본 실험은 사람의 치주인대세포에서 에스트로겐과 growth hormone(GH)이 상호 어떠한 작용을 하는지를 규명하는 것이 목적이다. 교정치료를 받고자 내원한 환자중에서 건강한 20대 여자환자들의 상하악 제1소구치를 발거하여 치근의 중간1/3부위에서 긁어모은 치주인대 조직을 배양하여 치주인대세포를 얻었다. 사람의 치주인대세포의 분열증식에 대한 $17{\beta}$-estradiol과 hGH의 효과를 평가하고, 치주인대세포에 $17{\beta}$-estradiol을 전처리한 후 치주인대세포의 분열증식에 미치는 hGH의 효과 변화를 평가하였으며, 치주인대세포에 $17{\beta}$-estradiol을 전처리 하였을 경우 치주인대세포에 서 hGH receptor치 발현 양상의 변화를 평가하여 다음과 같은 결론을 얻었다. 1. $17{\beta}$-estradiol이나 humangrowth hormone의 단독처리는 사람의 치주인대세포의 분열종식에 큰 영향을 주지 않는다. 2. $17{\beta}$-estradiol $10^{-12}M$로 전처리한 후 hGH를 투여한 경우 hGH의 농도에 관계없이 사람의 치주인대세포의 분열증식을 증가시킨다. 3. 사람의 치주인대세포에는 hGH receptor가 없으나, $17{\beta}$-estradiol $10^{-12}M$로 5시간 이상 처리하면 hGH receptor가 발현된다. 4. hGH이 사람의 치주인대세포의 분열증식에 미치는 효과는 hGH receptor의 발현과 관련이 있으며 $17{\beta}$-estradiol의 전처리가 치주인대세포에서의 hGH receptor의 발현에 기여함으로써 hGH이 치주인대세포에 작용할 수 있도륵 해준다.비, 상악의 대구치간 너비, 구개정 높이가 상관관계가 있었다.에서 치 아이동은 정상상태보다 촉진될 가능성이 있음을 시사한다.따라 평균마찰력은 유의하게 증가하였다. 6. 브라켓간 거리의 변화에 대한 마찰력의 차이는 교정선의 재질에 따라 차이가 나며, 스테인레스 스틸 교정선은 브라켓간 거리가 감소할 때 마찰력이 유의하게 변화하지 않았으나, NiTi 교정선의 경우는 유의하게 증가하였다. 7. 브라켓내의 교정선의 이동속도에 따라 마찰력은 유의하게 변화하지 않았다. 이상의 결과로 볼 때 교정 치료동안 적정 교정력을 유지하기 위해 자가결찰브라켓, 스테인레스 스틸 교정선과 탄성모듈결찰법을 사용하는 것이 유리하며, 치료시기에 따라서 요구하는 마찰력이 다르므로 상황에 따라 재료를 선택하는 것이 중요할 것으로 사료된다.$, B군에서 $4.3\%$ 감소하였다. 7. 수술로 인한 얼굴폭의 변화는 무시할만한 것으로 판단되나 수술후 얼굴의 길이가 짧아져 상대적으로 얼굴의 폭이 넓어보일 수 있으므로 수술시 이에 대한 충분한 고려 및 환자에 대한 설명이 있어야 한다.로는 4-6세군 ($27.5\%$), 6-8세군 ($19.6\%$), 2-4세군 ($13.7\%$)이 $60\%$이상을 차지하여 초등학교 취학 전에 구순의 반흔을 제거하려 함을 알 수 있었다. 8. 비변형 교정수술시기로는 0-2세군 ($7.1\%$), 2-4세군 ($14.3\%$), 4-6세군 ($21.4\%$), 6-8세군 ($14.3\%$)으로 초등학교 취학이전이 $57.1\%$로서 최근의 조기 치료경향을 반영하는 것으로 보인다. 9. 인두피판술은 평균 6세에 시행되었으며, 수술 시행 시기별의 차이를 보이지 않고 고른 분포를
The sex steroid hormone estradiol-$17{\beta}(E_2)$ mediate their biological effects on development, differentiation and maintenance of reproductive tract and other target tissue through gene regulation by nuclear steroid receptors. Although the importance of $E_2$ in many physiological process has been reported, but little is known about the effects of $E_2$ on primary cultured chicken hepatocyte. therefore, in the present study, we have examined the effect of $E_2$ on cell proliferation and it's related signal cascades. $E_2$ increase $[^3H]$-thymidine incorporation in time-(${\leq}8hr$) and dose-($10^{-10}M$)dependent manner and treatment of $E_2$ increased the phosphorylation of p44/43 MAPKs(p44/42 mitogen-activated protein kinase) and JNK(c-Jun N-terminal kinase) in a time dependent manner. In addition, PD98059(p44/42 blocker, $10^{-5}M$), SP600125(JNK blocker, $10^{-6}M$) blocked the estrogen-induced increase in $[^3H]$-thymidine incorporation. In conclusion, $E_2$ stimulates the proliferation of primary cultured chicken hepatocytes and this action is mediated by p44/42 MAPKs and JNK signal transduction pathway.

