Objectives: Galgeun-tang (GGT, gegen-tang, kakkon-to), an herbal formula, is used to treat the common cold, fevers, headaches, hangovers and neck and upper back stiffness. The drugs currently used to treat atopic dermatitis (AD) are limited by the significant adverse effects associated with their long-term usage. The need to efficiently manage the AD response while reducing side effects has led to the development of alternative remedies. Methods: To assess the effects of GGT on AD, the anti-inflammatory and anti-AD properties of GGT were evaluated in both in vitro and in vivo systems. Results: Nitric oxide (NO) and histamine production on lipopolysaccharide (LPS)-treated RAW264.7 cells and phorbol-12 myristate 13-acetate (PMA)/A23187-treated MC/9 cells, respectively, were inhibited by GGT. GGT reduced thymus and activation-regulated chemokine (TARC/CCL17) release on TNF-${\alpha}$/IFN-${\gamma}$ stimulated HaCaT cells in a dose-dependent manner. GGT reduced both plasma levels of IgE and histamine and the dermatitis score in house dust mite induced atopic dermatitis-like lesions on NC/Nga mice. However, there were no significant histopathological differences observed between the GGT group and the AD-induced group, such as AD-like lesions in the dorsal skin or ear or mast cell infiltration in the dorsal skin. Conclusions: These results indicate that GGT inhibits chemokine production by keratinocytes and the atopic dermatitis response in NC/Nga mice, suggesting that GGT may be useful as a therapeutic remedy for treating AD and allergic inflammation-related diseases.
Objectives Hataedock is a Korean herbal medical oral treatment that removes fetal toxic heat and meconium from new born babies. The purpose of this study is to evaluate whether Hataedock treatment of Duchi extracts has anti-inflammation effects on AD (Atopic Dermatitis)-induced NC/Nga mice. Methods After Hataedock treatment of Duchi extracts on days 0, 3-week-old NC/Nga mice were sensitized on days 28, 35, 42 by exposure of DNFB (dinitrochlorobenzene) and were induced to have AD. Immunohistochemistry of NF-${\kappa}B$ p65, iNOS, COX-2 and TUNEL assay of apoptotic body was used to identify changes of skin damages and anti-inflammation effects. Results The alleviate effect of the skin damage and angiogenesis was observed in DT group. The damage of stratum corneum, hyperplasia, edema, infiltration of lymphocytes and distribution of capillary were decreased in DT group. Also, the study results suggested that Hataedock treatment of Duchi extracts in DT group remarkably downregulated levels of NF-${\kappa}B$ p65 by 70% (p < 0.001), as well as COX-2 by 51%, iNOS by 62% (p < 0.001). Additionally, Hataedock treatment of Duchi extracts in DT group up-regulated apoptosis of inflammatory cells by 68% in atopic dermatitis-like skin lesion. Conclusions From the study results, we observed that Hataedock treatment of Duchi extracts alleviates AD through diminishing various inflammatory cytokines in the skin lesions, which are involved in the initial steps of AD development. It is anticipated to have potential applications for prevention and treatment of atopic dermatitis.
We examined the effects of essential oil from Thujopsis dolobrata Sieb. et Zucc. var. hondai Makino (EOTD) (Cupressaceae) on atopic dermatitis (AD)-like skin lesions in NC/Nga mice. Treatment with EOTD twice daily for two weeks ameliorate AD-like skin lesions induced by DNCB (2,4 dinitrochlorobenzene), and clinical scores were reduced to 7.29, 7.07, and 4.5 points in the groups treated with 1.5%, 3.0%, and 6.0% extract (p<0.01) respectively, from the 15.0 score obtained using vehicle. EOTD inhibited the infiltration of mast cells into the AD-like skin lesion in NC/Nga mice (p<0.01) and also reduced serum histamine and IgE levels (p<0.05). Furthermore, it dose-dependently inhibited the release of beta-hexosaminidase from rat basophilic leukemia RBL 2H3 cells. These results indicate that EOTD reduces AD-like skin lesions by inhibiting the production of IgE and histamine, and, thus, IgE-mediated degranulation.
