Phylogenetic Relationships Using ITS2 Sequence and RAPD-PCR Data from Four Species of Korean Pseudo-nitzschia (Bacillariophyceae) (ITS2 부위의 염기서열 및 RAPC-PCR에 의한 Pseudo-nitzschia 4종의 유연관계)

  • A portion of ribosomal internal transcribed spacer (ITS) 2 was sequenced from the samples of Pseudo nitzschia (P. deticatissima, P. multiseries, P. pungens and P. subfraudulenta) to investigate the genetic characteristics by measuring tile magnitude of genetic diversity and the degree of similarity coefficient using random amplified polymorphic DNAs (RAPD)-PCR patterns. The phylogenetic trees inferred from the genetic distance analyses showed the placement of P. delicatissima formed a quite long distance from p. P. multiseries, P. pungens, and even P. subfraudulenta. The phylogenetic tree from RAPD patterns showed that P. multiseries and P. pungens had dissimilarity coefficient of 0.31, while P. delicatissima and three species of Pseudo-nitzschia had that of 0.81. It is likely thought that the genetic position of P. delicatissima formed far from P. multiseries, P. punges, and P. subfraudulenta. These results imply that ITS2 region is expected to support a useful molecular characters for recognizing at the species level and for even discriminating P. multiseries from P. pungens. RAPD method also will be used to differentiate the species of Pseudo-nitzschia in a short time.

Potentially toxic Pseudo-nitzschia species in Tongyeong coastal waters, Korea (통영 연안의 잠재독성 Pseudo-nitzschia 출현종)

  • Several species of the genus Pseudo-nitzschia produce the neurotoxin domoic acid (DA) known to be responsible for amnesic shellfish poisoning. In spite of the potentially toxic effects on marine ecosystem, even the representative Pseudo-nitzschia species occurring in Korean coastal waters have not been clearly reported. Plankton samples from several outer coastal sites of Tongyeong were collected fortnightly from May to November 2008 and the presence of diatoms of the genus Pseudo-nitzschia was examined using light and scanning electron microscopy. Thirteen species were observed, including P. americana, P. brasiliana, P. caciantha, P. calliantha, P. cuspidata, P. delicatissima, P. micropora, P. multiseries, P. multistriata, P. pseudodelicatissima, P. pungens, P. subfraudulenta, and P. subpacifica. The number of Pseudo-nitzschia species observed were only four in May, which was minimum during this survey, and then gradually increased attaining maximum, twelve, in September. After September it began to decrease again and got to five in November. Of these, P. americana, P. brasiliana, P. caciantha, P. calliantha, P. micropora, and P, pseudodelicatissima are new records for the Korean coastal waters and P. calliantha, P. cuspidata, P. delicatissima, P multiseries, P. multistriata, and P. pungens have been reported as DA producers around the world, but the potential toxicity of these species was not ascertained in Tongyeong area.

The Morphological Characteristics of Pseudo-nitzschia cf. brasiliana Observed in Korea Strait, 2010 (2010년 국내 남해 해역에 출현한 Pseudo-nitzschia cf. brasiliana의 형태 특성)

  • Several species of the genus Pseudo-nitzschia produce the neurotoxin domoic acid (DA) known to be responsible for amnesic shellfish poisoning. Recently off the coast of Geomundo and in Exclusive Economic Zone (EEZ) of Korea Strait, P. cf. brasiliana predominated in summer, 2010. As P. cf. brasiliana was new to Korea, we describe the morphological characteristics of the species. It was observed that cells were linear to lanceolate in valve and girdle view, $51{\sim}76{\mu}m$ long and $1.9{\sim}2.6{\mu}m$ wide. In the central part a large interspace was absent. The number of fibulae and striae in $10{\mu}m$ were 30~37 and 31~37. Each stria contained 2~3 rows of poroids, with 10 to 14 poroids in $1{\mu}m$ length. These morphological features of P. cf. brasiliana were found to be similar with the P. brasiliana, except with the fact that P. cf. brasiliana carred more numbers of fibulae/striae in $10{\mu}m$ and poroids in $1{\mu}m$. Based on our observation, we suggest that P. cf. brasiliana is a newly identified Pseudo-nitzschia species or a variety of P. brasiliana.

