Procedures in Establishing Residue Limits of Pesticides on Food Crops in Codex Alimentarius Commission and Foreign Countries (국제기구 및 외국에서 농산물중 농약잔류 허용기준의 설정절차)

  • This study was undertaken to find out necessary measures to improve the tolerance setting system of pesticide residues on food crops in Korea which is scientifically reasonable and harmonizable with international standards. Information on tolerance setting systems of pesticide residues by Codex Alimentarius Commission, Joint FAO/WHO Meeting of Experts on Pesticide Residues, USA, EU, Japan and Taiwan was collected and analyzed. On the basis of information in the above countries, necessary actions to be taken by the Korean regulatory authorities were recommended with respect to priority setting, maximum residue limits (MRLs) setting based on field residue data, group MRLs, minor crop problems, quantitation limit and dietary intake assessment.

Monitoring of Pesticide Residues in Vegetables Collected in Chungbuk, Korea (충북지역에 유통되는 일부 채소 농산물의 잔류농약 모니터링)

  • This study is being executed to investigate the pesticide residues for agricultural products on the markets in Chungbuk area from 2010 to 2012. The samples used were chard, perilla leaf, leek, spinach, crown daisy, marsh mallow, and winter grown cabbage, These were analyzed by GC/MSD and GC/ECD. Agricultural pesticide levels of spinach was 23.4%, winter grown cabbage 20.6%, chard 14.9%, marsh mallow 14.0%, perilla leaf 13.1%, leek 8.4%, and crown daisy 5.6%. Exceeds of residual pesticides were spinach, chard, leek, perilla leaf, and marsh mallow. Based on these results, a risk assessment was being conducted by used a percentage of acceptable daily intake (%ADI). The %ADI ranged from 0.00014% to 1.73910%, and these values was indicated to have no effects on human health.

  • Maximum residue limits (MRL) for pesticides in feed have been set to protect public health and produce safe livestock products. In vivo experiments to establish MRL are essential, as livestock are commonly used to obtain reliable in vivo quantitative information. Here, we aimed to evaluate whether small laboratory animals can replace or reduce monogastric livestock in experiments to quantify pesticide residues in vivo after oral consumption through feed. First, 24 pigs and rats were randomly assigned to four groups and fed 0, 3, 9, or 30 mg/kg of sulfoxaflor. After four weeks, serum, muscle, fat, liver, kidney, and small intestine samples were collected, and sulfoxaflor residues were analyzed using liquid chromatography - tandem mass spectrometry. Sulfoxaflor residues in pig tissues were significantly correlated with those in rat tissues. Model equations were formulated based on the residual sulfoxaflor amount in pig and rat tissues. The calculated and measured sulfoxaflor residues in pigs and rats showed more than 90% similarity. Sulfoxaflor did not affect body weight gain, feed intake, or the feed conversion ratio. Therefore, we concluded that pesticide residue quantification in vivo to establish MRL could be performed using small laboratory animals instead of livestock animals. This would contribute to obtaining in vivo pesticide residue information and reducing large-scale livestock animal experiments.

Establishment of the Korean total diet study (TDS) model in consideration to pesticide intake (한국형 총식이섭취조사(Total Diet Study, TDS) 모델 확립을 위한 농약섭취수준에 대한 접근)

  • This study was carried out to establish Korean total diet study (TDS) model for estimating pesticide residue in food samples. In addition, pesticide residues of food samples were monitored by setting the standards of established Korean total diet study model. For monitoring, first step were selection of total 102 species food samples, second step were selection of total 70 species food samples, and third step were selection of total 12 representative diet and 109 species food samples. Ninety-eight pesticides were analyzed using $GC-{\mu}ECD$, GC-MS, and LC-MS/MS after QuEChERS sample preparation method. The residue levels in detected food samples were below the maximum residue limit (MRL). Establishment of the Korean total diet study model means providing safe food for consumers, secure the safety of food samples and provide ongoing information to agricultural producers about use of pesticides.

A Research on the Residual Pesticide Content of Dried Agricultural Products from Ulsan (울산지역 건조농산물 가공품의 잔류농약 분석)

  • This study was conducted to evaluate the safety of 208 types of residue pesticides on 150 dried agricultural products in Ulsan. The pesticide residues were detected using GC-MS/MS, LC-MS/MS, GC/ECD, GC/NPD, and LC/PDA. The detection rate was 19.3% (29 of 150 samples), and 28 types of pesticides were detected. Two of the 29 pesticide residues exceeded the maximum residue limit (MRL). Difenoconazole was detected in Ulleungdo aster, and chlorothalonil and chlorpyrifos were detected in chili pepper leaves. In the vegetable group, the frequency of pesticide residues was found to be the highest in dried leafy vegetables, followed by dried fruiting vegetables other than cucurbit vegetables, and then dried stalk and stem vegetables. The pesticide types detected in the commercial dried agricultural products were fungicide (60%), insecticide (23.8%), and acaricide (16.3%). In the validation study, the values of limit of detection (LOD), limit of quantitation (LOQ), coefficient of determination (R2), and recovery rate were in the range of 0.0001-0.0409 mg/kg, 0.0003-0.1241 mg/kg, 0.994-0.999, and 81.58-116.79%, respectively. The ratio of estimated daily intake (EDI)/acceptable daily intake (ADI) was 0.00002-0.31395%.

