The genomic mapping of Red Jungle Fowl (Gallus gallus), local Village Chicken, and broiler was carried out by random amplified polymorphism DNA (RAPD) technique. Two different sets of arbitrary primers were used (Operon OPA01-20 and Genemed GM01-50). All the genomes of the three species of chickens were amplified with OPA01-20 primers. The genomes of the Red Jungle Fowl and local Village Chicken were further amplified with GM01-50 primers. Analysis of the results based on band sharing (BS) and the molecular size of individually amplified DNA fragments showed that Red Jungle Fowl and local Village Chicken shared the species similarity of 66% with Operon primers 01-20, 64% between local Village Chicken and broiler, and 63% when DNA bands between Red Jungle Fowl and broiler were compared. With GM01-50, the BS between Red Jungle Fowl and local village chicken increased to 72%. The results showed that the local village chicken is more closely related to Red Jungle Fowl than to broiler in the genetic distance. On the other hand, broiler is 1% closer in genetic distance to local village chicken than to Red Jungle Fowl. The results also indicated that primers like OPA-7, 8 and 9 can be used as species specific DNA markers for these three species of chickens.

RAPD analyses using 60 OPERON primers and 13 URPs were performed in order to assess the genetic variation and frequency of polymorphisms in Korean salmonids. RAPDS were very reproducible and most useful at the sub-species level. In RAPD analysis, 138 polymorphic bands were detected between Oncorhynchus masou subspecies and 99 bands were generated in two types of rainbow trout. Estimated genetic distances between O. masou subspecies were 0.28794, and between wild rainbow trout and an albino mutant was 0.22786. Each species of salmonid was well characterized using URP 4R, the obtained bands could be useful as a species specific RAPD markers.

Three assay methods for quantification of the two galactose-operon mRNAs that only differ by 5 bases in their 5'-end are presented. The 5' ends of each mRNA were extended by ligating the 3'-end of the abundant 5S rRNA. This ligation extends the 5' ends of the two gal mRNAs long enough to be distinguished by the specific PCR primers in the following quantification reactions. Quantification of the corresponding cDNAs was performed either by primer extension assay or real-time qPCR. To circumvent the problem of the RNA ligation reaction (i.e. very low ligation efficiency), we devised a new method that employs real-time qPCR directly for the quantification of the gal transcripts which differ by 5 bases in their 5'-ends.
Bovine genome samples were collected from meat of three different beef breeds (Hanwoo, Holstein and imported beef breed) that are commercially merchandized in Korean beef market. Operon B (OPB)-kits were used as random primers (3, 7, 10, 11, 12, 14) in random amplified polymorphic DNA (RAPD) method on whole genome. Each primer provided characteristic bands that were highly polymorphic. Each single primer could provide relatively efficient polymorphic band patterns among breeds. However, use of two or more primers in combination is recommended to improve resolution of experiments with higher molecular weight bands of DNA. In our experiments, OPB-11 resolved well between beef cattle breeds and Holstein. And OPB-7, 12 and 14 could be combined with OPB-11 to identify Hanwoo beef from the other two kinds of beef.
Previously, we found the operon random primer (OP-5A) that is characteristic the genus Panax by randomly amplified polymorphic DNA (RAPD) analysis. However, OP-5A primer is limited to apply on the differentiation of only crude herbal plants. To construct more sensitive and unique primers on the genus Panax, ginseng-specific DNA profile (350 bp) that was amplified by OP-5A primer were inserted in a plasmid vector in the TA cloning method and sequenced. We designed the PCR primers (Forward: 5"-AGGGGTCTTGCTAT AGCGGAAC-3", Reverse: 5"-AGTCTTAATTTCATATTTTCGTATG-3") and identified the unique ginseng band (350 bp) in commercial granule products including ginseng extracts as well as crude ginseng plants by nascent PCR.(omitted)
Peaches (Prunus persica) are less popular than the fresh fruits, because their flesh gets soft faster. So many breeders focused on their aim to firmness. Other breeders focused on juiciness, flavor and aroma. Breeding requires much labor, time and money. To reduce these requirements, many scientists develop many SSR, CAPS and SCAR makers. New peach varieties bred in our National Institute of Horticultural & Herbal Science (NIHHS) such as, Cheonhong, Suhong and Harhong are yellow flesh cultivars and Yumyeong, Baekmijosaeng, Baekhyang, Jinmi, Soomee, Mihong, Misshong and Yumee are white flesh cultivars. These peach cultivars are planted in orchard of Korea. To assert breeding cultivar patents and prevent patent disputes, we detected cultivar-specific DNA fragment using 235 sets of Operon RAPD primers, analyzed 134 DNA sequences and constructed SCAR primers. To confirm the cultivar-specific SCAR markers, we applied candidate SCAR primers to 30 peach cultivars widely cultivated in Korea. These selected lines are included father and mother lines that were used to develop new varieties in NIHHS. Using fourteen SCAR primer sets, we characterized thirty cultivars selected. The SCAR marker is expected to serve as molecular evidence distinguishing different peach varieties.
Polymerase chain reaction (PCR) and Southern hybridization analyses were carried out to identify clones of silk worm thorn (Cudraria tricuspidata) and Rose of sharon (Hibiscus syriacus) which look like one tree with two ar three, branches or two or three different trees. For PCR five different PCR primers $(17{\sim}24\;nucleotides)$ are derived from CaMV 35S promoter, nopaline synthase terminator and coding region of thylakoid membrane protein gene. In the case of silk worm thorn, about 500 bp of PCR product was produced from DNAs of one tree or branch in the presence of 35S primer alone. Southern hybridization analysis of genomic DNAs hybridized with $^{32}P$ labeled PCR product showed that the same size of DNA fragments were hybridized with different intensities. In addition, PCR analyses using 20 different primers of OPERON 10-mer kits showed that only OPA01 primer produced PCR products of different size. These results indicate that two different trees of silk worm thorn combined to one tree. In the case of the Rose of Sharon, the same size of PCR products were produced from three different samples but Southern hybridization with the above PCR product as a probe did not show any hybridized bands. PCR analyses in the presence of OPERON 10-mers showed OPA04 and OPA13 produced different products including same sizes of products. These, results indicate that three different trees of the Rose of Sharon seem to be derived from the tree.

