Estimation of Genetic Parameters for Economic Traits in Korean Native Chickens (재래닭 경제 형질에 대한 유전모수 추정)

  • This study was conducted to estimate heritabilities, genectic and phenotypic correlations on economic traits in three strains of Korean Native chickens (KNC) : Black (B), Red Brown (RB) and Yellow Brown (YB). The data used in this study were collected from 2000 to 2007 in the National Institute of Animal Science and a total number was 14,421 birds. The effect of the strains analysis showed that the strain B were greater than the other strains for body weight at 150 days and 270 days. The number of egg production at 270 days of the strain YB was higher than the other strains. The estimated heritabilities age at $1^{st}$ egg, body weight at 150 days, egg weight at $1^{st}$ egg, body weight at 270 days, egg weight at 270 days and number of egg production at 270 days were 0.32, 0.50, 0.31, 0.43, 0.48 and 0.69 for strain B; were 0.42, 0.41, 0.19, 0.48, 0.44 and 0.76 for strain RB; were 0.44, 0.52, 0.24, 0.54, 0.51 and 0.66 for strain YB respectively, The genetic and phenotypic correlations were also estimated.

Egg Quality Traits and Their Correlations in 12 Strains of Korean Native Chicken (토종닭 12계통 난질의 일반능력 및 상관관계 분석)

  • In this study, sixteen egg quality traits, including egg weight, albumen weight, yolk weight, eggshell weight, albumen weight ratio, yolk weight ratio, eggshell weight ratio, yolk color, eggshell color, egg height, egg width, shape index, albumen height, Haugh unit, eggshell thickness, and eggshell strength were investigated in 12 strains of Korean native chicken using 600 eggs from birds at 38 weeks of age. Results showed that the Korean White Leghorn F strain had the highest egg weight of 62.7 g and the lowest (47.5 g) was observed in the Korean native chicken W strain. The Haugh unit was significantly different between strains. The mean Haugh unit for Korean native chicken ranged between 84.2 (L strain) and 76.0 (F strain), with an overall average of 79.9. Eggshells were the thinnest in Korean Rhode Island Red C and D strains (0.342 mm), whereas the highest eggshell thickness (0.393 mm) was observed in the Korean White Leghorn K strain. Korean Rhode Island Red C and D strains had low egg strength compared to that of other strains. Korean Cornish and Korean native chicken showed significant differences in egg quality traits between strains within breeds. Positive correlation coefficients were observed between egg weight and egg quality traits. The traits for color showed zero or low correlations with most egg quality traits. Egg shape index showed no correlation with most egg quality traits. Albumen height and the Haugh unit showed a positive correlation with albumen traits. Eggshell strength showed a positive correlation with eggshell traits.

Isolation and Characterization of Endocellulase-Free Multienzyme Complex from Newly Isolated Thermoanaerobacterium thermosaccharolyticum Strain NOI-1

  • An endocellulase-free multienzyme complex was produced by a thermophilic anaerobic bacterium, Thermoanaerobacterium thermosaccharolyticum strain NOI-1, when grown on xylan. The temperature and pH optima for growth were $60^{\circ}C$ and 6.0, respectively. The bacterial cells were found to adhere to insoluble xylan and Avicel. A scanning electron microscopy analysis showed the adhesion of xylan to the cells. An endocellulase-free multienzyme complex was isolated from the crude enzyme of strain NOI-1 by affinity purification on cellulose and Sephacryl S-300 gel filtration. The molecular mass of the multienzyme complex was estimated to be about 1,200 kDa. The multienzyme complex showed one protein on native PAGE, one xylanase on a native zymogram, 21 proteins on SDS-PAGE, and 5 xylanases on a SDS zymogram. The multienzyme complex consisted of xylanase, ${\beta}$-xylosidase, ${\alpha}$-L-arabinofuranosidase, ${\beta}$-glucosidase, and cellobiohydrolase. The multienzyme complex was effective in hydrolyzing xylan and corn hulls. This is the first report of an endocellulase-free multienzyme complex produced by a thermophilic anaerobic bacterium, T. thermosaccharolyticum strain NOI-1.

