경남지방에서 도라지 재배지역별 토양이화학적 특성과 사포닌 함량 (Soil Physico-Chemistry and Saponins Content of Platycodon grandiflorum Radix Cultured from Different Sites in Gyeongnam Province)

  • This study was carried out in order to survey the soil physico-chemical, morphology and content of saponins of 5 year-old Platycodon grandiflorum radix from different areas in Gyeongnam. Soil physical properties was showed 1.01-1.29 of bulk density of soil and soil hardness was increased with increasing soil depth, especially below the 30 cm, however it was maintained or small increased below the 40 cm. T-N and OM contents of top soil were highest at Geochang as 0.27% and 56.9 g/kg, respectively, compared to other areas. The root length was longer at Kimhea and Geochang as 33.8 and 33.7 cm, respectively, and fresh weight was heavy at Geochang as 208.5 g. The contents of saponin of P. grandiflorum radix was higher in fine root compare to main root. By region, content of saponins of P. grandiflorum radix were higher at Hapcheon than other area. Mg content was highly negatively correlated at p < 0.01 with platycodin D3, deapioplatycodin D, platycodin D, and deapioplatycodin D ($-0.499^{**}$, $-0.433^{**}$, $-0.421^{**}$, $-0.511^{**}$ respectively). These results suggest that no-fertilized Mg effected on the improvement of saponin contents in P. grandiflorum.

구증구포 도라지 Ethyl Acetate 분획물의 예쁜 꼬마선충 내의 항산화 효과 (Antioxidant Activity of Ethyl Acetate Fraction of the Guzeunggupo-procossed Platycodon grandiflorum A. De Candolle Roots in Caenorhabditis elegans)

  • Through Caenorhabditis elegans model system, the antioxidant activity of methanol extract of the guzeunggupo-processed Platycodon grandiflorum A. De Candolle (Campanulaceae) roots was calculated. Between the methanol extracts of guzeunggupo-processed and non-processed P. grandiflorum roots, the processed P. grandiflorum root showed higher DPPH radical scavenging effect than the non-processed one. The ethyl acetate soluble fraction of the methanol extract of the guzeunggupo-processed P. grandiflorum showed the best DPPH radical scavenging activity. The ethyl acetate fraction of the processed sample was measured for the activities of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species level. In addition, to confirm the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction of the processed sample, SOD-3 expression was measured using a transgenic strain (CF1553). Consequently, the ethyl acetate fraction of the processed sample, increased SOD and catalase activities, and decreased ROS accumulation in a dose-dependent manner. Furthermore, the ethyl acetate fraction of the processed sample-treated CF1553 worm showed higher SOD-3::GFP intensity than the control worm.

길경(桔梗)발효 추출물의 알레르기성 소양행동 억제효과연구 (Anti-pruritic Effect of Ethanol Extracts from Platycodon grandiflorum and its fermented production in Scratching Behavior Mouse Models)

  • Objectives : Platycodon Root is frequently used in traditional medicine to treat inflammatory diseases of the throat. The purpose of this study was to characterize the effect of the EtOH extract of fermented Platycodon grandiflorum on the ameliorative effects on the Antipruritic Effect of atopic dermatitis mouse model induced by compound 48/80 and ovalbumin (OVA)-induced allergic responses in mice. Methods : In the present study, we examined the anti allergic effect of Platycodon grandiflorum (PR) and its fermented production (FPR) in several mouse model. We measured acute ear edema in a mouse model caused by TPA and consecutively histological change of Ear tissue was observed by hematoxylin and eosin (H&E) staining. and also Scratching behaviors by compound 48/80 was investigated. The levels of allergic mediators such as immunoglobulin (Ig) E, and anti-oxidant markers such as SOD and MDA in the sera of OVA induced allergic mice were measured by enzyme-linked immunosorbent assay. Results : FPR inhibited compoud 48/80-induced scratching behavior in mice, as well as acetic acid-induced writhing in mice. The anti-scratching behavioral effect of FPR was more potent than PR. FPR extract significantly decreased the serum levels of IgE and MDA compared with those of OVA control group. Conclusions : These results indicate that Anti allergic effect of Platycodon grandiflorum is enhanced by fermentation with Saccharomyces cerevisae and FPR may be useful for protection from the itching reactions, which are IgE-mediated representative skin allergic diseases.