To analyze the genetic relationship 8 accessions of Ostericum koreanum Kitakawa, random amplified polymorphic DNA(RAPD) analysis was performed using 60 Operon primers. The 25 primers out of 60 random primers were amplified DNA by PCR using genomic DNA of O. koreanum. Eighty-five (49.1%) among 173 bands derived from 25 primers showed poly morphism, On the basis of similarity coefficient analysis by UPGMA, 8 accessions of O. koreanum Kitakawa could be classified into three groups at the similarity coefficient value of 0.71. Group I contained three accessions (Nam Gangwhal), Group II contained one accession (Nam Gangwhal) and Group III contained four accessions (Buk Gangwhal), The range of total genetic similarity coefficient value of 8 accessions of O. koreanum Kitakawa was $0.63{\sim}0.96$. Buk Gangwhal was flowered 18 to 26 days earlier than Nam Gangwhal, and Nam Gangwhal leaf stalk was thin and long as bolting rate high compared with Buk Gangwhal.
RAPD technique was employed for the genetic analysis of major Lilium cultivars and horticultural hybrids. As a result of RAPD with 10-mer random primers, total 107 bands were observed within 300bp and 2kb range, and the same band patterns were observed within the same cultivar for different primers. However, Casa Blanca in Orientals and Adelina in Asiatics showed different band patterns with others in the same. Cultivars within L. longiflorum showed different band patterns. RAPD markers produced with random primers OPA- 02, 03, 04, 14, 16 and 17 can be used for the classification of Lilium cultivars.

The genetic relationship among 48 Cymbidium cultivars was analyzed using randomly amplified polymorphic DNA (RAPD) with eighty 10 mers random primers (Operon Technologies) and twelve 20 mers random primers. Forty eight Cymbidium cultivars included 34 oriental Cymbidium, 7 hybrids, and 7 western Cymbidium. 407 (9.9 per primer) and 56 polymorphic bands (9.5 per primer) were generated by polymerase chain reaction with selected thirty 10 mers primers, and nine 20 mers primers, respectively. The polymorphic fragments ranged from 0.4 to 1.5 kb in size. The dendrogram was constructed by using the UPGMA clustering algorithm based on genetic similarity. Forty eight Cymbidium cultivars were classified into four major groups at similarity coefficient value of 0.638.