Oxidative stress has been implicated in cutaneous damage in various inflammatory skin diseases, including atopic dermatitis (AD). Atoberry is the herb medicine extract which is composed with Spirodelae Herba, Xanthii Fructus, Houttuyniae Herba, Taraxaci Herba, Retinervus Luffae Fructus, Platycodi Radix, and Scutellariae Radix. In this study, we investigated the antioxidant and anti-inflammatory effects of Atoberry in AD-like skin lesion NC/Nga mice. Murine AD-like skin lesions were made by painting Dermatophagoides farinse (Df) extract. Atoberry significantly increased electron donating ability (DPPH), nitrite scavenging (NO) and superoxide dismutase (SOD) activities in dose dependant. Topically applied Atoberry significantly reduced clinical severity score, ear thickness and histological grade in AD-like skin lesion NC/Nga mice. In addition, the serum levels of IgE, NO and prostaglandin E2 were significantly reduced by Atoberry. Futhermore, skin tissue levels of SOD, catalase and glutathione peroxidase (GPx) were significantly reduced by Atoberry. These results demonstrate that topical application of Atoberry may be improve the AD-like skin lesion by antioxidant and anti-inflammatory effects.
Objective : This Experimental study was done to investigate the Effect of Taklisodok-um and Hwangryunhaedok-tang(TH) on Atopic Dermatitis. Methods : We assessed effects of TH on the IgE, IL-4, IL-5, IL-6, IgM, IgG1, IFN-${\gamma}$ in vivo, on the IL-4, IL-5, CCR3 in the skin tissues of ear and dorsum with NC/Nga mice. And we assessed effects of TH on the COX-2, IL-$1{\beta}$, TNF-${\alpha}$, IL-6 with RAW 264.7 cell. Results and Conclusion : 1. IgE, IL-4, IL-5, IL-6, IgM, IgGl levels in the serum of TH treated NC/Nga mouse group were decreased compared to the untreated control mice. IFN-r showed a increase in the experimental group compared to the untreated control group. The spleen weight of TH treated NC/Nga mice was decreased compared to the untreated control group. 2. mRNA expression levels of IL-4, IL-5 and CCR3 in the skin tissues of TH treated NC/Nga mice were decreased, and expression levels of IL-6 in the skin tissues of TH treated NC/Nga mice were decreased compared to the untreated control group. IFN-${\gamma}$ mRNA expression levels were increased compared to the untreated control group. 3. Judged from that IL-$1{\beta}$, TNF-${\alpha}$, IL-6 express of gene, effect of inflammatory Cytokines revelation were decreased compared to the untreated control group. 4. Depend on the strength of TH, inflammatory RAW 264.7 in the serum of TH were inhibited compared to the untreated control serum that leaded a COX-2 activity model. 5. Histological observation of the ear and skin tissues showed that the extents of inflammation and infiltrated immune cells in the epidermis and dermis of TH treated NC/Nga mice were highly reduced compared to the untreated control group.
