의성지역의 약용작물 생산현황(生産現況) 및 경영분석(經營分析)을 위하여 약용작물 재배 농가 147호를 대상으로 재배실태와 작물별 소득(所得)을 조사(調査)한 결과는 다음과 같다. 1. 특화계수(特化計數)가 1이상인 작물은 작약(21.59), 산수유(14.82), 시호(7.90), 지모(6.41), 지황(3.96), 두충(3.44), 목단(2.28), 백지(1.51)순으로 나타났다. 2. 조사농가의 평균 토지이용면적(土地利用面積)은 1.92ha이었는데, 그중 밭이 1.35ha. 논은 0.56ha이었다. 농지분포(農地分布)는 농가당 평균 3.1곳에 10.2필지로 구성되어 있으며, 약용작물부문에 년간 노동투하(勞動投下)는 31.1%를 차지하였다. 3. 조사대상 지역에 재배되고 있는 약용작물은 36개 작물로 농가당 평균 2.4작물이었다. 대표적인 작물은 작약이었는데, 조사농가 147호 가운데 85농가가 참여하여 30.72ha를 재배하고 있었다. 4. 약용작물 재배실태를 보면, 작약, 시호, 택사, 길경, 황기, 감국 등은 재배면적이 줄어드는 추세(趨勢)이고 홍화는 새로운 소득작물(所得作物)로 재배되고 있으며, 더덕, 지모, 지황, 원황정, 백출 등은 재배 면적이 늘어났다. 5. 36개 약용작물중 10a당 평균소득이 높은 작물은 홍화(2,775천원), 원황정 (2,706천원), 일천궁(2,573천원) 등의 순이었으나 재배농가에서는 농가여건에 부응하면서 위험부담이 적고 가격안정(價格安定)이 기대되는 작물을 선택하여 농가 수준에서 부가가치(附加漂値)를 최대한 증대(增大)시켜야 할 것으로 생각되었다.
본 연구에서 목단피로부터 분리한 gallic acid(GA)의 항암 활성을 평가하였다. 목단피로부터 GA는 스펙트로 분석법(ESI-MS, $^1H$-NMR, and $^{13}C$-NMR)에 의해 구조를 밝혔으며, 항암활성은 마우스에 결장암을 유도시킨 후 하루에 한번 gallic acid(GA; 20, 100 mg/kg p.o)을 14일 동안 투여 한 후 암세포 크기를 측정하였다. 또한 GA투여 후 체중변화, 급성독성, 간과 비장의 무게 변화와 간의 생화학적 지표를 측정하였다. 결과로써 GA 처리군의 경우 체중 변화 및 급성 독성 없이 항암제 처리군과 비슷하게 암의 크기가 유의적으로 감소하였다(p<0.05). 또한 암의 유도에 의해 증가된 간과 비장의 무게 및 GSH, ALT, AST는 GA 처리에 의해 유의적으로 감소하였으며, 암의 유도에 의해 감소된 GSH는 GA 투여에 의해 유의적으로 증가하였다(p<0.05). 이상의 연구 결과를 통해 GA는 항암제 개발을 위한 후보군으로 활용 가능하리라 사료된다.
흰가루병은 거의 모든 식물재배에서 나타나며, 다양한 곰팡이에 의해 발병한다. 본 연구에서는 천연물 추출물을 이용한 식물병원성 곰팡이 제어가능성을 검토하기 위해, 보리 흰가루병에서 분리된 Fusarium graminearum을 대상으로 900여종의 천연물 추출물의 항진균 활성을 평가하였다. 먼저 포자 발아억제 활성평가 결과 산초, 당귀, 미국가막사리, 까마중, 오수유, 윤판나물, 미치광이풀, 때죽나무, 백선, 방기, 정향, 멍석딸기, 호장근, 천황련, 작약, 대황, 모란, 달맞이꽃, 대극, 개박하 추출물을 1차 선별하였으며, 균사 생육억제 활성 원재료 확보 용이성 및 실제 적용 가능성 등을 고려하여 산초 추출물을 최종 선정하였다. 산초 추출물 조제 시 methanol 추출이 효율적이며, 순차적 유기용매 분획물의 경우 물 잔류물에서 항진균 활성이 나타났다. 그러나 methanol 추출물에 비해 분획물의 활성이 다소 감소하여 다양한 물질이 보완적으로 항진균 활성을 나타냄을 추측할 수 있으며, 미약하지만 methanol 추출물 및 분획물에서 항진균 활성도 확인하였다. 토마토 묘목을 이용한 산초 추출물의 활성 분획물의 약해평가 결과 $1,000\;{\mu}g/ml$ 농도 처리 시에도 특이한 약해는 나타나지 않았다. 본 연구결과는 F. graminearum에 의해 나타나는 흰가루병 제어에 산초 추출물이 유용함을 제시하고 있다.
