Molecular Cloning and Characterization of a cis-Epoxysuccinate Hydrolase from Bordetella sp. BK-52

  • A cis-epoxysuccinate hydrolase (CESH) from Bordetella sp. BK-52 was purified 51.4-fold with a yield of 27.1% using ammonium sulfate precipitation, ionic exchange, hydrophobic interaction, molecular sieve chromatography and an additional anion-exchange chromatography. The CESH was stable in a broad range of temperature (up to $50^{\circ}C$) and pH (4.0-10.0) with optima of $40^{\circ}C$ and pH 6.5, respectively. It could be partially inhibited by EDTA-$Na_2$, $Ag^+$, SDS, and DTT, and slightly enhanced by $Ba^{2+}$ and $Ca^{2+}$. The enzyme exhibited high stereospecificity in D(-)-tartaric acid (enantiomeric excess value higher than 99%) with $K_m$ and $V_max$ values of 18.67 mM and $94.34\;{\mu}M$/min/mg for disodium cis-epoxysuccinate, respectively. The Bordetella sp. BK-52 CESH gene, which contained 885 nucleotides (open reading frame) encoding 294 amino acids with a molecular mass of about 32 kDa, was successfully overexpressed in Escherichia coli using a T7/lac promoter vector and the enzyme activity was increased 42-times compared with the original strain. It may be an industrial biocatalyst for the preparation of D(-)-tartaric acid.

  • Protocatechualdehyde (PA)는 항산화 활성과 항암 활성을 가진 페놀성 물질이다. Streptomyces lincolnensis M-20 균주에서 생산된 PA를 균주 상등액에서 분리, 정제하였다. 항산화 활성을 가진 PA가 구리 이온 존재 하에서는 산화촉진제로 작용하였다. 항산화 활성은 DPPH를 이용한 방법으로 측정하였으며, 구리 이온 존재 하에서 PA의 산화 촉진 작용은 pBR322 플라스미드의 DNA 절단 작용으로 측정하였다. DNA 손상으로 생성되는 활성산소 종의 확인은 활성 산소종의 포집자인 글루타치온에 의해 DNA 절단이 억제되는 것으로 확인하였다. PA와 구리 이온의 복합체 형성은 금속 이온의 킬레이트인 EDTA가 존재할 경우와 존재하지 않을 경우를 자외선/가시광선 분광학적 분석법으로 비교, 확인하였다.

Purification and Cloning of an Extracellular Serine Protease from the Nematode-Trapping Fungus Monacrosporium cystosporium

  • An extracellular protease (Mc1) was isolated from the nematode-trapping fungus Monacrosporium cystosporium by gel filtration, anion-exchange, and hydrophobic interaction chromatographies. This protease had a molecular mass of approximately 38 kDa and displayed an optimal activity at pH 7-9 and $56^{\circ}C$ (over 30 min). Its proteolytic activity was highly sensitive to the serine protease inhibitor PMSF (phenylmethylsulfonylfluoride, 0.1 mM), indicating that it belonged to the serine-type peptidase group. The Michaelis constant ($K_m$) and $V_max$ for substrate N-Suc-Ala-Ala-Pro-Phe-pNA were $1.67{\times}10^{-4}\;M$ and 0.6071 $OD_{410}$ per 30 s, respectively. This protease could degrade a broad range of substrates including casein, gelatin, BSA (bovine serum albumin), and nematode cuticle. Moreover, the enzyme could immobilize the free-living nematode Panagrellus redivivus and the pine wood nematode Bursaphelenchus xylophilus, suggesting that it might playa role in infection against nematodes. The encoding gene of Mc1 was composed of one intron and two exons, coding for a polypeptide of 405 amino acid residues. The deduced amino acid sequence of Mcl showed 61.4-91.9% identity to serine proteases from other nematode-trapping fungi. Our results identified that Mcl possessed biochemical properties including optimal reaction condition and substrate preference that are different from previously identified serine proteases.

