Plant Regeneration Via Direct Adventitious Roots from Free Root Segments of Ulmus davidiana Planch (당느릅나무(Ulmus davidiana Planch) 부정근 절편으로 부터 부정아 유도를 통한 식물체 생산)

  • Micropropagation of Ulmus davidiana Planch was established via adventitious shoot formation from the segments of adventitious roots. Adventitious roots were produced directly from root segments of seedlings on a 1/2 SH medium plus various concentrations of IBA. The maximum growth of adventitious roots was observed in the presence of 2.0 mg/L IBA. After the segments of adventitious roots were cultured on various cytokinins (zeatin, 2-iP, BA, kinetin) and cytokinins plus auxin (IBA), formation of adventitious shoot was investigated. Among cytokinin treated, kinetin was the most effective on both adventitious shoot induction and number of shoots. Especially, 2.0 mg/L kinetin was the best to increase adventitious shoot induction (95.8%) and a number of shoots (8.4). Adventitious shoots were rooted on 1/2 WPM medium and the plantlets were acclimated 100% on composed soil (peatmoss : vermiculite 1 : 1).

  • Poly(vinyl chloride)-g-poly(oxyethylene methacrylate) (PVC-g-POEM) graft copolymer was synthesized via atom transfer radical polymerization (ATRP) and used as an electrolyte for electrochromic device. Plasticized polymer electrolytes were prepared by the introduction of propylene carbonate (PC)/ethylene carbonate (EC) mixture as a plasticizer. The effect of salt was systematically investigated using lithium tetrafluoroborate ($LiBF_4$), lithium perchlorate ($LiClO_4$), lithium iodide (LiI) and lithium bistrifluoromethanesulfonimide (LiTFSI). Wide angle X-ray scattering (WAXS) and differential scanning calorimetry (DSC) measurements showed that the structure and glass transition temperature ($T_g$) of polymer electrolytes were changed due to the coordinative interactions between the ether oxygens of POEM and the lithium salts, as supported by FT-IR spectroscopy. Transmission electron microscopy (TEM) showed that the microphase-separated structure of PVC-g-POEM was not greatly disrupted by the introduction of PC/EC and lithium salt. The plasticized polymer electrolyte was applied to the electrochromic device employing poly(3-hexylthiophene) (P3HT) conducting polymer.

  • This study was conducted to see what types of bovine rotaviruses were isolated at Jedong farm in Jeju province and Seohwa farm in Chungnum province. The results were as follows. 1. Rotavirus was positively detected in 18 out of 39 fecal samples from calves with diarrhea in Jeju province and in 13 out of 18 fecal samples from calves with diarrhea in Chungnam province. 2. The electropherotype pattern of dsRNA for 31 viruses was shown to be 4 : 2 : 3 : 2 type like traditional group A and the imigration pattern of dsRNA was the long type like NCDV (G6), JBR (G6), B223 (G10) and KK3 (G10). 3. The serotypes of the 18 viruses of Jedong and 9 viruses of Seowha were shown to be group A, subgroup I, G6, and P1 by ELISA and PCR analyses. The serotypes of S-2, S-6, S-9 and S-12 viruses of Seowha were shown to be group A, subgroup I, G10, but was not shown to be P type. 4. The partial nucleotide sequence of VP4 of S-8 was 97% homology with that of BRV 033. VP4 of J-10 showed 96% homology with that of BRV 033 in nucleotide sequence.

A Review of Geochemical Factors Governing the Phase Transformation of Birnessite (버네사이트 상변화 반응의 지화학적 반응 조절인자 연구)

  • Birnessite is one of the dominant Mn (oxyhydr)oxide phases commonly found in soil and deep ocean environments. It typically occurs as nano-sized and poorly crystalline aggregates in the natural environment. It is well known that birnessite participates in a wide variety of bio/geochemical reactions as a reactive mineral phase with structural defects, cation vacancies, and mixed valences of structural Mn. These various bio/geochemical reactions control not only the fate and transport of inorganic and organic substances in the environment, but also the formation of diverse Mn (oxyhydr)oxides through birnessite transformation. This review assessed and discussed about the phase transformation of birnessite under a wide range of environmental conditions and about the potential geochemical factors controlling the corresponding reactions in the literature. Birnessite transformation to other types of Mn (oxyhydr)oxides were affected by dissolved Mn(II), dissolved oxygen, solution pH, and co-existing cation (i.e., $Mg^{2+}$). However, there still have been many issues to be unraveled on the complex bio/geochemical processes involved in the phase transformation of birnessite. Future work on the detail mechanisms of birnessite transformation should be further investigated.