길경 재배년수, 수확시기 및 건조방법에 따른 조사포닌 함량 (Crude Saponin Contents of Platycodon grandiflorum(Jacq.) A.DC.)

  • 길경의 품질평가 기준으로 활용하고 있는 crude saponin 함량 변화를 밝히고 고품질 약재생산 및 가공을 위한 기초자료를 얻고자 생육년수, 부위, 수확시기, 및 건조방법별로 조사포닌 함량을 분석한 결과는 다음과 같다. Crude saponin 함량은 생육년수가 증가함에 따라 점점 감소하였다. 부위별로는 미부에서 2.74%로 두부 1.65%보다 1.6배 높았으며, 외피가 내심보다 1.8배 높았다. 수확시기별로는 3월과 12월에 수확했을 때가 각각 2.82, 2.74%이었으며 6월과 9월은 각각 2.12, 2.09%이었다. 건조방법별로는 증건했을 때 가 1.65%로 가장 낮았고 양건, 음건, 열풍건 및 원적외선 건조에서는 차이가 없었다.

Comparative proteome analysis of diploid and tetraploid root in Platycodon grandiflorum

  • In spite of the potential medicinal significance and a wide range of pharmacologic properties of Platycodon grandiflorum, the molecular mechanism of its roots is still unknown. The present study was conducted to profile proteins from 3, 4 and 5 months aged diploid and tetraploid roots of Platycodon grandiflorum using high throughput proteome approach. Two-dimensional gels stained with CBB, a total of 68 differential expressed proteins were identified from the diploid root out of 767 protein spots using image analysis by Progenesis SameSpot software. Out of total differential expressed spots, 29 differential expressed protein spots (${\geq}2-fold$) were analyzed using LTQ-FTICR MS whereas a total of 24 protein spots were up-regulated and 5 protein spots were down-regulated. On the contrary, in the case of tetraploid root, a total of 86 differential expressed proteins were identified from tetraploid root out of 1033 protein spots of which a total of 39 differential expressed protein spots (${\geq}2-fold$) were analyzed using LTQ-FTICR MS whereas a total of 21 protein spots were up-regulated and a total of 18 protein spots were down-regulated. It was revealed that the identified proteins from the explants were mainly associated with the nucleotide binding, oxidoreductase activity, transferase activity. Taken together, the identified proteins may be helpful to identify key candidate proteins for genetic improvement of plants.

Proteome characterization of the liquid cultured tetraploid roots in Platycodon grandiflorum

  • The roots of Platycodon grandiflorum are commonly used for treating bronchitis, asthma, tuberculosis, diabetes, and other inflammatory diseases. Since the molecular mechanism underlying the roots of the plant is unclear. Therefore, the present study was conducted to profile proteins from liquid cultured tetraploid roots of Platycodon grandi orum fl using high throughput proteome approach. Two-dimensional gels stained with CBB, a total of 659 differentially expressed proteins were identified from the liquid medium cultured tetraploid roots of which 32 proteins spots (${\geq}1.5-fold$) were sorted for mass spectrometry analysis. Out of these 32 proteins, a total of 15 proteins were up-regulated such as Serine carboxypeptidase-like 27, Transcription factor bHLH150, 60 kDa jasmonate-induced protein, Cytosolic Fe-S cluster assembly factor NBP35, Regulatory associated protein of TOR 2 and a total of 17 proteins were down-regulated such as Protein G1-like2, Phenylalanine ammonia-lyase, Fructokinase-2, Trihelix transcription factor GT-3a, Guanine nucleotide-binding protein alpha-1 subunit. However, the frequency distribution of identified proteins was carried out within functional categories based on molecular functions, cellular components, and biological processes. Functional categorization revealed that the most of the identified proteins from the explants were mainly associated with the nucleic acid binding, oxidoreductase, transferase activity, protein binding and hydrolase activity. In addition, the proteomic feedback of tetraploid roots of P. grandiflorum may potentially be used to understand the characteristics of proteins and their functions.