Purpose: The aims of this study were as follow: 1) to de scribe the expression status of estrogen receptor-${\alpha}$ and -${\beta}$ mRNAs in five gastric carcinoma cell lines; 2) to evaluate in vitro the effects of $17{\beta}$-estradiol and estrogen receptor antagonists on the proliferation of the cell lines. Materials and Methods: Detection of estrogen receptor-${\alpha}$ and estrogen receptor-${\beta}$ mRNA in five human gastric cancer cell lines (AGS, KATO III, MKN28, MKN45 and MKN74) was made by the reverse transcription-polymerase chain reaction system. To evaluate the effect of $17{\beta}$-estradiol and estrogen receptor antagonists on the proliferation of gastric cancer cell line, the cell lines which expressed both es trogen receptors were chosen and treated with $17{\beta}$-estradiol and estrogen receptor antagonists (methyl-piperidino-pyrazole and pyrazolo [1,5-a] pyrimidine). Cell proliferation was assessed with the methylthiazol tetrazolium test. Results: Estrogen receptor-${\alpha}$ and estrogen receptor-${\beta}$ mRNAs were expressed in three (KATO III, MKN28 and MKN45) and all of the five gastric cancer cell lines, respectively. At higher concentrations, $17{\beta}$-estradiol inhibited cell growth of MKN28, MKN45 and KATO III cell lines. Neither estrogen receptor-${\alpha}$ nor estrogen receptor-${\beta}$ antagonist blocked the anti-proliferative effect of $17{\beta}$-estradiol. Conclusions: Our results indicate that estrogen receptor-${\beta}$ mRNAs are preferentially expressed in gastric cancers and also imply that hormone therapy rather than estrogen receptor blockers may be a useful strategy for the treatment of estrogen receptor-${\beta}$ positive gastric cancer. Its therapeutic significance in gastric cancer are, however, limited until more evidence of the roles of estrogen receptors in the gastric cancer are accumulated.
The mechanisms of $estradiol-17{\beta}$ regulating growth of both normal and neoplastic cells are not clear until now. In studies using various estrogen-dependent breast cell lines, it is recently known that estrogen controls the cell growth by regulating the expression of growth factors and/or their receptors. In the present study, we investigated the effects of $estradiol-17{\beta}$on cell growth and IGF-I binding sites using primary cultured renal proximal tubule cells. We have obtained results as follows : $Estradiol-17{\beta}(10^{-9})$ has stimulatory effects in cell growth. Cotreatment of $estradiol-17{\beta}(10^{-9}M)$ and $IGF-I(5{\times}10^{-8}M)$ significantly increased the growth of primary rabbit renal proximal tubule cells compared to that of $estradiol-17{\beta}$ or IGF-I alone treated cells. In binding studies, we found that the binding of $^{125}IGF-I$ on cell membranes was incubation time- and temperature-dependent. Incubation at $37^{\circ}C$ results in higher binding of $^{125}IGF-I$ than that of $23^{\circ}C$ or $4^{\circ}C$. Maximum binding was observed at $37^{\circ}C$ between 30 and 60 minutes. The binding of $^{125}IGF-I$ to both control and $estradiol-17{\beta}-treated$ cells was inhibited by unlabelled $IGF-I(10^{-8}{\sim}10^{-12}M)$ in a concentration-dependent manner. However, EGF did not compete for $^{125}IGF-I$ binding at $10^{-8}{\sim}10^{-12}M$. IGF-I binding to the membranes from both control and $estradiol-17{\beta}-treated$ cells was also analyzed. We found that $estradiol-17{\beta}-treated$ cells exhibited higher binding activity for IGF-I. When $estradiol-17{\beta}$ or tamoxifen alone, or $estradiol-17{\beta}$ and tamoxifen cotreated cells were compared, the binding ratio of $^{125}I-IGF-I$ of $estradiol-17{\beta}-treated$ cell was significantly increased but was similar to control in both $estradiol-17{\beta}$ and tamoxifen cotreated cell. These results suggest that $estradiol-17{\beta}$ in part controls cell proliferation by regulating the expression of IGF-I receptors in primary rabbit renal proximal tubule cells.