Agrobacterium-mediated Transformation via Somatic Embryogenesis System in Korean fir (Abies koreana Wil.), A Korean Native Conifer

  • This study was conducted to establish an efficient transformation system by using somatic embryogenesis in an important Korean native conifer, Korean fir (Abies koreana). Embryogenic masses were induced from mature zygotic embryos of the Korean fir on Schenk and Hildebrandt medium, which was supplemented with thidiazuron. For genetic transformation, the embryogenic masses were co-cultivated with a disarmed Agrobacterium tumefaciens strain C58/pMP90 containing the plasmid vector pBIV10 or LBA4404 containing the plasmid vector MP90. Both vectors contain the kanamycin resistance and beta-glucuronidase (GUS) reporter genes. A total of 48 lines of embryogenic masses were selected on mLV medium containing $50{\mu}g/mL$ of kanamycin after 4 weeks of culture, following 3 days of co-cultivation with A. tumefaciens strain C58/pMP90 carrying pBIV10 (none of the lines was cultivated with strain LBA4404 carrying MP90). Quantitative real-time PCR was performed, and high levels of GUS transcripts were observed in the 48 putative transgenic lines; however, the control (non-transgenic line) showed negative results. Results of histochemical staining showed that the expression of the GUS reporter gene was observed in somatic embryos that developed from the embryogenic masses of all 48 lines. Stably transformed cultures were successfully produced by co-cultivation with A. tumefaciens strain C58/pMP90 carrying pBIV10 in Korean fir. Here, we have reported an Agrobacterium-mediated gene transfer protocol via somatic embryogenesis that may be helpful in developing breeding and conservation strategies for the Korean fir.

Performance of Laying Period of Two-way Crossbreed Parent Stock to Produce Laying-type Korean Native Commercial Chickens (산란 전용 토종 실용계 생산을 위한 2원교배 종계의 산란 능력 검정)

  • This study was carried out to investigate the performance of laying period of two-crossbred of Korean native chickens for producing laying hens. A total of four hundred eighty female 2-crossbred chicks that restored strains and were aboriginal at National Institute of Animal Science. There were four crossbreds (4 replications/crossbred, 30 birds/replication) as A) C strain ${\times}$ Y strain, B) C strain ${\times}$ L strain, C) C strain ${\times}$ G strain, and D) C strain ${\times}$ W strain, respectively. There were no significant difference on age at first egg among crossbreds (P>0.05). Egg weight and body weight of B crossbred at first egg was higher than other crossbreds (P<0.05). Body weight of B crossbred was the highest 20 to 72 weeks (P<0.05), and C and D crossbreds were lower compared to A and B crossbreds (P<0.05). Average feed intake of B crossbred was the highest among crossbreds (P<0.05), and that of A crossbred was higher compared to C and D crossbreds (P<0.05). Weekly feed intake of four crossbreds decreased from 50 weeks. Weekly egg weight of B crossbred was the highest and that of D crossbred was the lowest among crossbreds (P<0.05). Feed conversion ratio of A and B crossbreds was lower than that of C and D crossbreds. Hen-house egg production of C crossbred was the highest among crossbreds until 26 weeks old (P<0.05), but there was no significant difference among crossbreds from at the age of 26 weeks (P>0.05). Hen-day egg production decreased after at the age of 38 weeks. Weekly egg production of A and B crossbreds was higher compared to C and D crossbreds at the age of 68~72 weeks (P<0.05). These results suggested the basic data on the record of laying period of 2-crossbreed Korean Native Chickens for producing laying hens.

Performance of Middle Laying Period of Large-Type Korean Native Ducks (토종오리 대형종의 산란중기(30~50주령) 생산 능력)

  • This work was carried out to investigate the performance of large-type Korean native ducks at middle laying phase (30 to 50 weeks). A total of one hundred and sixty eight laying ducks were divided into A and B strains (6 replications/strain, 14 birds/replication and fed practical diet, for 20 weeks). There was no significant difference in body weight, daily feed intake and average egg weight. With increasing rearing period, mean body weight tended to be decreased and showed the lowest value (2,649 g) at 42 weeks of age (P<0.05). Similar trend was observed for weekly feed intake and average egg weight which decreased with passing weeks and was lowest (190.5 g and 83.8 g, respectively) at the age of 42~46 weeks (P<0.05). Daily egg production of B strain was higher than that of A strain at the age of 46~50 weeks (P<0.05). Weekly egg production of A and B strains commenced to decrease from the age of 42 weeks, however, B strain recovered at 46~50 weeks of age (P<0.05). At the age of 30~38 weeks, the egg production in B strain was higher than A strain (P<0.05), whereas, there was no significant difference between both the strains during 38~50 week. Feed conversion ratio of A strain was higher than that of B strain at 46~50 weeks of age (P<0.05). Weekly feed conversion was highest at the age of 42~46 weeks (P<0.05). In conclusion, the present results provides the basic information of the performance record of large-type Korean native ducks for the middle laying phase.