Mudanpi (Cortex Moutan Radicis; the root cortex of Paeonia suffruticosa Andrews) is an important Chinese crude drug used in many oriental prescriptions. 1,2,3,4,6-Penta-O-galloyl-beta-D-glucose (PGG), a major component of this crude drug, has been shown to possess potent antioxidant, anti-mutagenic and anti-proliferative effects. In this study, I examined whether PGG could protect Neuro 2A cells, a kind of neuronal cell lines, from oxidative damage through the induction of HO-1 expression and HO activity. Exposure of Neuro 2A cells to PGG (10-50μM) resulted in a concentration- and time-dependent induction of HO-1 mRNA, and protein expressions and heme oxygenase activity. PGG protected the cells from hydrogen peroxide-induced cell death. The protective effect of PGG on hydrogen peroxide-induced cell death was abrogated by zinc protoporphyrin IX (ZnPP IX), a HO inhibitor. These results indicate that PGG is a potent inducer of HO-1 and HO-1 induction is responsible for the PGG-mediated cytoprotection against oxidative damage.
We tested Trichoderma harzianum as a biocontrol agent for Rhizoctonia solani AG2-1, using six natural antifungal materials to improve its efficacy. Among the six materials tested, peony (Paeonia suffruticosa) root bark (PRB) showed the strongest antifungal activity against R. solani AG2-1, and was not antagonistic to T. harzianum. Scanning electron microscopy showed that treatment with PRB extract resulted in shortened and deformed R. so/ani AG2-1 hyphal cells. The control of radish damping-off caused by R. so/ani AG2-1 was greatly increased by combined treatments of T. harzianum and PRB, as compared with either of the two treatments alone, with the control effect increased from 42.3-51.5% to 71.4-87.6%. The antifungal compound in PRB, which was isolated in chloroform and identified as paeonol by mass spectrometry, $^1H$ NMR, and $^{13}C$ NMR analyses, inhibited the growth of R. so/ani AG2-1 but not that of T. harzianum. Thus, PRB powder or extract may be used as a safe additive to T. harzianum to improve the control of the soil borne diseases caused by R. so/ani AG2-1.
The extracts from 40 different traditional medicinal herbs were used to investigate the antimicrobial activities against Escherichia coli. Among them, the extracts from Paeonia suffruticosa (PS), Siegesbeckia orientalis (SO), Schizandra chinensis (SC), Caesalpinia sappan (CS) and Rhus javanica (RJ) exhibited high antimicrobial activities against E. coli, Minimum inhibitory concentrations (MIC) of the RJ extract against E. coli were 0.8 mg/ml. After heating treatment of these extracts, the antimicrobial activities against E. coli were significantly reduced in case of the CS extract. After alkaline or acid treatment of these extracts, the antimicrobial activities against E. coli were significantly increased in the PS extract but reduced in both SO and RJ extract. Since extracts from RJ and CS exhibited the highest antimicrobial activities, bacterial growth-inhibiting activities against E. coli by these two extracts were further examined. Optical density at 620 nm after 24 hours incubation of E. coli in the presence of 100, 300 or 500 ppm of RJ extract ranged from 0.1 to 0.2 compared to 0.35~0.65 in the absence of RJ extract, indicating that growth of E. coli was significantly inhibited within 24 hours by the addition of at least 100 ppm of RJ extract. Optical density at 620 nm after 24 hours incubation of E. coli in the presence of 300 or 500 ppm of CS extract ranged from 0.01 to 0.25 compared to 0.5~0.55 in the absence of CS extract, indicating that growth of E. coli was also significantly inhibited within 24 hours by the addition of at least 300 ppm of CS extract. In conclusion, these findings suggest that extracts from RJ and CS may play important roles for antimicrobial activities against E. coli causing various animal diseases.
