Effect of Rubus coreanus Miquel on the Specific Immune Response in Mice (복분자가 생쥐의 특이적 면역반응에 미치는 영향)

  • The specific immune response of unripened fruits and ripened fruits of Rubus coreanus Miquel was examined in BALB/C mice. The 70% ethyl alcohol extracts (20 or 100 mg/kg) of unripened fruits (RCE-I) and of ripened fruits (RCE-II) were administered p.o. once a day for 7 days. RCE-I and RCE-II decreased the viability of thymocytes, but increased the viability of splenocytes. Also, RCE-I enhanced the population of helper T and cytotoxic T cells in thymocytes and RCE-II enhanced the population of helper T cells. Furthermore, RCE-I and RCE-II decreased the population of B220/sup +/ cells and Thy1/sup +/ cells and helper T cells in splenocytes. In a general way, the immunosuppressive action of RCE-I was more potent than those of RCE-II. These results suggest that RCE-I and RCE-II have a regulative action of immune response via decrease the viability of thymocytes and increase the viability of splenocytes.

Rumex crispus Suppresses Type I Hypersensitive Immune Response (소리쟁이(Rumex crispus) 추출물의 제1형 알레르기 반응 억제 효과)

  • Rumex crispus is known to have anticancer, antioxidant, antibacterial, and bone loss inhibitory activities. Mast cells are critical immune cells that induce a type 1 IgE-mediated allergic reaction. However, there are no reports of inhibitory effects of Rumex crispus on mast cells and allergic reactions. In this study, we performed some experiments to investigate whether Rumex crispus ethanol extract(RCE) has any inhibitory effect on antigen-induced type I allergic response in vitro and in vivo. RCE inhibited degranulation of IgE-mediated mast cells(IC50, ~57 ㎍/ml) and cytokine production such as TNF-α and IL-4 in a dose-dependent manner. In vivo, RCE significantly inhibited passive cutaneous anaphylaxis(PCA)(ED50, ~198 mg/kg) in mice. Furthermore, RCE inhibited degranulation of MCs in ear tissue of mice with PCA. Mechanism studies showed that RCE inhibited the activation of Syk and Syk-dependent pathway such as LAT, PLC-γ, Akt, and MAP Kinase. Our results demonstrate for the first time that RCE inhibits type I hypersensitive response by suppressing the activity of Syk in mast cells, thereby reducing degranulation and cytokine production. Taken together, RCE could be used as a novel therapeutic material to suppress allergic diseases.

Effect of Unripened fruits and Ripened fruits of Rubus coreanus Miquel on Murine Peritoneal Macrophages (복분자 미숙과 및 성숙과가 생쥐의 복강 Macrophages에 미치는 영향)

  • The purpose of this research was to investigate the effects of unripened fruits and ripened fruits of Rubus coreanus Miquel on murine peritoneal macrophages. The 70% ethyl alcohol extracts (20 or 100 mg/kg) of unripened fruits (RCE-I) and of ripened fruits (RCE-II) were administered p.o. once a day for 7 days to mice. RCE-I and RCE-II decreased the phagocytic activity of murine peritoneal macrophages and the production of nitric oxide. Also, RCE-I and RCE-II increased the production of tumor necrosis factor- a from peritoneal macrophages. In general, the immuno-suppressive action of RCE-I on macrophages was more potent than those of RCE-II. These results suggest that the fruits of Rubus coreanus Miquel regulates the non-specific immune response via decrease of phagocytic activity and increase of production of tumor necrosis factor- a from murine peritoneal macrophages.

Can distinction between the renal cortex and outer medulla on ultrasonography predict estimated glomerular filtration rate in canine chronic kidney diseases?

