A unicellular red microalga was isolated from environmental freshwater in Korea, and its morphological, molecular, and biochemical properties were characterized. Morphological analysis revealed that the isolate was a unicellular biflagellated green microalga that formed a non-motile, thick-walled palmelloid or red aplanospore. To determine the taxonomical position of the isolate, its 18S rRNA and rbcL genes were sequenced and phylogenetic analysis was performed. We found that the isolate was clustered together with other related Haematococcus strains showing differences in the rbcL gene. Therefore, the isolated microalga was classified into the genus Haematococcus, and finally designated Haematococcus sp. KORDI03. The microalga could be cultivated in various culture media under a broad range of pH and temperature conditions. Compositions of the microalgal cellular components were analyzed, and its protein, carbohydrate, and lipid compositions were estimated to be 21.1 ± 0.2%, 48.8 ± 1.8%, and 22.2 ± 0.9%, respectively. In addition, D-glucose and D-mannose were the dominant monosaccharides in the isolate, and its amino acids were composed mainly of aspartic acid, glutamic acid, alanine, and leucine. Moreover, several polyunsaturated fatty acids accounted for about 80% of the total fatty acids in Haematococcus sp. KORDI03, and the astaxanthin content in the red aplanospores was estimated to be 1.8% of the dry cell weight. To the best of our knowledge, this is the first report of an Haematococcus sp. isolated from Korea, which may be used for bioresource production in the microalgal industry.
은연어 해상 사육 종묘의 나이에 따른 성장을 알아보기 위하여 1988년 12월 17일 부터 이듬해 4월 26일 까지 평균수온 $9.5\~14.5^{\circ}C$의 조건을 가진 경남 통영군 산양면 저도에 위치한 해상 가두리에서 실험을 실시하였다. 1 차 실험에서 평균 체중 167.0 g 되는 0+ 세어는 66일 후 633.1 g으로 성장하여 1 일 성장률 $2.0\%$, 사료계수 1.1을 나타냈다. 반면, 평균체중 396.0 g되는 1+ 세어는 854.6 g으로 성장하여 1 일 성장률 및 사료계수 각각 $2.0\%$ 및 1.1을 기록하였다. 2차 실험에서는 0+ 세어가 64일 후 1171.9 g으로 성장하여 1일 성장률 $1.0\%$, 사료계수 1.5로 나타났고, 1+ 세어는 1239.7 g으로 성장하여 1 일 성장률 및 사료계수 각각 $0.7\%$ 및 2.6을 기록하였다. 은연어의 해상 사육에 있어서 그 크기가 증가함에 따라 1 일 성장률 및 사료효율이 급격히 감소하는 것으로 나타났다. 실험 기간 동안 0+ 재어가 사료섭취율이 훨씬 우수하여 0+ 세어가 해상사육 종묘로 적합한 것으로 나타나 당년에 해상 사육용 종묘를 smolt 될 수 있도록 할 필요가 있다.
Mycosporine-like 아미노산(MAAs)은 UV 흡수물질이며, 다양한 해양생물들은 MAAs의 합성과 축적을 통하여 환경 자외선의 직 간접적인 영향을 감소시키는 기능을 진화시켜 왔다. 이 연구에서 우리는 radiofrequency(RF) 발생장치를 제작하였고, 이를 미세조류 배양에 적용하였다. $0.35{\pm}0.05$ mHz의 RF를 Chlamydomonas sp. 배양기에 공급하였고, 정해진 시간(0, 0.5, 1 및 2 시간)에 시료를 채취하였다. MAAs 생합성 관련 유전자인 dehydroquinate synthase homolog (DHQS-like)와 nonribosomal peptide synthetase homolog (NRPS-like) 유전자를 Chlamydomonas sp.로부터 클로닝하였고, RF 노출에 대한 유전자 발현을 qRT-PCR을 이용하여 분석하였다. 연구결과 RF에 노출된 Chlamydomonas sp.의 DHQS-like와 NRPS-like 유전자의 발현은 노출 1시간에 각각 1.46 배 및 1.19 배 증가하였다. 이러한 결과는 DHQS-like와 NRPS-like 유전자가 Chlaydomonas sp.의 MAAs 생합성을 진단할 수 있는 좋은 바이오마커 후보가 될 수 있음을 의미한다.

