The aim of this study was to analyze the antioxidant properties of different cultivars of grape skin extract in an in vitro system. The extracts were prepared from eight grape cultivars: 'Campbell Early' (CE), 'Kyoho' (K), 'New Kyoho' (NK), 'Muscat of Alexandria' (MOA), 'Seibel' (S), 'Morgen Schow' (MS), 'Gold Finger' (GF), and 'Meru' (M). The total phenolic acid contents were highest in MS and K. Resveratrol content was high in NK (50.88 mg/l g of coat), and quercetin content was significantly higher in K (0.68 mg/l g of coat) than in the other grape species (0.21-0.44 mg/l g of coat). The K and MS grape species, in which total phenol content was comparatively high (K: $24.15\;{\mu}g/mL$, MS: $25.52\;{\mu}g/mL$), also showed a high level of electron donating activity (K, 53%; MS, 59%). The hydrogen radical scavenging activity of M (50.36%) was significantly higher than the other grape species, including the S (50.21%), MS (49.43%), and K (49.06%) cultivars. Antioxidant activity varied depending on grape species, but overall it was highest in the MS and K cultivars.
Mulberry (oddi) is one of the most popular functional foods with many physiological components. This study investigated and compared the nutritional compositions and physiological activities of four mulberry fruits from Morus albo L. including Cheongilppong, Iksuppong, Suwonppong and Cheongsuppong (a new cultivar from Chungcheongbuk-do). To analyze the nutrient contents, mulberry fruits were freeze-dried. The results showed that the proximate compositions of the four mulberry cultivars ranged from 9.61~14.11% for moisture, 8.28~11.90% for crude protein, 3.70~4.86% for crude ash, 4.28~5.54% for crude lipid and 7.46~10.78% for crude fiber. The above proximate contents of mulberry cultivars were not significantly different. However, Cheongsuppong had the highest content of reducing sugar, 74.7%. Chungsuppong and Iksuppong showed higher contents of total polyphenol and anthocyanin than other mulberry cultivars, while all mulberry cultivars showed high antioxidant activities. Hypoglycemic effect had a slightly higher level in Suwonppong and Cheongsuppong than in the other samples. Taken together, the new cultivar "Cheongsu" oddi can be suggested as a potential source of functional food.

Genetic improvement in potato can be carried out through several approaches, as sexual crosses, somatic hybridization, mutation and genetic engineering. Although the approach is different, but the goal is the same, to get a superior cultivar. Mutation and genetic engineering are very interesting methods for genetic improvement of potato plants. Mutation by gamma-ray irradiation have been performed to get some new potato cultivars which are more resistant to disease and have higher productivity. We have carried out a mutation of some potato cultivars and obtained some excellent clones to be potentially released as new superior cultivars. By the mutation method, we have released one potato cultivar for the French fries industry, and we registered one cultivar of potato for chips, and two cultivar for vegetable potatoes. Actually we are doing multi-location trial for three clones to be released as new cultivars. Through genetic engineering, several genes have been introduced into the potato plant, and we obtained several clones of transgenic potato plants. Transgenic potato plants containing FBPase gene encoding for fructose bisphosphatase, have a higher rate of photosynthesis and higher tuber productivity than non-transgenic plants. This result suggests that FBPase plays an important role in increasing the rate of photosynthesis and potato tuber productivity. Some transgenic potatoes containing the Hd3a gene are currently being evaluated for their productivity. Over expression of the Hd3a gene is expected to increase tuber productivity and induce flowering in potatoes. Transgenic potato plants containing MmPMA gene encoding for plasma membrane ATPse are more tolerant to low pH than non-transgenic plants, indicating that plasma membrane ATPase plays an important role in the potato plant tolerance to low pH stress. Transgenic potato plants containing c-lysozyme genes, are highly tolerant of bacterial wilt diseases caused by Ralstonia solanacearum and bacterial soft rot disease caused by Pectobacterium carotovorum. Expression of c-lyzozyme gene plays an important role in increasing the resistance of potato plants to bacterial diseases.
