Phellinus linteus WI-001 균사체의 고밀도 배양을 위한 배양학적 특성 연구 (Studies on Cultural Characteristics for High Density Fermentation of Phellinus linteus WI-001)

  • 강력한 함암작용을 나타내는 단백다당체를 생산하는 Phellinus linteus WI-001 균주를 대상으로 배양학적 특성 및 초기 배양배지의 탄소원인 glucose 농도에 따른 균사체 생산성을 조사하였다. 배양중 최적 pH와 온도는 각각 5.5와 28$^{\circ}C$인 것으로 나타났다. 특히 pH의 경우 전 배양기간에 걸쳐 5.5에서 6.5의 비교적 좁은 범위에서 변화하였다. 초기 glucose 농도를 0.5%, 1%, 2%, 3% 및 4%에서 균사체 생산량 및 세포비성장속도를 조사한 결과 최종 균사체 생산량은 glucose 농도가 증가할수록 높게 나타났으나, 세포비성장속도가 가장 빠르게 나타나는 초기 1% glucose 조건하에서 배양하다가 추가로 glucose를 간헐적 또는 지속적으로 공급하는 전략으로 생산성 향상을 시도하였다. 그 결과 회분식 배양에 비해 간헐적 공급방식에서는 28% 지속적 공급방식에서는 42%의 수율이 증가되었다.

상황버섯(Phellinus linteus) 균사체로부터 항혈전 생산의 최적화 (Optimization of Anticoagulant Production from Phellinus linteus Mycelia)

  • 각종 버섯에서 항혈전 활성균주를 선별하기 위해 보관 균주 50여종과 임업협동조합에서 분양 받은 20여종의 균주를 대상으로 항혈전 활성을 검색한 결과, 항혈전 활성이 167 sec를 나타낸 상황버섯(Phellinus linteus)을 선별하였다. 본 균주가 생산하는 항혈전 활성의 본체를 파악하기 위해 pronase 처리와 periodate 산화를 행한 결과 pronase로 처리한 시료는 무처리군과 비교하여 차이가 없었던 반면 periodate로 산화시킨 시료는 항혈전 활성이 크게 감소함에 따라 P. linteus가 생산하는 항혈전 활성의 본체는 다당에 기인되는 것으로 추정되었다. 항혈전 생산을 위한 최적 배양조건은 soluble starch 3.0%, peptone 0.1%, $MgSO_4{\cdot}7H_2O$ 0.1%, $K_2HPO_4$ 0.1%, 초기 pH 7.0, 배양온도 $30^{\circ}C$ 및 교반속도 150 rpm이었다. 상기의 최적 배양조건에서 jar fermentor로 10일 배양하였을 때 390 sec의 항혈전 활성과 7.5 mg/mL의 균사체 생육을 나타내었다.

  • The medicinal fungus Phellinus linteus, in the family Hymenochaetaceae, has been used as a traditional medicine for the treatment of various diseases. In this study, the chemical constituents of the culture broth of P. linteus were investigated. P. linteus was cultured in potato dextrose broth medium, and the culture broth was extracted with ethyl acetate. The ethyl acetate-soluble portion was concentrated and subjected to ODS column chromatography, followed by Sephadex LH-20 column chromatography. Six compounds (1~6) were purified by preparative reversed-phase high-performance liquid chromatography. Spectroscopic methods identified their structures as caffeic acid (1), inotilone (2), 4-(3,4-dihydroxyphenyl)-3-buten-2-one (3), phellilane H (4), (2E,4E)-(+)-4'-hydroxy-${\gamma}$-ionylideneacetic acid (5), and (2E,4E)-${\gamma}$-ionylideneacetic acid (6). Compounds 1, 2, and 3 exhibited potent dose-dependent antioxidant activity.

Neuraminidase Inhibitors from the Fermentation Broth of Phellinus linteus

  • During a search for neuraminidase inhibitors derived from medicinal fungi, we found that the fermentation broth of Phellinus linteus exhibited potent neuraminidase inhibitory activity. Through bioassay-guided fractionation, two active compounds were purified from the ethyl acetate-soluble portion of the fermentation broth of P. linteus. These structures were identified as inotilone (1) and 4-(3,4-dihydroxyphenyl)-3-buten-2-one (2) by spectroscopic methods. Compounds 1 and 2 inhibited H1N1 neuraminidase activity with $IC_{50}$ values of 29.1 and 125.6 $125.6{\mu}M$, respectively, in a dose-dependent manner. They also exhibited an antiviral effect in a viral cytopathic effect reduction assay using MDCK cells. These results suggest that compounds 1 and 2 from the culture broth of P. linteus would be good candidates for the prevention and therapeutic strategies towards viral infections.

