Serotonin (5-hydroxytroptamine, 5-HT) has been shown to affect the induction of long-term potentiation (LTP) in the cortex such as the hippocampus, the visual cortex and the prefrontal cortex. Fluoxetine, as a selective serotonin reuptake inhibitor, is used in the management of a wide variety of psychological diseases. To study the effect of fluoxetine on the induction of LTP, we recorded the field potential in layer II/III of the frontal cortex from 3-wk-old. LTP was induced in horizontal input by theta burst stimulation (TBS). TBS with two-folds intensity of the test stimulation induced LTP, which was blocked by application of D-AP5 $(50 {\mu}M)$, an NMDA receptor antagonist. Whereas bath application of 5-HT $(10 {\mu}M)$ inhibited the induction of LTP, treatment with the 5-HT depleting agent, para-chloroamphetamine $(PCA,\;10{\mu}M)$, for 2hr did not affect the induction of LTP. Bath application of fluoxetine (1, 3, and $10 {\mu}M)$ suppressed the induction of LTP in concentration-dependent manner, however, fluoxetine did not inhibit the induction of LTP in 5-HT-depleted slices. These results indicate that fluoxetine may inhibit the induction of LTP by modulating serotonergic mechanism in the rat frontal cortex.
Objectives : We aimed to identify the antidepressant effect of liver tonification acupuncture treatment (ACU (LT); KI10, LR8, LU8, LR4) and four gate acupuncture treatment (ACU (FG); LI4, LR3) and its brain neural activity in the normal and chronic restraint stress (CRS)-induced mouse model. Methods : Firstly, normal mice were given ACU (LT) or ACU (FG) and the c-Fos expressions in each brain region were analyzed to examine brain neural activity. Secondly, CRS was administered to mice for 4 weeks, then ACU (LT) or ACU (FG) was performed for 2 weeks. The depression-like behavior was evaluated using open field test (OFT) before and after acupuncture treatment. Then, the c-Fos expressions in each brain region were analyzed to examine brain neural activity. Results : In normal mice, ACU (FG) regulated brain neural activities in the hypothalamus, hippocampus, and periaqueductal gray. ACU (LT) changed more brain regions in the prefrontal cortex, insular cortex, striatum, and hippocampus, including those altered by ACU (FG). In CRS-induced model, ACU (LT) alleviated depression-like behavior more than ACU (FG). Also, brain neural activities in the motor cortex area 2 (M2), agranular ventral part and piriform of insular cortex (AIV and Pir), and cornu ammonis (CA) 1 and CA3 of hippocampus were changed by ACU (LT), and those of AIV and CA3 were also changed by ACU (FG). As in normal mice, ACU (LT) resulted in changes in more brain regions, including those altered by ACU (FG) in CRS model. M2, Pir, and CA1 were only changed by ACU (LT) in depression model, suggesting that these brain regions reflect the specific effect of ACU (LT). Conclusions : ACU (LT) relieved depression-like behavior more than ACU (FG), and this acupuncture effect was associated with modulation of brain neural activities in the motor cortex, insular cortex, and hippocampus.
Since the early 2000s, there has been a continued interest in lie detection using functional magnetic resonance imaging (fMRI) in neuroscience and forensic sciences, as well as in newly emerging fields including neuroethics and neurolaw. Related fMRI studies have revealed converging evidence that brain regions including the prefrontal cortex, anterior cingulate cortex, parietal cortex, and anterior insula are associated with deceptive behavior. However, fMRI-based lie detection has thus far not been generally accepted as evidence in court, as methodological shortcomings, generalizability issues, and ethical and legal concerns are yet to be resolved. In the present review, we aim to illustrate these achievements and limitations of fMRI-based lie detection.
Purpose: Some of the adolescent drinks more sugar-sweetened beverages. However, there is little evidence on the effect of eating behavior on emotional state and neurochemical changes under stress, especially on the levels of typical inhibitory neurotransmitters and gamma-aminobutyric acid. This article demonstrates that sucrose or saccharin drink reduces stress-related behavior responses and GABAergic deficits in adolescent rats. Methods: We randomly assigned 7-weeks-old Sprague-Dawley male rats to three groups: control group (Control), restraint stress only group (Stress), and restraint stress with unrestricted access to saccharin solution (Saccharin) and sucrose solution (Sucrose) as a positive control. We evaluated both anxious and depressive moods using an open field test and forced swim test, respectively. Using western blot analyses, the expression of a GABA-synthesizing enzyme, glutamate decarboxylase-67 (GAD67) and GABAergic markers, including calbindin and parvalbumin was assessed in the prefrontal cortex and the amygdala. Results: We found that both the drinks alleviated anxiety and depressive moods, induced significant attenuation in GAD67 level, and reduced calbindin level under stress in the prefrontal cortex and the amygdala. Conclusion: The results provide an understanding of the effect of sucrose or saccharin drink on stress-related responses. We propose the consumption of sweet drinks as a plausible strategy to alleviate stress-related alterations in adolescents.