This study was designed to determine the effects of the insulin-like growth factor (IGF-1) and estradiol $17-{\beta}$ on the in vitro proliferation of stromal vascular cell from Hanwoo omental, subcutaneous, intermuscular and intramuscular adipose tissues. Cells were cultured in M199+20% newborn calf serum and the proliferation of cells was measured by direct microscopic cell counting and change of genomic DNA amount. Cell numbers increased slightly over the first 72 hour of culture and then increased greatly, regardless of adipose tissue depots. In IGF-1 treatment, the number of omental preadipocytes maintained highest level from the beginning to the 20th day of culture. However, in estradiol-$17{\beta}$ treatment, those tended to be lower than the control from the beginning of culture and significantly lower at the 24th day. When IGF-1 was added to subcutaneous preadipocytes, the numbers of cells were higher from 11th day than those from other treatments, although there was no statistical significance. For intermuscular preadipocytes treated with IGF-1, its numbers were significantly (p<0.05) higher at 11th day, and in the other days it showed a similar tendency to those of the subcutaneous tissue. In this experiment, preadipocytes were taken from 24 month old fully matured steers and the highest proliferation rate was shown in intramuscular tissue followed by those of subcutaneous preadipocytes. Addition of $5{\mu}M$ estradiol-$17{\beta}$ to the growth medium failed to promote the replication of Hanwoo preadipocytes, as indicated by direct cell counts and total genomic DNA content. As the culture period proceeded, the amounts of DNA were increased, but the patterns of increment were not consistent with the results of cell numbers.
The purpose of this study is to evaluate the expression ofIGF-I, considered as the mediator of action of estrogen, and IGF-IA and IGF-IB, alternative slicing form of IGF-I, using $17{\beta}-estradiol$ in MC3T3-E1 cells. We observed the effect on type I collagen and osteopontin gene expression and DNA synthetic activity of MC3T3-E1 cells, added by estrogen, IGF-I and combination and the interactionon proliferation and differentiation of MC3T3-E1 cells. The results were as follows :RT-PCR experiment for observing timedependantIGF-I gene expression patternshowed IGF-IA and IB gene expression in both of control and test group. In these IGF-IA gene expression was appeared predominantly. In control, IGF-I geneexpression level was maintained until 24hr and then decreased gradually. In testgroup, IGF-I gene expression level increased as time goes by. Experiment measuring DNA synthetic activity, as it is added by $17{\beta}-estradiol$, IGF-I and combination, showed that first day , there was the tendency of more increase of synthetic activity in all test group than control but no statical significance(P>0.05), and third day, there was more increase of DNA synthetic activity in $17{\beta}-estradiol$ group and combination group and it was statically significant. (P<0.005) Experiment for observing type I collagen gene expression pattern showed more increase of expression in $17{\beta}-estradiol$ group than control and no significant difference in IGF-I group and combination group. Experiment for observing osteopontin gene expression pattern showed no significant difference in control and test group. In conclusion, $17{\beta}-estradiol$ in MC3T3- E1 cells increased IGF-I gene and DNA synthetic activity simultaneously, therefore it appeared that IGF-I is related to the action of estrogen. Combination treatment of IGF-I and $17{\beta}-estradiol$ has effect on cell proliferation but this effect is lower than IGF-I or $17{\beta}-estradiol$ alone. However, combination treatment has not great effect on type I collagen or osteopontin gene expression thus little effect of cell differentiation.