  • Background: Quantitative evaluation of renal cortical echogenicity (RCE) has been tried and developed in human and veterinary medicine. Objectives: The objective of this study was to propose a method for evaluating RCE quantitatively and intuitively, and to determine associations between ultrasonographic renal structural distinction and estimated glomerular filtration rate (eGFR) in canine chronic kidney disease (CKD). Methods: Data were collected on 63 dogs, including 27 with normal kidney function and 36 CKD patients. Symmetric dimethylarginine and creatinine concentrations were measured for calculating eGFR. RCE was evaluated as 3 grades on ultrasonography images according to the distinction between the renal cortex and outer medulla. The RCE grade of each kidney was measured. Results: There was a significant difference in eGFR between the group normal and CKD (p < 0.001). As mean of RCE grades (the mean values of each right and left kidney's RCE grade) increases, the proportion of group CKD among the patients in each grade increases (p < 0.001). Also, severity of RCE (classified as "high" if any right or left kidney evaluated as RCE grade 3, "low" otherwise) and eGFR is good indicator for predicting group CKD (p < 0.001). Conclusions: The degree of distinction between the renal cortex and the outer medulla is closely related to renal function including eGFR and the RCE grade defined in this study can be used as a method of objectively evaluating RCE.

Antioxidant activity and hepatic lipids improvement effects of Rubus coreanus in high-fat diet-fed rats (복분자(Rubus coreanus) 추출물의 라디칼 소거활성 및 고지방 식이에 대한 흰쥐의 간 지질 개선 효과)

  • This study was investigated the effects on the radical scavenging activities and hepatic lipid levels in rats fed a high-fat diet (HFD) by the 70% ethanolic extract from Rubus coreanus. The Rubus coreanus extract (RCE) was administered through the gastrointestinal tract at 150 mg/kg BW/day for 5 weeks (HFD+RCE). Total phenol content was 157.93 mg gallic acid/g in the RCE. Scavenging activities to DPPH, ABTS, an d hydroxyl radicals by RCE significantly increased in a does-dependent manner of $50{\sim}500{\mu}g/mL$ concentration. DPPH and ABTS radical scavenging activities were over the 70% in the $500{\mu}g/mL$ concentration. During the five weeks study, the food efficiency ratio was no significant differences between the HFD and HFD+RCE. Liver weight of HFD+RCE (3.75 g/100 g BW) was significantly decreased compared to those of the HFD. Hepatic triglyceride and total cholesterol levels of HFD+RCE were significantly lower compared to those of the HFD. The hepatic TBARS contents in the HFD+RCE was significantly decreased to the HFD, radical scavenging activity was markedly increased in HFD+RCE as normal group. In histological observation of liver tissues, the hepatocytes of HFD+RCE appeared the reduced aspects on the size and numbers of lipid droplets in cytoplasm compared to the HFD. Our results suggest that RCE could have antioxidant activities and hepatic hypolipidemic effects by its phenolic compounds.

  • We describe methods for depositing high temperature superconducting films on textured metal tapes by reactive co-evaporation (RCE). We discuss how RCE can be used to deposit on moving tape in a continuous fashion in a Garching-style process. Results are presented on films deposited by RCE at Los Alamos on IBAD-MgO textured tapes. The performance achieved, attaining over 500A/cm-width in self-field at 75.5 K, is competitive with the best results obtained by other processes for coated conductors. Tape production throughput is critical for the economics of the process and high deposition rates achieved in RCE are attractive for this. We present a detailed cost analysis model for HTS deposition using an RCE Garching process. The results indicate that HTS deposition can cost $<$5/kA{\cdot}m$ in a scaled up manufacturing environment.

Anti-osteoporotic Effects of Unripe Fructus of Rubus coreanus Miquel in Osteoblastic and Osteoclastic Cells

  • Osteoporosis is a progressive bone disease characterized by low bone mass which is caused by disturbance in the balance between the activities of osteoblasts and osteoclasts. Postmenopausal osteoporosis is one of the most common disorders in women after menopause, which is linked to an estrogen deficiency and characterized by an excessive loss of trabecular bone. Rubus coreanus has been used for their various pharmacological properties in Asia as a traditional medicine. To investigate the effect of unripe fruits of R. coreanus 30% ethanol extract (RCE) on osteoblast-like cells (MG63) differentiation, we examined the effects of RCE on in vitro osteoblastic differentiation markers, alkaline phosphatase (ALP) activity and receptor activator of nuclear factor ${\kappa}$-B ligand (RANKL) and osteoprotegerin (OPG) expression. The high concentration (50 and $100{\mu}g/mL$) of RCE markedly increased ALP activity, whereas decreased the RANKL/OPG. We also investigated the effect of RCE on M-CSF plus RANKL-induced differentiation of pre-osteoclast cells (RAW 264.7). RCE treatment remarkably inhibited M-CSF/RANKL-induced formation of osteoclast-like multinuclear cells from RAW 264.7 cells. Moreover, the inhibitory effect of RCE was reduced by selective estrogen receptor-${\alpha}$ antagonist. Our research suggests that suggested that unripe fruits of R. coreanus may act beneficial effects on bone mass by regulating both osteoblast and osteoclast.