  • 재배 상황버섯의 유산균 증식 효과를 측정한 결과 B. breve에 대한 증식 효과와 배양배지의 pHwj하 효과가 우수했다. 사람 및 흰주의 장내 세균을 버섯추출물 함유 배니에서 배양시 배지의 pH저하 효과가 우수할뿐만아니라 ${\beta}-glucosidase$의 효소 활성을 억제하였다. 그외에도 대장암 및 방광암 등의 원이효소로 주목되고 있는 사람 및 흰쥐의 장내 세균총 효소인 ${\beta}-glucosidases$ 및 tryptophanase 효소 활성을 억제하였다. 이외에도 인슐린 비의존형 당뇨병의 치료제로 사용되는 acarbose보다는 약하지만 maltase, sucrase 및 알파아밀라제를 효과적으로 저해하였다.

  • 상황버섯 자실체, 액체배양 균사체 및 배양액으로부터 고분자물질(polymer)을 추출하여 면역활성을 anti-complementary assay 방법으로 측정하였다. 자연산과 인공재배 자실체로부터 생산된 polymer의 면역활성은 각각 65.77%와 63.94%로 나타나 큰 차이를 보이지 않았다. 자실체, 균사체 및 배양액으로부터 추출한 polymer의 면역활성 측정결과, 균사체는 41.95%, 배양액은 21.87%로 자실체 보다 낮게 나타났다.

  • Phellinus linteus (Berk. & M.A. Curtis) Teng, commonly referred to as Sangwhang in Korea, is a well-known species of the genus Phellinus, which attracts great attention due to its phamarcological values. P. linteus has been reported to produce anti-tumor, anti-angiogenic, anti-mutagenic and immunomodulatory activities in vivo and in vitro. However, despite extensive biochemical studies on P. linteus, the wine produced by P. linteus fermentation (WPLF) has poorly investigated. In the present study, it was compared the in vitro cytotoxic effects of WPLF with ethanol as positive control. WPLF as well as ethanol induced the inhibition of cell proliferation and morphological changes in both HepG2 and A549 cells in a concentration-dependent manner, however, WPLG treatment has less cytotoxic effects than ethanol treatment. These cytotoxic effects were associated with the induction of apoptotic cell death, but, WPLG treatment has less apoptotisis inducing effects than ethanol treatment.

Development of Species-specific Primers for Rapid Detection of Phellinus linteus and P. baumii

  • Genus Phellinus taxonomically belongs to Aphyllophorales and some species of this genus have been used as a medicinal ingredients and Indian folk medicines. Especially, P. linteus and morphological-related species are well-known medicinal fungi that have various biological activities such as humoral and cell-mediated, anti-mutagenic, and anti-cancer activities. However, little is known about the rapid detection for complex Phellinus species. Therefore, this study was carried out to develop specific primers for the rapid detection of P. linteus and other related species. Designing the species-specific primers was done based on internal transcribed spacer sequence data. Each primer set detected specifically P. linteus (PL2/PL5R) and P. baumii (PB1/PB4R). These primer sets could be useful for the rapid detection of specific-species among unidentified Phellinus species. Moreover, restriction fragment length polymorphism analysis of the ITS region with HaeIII was also useful for clarifying the relationship between each 5 Phellinus species.

Neuraminidase Inhibitors from the Culture Broth of Phellinus linteus

  • During the search for neuraminidase inhibitors from medicinal fungi, we found that the culture broth of Phellinus linteus exhibited potent inhibitory activity. Solvent partition, Sephadex LH-20 column chromatography, and high-performance liquid chromatography (HPLC) were performed for purification of two active substances from the culture broth. According to $^1H$ NMR measurements and comparison of HPLC retention times with those of authentic compounds, their chemical structures were identified as hispidin and hypholomine B. Compounds (hispidin) 1 and 2 (hypholomine B) inhibited neuraminidase, with $IC_{50}$ values of 13.1 and 0.03 ${\mu}M$, respectively.

Prolyl Endopeptidase Inhibitory Activity of Two Styrylpyranones from Phellinus linteus

  • Two styrylpyranones (1 and 2) were isolated from the $CH_2Cl_2$-soluble fraction of Phellinus linteus and their structures were established by extensive Nuclear magnetic resonance (NMR) spectral data as inoscavin E and C. Both compounds showed significant prolyl endopeptidase inhibitory activity with $IC_{50}$ values of $4.26{\pm}0.14$ and $4.08{\pm}0.04{\mu}M$ and $K_i$ values of $1.50{\pm}0.02$ and $1.43{\pm}0.03{\mu}M$, respectively. They also exhibited antioxidant capacities against the ABTS radical system with $EC_{50}$ values of $6.47{\pm}0.05$ and $7.64{\pm}0.06{\mu}M$, respectively.