우리나라에서 표고버섯은 소비자의 선호도가 매우 높고, 전체 임산버섯 생산량의 약 97.7%를 차지하고 있는 매우 중요한 단기소득임산물중 하나이다. 이러한 표고버섯은 최근 신품종의 개발이 활발히 이루어지고 있어, 육종가의 권리 보호를 위하여 품종을 구분할 수 있는 분자마커 개발이 요구되고 있다. 본 연구에서는 신품종 '산마루2호'를 37개의 표고버섯 품종으로부터 구분할 수 있는 CAPS 마커를 개발하였다. '산마루2호'의 유전체에서 Scaffold 2번 1803483에 위치한 단일염기 다형성(SNP)이 확인되었고, 이 SNP를 포함하여 증폭한 DNA는 제한효소 Hhal에 의하여 특이적으로 절단되지 않아 다른 표고버섯 품종들과 구분되었다.
Background: Korean ginseng (Panax ginseng) is a well-known medicinal plant of Oriental medicine that is still in practice today. Until now, a total of 11 Korean ginseng cultivars with unique features to Korean ginseng have been developed based on the pure-line-selection method. Among them, a new cultivar namely G-1 with different agricultural traits related to yield and content of ginsenosides, was developed in 2012. Methods: The aim of this study was to distinguish the new ginseng cultivar G-1 by identifying the unique single-nucleotide polymorphism (SNP) at its 45S ribosomal DNA and Panax quinquefolius region than other Korean ginseng cultivars using multiplex amplification-refractory mutation system-polymerase chain reaction (ARMS-PCR). Results: A SNP at position of 45S ribosomal DNA region between G-1, P. quinquefolius, and the other Korean ginseng cultivars was identified. By designing modified allele-specific primers based on this site, we could specifically identified G-1 and P. quinquefolius via multiplex PCR. The unique primer for the SNP yielded an amplicon of size 449 bp in G-1 cultivar and P. quinquefolius. This study presents an effective method for the genetic identification of the G-1 cultivar and P. quinquefolius. Conclusion: The results from our study shows that this SNP-based approach to identify the G-1 cultivar will be a good way to distinguish accurately the G-1 cultivar and P. quinquefolius from other Korean ginseng cultivars using a SNP at 45S ribosomal DNA region.
Research was initiated to investigate early establishment characteristics and germination pattern of Kentucky bluegrass (KB, Poa pratensis L.) under natural room temperature and to analyze differences between alternative and natural conditions for a practical application. Fifteen cultivars of KB were evaluated in the study. Daily and cumulative germination pattern were measured and analyzed on a daily basis. Differences were observed in germination percentage, germination pattern, days to the first germination and days to 75% germination. Germination percentage was variable with cultivars at the end of study. It was 75.75 to 90.25% under natural condition. There were considerable variations in early germination characteristics and germination pattern among KB cultivars according to germination conditions. The first germination in all entries tested was initiated between 6 and 9 DAS (days after seeding) under ISTA (International Seed Testing Association) condition. But it was observed between 9 and 12 DAS under natural condition, being 3 to 5 days later as compared with ISTA condition. On the first date of a germinating process, the germination rate was 0.25% to 25.00% under ISTA condition, while 0.50% to 13.25% under natural condition. Days to the 75% germination were 15.08 to 28.80 DAS under ISTA condition and 17.78 to 28.75 DAS under natural condition. Midnight II, Excursion and Midnight were the fastest cultivars under ISTA condition, while Odyssey the fastest one under natural condition. Regardless of germination condition, the slowest cultivar was Voyager II, being over 4 weeks to 75% germination percentage. For the most cultivars, days to 75% germination under natural room temperature condition were 1 to 8 days slower than under ISTA condition. From this study information on differences in germination characteristics and pattern among cultivars would be usefully applied for KB establishment such as golf course construction. Prior to on-site seeding, a comprehensive germination test is practically recommended due to a possible decline in germination capability during domestic distribution and storage after import.