  • The nutritional requirements for Prevotella sp. 4PCCNB2 isolated from the rumen of a native goat in Korea and those of the ATCC 19189 strain isolated from the bovine rumen were investigated. The two strains grew well with ammonium sulfate as the sole added nitrogen source. However, neither a complex of amino acids nor casein hydrolysate effectively replaced ammonium sulfate. Biotin, p-aminobenzoic acid, and vitamin $B_12$ were essential to culture the ATCC 19189 strain. Unlike the ATCC 19189 strain, however, $B_12$ was only stimulatory for the growth of the 4PCCNB2 strain. The 4PCCNB2 strain grew well in the basal medium without an individual acid such as acetic acid or valeric acid. In contrast, either acetic or valeric acid was absolutely required for the growth of the ATCC 19189 strain.

  • The significant strain effects were investigated In all the major economic traits in Korean native chicken. The highest Least Square(LS) means of the body weights at 150 and 270days of age were observed in Black strain as 1, 594.38 and 1, 911.57 g. The earliest LS mean of the ages at first egg was 146.88 days in White strain. The heaviest LS means of egg weights at the first egg and 270 days of age were observed in Red brown strain as 33.20 and 50.74 9. The LS mean of the largest number of egg production was 79.50 eggs in Yellow strain.

Performance of Growing Period of Large-Type Korean Native Ducks (토종오리 대형종의 육성기 능력)

  • This work was carried out to investigate the performance of growing period of large-type Korean native ducks. A total of four hundred twenty female and male duck chicks generated from A and B strains of ducks. Groups were four crossbreds (3 replications/crossbred, 35 birds/replication) as $2{\times}2$ factors (2 strains, A and B; 2 genders, female and male). On the body weight, male of B strains was the highest among 4 treatments after the age of 4 week (P<0.05). Body weight of B strain was higher than that of A strain at all period (P<0.05). Body weight of male was higher compared to that of female after the age of 4 week (P<0.05). On the body weight gain, male of B strain was the highest and female of A strain was the lowest among 4 treatments at 0~8, 0~12, 0~16 and 0~20 weeks (P<0.05). Body weight gain of male was higher than that of female after the age of 4 week (P<0.05). On the feed intake, male of B strain was the highest among 4 treatments from the age of 0 week and to the age of 4, 12, 16 and 20 week (P<0.05). Feed intake of male was higher than that of female at 0~12, 0~16 and 0~20 weeks (P<0.05). On the feed conversion ratio, male of B strain was the highest among other treatments 0~4 weeks and female of A strain was at 0~8 and 0~12 weeks (P<0.05). Feed intake of female was higher compared to that of female at 0~8 weeks and male was high at 0~20 weeks. These results provided the basic data on the record of growing phase of large-type Korean native ducks.

Screening and Isolation of Chitinase and Chitosanase Producing Microbes from the Feces of Korean Native Calves Medicated DFMs Including Chitin (키틴함유 DFMs 급여 한우송아지 분변내 키틴 및 키토산분해효소 생산 미생물 선발 및 동정)

  • This study was carried out to screen and identify the chitinase and chitosanase producing microorganisms from the feces of calves medicated DFM sincluding chitin in order to do the immune fortification of Korean Native calves. Ten isolates were grown in the medium containing chitin and chitosan that had more than $10^5$ cfu/g in feces. Among these 10 strains, 2 strains (HANDI 110 and HANDI 309) had the chitinase activities and 2 strains (HANWOO and HANYOO) had the chitosanase activities in calves' feces. They showed no reaction in hemolysis tests by utilizing chitin and chitosan. The results from morphological, physicochemical and genetical identification indicated the HANDI 110 as a strain of Escherichia fergusonii, HANDI 309 was identified as a strain of Acinetobacter parvus, HANWOO was identified as a strain of Comamonas koreensis, and HANYOO as a strain of Chryseobacterium indologenes.