목적 : 본 연구에서는 감정조절에 대한 신경학적 메커니즘을 이해하고, 감정조절과 감각처리의 연관성에 대해 알아보고자 한다. 본론 : 신경학적으로 감정조절은 주로 둘레계의 구조물에 의해 처리되고 특히, 편도의 바닥-바깥쪽 그룹이 주요한 역할을 한다. 바닥-바깥쪽 그룹은 감각의 중계소인 시상과 직 간접적으로 연결되어 감각 정보에 대한 처리를 함께 한다. 이 감각 정보는 눈-안쪽 앞 이마엽과 연결되어 감각처리 및 감정과 관련된 복잡한 행동을 하게 한다. 시상, 편도, 눈-안쪽 앞 이마엽의 연결 회로로 인해 부적절한 감각처리는 감정조절과 행동에 어려움을 발생시킬 수 있다. 자폐스펙트럼장애와 주의력결핍과잉행동장애 아동에서 편도 및 앞 이마엽의 비정상적인 처리에 대한 신경학적 연구가 보고되고 있다. 결론 : 자폐스펙트럼장애와 주의력결핍과잉행동장애 아동은 신경계의 상태가 다르고 이는 감각처리와 감정조절, 행동에서 다양한 양상을 나타낸다. 치료사는 아동의 행동에 영향을 미치는 감각처리와 감정조절에 대한 신경학적 메커니즘을 이해하고, 치료적 적용을 할 필요가 있다.
Objective : This study is accomplished in order to investigate the effect of banggibongnyeongtang on the immunohistological change in LPS-induced depression rats to confirm the histological result of the previous behavioral and biochemical effect. Methods : LPS 5 ㎍ was injected to lateral ventricle and experimental groups were administered BBT intraperitoneally. The concentration of 5-HT in the Medial Prefrontal Cortex, Striatum, Hippocampus, Amygdala was measured by ELISA. IL-1β, TNF-α mRNA and BDNF mRNA expression in the hippocampus was examined by RT-PCR. Result : BBT enhanced 5-HT concentration at all part of brain but no significantly difference at medial prefrontal cortex and striatum. LPS+BBT400 group increased 5-HT concentration significantly than LPS group at hippocampus and amygdala (p<0.05). BBT decreased IL-1β mRNA expression dose dependently but only with significantly decrease in LPS+BBT400 group than LPS group's in Hippocampus (p<0.05). But BBT did not decrease TNF-α mRNA expression significantly in Hippocampus. BBT increased the expression of BDNF mRNA at hippocampus and LPS+BBT400 group significantly increased comparing with LPS group does (p<0.05). Conclusion : It is postulated that the anti-depressant effect of BBT can be validated through the anti-inflammatory effect, 5-HT concentration increase, and the neuro-protective effect mediated by BDNF by combining the results of the previous report about the behavioral and biochemical effect.
Objective: Physical activity restriction in sows may lead to behavioral abnormalities and affective disorders. However, the psychophysiological state of these sows is still unclear. As miRNAs can be used as effective markers of psychopathy, the present study aimed to assess the difference in microRNA expression between the long-term activity restricted sows and activity free sows, thus contributing to the understanding of abnormal sow behavior. Methods: Four dry sows (sixth parity, Large${\times}$White genetic line) were selected from activity restricted crates (RC) or activity free pens (FP) separately. microRNAs in the ventromedial prefrontal cortex (vMPFC) and serum were examined using real-time polymerase chain reaction, and the correlation between the miRNAs expressed in the vMPFC and serum was evaluated. Results: miR-134 (1.11 vs 0.84) and miR-1202 (1.09 vs 0.85) levels were higher in the vMPFC of the RC sows than in the FP sows (p<0.01). Furthermore, miR-132 (1.27 vs 1.08) and miR-335 (1.03 vs 0.84) levels were also higher in the RC sows than in FP sows (p<0.05); however, miR-135a, miR-135b, miR-16, and miR-124 levels were not different (p>0.05). The relative expression of serum miR-1202 was higher in the RC sows than in the FP sows (1.04 vs 0.54) (p<0.05), and there was a strong correlation (R = 0.757, p<0.05) between vMPFC and Serum levels of miR-1202. However, no significant difference was observed in miR-16 levels in the serum of the RC sows and FP sows (p>0.05). Conclusion: This pilot study demonstrates that long-term activity restriction in sows likely results in autism or other complex psychopathies with depression-like behaviors. These observations may provide new insights for future studies on abnormal behavior in sows and contribute to research on human psychopathy.