A bacterial pathogen, Pseudomonas syringae pv. actinidiae (Psa), is a causal agent of kiwifruit bacterial canker worldwide. Psa biovar 3 (Psa3) was first detected in 2011 at an orchard in Dodeok-myeon, Goheung-gun, Jeonnam Province in Korea. In this study, we present the results of an epidemiological study regarding Psa3 occurrence on kiwifruit orchards in Korea for the period of 2013 to 2015. Since the first detection of Psa3 in 2011, there was no further case reported by 2013. However, Psa3 was rapidly spreading to 33 orchards in 2014; except for three orchards in Sacheon-si, Gyeongnam Province, most cases were reported in Jeju Island. Entering 2015, bacterial canker by Psa3 became a pandemic in Korea, spreading to 72 orchards in Jeju Island, Jeonnam, and Gyeongnam Provinces. Our epidemiological study indicated that the first Psa3 incidence in 2011 might result from an introduction of Psa3 through imported seedlings from China in 2006. Apart from this, it was estimated that most Psa3 outbreaks from 2014 to 2015 were caused by pollens imported from New Zealand and China for artificial pollination. Most kiwifruit cultivars growing in Korea were infected with Psa3; yellow-fleshed cultivars (Yellow-king, Hort16A, Enza-gold, Zecy-gold, and Haegeum), red-fleshed cultivars (Hongyang and Enza-Red), green-fleshed cultivars (Hayward and Daeheung), and even a kiwiberry (Skinny-green). However, susceptibility to canker differed among cultivars; yellow- and red-fleshed cultivars showed much more severe symptoms compared to the green-fleshed cultivars of kiwifruit and a kiwiberry.
Tomato spotted wilt virus (TSWV) causes one of the most destructive viral diseases that threatens global tomato production. Sw-5b was reported as the resistance gene effective against TSWV. The objective of this research was to develop a single-nucleotide polymorphism (SNP) marker to distinguish tomato cultivars resistant to TSWV from susceptible cultivars for marker-assisted breeding. First, we determined genotypes for TSWV resistance in 32 commercial tomato cultivars using the previously reported Sw-5b gene-based marker. Then, DNA sequences of Sw-5b alleles in tomato cultivars showing resistant or susceptible genotypes were analyzed; a single SNP was found to distinguish tomato cultivars resistant to TSWV from susceptible cultivars. Based on the confirmed SNP, a SNP primer pair was designed. Using this new SNP sequence and high-resolution melting analysis, the same 32 tomato cultivars were screened. The results were perfectly correlated with those from screening with the Sw-5b gene-based marker. These results indicate that the SNP maker developed in this study will be useful for better tracking of resistance to TSWV in tomato breeding.
본 연구에서는 국내에 유통되고 있는 40개 표고 품종들로부터 산백향과 설백향의 구분이 가능한 CAPS 마커를 개발하였다. 제한효소 Hha I 과 HpyCH4IV를 이용한 밴드 패턴 분석을 통해 각각 산백향과 설백향을 다른 균주들과 구분하여 구별성을 확보할 수 있었다. 본 연구에서 개발된 CAPS 마커는 표고의 품종들 간에 유전적 다양성을 부여함으로써, 품종을 보호할 수 있는 분자생물학적 근거가 될 수 있다. 이로써 향후 유전자원에 대한 국가간 분쟁을 미연에 방지할 수 있을 것이다.
벼 도열병은 안정적인 쌀 생산을 위협하는 대표적인 병으로, 도열병 방제에는 저항성 품종의 육종이 가장 효과적으로 이용되었다. 병원균의 꾸준한 수집과 이들을 이용한 저항성 검정 테스트는 신품종 육성 프로그램에서 중요한 과정이다. 1985년 이래 우리나라의 도열병균은 8개의 판별품종을 이용한 레이스 판별시스템을 구축하여 분류하였다. 그러나 기존의 판별품종은 유전자형이 연구된 바 없어 새로운 레이스 출현이나 병 저항성 붕괴 등에 대하여 과학적으로 분석하여 신속하게 대처하기가 어려웠다. 따라서 이 연구에서는 유전자형이 밝혀진 단인자 저항성 계통과 우리나라에서 수집된 200개 균주의 병원성 반응을 분석하여, 새로운 판별 품종 시스템의 필요성을 제시하였다. 또한, 24개의 단인자 저항성 계통의 병원성 반응을 통해 9개의 대표 저항성 유전자를 선발하였으며, 이에 따른 10개의 대표균주 그룹과 그 저항성 반응을 인덱스화 하고 30개의 한국형 대표균주를 선발하였다. 이 연구는 향후 도열병 저항성 검정 및 벼 신품종 육종에 기초자료로서 활용될 수 있을 것이다.