Streptomycetes are Gram-positive microorganisms producing secondary metabolites through unique physiological differentiation [4]. The microbes show unusual morphological differentiation to form substrate mycelia, aerial mycelia, and arthrospores on solid medium [19]. Substrate mycelium growth is sustaining with sufficient nutrients in the culture medium. The concentration of a specific individual substrate in the culture environment is the most important extracellular factor allowing vegetative mycelia growth, where extracellular hydrolytic enzymes participate in the utilization of waterinsoluble substrates. However, with starvation of nutrients in the culture medium, the vegetative mycelia differentiate to aerial mycelia and spores. It has been considered that shiftdown of essential nutrients for mycelia growth is the most important factor triggering morphological and physiological differentiation in Streptomyces spp. Since proteineous macromolecule compounds are the major cellular components, these are faced to endogenously metabolize following a severe depletion of nitrogen source in culture nutrients (Fig. 1). Various proteases were identified of which production was specifically related with the phase of mycelium growth and also morphological differentiation. The involvement of proteases and protease inhibitor is reviewed as a factor explaining the mycelium differentiation in Streptomyces spp.

Although soft tunic syndrome (STS) in the ascidian is a serious disease, helpful measures have yet not been established. It was examined in this study by applying aniti-parasitic drugs to eradicate the causative protozoa Azumiobodo hoyamushi from infected ascidians. Formalin was synergistic in killing parasites in vitro when co-treated with hydrogen peroxide ($H_2O_2$) or bronopol, but not with chloramine-T or povidone-iodine (PVP-I), when tested with in vitro parasite culture. The synergistic effects did not change when $formalin-H_2O_2$ (or bronopol) ratios were changed. It was found that treatment periods less than 60 min achieved a sub-maximal efficacy. Increasing drug concentration while keeping 30 min period improved anti-parasitic effects. Anti-parasitic effects of $formalin(F)+H_2O_2$(H) were also assessed in an in vivo STS model infected with cultured parasites. It was observed that combined 50 (40F + 10H) and 100 (80F +20H) ppm were effective in partially preventing STS-caused mortality. In horizontally transmitted artificial STS model, significant prevention of ascidian mortality was also observed after 50 ppm. Marked reduction of living parasites were noted after drug treatments in vivo. The results provide a highly useful basis to develop a preventive or treatment measure against the currently uncontrollable STS in the ascidian.
Recently, we reported violapyrones B, C, H and I, unusual 3, 4, 6-trisubstituted ${\alpha}-pyrones$ derivatives, from the culture broth of the marine Streptomyces sp. 112CH148. In previous studies, violapyrones have been shown to have antibacterial and antitumor activities. However, the anti-inflammatory effect of violapyrones has not been reported yet. As part of our ongoing study for the discovery of bioactive metabolites from marine microorganisms, we found that violapyrones also have anti-inflammatory activity. In this study, we investigated the effect of violapyrones on LPS-induced inflammatory responses in vitro. Violapyrones B and C did not affect the viability of RAW 264.7 cells at concentrations up to $25{\mu}M$. However, violapyrones B and C inhibited the production of NO compared to the LPS-induced control. In addition, violapyrones B and C down-regulated the expression of iNOS protein in LPS-stimulated RAW 264.7 cells. To the best of our knowledge, this is the first report on the anti-inflammatory activity of violapyrones B and C.