When situated in green landscapes, water bodies play a crucial role in positively influencing mood and mental health, yet research on the cognitive mechanisms underlying these therapeutic effects is lacking. This study is intended to examine differences in brain function among adult males exposed to forest landscapes with or without water bodies. The wooded landscapes included views of a waterfall, a valley, and a forest without water. The control group was exposed to a local urban landscape. Twelve adult males participated in a field experiment in which prefrontal cortex (PFC) activity was measured using near-infrared spectroscopy (NIRS). In the experiment, participants engaged in low-intensity walking in three forested areas with similar vegetation and climatic conditions. Participants showed significant differences in left PFC activity depending on whether they were in the three forested landscapes or in the control landscape (P < 0.01). An analysis of variance (ANOVA) confirmed that significantly lower left PFC activity was recorded in the wooded landscape containing a water view . Notably, the lowest PFC values recorded in the landscape with a waterfall view suggest that landscapes with dynamic water flow may be associated with greater therapeutic benefits in terms of PFC activity than static landscapes. Our results underscore that water is a critical aspect of a landscape due to its therapeutic benefits and should be incorporated in the planning and design of green spaces for health promotion.
이 연구의 목적은 교사의 교수-학습 방식(범주생성형 vs. 범주이해형)이 학생의 분류능력에 어떤 영향을 미치는지를 두뇌 수준에서 규명하는 것이다. 이를 위해 대학교에 재학 중인 24명의 건강한 오른손잡이 학생들이 이 연구에 참여하였다. 이들은 생성 집단과 이해 집단의 두 집단으로 나뉘어 12주간의 서로 다른 학습프로그램을 체험하였다. 연구 참여자들의 과제수행 과정에서의 두뇌활성을 측정하기 위하여 학습프로그램 경험 전후에 fMRI 측정과 지필 검사를 실시했다. 연구결과에 따르면, 분류범주생성 과제의 수행에서 생성집단은 교수-학습 경험 후 대뇌피질 영역과 기저핵 영역이 함께 증가되었으며, 선택-갈등과 관련된 전두피질 영역과 해마옆이랑의 활성감소가 나타났다. 반면 이해집단은 의미 있는 활성변화가 나타나지 않았다. 분류범주이해 과제의 수행에서는 이해집단이 교수-학습 경험 후 대뇌피질 영역과 해마옆이랑의 활성증가가 있었으며, 우측 전두피질부와 소뇌의 활성감소가 있었다. 이와 같은 사실은 특정 교수-학습 양식에 의해 학생이 경험하는 학습양식은 이와 관련된 학생의 특정 두뇌 시스템의 발달을 강화 혹은 약화 시킬 수 있게 된다는 것을 보여준다. 아울러 이러한 연구결과는 두뇌 맞춤형 과학적 분류 학습프로그램 개발을 위한 근거자료로 활용될 수 있을 것이다.
본 연구에서는 과학고학생(n=8)과 외국어고학생(n=5)을 대상으로 지능과제 수행 시 뇌활동성을 분석하였다. 두 집단은 지능점수가 상위 1% 안에 드는 높은 지능을 지닌 학생들이었다(과학고집단: RAPM 평균점수=34.0, WAIS 평균 IQ=139.6; 외국어고집단: RAPM 평균점수=33.8, WAIS 평균 IQ=147.2). 분석결과 두 집단은 지능과제 수행 시 공통적으로 좌측과 우측의 전두엽, 두정엽, 전대상피질에서 활동성을 보였다. 과학고집단은 도형 작업기억과 관련된 우측 두정엽에서 가장 높은 활동성을 보인 반면, 외국어고집단은 언어 작업기억과 관련된 좌측 전두엽에서 가장 높은 활동성을 보였다. 두 집단 간의 뇌활동성 차이를 분석한 결과 외국어고집단은 과학고집단에 비해 혀와 입술의 운동과 관련된 좌측 중심전이랑에서 높은 활동성을 보였다. 이러한 결과는 지능과제 수행 시 과학고학생은 도형 관련 영역이 주요하게 관여하는 반면, 외국어고학생은 언어 관련 영역이 주요하게 관여하는 것을 보여주는 것으로 동일과제임에도 불구하고 관여하는 주 사고체계가 영재성을 보이는 분야에 따라 서로 다를 수 있음을 시사한다.