Effect of Deep Sea Water on Phase I, Phase II and Ornithine Decarboxylase. (Phase I, phase II 효소 및 ornithine decarboxylase에 미치는 해양심층수의 영향)

  • Deep sea water was tested for cancer chemopreventive activity by measuring the activities of ${\beta}-$ naphthoflavone $({\beta}-NF)-induced$ cytochrome P 450 1A2 (CYP 1A2), quinone reductase (QR) and glutathione-S-transferase (GST), glutathione (GSH) levels, and ornithine decarboxylase (ODC) activity. The in vitro incubation of rat liver microsome with deep sea water (a hardness range of $100{\sim}1,000$) showed a hardness-dependent inhibition of CYP 1A2 activity. QR and GST activities were induced about $1.1{\sim}1.2$ fold with the treatment of deep sea water in murine hepatoma Hepa 1clc7 cells. In addition GSH levels were increased $1.3{\sim}1.4$ fold in a hardness range of $100{\sim}1,000$. The deep sea water showed 20.3 and 35.0% inhibition of 12-O- tetradecanoylphorbol-13-a-cetate (TPA)-induced ODC activity at hardness 800 and 1,000, respectively. Therefore, deep sea water is worth further investigation with respect to cancer chemoprevention or therapy.

Cultural Characteristics for Inducing Fruting-body of Isaria japonica (눈꽃동충하초의 자실체 유도를 위한 배양조건)

  • To obtain basic data for mass production of Isaria japonica, cultural characteristics of japonica were investigated by using liquid, solid media and silkworms pupa. Mycelia grew favorably at the temperature of $23{\sim}28^{\circ}C$ on MYG medium with pH 7.0. The fruiting-body of I. japonica was induced below $20^{\circ}C$ in MYG liquid medium (Malt yeast glucose) under fluorescent light. In MYG basal medium mixed with pupal powder of silkworms, the fresh weight of fruiting-bodies was increased with increasing concentration of pupal powder. The highest yield of fruiting bodies was obtained in carbon-rich medium supplemented with pupal powder of silkworm. Also, fruiting-bodies of I. japonica were produced massively on the silkworm pupa placed on the stainless tray in the shortest time. The structure and shape of fruiting-bodies were coral-like, many-branched types with numerous conidiospores.

Determination of thyroid hormones in plasma samples by high performance liquid chromatograph/diode array detector/electrospray ionization mass spectrometer (HPLC/DAD/ESI-MS를 이용한 혈장시료 중 갑상선 호르몬 분석)

  • An analytical method for the determination of thyroid hormones in plasma samples has been studied by solid-phase extraction and high-performance liquid chromatography/diode array detector (DAD)/electrospray ionization (ESI)-mass spectrometer. Seven thyroid hormones were successfully separated by gradient elution on the reverse phase Hypersil ODS column (4.6 mm I.D., 250 mm length, particle size $5{\mu}m$) with ammonium formate buffer and acetonitrile. In addition, these compounds were confirmed by UV spectra and ESI-mass Spectra. The extraction recoveries of thyroid hormones in the plasma sample (at pH 3) were in the range of 74.5-115.7 % with solid-phase extraction by C18, followed by elution with 4 mL of methanol. The calibration curves showed good linearity with the correlation coefficients ($r^2$) varying from 0.9939 to 0.9978 and the detection limits of all analytes were obtained in the range of 20-50 ng/mL (38.1-162.8 pmol/mL). As a result, thyroxine was found in the range of 50.98-112.97 ng/mL in normal plasma samples.

