The Root from Heracleum moellendorffii Exerts Anti-Inflammatory Activity via the Inhibition of NF-κB and MAPK Signaling Activation in LPS-Stimulated RAW264.7 Cells

  • Although the roots of Heracleum moellendorffii (HM-R) have been long treated for inflammatory human diseases, scientific evidence for the anti-inflammatory activity of HM-R is not sufficient. In this study, we investigated anti-inflammatory activity and mechanism of action of HM-R in LPS-stimulated RAW264.7 cells. HM-R blocked LPS-induced NO and PGE2 production, but not HM-L. HM-R inhibited LPS-induced overexpression of iNOS, COX-2, $IL-1{\beta}$ and IL-6 in RAW264.7 cells. HM-R inhibited LPS-induced $NF-{\kappa}B$ signaling activation through blocking $I{\kappa}B-{\alpha}$ degradation and p65 nuclear accumulation. In addition, HM-R inhibited MAPK signaling activation by attenuating the phosphorylation of ERK1/2, p38 and JNK. Furthermore, HM-R inhibited attenuated LPS-mediated overexpression of the osteoclast-specific factors such as NFATc1, cathepsin K, MCP-1 and TRAP. These results indicate that HM-R may exert anti-inflammatory activity by inhibiting $NF-{\kappa}B$ and MAPK signaling activation. From these findings, HM-R has potential to be a candidate for the development of chemopreventive or therapeutic agents for the inflammation and inflammatory diseases.

Antifungal Activity of the Methanol Extract of Myristica malabarica Fruit Rinds and the Active Ingredients Malabaricones Against Phytopathogenic Fungi

  • In a search for plant extracts with in vivo antifungal activity for plant diseases, we found that the methanol extract of Myristica malabarica fruit rinds effectively suppressed the development of several plant diseases. The methanol extract exhibited potent 1-day protective activity against rice blast, tomato late blight, wheat leaf rust and red pepper anthracnose. It also showed 7-day and 4-day protective activities against the plant diseases. Three antifungal resorcinols were isolated from the methanol extract of M. malabarica fruit rinds and identified as malabaricones A(MA), B(MB), and C(MC). Inhibitory activity of the three resorcinols against mycelial growth of plant pathogenic fungi varied according to compound and target species. All three compounds effectively reduced the development of rice blast, wheat leaf rust and red pepper anthracnose. In addition, MC was highly active for reducing the development of tomato late blight. This is the first report on the antifungal activities of malabaricones against filamentous fungi.

Incidence and sero-surveillance of feline viruses in Korean cats residing in Gyeonggi-do

  • Incidences of major feline viral diseases provide basic information for preventing viral disease in cats. Despite the growing interest in feline viral diseases, sero-surveillances have been lacking. In this study, we analyzed the diagnoses of feline viral diseases and conducted a sero surveillance of feline panleukopenia virus (FPV), feline calicivirus (FCV), feline herpesvirus-1 (FHV-1), and feline infectious peritonitis virus (FIPV) in Korean cats. Of the 204 confirmed cases since 2015, the numbers of diagnoses for FPV, FIPV, FCV, feline influenza virus, and FHV-1 were 156, 32, 12, 3, and 1 case, respectively. In total, 200 sera, collected between 2019 and 2021, were screened for the presence of antibodies against FPV, 2 FCVs, FHV-1, and FIPV using a hemagglutination inhibition test and a virus-neutralizing assay (VNA). The overall seropositive rates in cats tested for FPV, the 2 FCVs, FHV-1, and FIPV were 92.5%. 42.0%, 37.0%, 52.0%, and 14.0%, respectively. A low correlation (r = 0.466) was detected between the VNA titers of 2 FCV strains. The highest incidence and seropositive rate of FPV reveal that FPV is circulating in Korean cats. The low r-value between 2 FCVs suggests that a new feline vaccine containing the 2 kinds of FCVs is required.

수종 식물의 Plasmin 저해 활성 검색 (Plasmin Inhibitory Activity of Medicinal Plants)

  • Binding of urokinase-type plasminogen activator to its cellular receptor accelerate production of plasmin from plasminogen on the cell surface. Plasmin can digest extracellular matrix components and basement membranes through activating certain proMMPs, which is related to the invasiveness to the cells. Plasmin also acts the regulation of blood coagulation and relates closely to cardiovascular diseases such as stroke and coronary occlusion. Therefore, its inhibitors may be useful as antimetastatic agents and to the treatment of cardiovascular diseases. To search for plasmin inhibitors from plant resources, we screened plasmin inhibitory activities with 76 methanol extract of plant species. Among them, three plant samples showed strong inhibitory activities (>70%) and thirteen plant samples showed more than 50% inhibitory activities of plasmin. Their inhibitory activities were not directly related with uPA inhibitory activites and cell viability.

Aphelenchus avenae에 의한 인산 토양병의 억제효과 (Effects of Aphelenchus avenae on Suppression of Soilborne Diseases of Ginseng)

  • The monoxenic culture of the fungivorous nematode, Aphelenchus avenae, was applied for the control of soil-borne ginseng pathogens such as Fusarium solani and Rhizoctonia solani. Fungivorous nematode populations were measured in a field to examine relationships between the nematode populations and suppression of ginseng root diseases. Inoculation of A. avenae (5000 nematodes per petri-dish) reduced the colonization of the Fusarium mycelium on root discs of ginseng and carrot by 80.0% and 60.5%, respectively. A. avenae also significantly reduced the occurrence of damping-off of ginseng by R. solani pathogenic to ginseng, and no plant damage by the nematode was noted. In a 3-year-old ginseng field infested with Cylindrocarpon destructans, plant missing caused by root rot positively correlated to the density of potato rot nematode, Ditylenchus destructor, but it was reduced with the population of A. avenae, suggesting that A. avenae might inhibit the occurrence of ginseng root rot.

