Effect of crude ginseng saponin and raw ginseng juice on the growth of ginseng root rot organisms, Fusarium solani and Erwinia carotovora (인삼 조 Saponin과 조즙액이 인삼근부병균 Fusarium solani와 Erwinia carotovora의 생육에 미치는 영향)

  • Effect of crude saponin and raw ginseng juice on root rot pathogens such as Fusarium solani and Erwinia carotovora for there growth or spore germination was investigated. Macroconidial germination of F. solani was decreased as the incrasee of the crude saponin concentration. especially, percentage of the germination was remarkably reduced when the concentration was more than 500ppm. The spore production of F. solani was also reduced as the increase of the crude saponin concentration and this phenomenon was particularly profound on a solid medium. Mycelial growth was decreased when the crude saponin was added, while the effect of the concentration was not apparently significant. The higher concentration of raw ginseng juice is, the more the growth of F. solani. Growth of E. carotovora was enhanced by crude ginseng saponin and raw ginseng juice. The crude ginseng saponin stimulated the growth of E. carotovora as the increase of the concentration, while more growth of the bacteria obtained at $1\%$ raw ginseng juice added.

Effects of Red Ginseng-Ejung-tang and White Ginseng-Ejung-tang Water Extract on Hydrogen Peroxide Production in RAW 264.7 Cells (백삼(白蔘)과 홍삼(紅蔘)이 포함된 이중탕(理中湯)의 마우스 대식세포 내 hydrogen peroxide 생성에 미치는 영향)

  • The purpose of this study is to investigate whether the intracellular hydrogen peroxide productions of mouse macrophage RAW 264.7 are modulated by Red Ginseng-Ejung-tang water extract (ER) and White Ginseng-Ejung-tang water extract (EG). Red Ginseng-Ejung-tang were composed of Red Ginseng, Atractylodes rhizome white, Zingiberis Rhizoma Siccus, and Glycyrrhizae Radix. White Ginseng-Ejung-tang were composed of White Ginseng, Atractylodes rhizome white, Zingiberis Rhizoma Siccus, and Glycyrrhizae Radix. The intracellular hydrogen peroxide productions were measured by dihydrorhodamine 123 assay with spectrofluorometer (excitation 485 nm; emission 535 nm). For 4, 20, 24, 44, 48, 68, and 72 h incubation, ER significantly increased hydrogen peroxide productions of RAW 264.7 at the concentration of 25, 50, 100, and $200{\mu}g/mL$ (P <0.05). EG for 4, 20, 24, 44, and 48 h incubation significantly increased hydrogen peroxide productions of RAW 264.7 at the concentration of 25, 50, 100, and $200{\mu}g/mL$ (P <0.05). For 68 and 72 h incubation, EG at the concentration of 50, 100, and $200{\mu}g/mL$ significantly increased hydrogen peroxide productions in RAW 264.7 (P <0.05). These results suggest that ER and EG have the immune-enhancing properties related with their increasing effects on the intracellular hydrogen peroxide production of macrophage.

Optimal Analytical Conditions for Panax Ginseng Ginsenosides using HPLC and Ginsenosides Content Analysis of Red Ginseng Products and their Raw Materials (HPLC를 이용한 인삼 진세노사이드의 최적 분석 조건 및 홍삼 제품과 원료삼의 진세노사이드 함량 분석)

  • The analytical conditions of ginsenosides were optimized using high performance liquid chromatography (HPLC). The optimal analysis conditions were set up from the experiments using different gradient conditions, and the ginsenosides contents of red ginseng products and their raw materials were analysed. Red ginseng contained 0.29% Rg1, 0.82% Rb1, 0.38% Rc, 0.32% Rb2, 0.11% Rd, showing the highest ginsenoside contents. Among the red ginseng products, red ginseng extract showed the highest content. The ginsenoside contents were varied according to the raw material type and product type. Re and Rb1 contents were the highest in most raw materials and products.

Anti-inflammatory Activity of Solvent Fractions from Ginseng Berry Extract in LPS-Induced RAW264.7 Cells (인삼열매추출물의 용매분획물이 LPS로 유도된 RAW264.7 세포에 대한 항염활성)

  • Anti-inflammatory activity of the extracts of ginseng berry (GBE) was investigated through the evaluation of its inhibitory effect on the production of inflammatory meditator, nitric oxide(NO), tumor necrocis factor-alpha (TNF-${\alpha}$), interleukin-6 (IL-6) in LPS-induced RAW264.7 macrophage cells. GBE was fractionated using n-hexane, chloroform, ethylacetate, buthanol and $H_2O$, sequentially. RAW264.7 cells were induced $100ng/m{\ell}$ of lipopolysaccharide (LPS) and treated with 0, 1.6, 8, 40 and $200{\mu}g/m{\ell}$ of GBE fractions. LPS-induced NO production on all of GBE fractions was inhibited with increasing added concentration of GBE fractions. Chloroform fraction of GBE was the most effective in inhibiting LPS-induced TNF-${\alpha}$ production. Hexane, chloroform and $H_2O$ fractions of GBE exhibit strong inhibition LPS-induced IL-6 production. Especially, $H_2O$ fractions of GBE was the most effective in inhibiting LPD-induced IL-6 production without significant cytotoxicity in RAW264.7 cells, and reduced the activation of mitogen-activated protein kinases (MAPK) and IkB phosphorylation. These results indicate that $H_2O$ fractions of GBE exhibits strong anti-inflammatory effects by inhibition of NF-kB by inhibition of p-38 on MAPK and IkB phosphorylation.