Binding of Lichen Phenolics to Purified Secreted Arginase from the Lichen Evernia prunastri

  • Secreted arginase from Evernia prunastri thallus has been purified 616-fold from the incubation medium. Purified arginase was resolved as only one peak in a capillary electrophoresis with a pI value of 5.35. The protein contained high amounts of acidic amino acids, such as Asx and Glx, and a relatively high quantity of Ser and Gly. The molecular mass of native, purified arginase was estimated as about 26 kDa by SE-HPLC. Substrate saturated kinetic showed a typical Michaelis-Menten relationship with a K_m value of 3.3 mM L-arginine. Atranorin behaved as a mixed activator of the enzyme (apparent $K_m$ = 0.96 mM); whereas evernic and usnic acid were revealed as non competitive inhibitors (apparent $K_m$ values were 3.16 mM and 3.05 mM, respectively). Kinetics of atranorin binding indicated that saturation was reached from 0.18 ${\mu}mol$ of the total atranorin and the occurrence of multiple sites for the ligand. This agrees with a possible aggregation of several enzyme subunits during the interaction process. A value of binding sites of about 12 was obtained. The binding of evernic acid was saturated from 23 nmol of total phenol. The number of binding sites was about 5. The loss of the binding ability of evernic acid could be interpreted as a single negative cooperatively. Usnic acid behaves in a similar way to evernic acid, although the binding saturation occurs at $0.14\;{\mu}moles$ of the ligand. This binding appears to be unspecific, and has 28 usnic acid binding sites to the protein.

HST Pixel Analysis of NGC 5194

  • We report the HST pixel analysis results of the interacting face-on spiral galaxy, NGC 5194 (M51), which is the first step of a new observational research project, PANCluG (Pixel Analysis of Nearby Cluster Galaxies). We derive several quantities describing the pixel color-magnitude diagram (pCMD) of NGC 5194, suitable for future comparisons with the pCMDs of other galaxies. We investigate the spatial distribution of pixel stellar populations, finding that the spiral arm pattern and the tidal interaction with NGC 5195 significantly affect the stellar populations in NGC 5194. We find that the pixels corresponding to the central active galactic nucleus (AGN) of NGC 5194 show a very tight sequence in the bright-end of the pCMD, of which spatial distribution seems to agree with the AGN torus region.

The Effect of Dietary Selenium Source and Vitamin E Levels on Performance of Male Broilers

  • Selenium and vitamin E are micronutrients essential for normal health and maintenance in poultry. They are necessary in preventing free radical damage to phospholipid membranes, enzymes and other important molecules. Two experiments were conducted in a semi-commercial environment to examine the effect of Se source and vitamin E level in diet on broiler performance and meat quality. Increasing vitamin E from 50 IU to 100 IU did not affect growth performance of broilers although the 24 h drip-loss was tended to be reduced (p=0.06). There was an interaction between vitamin E and the source of Se in glutathione peroxidase activity (GSH-Px) and Se concentration in excreta. Increasing vitamin E from 50 IU to 100 IU elevated GSH-Px and Se concentration in excreta by 42 IU/g Hb and 0.9 ppm for the organic Se group, respectively, but reduced GSH-Px and Se concentration in excreta by 16 IU/g Hb and 1.3 ppm for inorganic group, respectively. Vitamin E played no role in the feather coverage of the birds when scored on day 37. Organic Se is more effective in improving feather score and 24 h drip-loss, with a markedly higher deposition rate in breast muscle and a lower excretion rate in the excreta (p<0.05) compared to the inorganic Se source. Both vitamin E and the source of Se did not affect (p>0.05) the energy utilisation by birds.

Purification and Characterization of Protein Phosphatase 2A from Petals of the Tulip Tulipa gesnerina

  • The holoenzyme of protein phosphatase (PP) from tulip petals was purified by using hydrophobic interaction, anion exchange and microcystin affinity chromatography to analyze activity towards p-nitrophenyl phosphate (p-NPP). The catalytic subunit of PP was released from its endogenous regulatory subunits by ethanol precipitation and further purified. Both preparations were characterized by immunological and biochemical approaches to be PP2A. On SDS-PAGE, the final purified holoenzyme preparation showed three protein bands estimated at 38, 65, and 75 kDa while the free catalytic subunit preparation showed only the 38 kDa protein. In both preparations, the 38 kDa protein was identified immunologically as the catalytic subunit of PP2A by using a monoclonal antibody against the PP2A catalytic subunit. The final 623- and 748-fold purified holoenzyme and the free catalytic preparations, respectively, exhibited high sensitivity to inhibition by 1 nM okadaic acid when activity was measured with p-NPP. The holoenzyme displayed higher stimulation in the presence of ammonium sulfate than the free catalytic subunit did by protamine, thereby suggesting different enzymatic behaviors.

