Morphological Characteristics of Decomposition and Browning of Oak Sawdust Medium for Ground Bed Cultivation of Lentinula edodes (표고 지면재배용 참나무 톱밥배지의 분해와 갈변의 형태적 특성)

  • This study investigated the internal and external morphological characteristics of decomposition and browning of oak sawdust medium for ground bed cultivation of Lentinula edodes. Within fifty days after L. edodes inoculation, surface hyphae on the bed browned. In 110 days, the fungal hyphae occupied and decomposed wood fibers, vessels and parenchymatous cells from the inside as white profuse hyphal mass was amorphously dissolving the saw dust particles from the outer surface. Most of the white hyphal bed surface became cleanly brown, however, some colony surface became blackened and slimy with contaminating bacteria, hyphae and spores. The brown layer was ca. 0.34 mm thick with highly dense and white hyphal mass beneath, whereas the blackened layer was ca. 1.17 mm thick with shrunken hyphae and less decomposed sawdust particles beneath. The surface hardness of the brown surface was ca. $0.73kgf/cm^2$, soft and resilient, while that of the blackened was ca. $0.91kgf/cm^2$, hard and nonresilient. By 150 days Lentinula edodes mushrooms fruited only on the brown surface and not on the blackened medium.

Investigation of the shift trends of temperature and humidity in greenhouses for oak mushroom cultivation (최근 국내 표고 재배사의 온도, 습도 변화 성향 조사)

  • The temperature and humidity of 49 greenhouses for oak mushroom cultivation were investigated for 5 years to analyze the trends in the change of these parameters according to the climate change in Korea. The 5-year average temperature and humidity were $24.7^{\circ}C$ and 60.5%, respectively, in sawdust media-based cultivation houses and $24.4^{\circ}C$ and 60.0%, respectively, in log-bed cultivation houses. The average temperature in the summer was $29.8^{\circ}C$ in 2016, $29.1^{\circ}C$ in 2017, and $33.3^{\circ}C$ in 2018 in the log-bed cultivation houses and $26.8^{\circ}C$ in 2016, $20.4^{\circ}C$ in 2017, and $24.2^{\circ}C$ in 2018 in the sawdust media-based cultivation houses. During the investigation, temperatures over $30^{\circ}C$ were detected in one cultivation house in spring and five such houses in summer. When classifying by cultivation type, temperatures over $30^{\circ}C$ were found in five log-bed cultivation houses and temperatures less than $20^{\circ}C$ were found in four log-bed cultivation houses in fall. This study shows that log-bed cultivation houses for oak mushroom need to be modified to cope with the climate change.

Selection of parental strain on the sawdust cultivation and mycelial growth and cultural characteristics of Lentinula edodes hybrid strains (표고 톱밥재배용 모균주 선발과 교배균주의 균사배양 및 생육 특성)

  • In order to breed new strains of oak mushroom, Lentinula edodes, on the sawdust cultivation, we collected 10 from Korea, Taiwan and China respectively and examined somatic incompatibility, morphological features of fruiting body and productivity. Four strains(SANJO701HO, FMRI2534, FMRI2337 and FMRI2613) shown remarkable results, were confirmed with parental strains. 80 monokaryotic strains derived from the selected 4 parental strains, were selected and 117 hybrid strains were made by mono-mono mating. Aslo, Physiological characteristics were investigated. Average of the mycelial growth of hybrid strains of mating combination SANJO701HO-FMRI2337 was approximately 10% lower than other mating combinations. Overall, This study was founded to develop new varieties of L. edodes. The productivity of new 117 strains on sawdust cultivation needs continued research.

Cultural Characteristics of Pycnoporus coccineus and P. cinnabarinus (간버섯과 주걱간버섯의 배양특성)

  • Basic studies on the cultural characteristics of Pycnoporus coccineus and P. cinnabarinus were performed. They exhibited $30{\sim}40{\circ}C$ optimal temperature ranges and optimal pH ranges of $5{\sim}6$. Among 6 media, they were good at mycelial growths on PDA, LBA and YMA.P. coccineus grew more than P.cinnabarinus on the same medium. Among 10 sawdust media, they were good at mycelial growth on three oak trees and Alnus hirsuta. However, the sawdust of Castanea crenata was bad at mycelial growth. Among 3 coniferous trees, Larix leptolepis showed better growth than the other trees such as Pinus densiflora and P. koraiensis. The fruit body production P. coccineus was about twice better than P. cinnabarinus on Quercus spp. sawdust cultivation.

Effect of brewers dried grain as a nutrient supplement in plastic vinyl bag cultivation of maitake (Grifola frondosa S. F. Gray) (잎새버섯 봉지재배시 영양원으로서 맥주박 효과)

  • This study was performed to determine most appropriate mixed ratio of brewers dried grain to wheat bran as a nutrient supplement in the plastic vinyl bag cultivation of Grifola frondosa. In the examination of an appropriate nutrient source, the IV substrate formulation (80 : 10 : 10, oak sawdust : wheat bran : brewers dried grain) resulted in 71.6 days to harvest and the highest yield of 142.6 g/bag. On the other hand, the investigation of the optimal mixing ratio of beer waste to wheat bran showed that the III substrate formulation (80 : 5 : 15, oak sawdust : wheat bran : brewers dried grain) resulted in the least days to harvest (61.8 days), with the highest yield of 140.8 g/bag.