냉이에 발생하는 진균성 병해 (Fungal Diseases of Capsella bursa-pastoris in Korea)

  • 주요 봄철 채소의 하나인 냉이에 발생하는 병해를 조사한 결과 다음의 4가지 병해가 발견되었다. 흰녹가루병은 주로 근생엽에 발생하나 때로 중기와 꽃대에도 발생하여 피해를 주었으며, 병원균은 Albu해 candida로 동정되었다. 노균병은 주로 군집밀도가 높은 상태에서 발생하였고 피해도 심했는데, 병원균은 Peronospora parasitica로 동정되었다. 흰무늬병은 드물게 발생하였고 피해도 거의 없다고 판단되는데, 병원균은 Pseudocercosporella capsellae로 동정되었다. 그리고 흰가루병은 주로 그늘에서 자란 개체에서만 발생하였는데, 병원균은 Erysiphe cichoracearum으로 동정되었다. 이들 4가지 병해는 모두 한국에서 처음기록되는 것이다. 이 중 흰녹가루병과 노균병은 자연상태에서 발생이 심하였을 뿐만 아니라 여러 가지 발병환경을 고려해 볼 때 시설재배에서 많이 발생할것으로 예상되므로 시설재배에서는 철저한 관리가 요구된다.

  • The ongoing global pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has not only influenced over 1.26 billion people but also caused 2.77 million deaths worldwide (as of March 28, 2021). The vaccination could be the most efficient strategy to prevent SARS-CoV-2 infection. However, the continuous emergence of novel variants such as VUI-202012/01 (United Kingdom) and 501.V2 (South Africa) raises huge concerns about the effectiveness of the vaccine designed to target the original virus strain. Since ancient times regardless of the East and West, the plants which refered in this presentation have been consumed not only as food but also as a natural medicine to treat diverse diseases including infectious diseases. Importantly, these plants contain secondary metabolites that display antiviral activity involved in the inhibition of viral adsorption, penetration, and replication. Also, plant-derived natural medicines are expected to have a wider range of efficacy and fewer side effects than synthetic medicine, discovering novel plant-based viral agents would be a promising strategy to fight against SARS-CoV-2.

Screening rhizobacteria for biological control of root rot and Phytophthora blight on glnseng.

  • Ginseng (Panax ginseng) is one of the most widely cultivated medicinal herbs in Korea. During 3 or 5 years cultivation of ginseng, yield losses can reach as high as 30-60% due to numerous diseases in Korea. Among 106 Bacillus strains isolated from various plant internal roots, we selected three promising biocontrol agents by screening against root rot caused by Cylindrocarpon destructan in a greenhouse. Preinoculation of selected isolates to seed or one-year-old root resulted in stimulation of shoot and/or root growth of seedlings, and control of root rot in infested soils with Cylindronrpon destructans (P=0.05). Furthermore, drenching of selected isolates on seedling-growing pots reduced the incidence of Phytophthora blight when the seedlings were challenged with zoospores of Phytophthora cactorum (P=0.05). However, isolates B1141 and B1142 did not show any antifungal activity against various soilborne pathogens while B1146 did in vitro. Our results provide an insight that rhizobacteria can induce resistance against various plant diseases on ginseng even if any resistant breeds have been unknown on ginseng yet.

Biocontrol Efficacy of Formulated Pseudomonas chlororaphis O6 against Plant Diseases and Root-Knot Nematodes

  • Commercial biocontrol of microbial plant diseases and plant pests, such as nematodes, requires field-effective formulations. The isolate Pseudomonas chlororaphis O6 is a Gram-negative bacterium that controls microbial plant pathogens both directly and indirectly. This bacterium also has nematocidal activity. In this study, we report on the efficacy of a wettable powder-type formulation of P. chlororaphis O6. Culturable bacteria in the formulated product were retained at above $1{\times}10^8$ colony forming units/g after storage of the powder at $25^{\circ}C$ for six months. Foliar application of the diluted formulated product controlled leaf blight and gray mold in tomato. The product also displayed preventative and curative controls for root-knot nematode (Meloidogyne spp.) in tomato. Under laboratory conditions and for commercially grown melon, the control was at levels comparable to that of a standard commercial chemical nematicide. The results indicated that the wettable powder formulation product of P. chlororaphis O6 can be used for control of plant microbial pathogens and root-knot nematodes.

A Review of Detection Methods for the Plant Viruses

  • The early and accurate detection of plant viruses is an essential component to control those. Because the globalization of trade by free trade agreement (FTA) and the rapid climate change promote the country-to-country transfer of viruses and their hosts and vectors, diagnosis of viral diseases is getting more important. Because symptoms of viral diseases are not distinct with great variety and are confused with those of abiotic stresses, symptomatic diagnosis may not be appropriate. From the last three decades, enzyme-linked immunosorbent assays (ELISAs), developed based on serological principle, have been widely used. However, ELISAs to detect plant viruses decrease due to some limitations such as availability of antibody for target virus, cost to produce antibody, requirement of large volume of sample, and time to complete ELISAs. Many advanced techniques allow overcoming demerits of ELISAs. Since the polymerase chain reaction (PCR) developed as a technique to amplify target DNA, PCR evolved to many variants with greater sensitivity than ELISAs. Many systems of plant virus detection are reviewed here, which includes immunological-based detection system, PCR techniques, and hybridization-based methods such as microarray. Some of techniques have been used in practical, while some are still under developing to get the level of confidence for actual use.