Mesoscopic numerical analysis of reinforced concrete beams using a modified micro truss model

  • Concrete is a heterogeneous material consisting of coarse aggregate, mortar matrix and interfacial zones at the meso level. Though studies have been done to interpret the fracture process in concrete using meso level models, not much work has been done for simulating the macroscopic behaviour of reinforced concrete structures using the meso level models. This paper presents a procedure for the mesoscopic analysis of reinforced concrete beams using a modified micro truss model. The micro truss model is derived based on the framework method and uses the lattice meshes for representing the coarse aggregate (CA), mortar matrix, interfacial zones and reinforcement bars. A simple procedure for generating a random aggregate structure is developed using the constitutive model at meso level. The study reveals the potential of the mesoscopic numerical simulation using a modified micro truss model to predict the nonlinear response of reinforced concrete structures. The modified micro truss model correctly predicts the load-deflection behaviour, crack pattern and ultimate load of reinforced concrete beams failing under different failure modes.

Integral effect test for steam line break with coupling reactor coolant system and containment using ATLAS-CUBE facility

  • To improve safety analysis technology for a nuclear reactor containment considering an interaction between a reactor coolant system (RCS) and containment, this study aims at an experimental investigation on the integrated simulation of the RCS and containment, with an integral effect test facility, ATLAS-CUBE. For a realistic simulation of a pressure and temperature (P/T) transient, the containment simulation vessel was designed to preserve a volumetric scale equivalently to the RCS volume scale of ATLAS. Three test cases for a steam line break (SLB) transient were conducted with variation of the initial condition of the passive heat sink or the steam flow direction. The test results indicated a stratified behavior of the steam-gas mixture in the containment following a high-temperature steam injection in prior to the spray injection. The test case with a reduced heat transfer on the passive heat sink showed a faster increase of the P/T inside the containment. The effect of the steam flow direction was also investigated with respect to a multi-dimensional distribution of the local heat transfer on the passive heat sink. The integral effect test data obtained in this study will contribute to validating the evaluation methodology for mass and energy (M/E) and P/T transient of the containment.

Comparison of Methodologies to Quantify Phytate Phosphorus in Diets Containing Phytase and Excreta from Broilers

  • The use of a suitable methodology to quantify the phytate phosphorus ($P_{phy}$) content in both the feed and the excreta from broilers is required to enable accurate calculation of the catalytic efficiency of the phytase supplemented in the feed. This study was conducted to compare 2 analytical methodologies (colorimetry and also high-performance liquid chromatography with a refractive index detector) in order to calculate the phytase efficiency by utilizing the results from the methodology that was shown to be the most appropriate. One hundred and twenty broilers were distributed in a $(4+1){\times}2$ factorial arrangement, corresponding to 4 diets that were equally deficient in P supplemented with increasing levels of phytase (0, 750, 1,500, and 2,250 units of phytase activity - FTU - per kg of feed) plus 1 positive control diet without phytase, supplied to male and female birds. The result indicated that the colorimetric methodology with an extraction ratio of 1:20 (mass of sample in g:volume of the solvent extractor in mL) was shown to be the most adequate. There was no interaction between the phytase level and the sex of the broilers (p>0.05). Males consumed 12% more $P_{phy}$ than did females (p<0.01), but the sex of the broilers did not affect (p>0.05) the excretion and retention coefficient of $P_{phy}$. The increase in the phytase level of the diet reduced (linear, p<0.01) the $P_{phy}$ excretion. The greatest $P_{phy}$ retention was estimated at 87.85% when the diet contained 1,950 FTU/kg (p<0.01), indicating that it is possible to reduce the inorganic P in the formulation at an amount equivalent to 87.85% of the $P_{phy}$ content present in the feed, which, in this research, corresponds to a decrease in 2.86 g of P/kg of the feed.