Ricinus communis extract inhibits the adipocyte differentiation through activating the Wnt/β-catenin signaling pathway

  • Ricinus communis, belongs to the family Euphorbiaceae, has been known as medicinal plants for treatment of inflammation, tumors, antidiabetic, hepatoprotective and laxative. Compared to many pharmacological studies, the effect of R. communis extract on regulating adipogenesis as therapeutic drug for treating obesity has not been reported. R. communis extract (RCE) was investigated to determine its effects on the adipogenesis by monitoring the status of $Wnt/{\beta}-catenin$ signaling and factors involving the differentiation of adipocytes. The differentiation of 3T3-L1 cells monitored by Oil Red O staining was inhibited in concentration dependent manner by RCE. The luciferase activity of HEK 293-TOP cells containing pTOPFlash with Tcf4 response element-luciferase gene was increased approximately 2-folds by the treatment of RCE at concentrations of $100{\mu}g/mL$ compared to the control. Activation of the $Wnt/{\beta}-catenin$ pathway by RCE was further confirmed by immunocytochemical analysis which shows an increment of nuclear localization of ${\beta}-catenin$. In addition, safety of RCE was verified through performing neural stem cell morphology assay. Among the identified flavonoids in RCE, isoquercitrin was the most abundant. Therefore, these results indicate that the adipocyte differentiation was significantly reduced by isoquercitrin in R. communis. In this study, RCE suppresses the adipogenesis of 3T3-L1 cells via the activation of $Wnt/{\beta}-catenin$ signaling.

Extract of Rubus coreanus Fruits Increases Expression and Activity of Endothelial Nitric Oxide Synthase in the Human Umbilical Vein Endothelial Cells (복분자 추출물에 의한 내피세포 NO 합성효소의 활성과 발현 증가)

  • This study aimed to investigate the effects of water extract of Rubus coreanus (RCE) on the expression and activity of endothelial nitric oxide synthase (eNOS), as well as its signal transduction pathways in human umbilical vein endothelial cells (HUVECs). The specific inhibitors of NOS show RCE treatment increases NO production in HUVECs due to the up-regulation of eNOS rather than iNOS. The real-time expression level of eNOS mRNA was also increased upon RCE treatment in HUVECs. While a PKC-specific inhibitor, RO-317549, did not alter RCE-induced NO production in HUVECs, tamoxifen (estrogen receptor-specific inhibitor), PD98059 (ERK-specific inhibitor) and LY-294002 (PI3K/Akt-specific inhibitor) did have suppressive effects. Increased NO production by RCE seems to result from a higher level of active eNOS (pSer1177). Specifically, inhibition of ERK not only decreased the level of active eNOS, but also increased the inactive form of the enzyme (pThr495) in HUVECs. This study suggests that RCE treatment increases NO production in HUVECs due to the increased expression and activity of eNOS. It is also shown that RCE-induced eNOS activation occurs partly through the binding of RCE to the estrogen receptor, along with ERK and PI3K/Akt-dependent signal transduction pathways. In addition, the regulatory binding proteins of eNOS including Hsp90 and caveolin-1 were related to these effects of RCE on eNOS activity in HUVECs.

Design of Gas Classifier Based On Artificial Neural Network (인공신경망 기반 가스 분류기의 설계)

  • In this paper, we propose the gas classifier based on restricted column energy neural network (RCE-NN) and present its hardware implementation results for real-time learning and classification. Since RCE-NN has a flexible network architecture with real-time learning process, it is suitable for gas classification applications. The proposed gas classifier showed 99.2% classification accuracy for the UCI gas dataset and was implemented with 26,702 logic elements with Intel-Altera cyclone IV FPGA. In addition, it was verified with FPGA test system at an operating frequency of 63MHz.