Wild Ginseng Exerts Anti-inflammatory Effects via $NF-{\kappa}B$ inactivation in RAW 264.7 Cells (장뇌산삼의 $NF-{\kappa}B$ 억제를 통한 RAW 264.7 세포에서의 항염증 효과)

  • Inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 are important inflammatory mediators that have been implicated in pathogenesis of inflammation and certain types of human cancers. The present study was designed in order to determine whether Wild ginseng (Panax ginseng C. A. Mayer) could modulate $I{\kappa}B$-kinase (IKK), iNOS and COX-2 gene expression and its immune responses in RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS, 1 ${\mu}/m{\ell}$). Wild ginseng extract dose-dependantly (*0.5 - 2 ${\mu}/m{\ell}$) decreased the LPS-induced IKK, iNOS and COX-2 mRNA expression and its immune responses. Moreover, it inhibited unclear factor (NF)-${\kappa}B$ immune response by LPS. These data be likely to indicate that Wild ginseng may acts as inflammatory regulator and may be possible to develope a useful agent for inflammatory diseases.

The Inflammation-modulatory Effects of Ginseng Saponin and Polysaccharide on Activated RAW264.7 Cell-line (인삼 사포닌과 다당류 혼합물의 활성화된 RAW264.7 세포주에 대한 염증조절 효과)

  • It is well known that the numbers and functions of immune-associated cells are increased by saponins and polysaccharides in ginseng. In this study, the mixture of polysaccharide and saponin (MPS) from Panax ginseng is applied to LPS- activated RAW 264.7 cells. The production of NO and the gene expression of IL-6 and TNF-$\alpha$ are decreased in LPSactivated RAW 264.7 cells and the expression of arginase II and PD-1L genes is decreased in LPS-untreated macrophages. Therefore, the mixture of saponin and polysaccharide from Panax ginseng could be used in order to regulate immune responses.

  • This study aims to analyze the differences of ginseng product consumers and segment Korean fresh ginseng and red ginseng root markets based on attributes for the purchase. As a result of analyzing survey data, the red ginseng root consumers had different aspects from fresh ginseng consumers. According to the result of cluster analysis, the fresh ginseng consumers were subdivided into three segments (safety-oriented consumption cluster, label centered consumption cluster, and high involvement consumption cluster), while the red ginseng root consumers were subdivided into four segments (convenience-oriented consumption cluster, high involvement consumption cluster, raw material's safety-oriented cluster, and raw material's information importance cluster). ANOVA and Crosstab were conducted to investigate characteristics of each cluster.

  • We conducted this study to investigate anti-inflammatory possibilities of applying cosmetic material about extracts from red ginseng. For this we carried out biological active evaluation about anti-inflammatory by using extracts of red ginseng. In order to evaluate the anti-inflammatory effects of the samples in macrophages (RAW 264.7 cells), MTT assay was used to evaluate the toxicity of red ginseng extracts and the inhibitory activity of nitric oxide production and the expression levels of inflammation-related proteins and genes. The inhibitory activity of nitric oxide in the LPS-induced RAW 264.7 cells was 71.2% at 25 ㎍/ml concentration and western blot analysis showed that the expression of iNOS and COX-2 protein decreased in a concentration-dependent manner. These results suggest that extracts from red ginseng may have value as the potential cosmetic materials.

Effects of Extraction Temperature and Time on Saponin Content and Quality in Raw Ginseng (Panax ginseng) Water Extract (수삼의 추출 온도 및 시간이 물 추출액의 사포닌 함량 및 품질에 미치는 영향)

  • In this study, raw ginseng water extract solutions were analyzed to set up the functional saponin content and quality optimization condition. The highest saponin content among the total raw ginseng water extracts was $74.6\;mg/100\;m{\ell}$ which was extracted at $75^{\circ}C$ for 24 hours. In addition, the saponin content decreased according to the increased extraction temperature and time. The highest total content of $Rb_2$ and Re was $19.9\;mg/100\;m{\ell}$ at $75^{\circ}C$ for 12 hours which decreased according to the increased extracted temperature and time. The highest prosapogenin ($Rg_2\;+\;Rg_3\;+\;Rh_1$) content among the total raw ginseng water extracts was $28.6\;mg/100\;m{\ell}$ which was extracted at $85^{\circ}C$ for 36 hours. The reducing sugar content, sweetness and turbidity were increased according to the increased extraction temperature and time. But pH were decreased according to the increased extracted time.

Phenolic Acid Composition and Antioxidative Activity of Red Ginseng Prepared by High Temperature and High Pressure Process (고온고압 처리 홍삼의 페놀산 조성과 항산화 활성)

  • This study was conducted to develop HTHP ginseng (high temperature and high pressure ginseng) with improved antioxidative activity and phenolic acid composition by high temperature and high pressure process. The HTHP ginseng extract was analyzed for the total phenol content, DPPH radical scavenging activity and phenolic acid composition. The total phenol content was increased in HTHP ginseng (14.76 mg/g) compared to raw ginseng (3.59 mg/g) and red ginseng (3.93 mg/g). DPPH radical scavenging activities of HTHP ginseng, raw ginseng and red ginseng extracts were 4.8~78.4%, 1~47.4% and 1.8~56.5% at $1{\sim}100mg/m{\ell}$ concentration. Also ABTS radical scavenging activities of HTHP ginseng, raw ginseng and red ginseng extracts were 8.9~99.8%, 3.4~96% and 1.2~96.5% at $1{\sim}100mg/m{\ell}$ concentration. In HPLC analysis, amounts of measured phenolic acid of HTHP ginseng greatly increased than raw ginseng and red ginseng, but salicylic acid was not detected in HTHP ginseng. In addition, DPPH radical scavenging activity of phenolic acid from HTHP ginseng was increased. Consequently, we believe high temperature and high pressure process is better method than existing method to increase the bioactivity of ginseng.