아토피피부염 (AD)은 천식, 음식 알레르기, 비염 같은 전신아토피질환을 동반하는 만성재발성 피부염증질환이다. 아토피피부염과 관련된 IL-17의 임상적 역할은 다양한 조건에서 보고되고 있으며, 또한 건선 피부 상태에 깊숙이 관여하고 있다. IL-17은 각질세포 (keratinocytes)에서 과잉으로 생산되며, 아토피피부염의 말초임파구에서도 다량 생성됨을 세포내염색을 통하여 확인된 바 있다. 본 연구에서는 창이자 추출물 (XS-E와 XS-FL)이 NC/Nga 생쥐의 $CD4^+$ T 세포에서 유도된 Th17 세포의 분화억제 및 IL-17의 생산량 감소 효과에 대한 실험을 하였다. 그 결과 XS-E와 XS-FL을 처리한 섬유아세포에서 세포독성은 나타나지 않았고, 4일간 XS-E와 XS-FL에 동시배양 한 $CD4^+$ T 세포의 IL-17 생산량을 FACS로 분석한 결과 $100\;{\mu}g$/mL XS-E 처리군의 IL-17 생산량은 32.3%로 대조군에 비하여 2배 이상의 감소를 나타내었으며, $20\;{\mu}g$/mL XS-30% AFL (acetone XS-FL) 처리군의 Th17 세포는 19.6%으로 대조군에 비하여 3.5배 억제되었다. 또한 real-time PCR을 이용하여 IL-17A와 IL-22 mRNA의 유전자 발현량을 비교 분석한 결과, IL-17A와 IL-22 mRNA의 유전자발현의 RQ값은 XS-E와 XS-30% AFL를 처리한 실험군이 대조군에 비하여 유의성 있는 감소를 나타내었다(p < 0.01, p < 0.001). ELISA로 측정한 IL-17A 생산량은 XS-E와 XS-30% AFL를 처리한 실험군이 대조군에 비하여 현저히 감소 (p < 0.05, p < 0.001)하였다. Th17 세포의 증식을 알아보기 위하여, rIL-6와 TGF-$\beta$로 분화시킨 Th17 세포를 CFSE로 표지한 후 rIL-23 처리를 하여 4일간 배양하여 증식을 유도시켰다. 대조군의 Th17 세포 분열은4일 동안 4번에 걸쳐 비슷한 세포수의 증식이 일어나는 것을 CFSE를 통하여 확인하였고, XS-30% AFL 처리군은 CFSE의 형광 분포가 점점 감소하여 Th17 세포의 증식이 억제됨을 알 수 있었다.
아토피 피부염에서 유산균의 유익성에 대해서는 이미 많이 알려져 있고 우리나라 전통식품인 김치유산균의 유익성에 대한 연구도 많이 진행되었으나 아토피 피부염에서 김치 유산균의 효과에 관한 연구는 아직 미비한 실정이다. 본 연구에서는 아토피 동물 모델인 NC/Nga mice에 김치유산균에서 추출한 유산균(L.plantarum K8)의 파쇄물과 유산균 및 ${\gamma}$-리놀렌산 복합체(L.rhamnosus GG 생균, B. lactis Bb-12Lb 생균, L. plantarum K8생균, L. plantarum K8 파쇄물, ${\gamma}$-리놀렌산)를 경구 투여한 후 혈청 면역지표(IgE, IL-4, IL-5, IFN-${\gamma}$)와 비장 배양액에서 IgE 농도를 측정하였다. 실험동물의 체중 및 사료섭취량은 사육기간이 지남에 따라 증가하는 경향을 보였으며 각 군 간에 유의적인 차이는 관찰되지 않았다. 실험 시작 시 혈청 IgE 농도는 각 군 간에 차이가 나타나지 않았으나 실험 전 기간 4주 동안 유산균 파쇄물(DPF), 유산균 및 ${\gamma}$-리놀렌산 복합체(DPOC), 항히스타민(DAH)을 섭취한 군에서 증류수(DDW)만을 섭취군에 비하여 유의적으로 낮게 나타났다. 혈청 IFN-${\gamma}$ 농도는 각 군에서 유의적인 차이가 나타나지 않았으나 혈청 IL-4 농도는 유산균 및 ${\gamma}$-리놀렌산 복합체를 섭취한 군(DPOC)에서 대조군에 비하여 유의적으로 낮게 나타났다. 혈청 IL-5 농도는 유산균 파쇄물(DPF), 유산균 및 ${\gamma}$-리놀렌산 복합체(DPOC), 항히스타민(DAH)을 섭취한 군에서 모두 대조군에 비하여 유의적으로 낮았다. 비장 임파구 배양 상층액의 IgE 농도는 각 군에서 군 간 유의적인 차이가 나타나지 않았다. 결론적으로, 아토피 동물모델 NC/Nga mice에 유산균 파쇄물(DPF)과 유산균 및 감마리놀렌산 복합체(DPOC)의 경구투여는 혈청 IgE, IL-4, IL-5를 감소시키는 것으로 나타났으며, 김치로부터 추출한 프로바이오틱스를 아토피 환자에게 섭취시켰을 때 IgE, IL-4, IL-5 감소와 같은 긍정적인 효과를 볼 수 있을 것으로 기대한다. 여러 종류의 프로바이오틱스는 아토피의 증상완화 또는
면역 체계에 미치는 영향과 정확한 메카니즘을 밝히기 위하여 과학적인 연구가 필요한 것으로 사료된다.