The aim of this study was to analyze the proteome expressions of proliferating stromal vascular cells from Hanwoo omental, subcutaneous and intramuscular depots subjected to hormone deprivation and IGF-1, Estradiol-$17{\beta}$ addition. For hormone deprivation or addition studies, the cells were either grown in 10% charcoal-dextran stripped fetal bovine serum (CD-FBS) or in 10% FBS supplemented medium. Further, to analyze the effect of insulin like growth factor (IGF-1) and $17\beta$-Estradiol (E2), cells were grown in 10% CD-FBS containing IGF-1 (10 ng/ml) or E2 (10 nM). The results showed that hormone deprivation had a negative impact on proliferation among the cells from all depots without any growth difference. On comparison of proliferation levels, higher levels were observed in cells that were grown in 10% FBS than in 10% CD-FBS alone or with IGF-1/E2. Proteome expression from preadipocytes grown in hormone deprivation conditions were compared by 2D-DIGE and MALDIToF/ToF. A total of twelve different proteins were found to be differentially expressed under hormone deprivation conditions. Further, our proteomic analysis with DIGE under IGF-1 and E2 addition revealed four proteins with differential expression levels. Moreover, the results highlighted in this study offer a role for each differentially expressed protein with respect to their effect in positive or negative regulation on proliferation.
내분비 교란 물질 중에서 최근에 문제가 되고 있는 물질이 프탈레이트계통 물질로서 플라스틱의 가소제로 사용하고 있는 DEHP와 DBP에 대하여 사람유방암세포 이면서 에스트로젠 의존성을 가지고 있는 MCF-7 세포에서의 세포 증식 정도를 농도 별로 측정하여 두 물질의 에스트로젠 작용가능성에 대하여 조사하였다. 시험물질인 DEHP와 DBP의 MCF-7 세포의 최대 증식 시의 농도를 조사해 본 결과, DEHP는 $17{\beta}-Estradiol$에 비하여 100배정도 높은 농도인 $10^{-7}M$ 에서 최대 증식능력을 보였고, DBP는 10배정도 높은 $10^{-8}M$ 에서 최대 증식능력을 보였다. 최대 증식 능력을 보일 때의 양성대조물질인 $17{\beta}-Estradiol$와 증식 정도 차이를 비교하였을 때에는 DEHP는 양성대조군 대비 87.5%의 증식 정도를 나타내었고, DBP는 73.4%의 증식 정도를 나타내었다 결론적으로 DEHP와 DBP 두 물질 모두 MCF-7 세포의 증식에 영향을 주는 것으로 판단되었으며, 최대 작용농도에 있어서는 DBP>DEHP, 세포 증식 정도에 있어서는 DEHP>DBP 인 것으로 판단되어 진다.
에스트로겐은 조직세포의 유형과 이들의 생리적 상태에 따라 세포증식을 촉진 또는 억제 시킬 수 있으며, 주로 에스트로겐 수용체(ER)에 의해 매개되는 신호전달 경로를 통해 작용한다. 이 연구는 특히 유방암세포에서 ER에 대한 길항제로 잘 알려진 fulvestrant (Ful)가 CHO 세포주의 증식 및 세포사멸에 미치는 영향과 이들에 대한 $17{\beta}$-estradiol (E2)의 작용에 미치는 효과를 조사하고자 하였다. 이를 위하여 먼저 다양한 농도의 E2, Ful 및 E2 plus Ful의 처리기간에 따라 세포증식에 미치는 효과를 조사하였다. Cell proliferation 분석에서, 6-10일의 처리기간 동안 E2는 1 ${\mu}M$의 농도까지는 세포증식에 영향을 주지 않았지만, 15-40 ${\mu}M$에서는 처리기간의 증가에 따라 점진적으로 현저히 세포증식을 억제하였다. 흥미롭게도 Ful 또한 1 ${\mu}M$의 농도까지는 세포증식에 영향을 주지 않았지만, 10-40 ${\mu}M$에서는 농도 및 시간 의존적인 세포증식 억제효과를 보였다. 뿐만 아니라, Ful은 10일 동안 E2와의 혼합처리에서 E2에 의한 세포증식 억제효과를 감소시키기 보다는 오히려 증대시켰다. 따라서 Ful은 CHO 세포에서 E2의 항 증식작용에 대한 길항적 효과를 갖지 않음을 제시한다. 한편, E2 및 Ful에 의한 DNA 분절효과를 확인하기 위한 TUNEL assay에서 20 ${\mu}M$의 E2로 처리된 CHO 세포주에서는 DAPI로 염색된 거의 모든 핵에서 TUNEL 양성반응이 나타났으며, 40 ${\mu}M$ Ful 및 20 ${\mu}M$ E2 plus 40 ${\mu}M$ Ful로 처리된 CHO 세포주에서는 TUNEL 양성반응을 보였으나, E2 처리군과 비교하여 현저히 낮은 비율로 나타났다. 이러한 결과는 Ful이 CHO 세포에서 E2의 항증식작용에 대한 길항적 효과를 갖지 않으며, E2와 Ful에 의한 세포증식 억제와 DNA 분절을 통한 세포사멸 관련기전은 다른 신호전달 경로를 통해 매개됨을 시사한다.