Despite recent studies, relatively few are known about the diversity of fungal communities in the deep Atlantic Ocean. In this study, we investigated the diversity of fungal communities in 15 different deep-sea sediments from the South Atlantic Ocean with a culture-dependent approach followed by phylogenetic analysis of ITS sequences. A total of 29 fungal strains were isolated from the 15 deep-sea sediments. These strains belong to four fungal genera, including Aspergillus, Cladosporium, Penicillium, and Alternaria. Penicillium, accounting for 44.8% of the total fungal isolates, was a dominant genus. The antiaflatoxigenic activity of these deep-sea fungal isolates was studied. Surprisingly, most of the strains showed moderate to strong antiaflatoxigenic activity. Four isolates, belonging to species of Penicillium polonicum, Penicillium chrysogenum, Aspergillus versicolor, and Cladosporium cladosporioides, could completely inhibit not only the mycelial growth of Aspergillus parasiticus mutant strain NFRI-95, but also the aflatoxin production. To our knowledge, this is the first report to investigate the antiaflatoxigenic activity of culturable deep-sea fungi. Our results provide new insights into the community composition of fungi in the deep South Atlantic Ocean. The high proportion of strains that displayed antiaflatoxigenic activity demonstrates that deep-sea fungi from the Atlantic Ocean are valuable resources for mining bioactive compounds.
Differential gene expression profiling was carried out in the hepatic tissue of medaka fish, Oryzias latipes, after exposure to an organophosphorus pesticide (OPP), Iprobenfos (IBP), a widely used pesticide in agri- and fish-culture, using a medaka cDNA micro array. Twenty six kinds of differentially expressed candidate genes, with 15 and 11 induced and repressed in their gene expressions, respectively, were associated with cytoskeleton (3.8%), development (7.7%), immune (7.7%), metabolism (30.8%), nucleic acid/protein binding (42.3%) and reproduction (7.7%). Of these genes, changes at the transcription level of five were re-evaluated by real-time quantitative PCR (qRT-PCR). Considering the known function of authentic genes, the effects of IBP on the biological activity and pathological aspects in medaka fish were discussed. The identified genes could be used as molecular biomarkers for biological responses to OPPs contamination in an aquatic environment.
Organic pollution causes eutrophication and dystrophication, which occur when excessive amounts of organic matter enters seawater. Eutrophication can contaminate sediment and harm aquaculture. Polychaeta species have been shown to restore eutrophic sediment. In this study, we used peptone to simulate a eutrophic environment and detect the levels at which eutrophication became toxic to the polychaete Perinereis aibuhitensis. Peptone concentrations were 0, 100, 200, and 500 mg/L. The median lethal concentrations were 950.35 mg/L at 48 h, 340.34 mg/L at 72 h, and 120.22 mg/L at 96 h, which are much higher than those of other aquatic species. Polychaeta species are highly tolerant of eutrophication. During the 15-day long-term experiment, sediment loss on ignition, as well as seawater total organic carbon and total nitrogen all decreased significantly (P<0.05). However, $NH_4^+$ concentration increased with time. Perinereis aibuhitensis slowed the increment of $NH_4^+$ but could not prevent its increase. Our results indicate that this polychaete is helpful in the recovery of seawater and sediment from eutrophication.
The sea cucumber, Apostichopus japonicus, is becoming an important aquaculture species in China, Japan and Korea. The purpose of this study is to evaluate the biological performance of a tubular type of subsurface cage for sea cucumber grow-out by in-situ tests. The cage structure was constructed of PP (polypropylene) frames and PP tubes (${\phi}130mm{\times}24pieces$). A set of tests were conducted at a depth of 15 m near Seosaeng, Ulsan, Korea for 295 days (23 July, 2012 to 13 May, 2013). A total of 155 sea cucumbers were used and fed a mixed diet containing mud, mineral, wheat, fish meal, etc (3% of their body weight). At the end of the experiments, sea cucumber showed a higher survival rate in feed shelter (77.14%) rather than no feed shelter (64.71%). The specific growth rate (SGR) of sea cucumbers in feed condition (0.04% $day^{-1}$) was higher than that of no feed culture animals (-0.49% $day^{-1}$) during total experimental periods. The result was able to find a no significance difference in survival rate but a significance difference in SGR under feed and no feed treatments.
