hydrogen peroxide ($H_2O_2$) contents in serum, whole brain, cerebral cortex and cerebellum. The results showed that MDA content of pre-SWE treatment was decreased significantly in serum, mitochondrial and cytosolic fraction of whole brain and cerebellum (P<0.01). The $H_2O_2$ content of pre-SWE treatment was decreased significantly in mitochondrial fraction of whole brain, cerebral cortex and cerebellum (P<0.01). These results suggest that SWE plays an important role for inhibition of oxidative damage of cells as well as antioxidant effect, aging delay and cells protected from irradiation.
