Comparative Study of Enzyme Activity and Stability of Bovine and Human Plasmins in Electrophoretic Reagents, β-mercaptoethanol, DTT, SDS, Triton X-100, and Urea

  • Effects of common electrophoretic reagents, reducing agents ($\beta$-mercaptoethanol [BME] and DTT), denaturants (SDS and urea), and non-ionic detergent (Triton X-100), on the activity and stability of bovine plasmin (b-pln) and human plasmin (h-pln) were compared. In the presence of 0.1% SDS (w/v), all reagents completely inhibited two plns, whereas SDS (1%) and urea (1 M) denatured plns recovered their activities after removal of SDS by treatment of 2.5% Triton X-100 (v/v). However, reducing agents (0.1 M of BME and DTT) treated plns did not restore their activities. Based on a fibrin zymogram gel, five (from b-pln) and four (from h-pln) active fragments were resolved. Two plns exhibited unusual stability in concentrated SDS and Triton X-100 (final 10%) and urea (final 6 M) solutions. Two bands, heavy chain-2 (HC-2) and cleaved heavy chain-2 (CHC-2), of b-pln were completely inhibited in 0.5% SDS or 3 M urea, whereas no significant difference was found in h-pln. Interestingly, 50 kDa (cleaved heavy chain-1, CHC-1) of b-pln and two fragments, 26 kDa (light chain, LC) and 29 kDa (microplasmin, MP), of h-pln were increased by SDS in a concentration dependent manner. We also found that the inhibition of SDS against both plns was reversible.

COMPARISON OF PHYSICAL PROPERTIES AND EVOLUTION OF AKARI AND SPITZER 24 ㎛-DETECTED GALAXIES AT z = 0.4 - 2

  • We present physical properties of $24{\mu}m$ galaxies detected by AKARI and Spitzer and their evolution between redshifts 0.4 < z < 2. Using multi-wavelength data from X-ray to radio observations in NEP Deep Field (for AKARI) and Subaru/XMM-Newton Deep Field (for Spitzer), we derive photometric redshift, stellar mass, star-formation rate (SFR), dust extinction magnitude and rest-frame luminosities/colors of the $24{\mu}m$ galaxies from photometric SED fitting. We infer the SFRs from rest-frame ultraviolet luminosity and total infrared luminosity calibrated against Herschel photometric data. For both survey fields, we obtain complete samples with stellar mass of > $10^{10}M_{\odot}$ and SFR of > $30M_{\odot}/yr$ up to z = 2. We find that specific SFRs evolves with redshift at all stellar masses in NON-power-law galaxies (non-PLGs) as star-formation dominant luminous infrared galaxies (LIRGs). The correlations between specific SFR and stellar mass in the Spitzer and AKARI galaxy samples are well consistent with trends of the main sequence galaxies. We also discuss nature of PLGs and their evolution.

Electrical Characteristics of Resistive-Switching-Memory Based on Indium-Zinc-Oxide Thin-Film by Solution Processing (용액 공정을 이용한 Indium-Zinc-Oxide 박막 기반 저항 스위칭 메모리의 전기적 특성)

  • We investigated the rewritable operation of a non-volatile memory device composed of Al (top)/$TiO_2$/indium-zinc-oxide (IZO)/Al (bottom). The oxygen-deficient IZO layer of the device was spin-coated with 0.1 M indium nitrate hydrate and 0.1 M zinc acetate dehydrate as precursor solutions, and the $TiO_2$ layer was fabricated by atomic layer deposition. The oxygen vacancies IZO layer of an active component annealed at $400^{\circ}C$ using thermal annealing and it was proven to be in oxygen vacancies and oxygen binding environments with OH species and heavy metal ions investigated by X-ray photoelectron spectroscopy. The device, which operates at low voltages (less than 3.5 V), exhibits non-volatile memory behavior consistent with resistive-switching properties and an ON/OFF ratio of approximately $3.6{\times}10^3$ at 2.5 V.

  • An attempt has been made to analyze time series of $H_\alpha,\;H_\beta,\;and\; H_\gamma$ line profiles taken from a 3B/X6.1 flare which occurred on Oct. 27, 1991 in an active region, NOAA 6891. A total of 22 sets of $H_\alpha,\;H_\beta,\;and\; H_\gamma$ taken with a low and non-uniform time resolution of 10-40 seconds were scanned by PDS with absolute intensity calibration to derive the physical characteristics of the material in the flare chromosphere. Our . results are as follows: (1) The lower Balmer lines observed during the flare activity are broadened by Stark effect. (2) At the peak of the flare activity, the electron temperature of the Balmer line emitting region reaches up to 35000K and its geometrical thickness increases to a scale of $10^4km$, suggesting that high energy particles penetrate deep into the photospheric level.

Observational Evidence for the Coevolution between Supermassive Black Holes and Host Galaxies

  • (1) The correlation between the mass of supermassive black holes (SMBHs) and the properties of their host galaxies suggests that SMBHs and host galaxies are closely linked in their formation and evolution. While the exact origin of their relationship is still under debate, theoretical models often invoke feedback from active galactic nuclei as a crucial mechanism for establishing the BH-host correlation. In the first part of my talk, I will present our efforts to find observational sign of the AGN feedback in young luminous AGNs. (2) While intermediate-mass black hole (IMBH) is thought be cosmologically important class to understand the link between stellar mass black holes and SMBHs, it is extremely rare in the present-day Universe. In the second part of this talk, I will report a Gemini/GMOS-N IFU study of an ultraluminous X-ray source in NGC 5252, which is a possible candidate of an off-nuclear non-stellar black hole.