Objective: There is increasing evidence that chronic non-bacterial prostatitis is recognized to be a local inflammatory disease, and there is substantiating evidence to support the role of the inflammatory responses in its pathogenesis, and clinical value in the evaluation of therapeutic efficacy. Prunella vulgaris has been traditionally used in treatment of inflammatory diseases, including of scrofula, goiter, and allergy diseases. In this study, we investigated the effects of Prunella vulgaris on inflammatory cytokines and cytopathological alternation in the rat model of non-bacterial prostatitis induced by castration and $17{\beta}-estradiol$ treatment. Methods: Two-month-old rats were treated with $17{\beta}-estradiol$ after castration for induction of experimental non-bacterial prostatitis, which is similar to human chronic prostatitis in histopathological profiles. Prunella vulgaris as an experimental specimen, and testosterone as a positive control, were administered orally. The prostates were evaluated by histopathological parameters including the epithelial score and epithelial-stromal ratio for glandular damage, and the expression of inflammatory cytokine genes including the interleukin $(IL)-1{\beta}$, IL-5, IL-12, and tumor necrosis factor $(TNF)-{\alpha}$. Results: While prostates of control rats revealed severe acinar gland atrophy and stromal proliferation, the rats treated with Prunella vulgaris showed a diminished range of tissue damage. Epithelial score was improved in Prunella vulgaris over that of the control (P<0.05). The epithelial-stromal ratio was lower with Prunella vulgaris when compared to that of the control (P<0.05). In the reverse transcription-polymerase chain reaction (RT-PCR) of inflammatory cytokine genes, Prunella vulgaris inhibited the expression of $IL-1{\beta}$ and $TNF-{\alpha}$ genes, while it modulated the expression of IL-5, which is an anti-inflammatory cytokine. Conclusions: These findings suggest that Prunella vulgaris may protect the glandular epithelial cells and also inhibit stromal proliferation in association with the immune modulation including the suppression of inflammatory cytokines and promotion of anti-inflammatory cytokine. From theses results, we suggest that Prunella vulgaris could be a useful remedy agent for treating chronic non-bacterial prostatitis.

① 이용자는 회원가입 신청 또는 회원정보 변경 시 실명으로 모든 사항을 사실에 근거하여 작성하여야 하며,
허위 또는 타인의 정보를 등록할 경우 일체의 권리를 주장할 수 없습니다.
② 당 사이트가 관계법령 및 개인정보 보호정책에 의거하여 그 책임을 지는 경우를 제외하고 회원에게 부여된
ID의 비밀번호 관리소홀, 부정사용에 의하여 발생하는 모든 결과에 대한 책임은 회원에게 있습니다.

제 4 장 서비스의 이용
제 12 조 (서비스 이용 시간)

원칙으로 합니다. 단, 당 사이트는 시스템 정기점검, 증설 및 교체를 위해 당 사이트가 정한 날이나 시간에
서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만
이 경우 그 내용을 공지합니다.
제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