Effect of Cheonggukjang Made from Sword Bean on Serum Lipid Profile and Tissue Change in Rats

  • This study was to investigate serum triglyceride (TG), phospholipid (PL), and cholesterol concentrations after feeding the combined diets of prepared traditional Cheonggukjang (TC) or Cheonggukjang made from sword bean powder (CS), the feed, and high-fat cholesterols at certain ratio for 8 weeks in rats, and then confirmed the effects of the diets based on the changes in the tissues of livers, kidneys, and spleens. The serum TG concentrations were lower in CS group compared to that of in TC group. The increase of PL concentrations was greater in CS group than that of TC group. FChCS6 group (92% feed+2% cholesterol+6% CS) showed the lowest total-cholesterol (T-C), $60.1{\pm}8.5mg/dL$, indicating that CS was effective to lower the T-C. Both TG/PL and T-C/PL ratio tended to be decreased. As CS intake was effective in the reduction of low density lipoprotein-cholesterol level as well as the increase of high density lipoprotein-cholesterol level, it would be effective in the prevention of adult diseases and circulatory diseases. It was confirmed that there were no changes in organ appearance and tissues in the rats with CS intake. Therefore, CS intake showed superior functionalities than that of TC intake.

Monitoring and Risk Assessment of Pesticide Residues for Circulated Agricultural Commodities in Korea-2013 (국내 유통 농산물의 잔류농약 모니터링 및 위해평가-2013년)

  • The purpose of this study is the establishment of scientific processes for making food safety policies. Thus, we investigated pesticide residue level of the agricultural commodities from market, and performed risk assessment. Fifteen agricultural items are chosen based on the frequency of Korean consumption. The samples were collected from 9 cities where populations are more than one million. Total 283 active ingredients were monitoring ( total sample number =232). Single-analysis of target pesticides was for three kinds of possible growth regulators and the multicomponent analysis was for 280 kinds of pesticides, a total of 283 species were selected to perform the pesticide residues. Before monitoring the analytes, the improvements of the analytical methods were done by method validations under the CODEX analytical method development guidelines and can produce metrics that represent the international standards applied in accordance with the guidelines. In addition to residual pesticides detected during monitoring we compare the ADI to EDI values using detected result and dietary consumption data which is extracted from annual market basket survey. The 163 samples were non-detected in the total 232 samples so it means that every agricultural commodity will residual pesticides-free in 70.3%. The detected residual pesticides showed for a total of 69 cases (29.7%). Two of samples violate Korean MRL (0.9%). The ratio of EDI compared to ADI resulted in only from 0.00087 to 0.902%. In result, we can assume that all detected residual pesticides are very safe level and current policies of Korean pesticides control may be working.

Pesticide Residue Survey and Risk Assessment of Fruits in Daejeon (대전시 유통 과실류의 농약 잔류 실태 및 위해성 평가)

  • Pesticide residues in some fruits collected at Noeun wholesale market, Daejeon were surveyed and assessed its risk. In 120 samples, the detection rate of pesticide was 70.8% and the rate exceed MRL was 1.67%. Highly detection rate of commodities was apple, peach and grape. Organophosphorus, organochlorine and pyrethroid insecticide were found with similar rate relatively, but most of fungicides were procymidone(dicarboximides) and chlorothalonil(organochlorine). The order of detected pesticide were chlorothalonil, chlorpyrifos, fenvalerate, procymidone, deltamethrin and EPN. In order to assess risk estimated intake amount of each pesticide was compared to ADI. Although some peach were exceed MRL the total amount and uptake pesticides was less than 1.22% comparing to ADI.

Toxicity Assessment and Establishment of Acceptable Daily Intake of Fungicide Isotianil (살균제 Isotianil의 독성평가와 일일섭취허용량 설정)

  • Isotianil is a fungicide which has prevention effects against rice blast disease. In order to register this new pesticide, the series of toxicity data on animal testing were reviewed to evaluate its hazards to consumers and to determine its acceptable daily intake. Isotianil was almost excreted by urine and feces. It has low acute oral toxicity while has no skin toxicity and ocular irritation. Its skin sensitization was evaluated as slight. Genotoxicity of parent compound and metabolite was negligible. Chronic toxicity tests on rats and dogs showed changes of hematology, clinical biochemistry and liver weight. It had no reproductive and teratogenic effects. The estimation of Acceptable Daily Intake(ADI) is based on the lowest no-observed adverse effect level (NOAEL). The lowest NOAEL of 2.83 mg/kg bw/day was found in the twelve-months rats study. The NOAEL was based on increased liver weight and treatment-related effect on clinica chemistry finding at the nest higher dose level of 2.83 mg/kg bw/day. Therefore, it is considered appropriated to apply an uncertainty factor of 100 to the NOAEL 2.83 mg/kg bw/day from the rat study, resulting in an ADI of 0.028 mg/kg bw/day.

Acute and Chronic Exposure Assessment of Organophosphate Pesticides through the Consumption of Fruit Vegetables (과채류 섭취를 통한 유기인계 농약의 급성 및 만성노출평가)

  • In this study, we monitored the residues of organophosphate pesticides (OP) in eight fruit vegetables grown in Korea, and assessed risk levels of acute and chronic exposure of OP through the consumption of fruit vegetables. Chlorpyrifos, EPN, methidathion and phosphamidon in eight fruit vegetables were detected in this study. The results of cumulative assessment of national estimated short term intake for acute exposure of OP were due to the following order; eggplant, tomato, squash and cucumber. Total theoretical maximum daily intake calculated at first step of chronic exposure assessment for registered OP in Korea was 76.14%, compared with acceptable daily intake ADI) based on chlorpyrifos. In addition, total national estimated maximum daily intake calculated at second step of chronic exposure assessment was 13.949%, compared with ADI. Third chronic assessment was conducted by probabilistic approach using OP residues detected in eight fruit vegetables and showed that total exposure risk was very low, corresponding to 0.0001% compared to ADI. Based on those finding, the risk of organophosphate pesticides in fruit vegetables was considered quantitatively negligible. In future, further investigation to expand the target should be followed to do more accurate and detailed risk assessment.