Potentially Toxic Diatoms Pseudo-nitzschia fraudulenta and P. calliantha from Russian Waters of East/Japan Sea and Sea of Okhotsk

  • Potentially toxic diatoms Pseudo-nitzschia calliantha and P. fraudulenta were found in bottle samples of phytoplankton collected in Amurskii Bay (East/Japan Sea) and in the coastal waters of Sakhalin Island (East/Japan Sea and Sea of Okhotsk) in different seasons during 2002-2006. The mass development of these species occurred in October and November 2002 at water temperatures of $6-16^{\circ}C$ and salinities of 28.8-33.5 PSU. The highest concentrations of P. calliantha and P. fraudulenta were about $2{\times}10^5\;cells\;L^{-1}$ and $1.5{\times}10^5\;cells\;L^{-1}$, respectively. P. fraudulenta was found for the first time in the Russian waters of the East/Japan Sea. Morphological descriptions of these species based on observation with light and electron microscopy and information on their ecology are presented. Data on the geographical distribution of these species are supplemented.

The Rapid Differentiation of Toxic Alexandrium and Pseudo-nitzschia Species Using Fluorescent Lectin Probes

  • Since toxic Alexandrium catenella and non-toxic A. fraterculus are morphologically similar, they are difficult to discriminate under the light microscope. However, a novel technology, such as fluorescein isothiocyanate (FITC)-conjugated lectin probes enables easy and rapid differentiation. Toxic A. catenella bound seven different lectins, whereas the non-toxic A. fratercuzus did not bind Arachis hypogaea (PNA) lectin. In addition, Pseudo-nitrschia species in this study were also difficult to identify to species level with light microscope techniques, but it was possible to classify them using fluorescent lectins. Pseudo-nitzschia multistriata, P. subfraudulenta and P. pungens bound Canavalia ensiformis (ConA), whereas P. subpaclfica did not, and P. pungens also bound Ricinus communis (RCA). These results imply that lectin could be used as a critical tool in the differentiation of P. multistriata, P. subfraudulenta and P. pungens. However, P. subpacifica was not differentiated by the lectins tested. Therefore, it isconcluded that lectin probes are useful for discriminating toxic A. catenella from non-toxic A. fraterculus, and for the identification of some Pseudo-nitzschia species. In addition, this method has a great potential to speed and detection between non-toxic and toxic harmful algal blooms (HABs) in Korean biotoxin monitoring systems.

Morphological and Molecular Identification of Pseudo-nitzschia sp. Strain G3 Isolated from Northern Coast of Vietnam Based on ITS Region Sequences

  • For the first time in Vietnam, morphological and molecular studies of a species belonging to Bacillariophyceae collected in Northern coast of Vietnam are presented. Observations with microscope showed that this species belong to genus: Pseudo-nitzschia and seem like P. pungens. Sequence data from the partial 18S small subunit ribosomal RNA gene (18S rDNA) and the internal transcribed spacer 1 - 5.8S - internal transcribed 2 have been used to determine clearly and generate a phylogenetic framework of the obtained sequences to previously reported sequences in GenBank. These results allowed us to highlight described species of Bacillariophyceae in Northern coast of Vietnam. Furthermore, accumulation of molecular study would be helpful for the identification of scientific name of harmful algal species and further taxonomic studies in Vietnam.

Seasonal Changes in Phytoplankton Composition in Jinhae Bay, 2011 (2011년 진해만 식물플랑크톤 군집의 계절적 변화)

  • Geographic distribution and species composition of phytoplankton were investigated in Jinhae Bay for 12 months. Total 158 species were found including 95 bacillariophyceae, 58 dinophyceae, 1 euglenoids, and 4 dictyochaceae. The total cell numbers of phytoplakton ranged from $4.0{\times}10^3{\sim}9.0{\times}10^7\;cells{\cdot}L^{-1}$. Highest and lowest amounts of cells were found in July (Chl.a 13.19 $mg{\cdot}m^{-3}$) and March(Chl.a 3.44 $mg{\cdot}m^{-3}$), respectively. Leptocylindrus danicus and Pseudo-nitzschia spp. were dominant in spring, and Dactliosolen fragilissimus and Pseudo-nitzschia spp. were dominant in summer. Chaetoceros spp. and Skeletonema spp. were domonant in autumn and winter. The amounts of nitrogen and phosphorus were also measured during the survey. Higher ratio of nitrogen (DIN : DIP = 28 : 1) was found in the east Jinhae Bay compared to the ratio of the west Jnhae Bay that showed 14 : 1 of N : P ratio. The total amounts of nitrogen, phosphorus and Chl.a were also higher in the east Jinhae Bay compared to the west area. The geographic differences between the east and west of Jinhae Bay were also discussed along with phytoplankton compostion.