The inhibitors of prostaglandin $E_2\;(PGE_2)$ production and cyclooxygenase-2 (COX-2) activity have been considered as potential anti-inflammatory agents. In this study, we evaluated 9 compounds isolated from 5 Korean phytomedicinal plants (Spirea prunifolia, Paeonia suffruticosa, Salvia miltiorrhiza, Scutellaria baicalensis, and Artemisia capillaris) for the inhibition of $PGE_2$production and COX-2 expession in lipopolysaccharide (LPS)-stimulated human macrophages U937 cells. As a result, several compound such as prunioside A, penta-O-galloyl-beta-D-glucose, tanshinone IIA, baicalin, baicalein, wogonin, scopolatin, scoparone and decursinol showed potent inhibition of $PGE_2$production (50-70% inhibition at the test concentration of $10\;{\mu}M$). In addition, these compounds were also considered as potential inhibitors of COX-2 activity (45-73% inhibition at the test concentration of $10\;{\mu}M$). These active compound mediating COX-2 inhibitory activities are warranted for further elucidation of active principles for development of anti-inflammatory agents and these properties may contribute to the anti-atopic dermatitis activity.
We studied the total polyphenol content, DPPH radical scavenging activity, hydroxyl radical scavenging activity and ${\alpha}$-glucosidase inhibition of water extracts from 17 medicinal plants. Total polyphenol contents ranged from 10.0 (Coix lachryma-jobi L, CL) ~ 279.7 (Perilla sikokiana, PS)mg/g. The water extract from medicinal plants were evaluated for its free radical scavenging activities and compared with a commercial antioxidant, ascorbic acid. DPPH radical scavenging activity of Pyrus pyrifolia (PP), Chamaecyparis obtusa L. (COL), Chamaecyparis obtusa F. (COF), and PS were higher than positive control. Higher hydroxyl radical scavenging activity were shown in Acanthopanax senticosus (AS) and Cordyceps militaris (CM) than the other plants. The highest anti-${\alpha}$-glucosidase activity was observed in Cornus officinalis (CO) and Paeonia suffruticosa Andrews (PSA) water extracts. PSA showed not only the higher DPPH radical scavenging activity but also the anti-${\alpha}$-glucosidase activity. The results of our study that PP, COL, COF, PS, AS, CM, CO and PSA could be potential candidates for natural antioxidants.
This study was carried out to search the natural antioxidants from the 36 medicinal plants. $IC_{50}$ values of linoleic acid peroxidation inhibition to the BHA, BHT and ${\alpha}-tocopherol$ showed $7.2{\mu}g/ml$, $7.3{\mu}g/ml$, and $26.6{\mu}g/ml$, but hexane extract of Zingiber officinale showed $13.2{\mu}g/ml$, EtOAc extracts of Phellinus linteus, Morus alba, Zingiber officinale and butanol extract of Diospyros kaki showed $15.9{\mu}g/ml$, $16.9{\mu}g/ml$, $20.1{\mu}g/ml$, and $21.5{\mu}g/ml$ respectively. $IC_{50}$ values of DPPH radical scavenging activity of BHA, BHT and $\alpha$-tocopherol showed $14.9{\sim}27.7{\mu}g/ml$, but EtOAc extract of Paeonia suffruticosa showed the lowest $6.5{\mu}g/ml$ than those of other extracts, EtOAc extracts of Camellia sinensis, Salvia miltiorrhiz, and Phellinus linteus showed $8.0{\mu}g/ml$, $11.5{\mu}g/ml$, and $13.0{\mu}g/ml$ respectively.
Moutan cortex, the root bark of Paeonia suffruticosa Andrews (Paeoniaceae), has pharmacological effects such as anti-inflammatory, antiallergic, analgesic and antioxidant activities. We investigated a methanol extract of Moutan cortex for neuroprotective effects on neurotoxicity induced by amyloid ${\beta}$ protein ($A{\beta}$) (25-35) in cultured rat cortical neurons. Exposure of cultured cortical neurons to $10\;{\mu}M\;A{\beta}$ (25-35) for 24 h induced neuronal apoptotic death. Moutan cortex inhibited $10\;{\mu}M\;A{\beta}$ (25-35)-induced neuronal cell death at 30 and $50\;{\mu}g/m{\ell}$, which was measured by a 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay and Hoechst 33342 staining. Moutan cortex inhibited $10\;{\mu}M\;A{\beta}$ (25-35)-induced elevation of intracellular calcium concentration ($[Ca^{2+}]_i$), and generation of reactive oxygen species (ROS) which were measured by fluorescent dyes. Moutan cortex also inhibited glutamate release into medium induced by $10\;{\mu}M\;A{\beta}$ (25-35), which was measured by HPLC. These results suggest that Moutan cortex prevents $A{\beta}$ (25-35)-induced neuronal cell damage by interfering with the increase of $[Ca^{2+}]_i$, and then inhibiting glutamate release and ROS generation. Moutan cortex may have a therapeutic role in preventing the progression of Alzheimer's disease.