Effect of Peanut Seed Orientation on Germination, Seedling Biomass, and Morphology in an Oak Tree Sawdust Cultivation System

  • We performed seed germination tests to investigate the effects of seed sowing orientation on germination viability on peanut (Arachis hypogaea L.) sprouts. Specifically, we assessed the influence of seed sowing orientation on germination rate, seedling weight, and seedling length, as well as the seedling vigor index. The seeds were sown in oak tree sawdust at 3.0 cm depth. Four seed orientations were tested: vertical with the hypocotyl end down, vertical with the hypocotyl end up, horizontal with the hypocotyl end down, and horizontal with the hypocotyl end up. The mean seed germination percentages of the four seed orientations were significantly different (p < 0.01) and ranged from 25 to 91.7%. The vertical orientation with hypocotyl-end-down and hypocotyl-end-up orientations showed the highest (91.7%) and lowest (25%) germination rates, respectively. The vertical orientation with the hypocotyl end down produced the heaviest (4.9 g) seedlings and the longest hypocotyls (4.65 cm). This orientation also produced the longest true leaf + epycotyl (2.15 cm) and had the highest seedling vigor index (197.1). The seedlings had a straight growth pattern, whereas seedlings from seeds sown with the hypocotyl up had an awkward plumular hook shape. Taken together, to produce peanut sprouts, we recommend placing the seeds vertically with the hypocotyl end down because this orientation leads to a high germination rate, high biomass production, and high overall seedling quality.

  • Although facilities for the passive treatment of AMD (Acid Mine Drainage) are currently operating in Korea, their removal efficiency for heavy metals is relatively low in average (only 80%). Passive treatment system is composed of oxidation tank, SAPS (Successive Alkalinity Producing System), and wetland. In the treatment system adopted in korea, SAPS (Successive Alkalinity Producing System) plays a major role to remove about 65% of heavy metals through a precipitation. However, the efficiency of SAPS is limited due to the use of mushroom compost (MC) as a organic material and of limestone as a neutralizer. Therefore, this research was performed to search for alternative organic materials through the field test. We tested two types of mixed organic materials: 1) cow manure and spent oak (herein, CO) and 2) cow manure and sawdust (herein, CS). For comparison mushroom compost (herein, MC) was also tested. The result showed that the average Fe removal efficiency was 91.38% with CO, 85.19% with CS, and 91.58% with MC. Thus, CO can be effectively used as an alternative of MC in the SAPS system for heavy metals removal.

Fuel Characteristics of Pitch Pine and Mongolian Oak Pellets Fabricated with Coffee Waste and Used Frying Oil as an Additive Using a Pilot-scale Flat-die Pellet Mills (식품부산물인 커피박과 폐식용유를 첨가제로 사용하여 파일럿 규모의 평다이 성형기로 제조한 리기다소나무 및 신갈나무 펠릿의 연료적 특성)

  • This study evaluated the potential of coffee waste (CW) and used frying oil (UFO) as an additive in the production of pitch pine (PIP) and Mongolian oak (MOK) pellets. Ash contents obtained from CW and UFO were 0.5% and <0.1%, respectively. The calorific values of UFO (31.4 MJ/kg) and CW (26.3 MJ/kg) are higher than PIP (20.6 MJ/kg) and MOK (19.1 MJ/kg). For pellets fabricated using a pilot-scale flat-die pellet mill, regardless of fabricating conditions, moisture content (MC) and bulk density of PIP and MOK pellets satisfied the A1 wood pellet standard for residential and small-scale commercial uses, as designated by the National Institute of Forest Science (NIFOS) of the Republic of Korea. When CW was used as an additive, durability of PIP pellets made with 12%-MC sawdust and MOK pellets increased. The optimal conditions for producing PIP and MOK pellets could be by adding 20 mesh CW as an additive and the using of 12%-MC sawdust. However, durability of PIP pellets and ash content MOK pellets did not satisfy the A1 wood pellet standard of NIFOS. Thus, further research is needed to improve the properties of wood pellets with additives.

Appropriate temperature of antler-shaped Ganoderma lucidum using King Oyster Mushroom Cultivation System (큰느타리(King Oyster Mushroom) 재배시스템을 활용한 녹각영지버섯(Ganoderma lucidum) 적정 재배온도)

  • High-income mushroom crops require complex farming. Therefore, we conducted a test to identify the optimum temperature for the production of antler-shaped Ganoderma lucidum using the King Oyster mushroom cultivation facility. T-N showed 0.28% of oak sawdust and 2.2% of nutritional source. The pH of oak sawdust was 6.0, indicating weak acidity, and that of rice steel was 6.6, indicating neutrality in nutrition source. Study on the quality characteristics of mushrooms showed that the number of days at 25℃ were 5~6 and those at 30℃ were 3~5; the representative length at 25℃ was 57.5 mm and that at 30℃ was 92.2 mm; the biological weight at 25 ℃ was 43 g, which was greater than that at 30℃.

Compost Production using Vegetable Waste and Spent Oak Mushroom Substrate (SMS) (채소 부산물과 표고 수확후 배지를 활용한 퇴비 제조방법)

  • Spent mushroom substrate (SMS) has generally been used for the manufacture of animal feed and production of bio fuel. Limited research has been conducted in the utilization of SMS as a co-material for composting. Therefore, the main purpose of this study was to evaluate the feasibility of composting vegetable waste mixed with various ratios of SMS (30, 40, and 50%). The results showed that the C/N ratio decreased when both sawdust (from 22.0~28.8 to 17.7~20.4) and SMS (from 18.5~19.5 to 12.7~16.8) were applied for composing, owing to increased contents of nitrogen. A maturity test conducted using mechanical (Solvita) and germination tests revealed that both sawdust (92.0~101.9%) and SMS (87.8~89.2%) satisfied a criteria of maturity standard (70%). A correlation analysis between compost maturity and its chemical properties revealed that the C/N ratio and pH were the most dominant parameters for compost maturity. Overall, SMS could be utilized as a compost material and especially, vegetable waste mixed with SMS could provide sufficient nutrients for crop growth.