  • Pseudo-nitzschia 속은 현재까지 알려진 유독 적조생물 중 규조류로서 유일하게 독성을 가지는 종이며, 생활사가 짧기 때문에 영양염 공급이 충분하거나 수온 등 환경조건만 적당하면 언제든지 수 일 이내로 대규모 적조를 형성할 수 있다. 지금까지 분리된 종은 P. pungens, P. multiseries, P. cuspidata, P. delicatissima, P. subfraudulenta, P. subpacifica, P. multistriata로 그 중에서 P. multiseries가 기억상실성 패독인 domoic acid (DA)를 생성하는 종이지만, 형태적으로 P. pungens와 매우 유사하여 전자현미경의 형태로서 구분되기 때문에 본 종을 신속 탐색하기 위하여 전세계적으로 많은 연구를 하고 있는 실정이다. 특히 P. pungens와 P. multiseries는 대체적으로 동시다발적으로 4월과 10월에 일시적으로 발생하고 있다. The Correlation between Environmental Factors and Phytoplankton Communities in Spring and Summer Stratified Water-column at Jinhae Bay, Korea (진해만에서 춘계와 하계 성층기간 동안 환경요인과 식물플랑크톤 군집구조의 관계)

  • We surveyed 23 sites of Jinhae Bay in spring and summer 2010 in order to study the correlation between the variation of environmental factors, including salinity, temperature, and nutrients and the characteristics of phytoplankton community structures in summer stratification. Phytoplankton biomass was high in the surface water in summer; however, it was very low in the bottom water. The results showed a negative correlation between chlorophyll a (chl. a) and nutrients (silicate, nitrate, nitrite, ammonium, and phosphate) or nutrients ratio in summer; however, there was mostly a positive correlation between chl. a and these nutrients in spring. This inconsistent correlation between spring and summer was attributed to the phytoplankton community, because a diverse phytoplankton community has different nutrient uptake abilities. In addition, the results of CCA (canonical correspondence analysis) showed a negative correlation between phosphate and dominant species, including Chaetoceros spp., Skeletonema costatum-like spp., and Pseudo-nitzschia delicatissima in summer, but a strong positive correlation between DIN (dissolved inorganic nitrogen) and the dominant species, including Cryptomonas spp. and Pseudo-nitzschia multistriata in spring. Consistently, the dominant algal species in summer showed a relatively smaller size cells compared with those in spring, suggested that it may have related with the low nutrient levels at surface layer due to strong stratified water column of summer.

Inter-annual Variation of Phytoplankton Community Structure in Aquacultural Areas of Tongyeong, SE Coastal Waters of Korea (통영 양식장 해역의 식물플랑크톤 군집의 연간변동)

  • Phytoplankton community structure is one of the indicators that can explain the enviromnental characteristics of coastal waters. In this study, phytoplankton community structure and water quality of aquaculture area were investigated for understanding regional enviromnental characteristics. Thirty stations in aquaculture areas of Tongyeong, southeast coast of Korea, were investigated monthly from January to December, 2009. Phytoplankton community, meteorologic dada and enviromnent factors including temperature, salinity, transparency, nutrients and chlorophyll a were also examined. Chaetoceros spp. and unidentified small flagellates were dominant species in all the year round. Pseudo-nitzschia spp., Dictyocha spp., and Nitzschia longissima were dominant in June to October being summer season, and Skeletonema costatum, Thalassiosira spp., Eucamphia zodiacus, Akashiwo sanguinea, Gymnodinium spp. and Asterionella japonicus appeared as dominant species in the rest of months. Dinoflagellate blooms occurred 3 times in near Hansan Bay and around Saryang-do, and the highest chlorophyll a was found in Hansan Bay. Species diversity of phytoplankton was lower in Hansan and Womnum Bay, and diatom was more abundant than dinoflagellates in Mireuk-do waters. These results showed that phytoplankton community varied by the seasonal and geographical characteristics, and recent increase of water temperature and heavy rain may affect on phytoplankton community structure.
