Isolation and Charcterization of Micrococcus sp. HJ-19 Secreting Extracellular Protease (단백질 분해효소를 분비하는 Micrococcus sp. HJ19의 분리 및 특성)

  • Proteases are degradative enzymes which hydrolyze a peptide bond between amino acids and they are abundantly applied to commercial field. In order to screen new source of pretense, bacteria secreting extracellular pretense were isolated by enrichment culture from deep sea water samples of East Sea, Korea. A bacterium, named as HJ19, showed the best growth and the largest clear zone in plates supplemented skim milk at $30^{\circ}C$. The partial DNA sequence analysis of the 16S rRNA gene, phenotypic tests and morphology identified that this strain was In genus Micrococcus. The strain HJ19 could not grow at $10^{\circ}C$ but it started growth and showed pretense activity at $20^{\circ}C$. The optimal growth was at $37^{\circ}C$ and the maximal protease activity at $30^{\circ}C$ was about 480unit/ml.

  • $Si_{0.88}Ge_{0.12}$/Si heterostructure channels grown by RPCVD were employed to n-type metal oxide semiconductor field effect transistors(MOSFETs), and their electrical properties were investigated. SiGe nMOSFETs presented very high transconductance compared to conventional Si-bulk MOSFETs, regardless substantial drawbacks remaining in subthreshold-slope, $I_{off}$, and leakage current level. It looks worthwhile to utilize excellent transconductance properties into rf applications requesting high speed and amplification capability, although optimization works on both device structure and unit processes are necessary for enhanced isolation and reduced power dissipation.

Nuclear waste attributes of near-term deployable small modular reactors

  • The nuclear waste attributes of near-term deployable SMRs were assessed using established nuclear waste metrics, which are the DU mass, SNF mass, volume, activity, decay heat, radiotoxicity, and decommissioning LLW volumes. Metrics normalized per unit electricity generation were compared to a reference large PWR. Three SMRs, VOYGR, Natrium, and Xe-100, were selected because they represent a range of reactor and fuel technologies and are active designs deployable by the decade's end. The SMR nuclear waste attributes show both some similarities to the PWR and some significant differences caused by reactor-specific design features. The DU mass is equivalent to or slightly higher than the PWR. Back-end waste attributes for SNF disposition vary, but the differences have a limited impact on long-term repository isolation. SMR designs can vary significantly in SNF volume (and thus heat generation density). However, these differences are amenable to design optimization for handling, storage, transportation, and disposal technologies. Nuclear waste attributes from decommissioning vary depending on design and decommissioning technology choices. Given the analysis results in this study and assuming appropriate waste management system and operational optimization, there appear to be no major challenges to managing SMR nuclear wastes compared to the reference PWR.

Isolation, Identification, and Fermentation Characteristics of Bacillus sp. with High Protease Activity from Traditional Cheonggukjang (전통 청국장으로부터 protease 분비능이 우수한 Bacillus sp. 균주의 분리 동정 및 발효 특성)

  • Twenty one strains strongly producing protease were isolated from Korean traditional Cheonggukjang. Eight strains selected by sensory evaluation on Cheonggukjang prepared with isolated strains were identified with based on biochemical properties a and 16S rDNA sequencing. Identified strains were Bacillus subtilis MB4, and Bacillus amyloliquefaciens A1, A2, B1, MC1, SB2, SC1, and SD1. Protease activities, important strain selection factor, were higher in Cheongjukjang prepared with B. subtilis MB4 (179.6 Unit) and B. amyloliquefaciens SB2 (201.9 Unit) than commercial traditional Cheonggukjang (97.9 Unit). Sensory evaluation revealed Cheonggukjang prepared with B. subtilis MB4 had flavor very similar to commercial traditional Cheonggukjang. Cheonggukjang prepared with B. suhtilis MB4 (0.0006 Pa s) and commercial traditional Cheonggukjang (0.0002 Pa s) revealed lower viscosities than those of Cheonggukjang prepared with B. amyloliquefaciens SB2, MC1, B1, A1, SD1, A2, and SC1 (0.006 to 0.008 Pa s at 1001/s. Results show Cheonggukjang could be prepared using single strain of B. subtilis MB4, maintaining high protease activity and very similar sensory and viscosity qualities with those of commercial traditional Cheonggukjang.

Triplexer based on Filter Characteristics of CRLH Transmission Line and Triple-Band Amplifier Applications (CRLH 전송선로의 필터 특성을 이용한 트리플렉서와 삼중대역 증폭기에의 응용)

  • In this paper, we proposed the triplexer using unit-cell of CRLH transmission line which has a bandpass characteristic to reduce adjacent channel interference. The input impedance of triplexer with each channel filter is operated open-circuit at the resonance frequencies of other channels. Such property is due to the combination a series and parallel resonance circuits of CRLH-TL unit-cell. The measured triplexer results are showed a measured insertion loss of each channel, less than 1.5 dB, matching at each port, less than 15dB and isolation between channel, better than 25 dB. Also, to validate the triplexer, a small signal amplifier with triple-band is designed and tested. the measured amplifier results show good agreements with prediction.