  • 기존의 센서가 컴퓨팅과 통신기능을 갖추게 되면서, 분산된 센서가 통신망을 통해 전달하는 현장의 물리량 변화를 직관적으로 판단할 수 있게 돕는 재사용 가능한 데이터 시각화 컴포넌트의 수요가 높다. 본 연구에서는 라즈베리파이를 IoT센서로 보고, 분산된 복수의 IoT센서들로부터 현장의 온도 값을 웹 서버에서 모아, 사용자가 클라이언트 단말을 통해 현장의 온도변화 상황을 직관성 있게 판단할 수 있도록 돕는 회귀분석 시각화컴포넌트를 개발하였다. 본 연구에서는 이종분산의 IoT 환경에서 다양한 브라우저를 지원할 수 있도록 Non-ActiveX 기반 데이터 시각화 컴포넌트 개발을 논한다. 본 연구의 결과는 단일의 온도센서가 보고하는 온도값을 현장의 온도값이라고 대표할 수 없는 항온항습 시설의 온도 감시제어 응용에서 본 연구 결과가 널리 활용될 것으로 본다.

A Clinical Study of Pulmonary Tuberculosis in Diabetics (당뇨병에 동반된 폐결핵의 임상적 연구)

  • Background: An association between diabetes and tuberculosis has long been implied. The severity of diabetes appears to correlate with the degree of tuberculous activity. Methods: A retrospective chart review of 82 patients with active pulmonary tuberculosis in diabetics (DMTB) and 83 patients with active pulmonary tuberculosis in nondiabetics (Non-DMTB) admitted to the Kyung Hee Medical Center between January 1995 and December 1996 was underiaken. Results: The sex ratio of DMTB was 58 : 24, and that of Non-DMTB was 62 : 21. Male patients predominated in both groups. The highest incidence of DMTB was 6th and 7th decades and that of Non-DMTB was 3rd and 4th decades. In case which the tuberculosis developed after diagnosis of diabetes, the prevalence of pulmonary tuberculosis was the highest in diabetes for 5-10 years. On chest X-ray findings, the moderate advanced tuberculosis cases were the most common (60.9% in DMTTB and 50.6% in Non-DMTB). There was no relation between the degree of tuberculosis activity on chest x-ray(minimal, moderate, and far advanced tuberculosis) and presence of diabetes. The incidence of lower lung field tuberculosis in DMTB was significantly higher than Non-DMTB(p<0.05). The multiple lobe involvement was the predominant chest roentgenographic finding in both groups. There was no significant difference of treatment response between DMTB and Non-DMTB. There was no relationship between initial HbA1c and the severity of pulmonary tuberculosis on chest X-ray. During treatment of pulmonary tuberculosis in excellently and well controlled diabetes, the cure rate of pulmonary tuberculosis was significantly higher than the poorly controlled diabetes and the rate of treatment failure was significantly lower than poorly controlled diabetes. (p<0.05). Conclusion: Poor control of blood glucose is related with increased rate of treatment failure in pulmonary tuberculosis with diabetes mellitus. Further investigation will be needed to study the mechanisms of treatment failure in poorly controlled diabetics with pulmonary tuberculosis.

Surface Characteristics of Titanium/Hydroxyapatite Double Layered Coating on Orthopedic PEEK by Magnetron Sputtering System (마그네트론 스퍼터링 시스템을 이용한 정형외과용 PEEK의 타이타늄/하이드록시아파타이트 이중 코팅층의 표면 특성 분석)

  • In this study, we have fabricated pure titanium (Ti)/hydroxyapatite (HA) double layer coating on medical grade PEEK from magnetron sputtering system, an investigation was performed whether the surface can be had more improve bio-active for orthopedi/dental applications than that of non-coated one. Pure Ti and HA coating layer were obtained by a radio-frequency and direct current power magnetron sputtering system. The microstructures surface, mechanical properties and wettability of the pure Ti/HA double layer deposited on the PEEK were analyzed by scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDS), X-ray diffraction (XRD), nano-indentation, and contact angle test. According to the EDS and XRD results, the composition and crystal structure of pure Ti and HA coated surface were verified. The elastic modulus and hardness value were increased by pure Ti and HA coating, and the pure Ti/HA double layer coating surface has the highest value. The contact angle showed higher value for pure Ti/HA double layered coating specimens than that of non-coated (PEEK) surface.

Effect of Iron Species in Mesoporous Fe-N/C Catalysts with Different Shapes on Activity Towards Oxygen Reduction Reaction

  • Among the non-precious metal catalysts, iron-nitrogen doped carbon (Fe-N/C) catalysts have been recognized as the most promising candidates for an alternative to Pt-based catalysts for the oxygen reduction reaction (ORR) under alkaline and acidic conditions. In this study, the nano replication method using mesoporous silica, which features tunable primary particle sizes and shape, is employed to prepare the mesoporous Fe-N/C catalysts with different shapes. Platelet SBA-15, irregular KIT-6, and spherical silica particle (SSP) were selected as a template to generate three different kinds of shapes of the mesoporous Fe-N/C catalyst. Physicochemical properties of mesoporous Fe-N/C catalysts are characterized by using small-angle X-ray diffraction, nitrogen adsorption-desorption isotherms, and scanning electron microscopy images. According to the electrochemical evaluation, there is no morphological preference of mesoporous Fe-N/C catalysts toward the ORR activity with half-cell configuration under alkaline electrolyte. By implementing X-ray photoelectron spectroscopy analysis of Fe and N atoms in the mesoporous Fe-N/C catalysts, it is possible to verify that the activity towards ORR highly depends on the portions of "Fe-N" species in the catalysts regardless of the shape of catalysts. It was suggested that active site distribution in the Fe-N/C is one important factor towards ORR activity.