도라지 종자의 영양학적 특성 평가 (Evaluation of Nutritional Characteristics of Platycodon grandiflorum Seeds)

  • 본 연구에서는 도라지(Platycodon grandiflorum A. DC) 종자의 영양학적 특성을 평가하기 위해 일반성분, 조섬유, 환원당, 유리당, 유기산, 무기질 및 아미노산 함량을 분석하였다. 종자의 수분, 조단백질, 조지방, 조회분, 탄수화물 함량은 각각 6.97, 26.05, 27.46, 3.78, 35.74%였으며 조섬유 함량은 6.31%, 환원당 함량은 1.54%이었다. 종자에 존재하는 유리당으로는 sucrose가 1,661 mg/100 g, 유기산으로는 lactic acid가 1,224 mg/100 g 으로 가장 풍부하였으며, 인과 칼륨이 주요 무기질로서 700 mg/100 g 이상 존재하였다. 종자 100 g에서의 아미노산 조성을 분석한 결과, 글루탐산(3.45 g), 아르기닌(2.51 g), 아스파르트산(1.66 g), 루신(1.29 g), 라이신(1.10 g), 알라닌(1.05 g), 글리신(1.04 g) 순이었으며 그 외의 아미노산은 1 g 이하로 존재하였다.

Proteome Profiling Unfurl Differential Expressed Proteins from Various Explants in Platycodon Grandiflorum

  • Platycodon grandiflorum, commonly known as Doraji in Korea, has a wide range of pharmacologic properties, such as reducing adiposity and hyperlipidemia, and antiatherosclerotic effects. However, the mechanisms underlying these effects remain unclear. In order to profile proteins from the nodal segment, callus, root and shoot, high throughput proteome approach was executed in the present study. Two dimensional gels stained with CBB, a total of 84 differential expressed proteins were confirmed out of 839 protein spots using image analysis by Progenesis SameSpot software. Out of total differential expressed spots, 58 differential expressed protein spots (${\geq}$ 2-fold) were analyzed using MASCOT search engine according to the similarity of sequences with previously characterized proteins along with the UniProt database. Out of 58 differential expressed protein, 32 protein spots were up-regulated such as ribulose-1,5-bisphosphate carboxylase, endoplasmic oxidoreductin-1, heat stress transcription factor A3, RNA pseudourine synthase 4, cysteine proteinase, GntR family transcriptional regulator, E3 xyloglucan 6-xylosyltransferase, while 26 differential protein spots were down-regulated such as L-ascorbate oxidase precursor, late embryogenesis abundant protein D-34, putative SCO1 protein, oxygen-evolving enhancer protein 3. However, frequency distribution of identified proteins using iProClass databases, and assignment by function based on gene ontology revealed that the identified proteins from the explants were mainly associated with the nucleic acid binding (17%), transferase activity (14%) and ion binding (12%). In that way, the exclusive protein profile may provide insight clues for better understanding the characteristics of proteins and metabolic activity in various explants of the economically important medicinal plant Platycodon grandiflorum.

  • The ameliorating effect of the MeOH extract of the root of Platycodon grandiflorum on the ethanol-induced cognitive dysfunction in mice was investigated. The total MeOH extract was devided into two fractions, saponin rich fraction and non-saponin fraction. The mice with repeated administration of the total MeOH extract and those with saponin rich fraction both showed a markedly prolongεd latency time upon the step-through test (passive avoidance performance) performed after acute ethanol intoxication, while those with non-saponin rich fraction did not. The results suggest that the extracts from the root of Platycodon grandiflorum exert a beneficial effect on ethanol-induced memory impairment in mice and also suggest that these effects of the extracts might be ascribed to the saponin constituents.