  • In the sheep and cattle's rumen, facultative anaerobic cellulolytic bacteria were isolated by using Hungate's roll tube technique. In the 21 isolated species, one was screened by its strong cellulolytic activity and identified as Cellulomonas fimi C-14 by investigate morphological, cultural, physiological characteristics and electron microgram. Optimum conditions of the cell growth and enzyme production were pH 6.5 an $30^{\circ}C$, Thiamine and biotin support a good growth of C. fimi C-14. In the enzyme activities, Crystalline cellulose hydrolyzing activity, CMCase activity and ${\beta}-glucosidase$ activity were 20.6, 226.6 and 0.56$(unit{\times}10^3/ml)$ at pH 6.0, $40^{\circ}C$. By addition of fungal cellulase, enzyme activity was increased. Simultaneous Saccharification Fermentation is better than two step fermentation in ethanol yield with Saccharomyces cerevisiae DY2.

Isolation of a Phytase-Producing Bacillus sp. KHU-10 and Its Phytase Production

  • A bacterial strain producing high level of an extracellular phytase was isolated from cooked rice and identified as a strain of Bacillus sp. and designated as Bacillus sp. KHU-10. Optimum culture conditions were investigated for the maximum productivity of phytase by Bacillus sp. KHU-10. 1.0% Maltose and 1.0% peptone with 0.5% beef extract were the best carbon source and nitrogen source, respectively. The addition of $CaCl_2$, stimulated the enzyme productivity with concentration between 0.01% and 0.2%, in the medium. Although sodium phosphate increased the cell mass, the enzyme activity decreased. Calcium phytate and wheat bran containing phytate did not enhance the enzyme production. Under the optimum medium, the production of the phytase reached the highest level of 0.2 unit/ml after 4 days of incubation.

  • The aim of the present research program was to identify and develop strains of actinomycetes producing antifungal antibiotics which inhibit cell wall biosynthesis. 860 strains of Actinomycetes were isolated from various soil samples. Three isolates, EMS4, EMP22, and L234 were selected as the strains producing antifungal antibiotics inducing abnormal morphology against Penicillium cyclopium, Cryptococcus laurentii, and Aspergillus flavus, respectively. Taxonomic unit characters of the strains were tested and the data were analyzed numerically using TAXON program. EMS4, EMP22, and L234 were indentified to be a member of Streptomyces lavendulae, Streptomyces willmorei, and Streptomyces aburaviensis, respectively.

  • This paper presents a novel balanced mixer receiver front-end design based on a metamaterial structure applicable to differential-/common-mode excitation. This metamaterial structure functions as a leaky-wave antenna and provides in-trinsic common-mode suppression. Low LO leakage and high RF to LO isolation are achieved without additional filters for LO and RF paths. The metamaterial is based on a unit-cell which under a differential-mode excitation behaves like a composite right/left-handed(CRLH) metamaterial. In contrast, the metamaterial unit-cell is below cut-off under a common-mode excitation. Experimental results are used to verify the proposed metamaterial's differential-/common-mode characteristics. The metamaterial is integrated with a balanced mixer design resulting in an operation frequency range of $1.96{\sim}2.40$ GHz with an optimum mixer conversion loss of 21.1 dB at 2,4 GHz.

Antimicrobial Activity of Methyl Gallate isolated from the Leaves of Glochidion superbum Against Hospital Isolates of Methicillin Resistant Staphylococcus aureus

  • An antimicrobial compound has been isolated from the leaves of Glochidion superbum. The compound was determined as methyl 3, 4, 5-trihydroxybenzoate (methyl gallate), based on ultraviolet (UV), infrared (IR), nuclear magnetic resonance (NMR) and mass spectroscopy (MS) analysis. The isolated compound exhibited potent antimicrobial activity against three clinical isolates of methicillin resistant Staphylococcus aureus (MRSA) by qualitative agar disc diffusion method and quantitative broth dilution method. Agar disc diffusion was done in a dose-dependent manner for each bacterial isolate at disc potencies of 25, 50, 100, and $150{\mu}g/disc$. The zones of inhibition were on average equal to 12.27, 14.20, 15.43, and 24.17 mm respectively. The inhibition zones were compared with that of vancomycin disc at $30{\mu}g$ as a reference standard. The MIC and MBC values were $50{\mu}g/ml$ and $100{\mu}g/ml$ respectively. The results of anti MRSA activity were analyzed using one-way ANOVA with Turkey's HSD and Duncan test. In conclusion, methyl gallate which was isolated from G. superbum showed the inhibition activity against methicillin resistant S. aureus.