The aim of this study is to investigate the effect of Zizyphus jujuba var. spinosa Composite oil(ZjJ-C_oil) and Aralia cordata var. continentalis Composite oil(ARC-C_oil) to NC/Nga mice induced in Atopic dermatitis-like skin lesions by DNCB. NC/Nga mice which have been induced to Atopic dermatitis-like skin lesions by DNCB are divided into 4 groups, the first is the mice which have been spread with izyphus jujuba var. spinosa Composite oil(ZjJ-C_oil), the second is the mice which have been spread with Aralia cordata var. continentalis Composite oil(ARC-C_oil), the third is which have been spread with dexamethasone (Dexa.) 0.5% on their Atopic lesion, the last is the control group. Then We analyzed skin clinical score, blood sample of each group of measure state of the dorsal skin, the number of immunocytes, and resect the skin lesion to anlayze the state of cells. There are meaningful results of measuring the number of IgE, IL-4, IL-13, $IFN-{\gamma}$, IL-5, the total cells in ALN, dorsal skin, CD4+ Th, CD11b+/Gr-1+ in PBMCs, CD4+ Th, B220+/CD23+ in ALN, $CD4^+$, $CD8^+$ Th in dorsal skin, the level of COX-2, $TNF-{\alpha}$, $TGF-{\beta}1$, IL-4, IL-13 mRNA, the state of the skin lesion and cells in the group with ZjJ-C_oil, ARC-C_oil cream in comprarison with the control group.
아토피 피부염(Atopic dermatitis, AD)은 주로 5세 이전에 발병하여 심한 가려움을 동반하는 만성 염증성 피부질환이다. 본 연구의 목적은 NC/Nga 마우스를 AD 유사 증상에 대한 405 nm+ 850 nm LED 광선 치료의 효과를 조사하는 것이다. 마우스는 Normal (Vehicle), 아토피성 피부염 유발군 (CON), 405 nm+ 850 nm LED 광선 치료군 (LED)으로 나누어 난괴법을 이용하여 무작위 배치하여 실험을 진행하였다. LED 실험군은 하루 10분씩 405 nm+ 850 nm 파장의 LED 치료를 7일 간 실시하였다. LED 광선치료 연구는 Dermatics 점수의 개선을 측정하고 피부염으로 인한 표피조직 두께 감소를 관찰하였다. LED 광선 요법으로 인한 혈청 IL-1𝛽의 현저한 감소와 경피 수분 손실 및 혈청 IgE 농도 결과를 바탕으로 LED 광선 치료는 아토피 유발 생쥐의 정상적인 피부 상태 회복에 도움을 주었다. 본 연구 결과는 아토피 피부염 마우스 모델에서 적외선 영역의 광선과 blue light 영역의 광선의 동시 조사치료가 아토피 피부염 치료에 뛰어난 효능을 가지며, 두 파장의 LED의 동시 사용 가능성에 대하여 시사하였다.
Various related factors and tissue changes in vitro and in vivo were observed to investigate the efficacy of HYBH on atopic dermatitis. The results are described below. HYBH improved the atopic dermatitis symptoms by naked eye examination, and significantly decreased dermatitis clinical index at 14 weeks. HYBH significantly decreased CD4+/CD45+, CD4+, CD8+, CD3+/CD69+ immune cell ratios in PBMC by 28%, 16%, 30%, 26% and 22% respectively. HYBH significantly decreased CD11b+/Gr-1+, CD3 immune cell ratios in dorsal skin by 35.3% and 67.5% respectively. HYBH significantly decreased the expression of IL-4 and IFN-${\gamma}$ in spleen by 23% and 15% respectively. HYBH significantly decreased the production rate of IL-5, IL-13 and histamine in serum by 17%, 23%, and 8.8% respectively and increased IL-17 production by 17%. HYBH significantly decreased immunoglubulins IgG1 and IgE production in serum. The results above indicated that treatment of HYBH improved atopic dermatitis symptoms by anti-oxidant activity and immune modulation activity as a clinical evidence. Also, different fermentation conditions using various microbial strains should be accumulated as the clinical evidence for broad application in the future.

제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