  • The grid-based KIneMatic wave STOrm Runoff Model (KIMSTORM) by Kim (1998) predicts the temporal variation and spatial distribution of overland flow, subsurface flow and stream flow in a watershed. The model programmed with C++ language on Unix operating system adopts single flowpath algorithm for water balance simulation of flow at each grid element. In this study, we attempted to improve the model by converting the code into FORTRAN 90 on MS Windows operating system and named as ModKIMSTORM. The improved functions are the addition of GAML (Green-Ampt & Mein-Larson) infiltration model, control of paddy runoff rate by flow depth and Manning's roughness coefficient, addition of baseflow layer, treatment of both spatial and point rainfall data, development of the pre- and post-processor, and development of automatic model evaluation function using five evaluation criteria (Pearson's coefficient of determination, Nash and Sutcliffe model efficiency, the deviation of runoff volume, relative error of the peak runoff rate, and absolute error of the time to peak runoff). The modified model adopts Shell Sort algorithm to enhance the computational performance. Input data formats are accepted as raster and MS Excel, and model outputs viz. soil moisture, discharge, flow depth and velocity are generated as BSQ, ASCII grid, binary grid and raster formats.

Effect of Vinclozolin on the Onset of Puberty in Immature Female Rats (미성숙 암컷 흰쥐의 사춘기 개시에 미치는 Vinclozolin의 영향)

  • Vinclozolin(VCZ), a systemic dicarboximide fungicide, has been used in the control of diseases caused by microorganism of some species in fruits, vegatables and ornamental plants. Although VCZ itself is a very weak antagonist for androgen receptor binding, both melabolites M1 and M2 are effective antagonists. The present study was undertaken to examine whether prepubertal exposure to VCZ affects on the onset of puberty and the associated reproductive parameters such as hormone receptor expressions in female rats. VCZ(10 mg/kg/day) was administered daily from postnatal day 21(PND 21) through the day when the first vaginal opening(V.O.) was observed. Gross anatomy and weight of reproductive tissues were compared to test the VCZ's effects on the cell proliferation. Furthermore, histological studies were performed to assess the structural alterations in the tissues. To determine the transcriptional changes in progesterone receptor(PR), total RNAs were extracted and applied to the semi-quantitative reverse transcription polymerase chain reaction(RT-PCR). As a result, delayed V.O. was shown in the VCZ group(PND $34.00{\pm}1.22$) compared to the control group(PND $38.20{\pm}1.92$; p<0.01). VCZ treatment significantly decreased the wet weight of ovaries and uteri compared to the control group(p<0.01). Graafian follicles and corpora lutea were observed only in the ovaries from the control animals, while numerous primary, secondary follicles and small atretic follicles were observed in the ovaries from VCZ group. Similarly, hypotrophy of luminal and glandular uterine epithelium was found in the VCZ group. In the semi-quantitative RT-PCR studies, the transcriptional activity of PR in ovary(p<0.01) from VCZ group were significantly lower than those from the control group while in uterus were similar compared with the control group. The present studies demonstrated that the acute exposure to VCZ during the critical period of prepubertal stage could inactivate the reproductive system resulting delayed puberty in female rats.

The Design and Fabrication of Conversion Layer for Application of Direct-Detection Type Flat Panel Detector (직접 검출형 평판 검출기 적용을 위한 변환층 설계 및 제작)

  • Recently, Interest to the photoconductor, which is used to flat form X-ray detector such as a-Se, $HgI_2$, PbO, CdTe, $PbI_2$ etc. is increasing. In this study, the film layer by using the photoconductive material with particle sedimentation was fabricated and evaluated. The quantization efficiency of the continuous X-ray with the 70 kVp energy bandwidth was analyzed by using the Monte Carlo simulation. With the results, the thickness of film with 64 % quantization efficiency was 180 ${\mu}m$ which is similar to the efficiency of 500 ${\mu}m$ a-Se film. And $HIg_2$ film has the high quantization efficiency of 74 % on 240 ${\mu}m$ thickness. The electrical characteristics of the 239 ${\mu}m$ $Hgl_2$ films produced by particle sedimentation were shown as very low dark current(under 10 $pA/mm^2$), and high sensitivity(19.8 mC/mR-sec) with 1 $V/{\mu}m$ input voltage. The SNR, which is influence to the contrast of X-ray image, was shown highly as 3,125 in low driving voltage on 0.8 $V/{\mu}m$. With the results of this study, the development of the low-cost, high-performance image detector with film could be possible by replacing the film produced by particle sedimentation instead to a-Se detector.
