The Characteristics of Cultural Conditions for the Mycelial Growth and Fruiting Body Formation of Paecilomyces sinclairii (매미눈꽃동충하초(Paecilomyces sinclairii)의 균사 생장과 자실체 형성 조건의 특성)

  • The fruiting body of Paecilomyces sinclairii was collected in Baekyangsa, Jeollanam-Do, Korea. Cultural conditions for the mycelial growth and fruiting body formation were investigated. Its optimum mycelial growth was obtained at 25℃ and pH 8 on potato dextrose agar and Hamada media among the various media tested. The carbon and nitrogen sources for the optimum mycelial growth were dextrin and glutamine, respectively. The optimum C/N ratio was about 20:1 in case that 1% glucose was supplemented to the basal medium as a carbon source. The favorable mycelial growth was obtained from corn meal extract medium mixed with 30% (w/v) milk solution. The maximum fruiting body was formed in unpolished rice medium supplemented with 20% (w/w) silkworm pupae at $25^{\circ}C$ under 500lux.

Effect of solid grain media on the mycelial growth of Pleurotus ostreatus and its biofunctional activity (곡물배지가 느타리버섯의 균사배양과 생리활성에 미치는 영향)

  • Effect of solid grain media on the mycelial growth of Pleurotus ostreatus and its biofunctional activities were investigated using seven kinds of grains. Foxtail millet and barley were good for growth of hypha of P. ostreatus. However, growth of the mycelium was very slow in the solid grain media containing wheat, corn and brown rice. Mycelial growth of P. ostreatus according to water content of solid grain media was good at 25% to 30%. Mycelial growth of P. ostreatus according to heating-time and temperature of solid grain media was good for 30 min at $121^{\circ}C$. Anticancer activities against lung cancer cell line of the myceial extracts from P. ostreatus grown on several grain media were strong in the corn, defatted soybean, brown rice, barly and black bean in order. Fibrinolytic activities of the myelial extracts were strong in order defatted soybean, wheat, foxtail millet, barley, brown rice and black bean. The mycelial extracts were showed good antibacterial activities against Pseudomonas aeroginosa and Staphylococcus aureus.

Co-treatment with Origanum Oil and Thyme Oil Vapours Synergistically Limits the Growth of Soil-borne Pathogens Causing Strawberry Diseases

  • Vapours from origanum oil (O) and thyme oil (T) were applied to the four soil-borne strawberry pathogens Fusarium oxysporum f. sp. fragariae, Colletotrichum fructicola, Lasiodiplodia theobromae, and Phytophthora cactorum, causing Fusarium wilt, anthracnose, dieback, and Phytophthora rot, respectively. Increasing T vapour doses in the presence of O vapour strongly inhibited mycelial growths of the four pathogens and vice versa. When mycelia of F. oxysporum f. sp. fragariae and P. cactorum exposed to the combined O + T vapours were transferred to the fresh media, mycelial growth was restored, indicating fungistasis by vapours. However, the mycelial growth of C. fructicola and L. theobromae exposed to the combined O + T vapours have been slightly retarded in the fresh media. Prolonged exposure of strawberry pathogens to O + T vapours in soil environments may be suggested as an alternative method for eco-friendly disease management.

Optimal Condition of Mycelial Growth of Grifola frondosa (잎새버섯 균주의 균사체 생육 최적조건)

  • This study was carried out to obtain basic data on mycelial growth characteristics for an artificial cultivation of Grifola frondosa. Ten strains of G. frondosa were collected from Korea, China and Japan and investigated its optimal culture condition. Among four kinds of mushroom culture media, PDA medium was selected as the suitable culture medium. The optimal conditions for the mycelial growth of G. frondosa in PDA medium were $25^{\circ}C$ and $4{\sim}5$ of pH, respectively. The carbon and nitrogen sources for the optimum mycelial growth were fructose and peptone, respectively, and the highest mycelial growth was observed when the C/N ratio was $10{\sim}20$.

  • The optimal conditions for mycelial growth of P. linteus ASI 26011 were 25-30$^{\circ}C$ and pH 6.0, respectively. The mycelial growth of P. linteus was excellent on MCM medium. In case of carbon sources, the mycelial growth of P. linteus was best on the culture media that were contained with sucrose, mannose and glucose. Potassium nitrate and sodium nitrate were good for the mycelial growth of P. linteus as a nitrogen source. For comparison of the mycelial colonization of P. linteus on logs, several techniques of inoculation were tested; the sterilized short log inoculation, drilling inoculation and log-end sandwich inoculation. The mycelial colonization of P. linteus on logs was good in the treatment of sterilized short log inoculation, but poor in the traditional methods such as drilling inoculation and log-end sandwich. The initial mycelial growth and the full mycelial colonization of P. linteus were the best on 20 cm logs under the condition of 42% of moisture content in log. Also the initial mycelial growth of P. linteus was accelerated over 12 hours of sterilization. Burying method of logs after 5-6 months of incubation was the best for formation of basidiocarp of P. linteus. The formation of fruiting body of P. linteus was quite good in the cultivation house at the 31-35$^{\circ}C$ and over 96% of relative humidity.

  • Saesongi (Pleurotus eryngii) was cultivated in both potato dextrose agar (PDA) and sawdust media supplemented with Ca salts. The addition of Ca phosphate and Ca carbonate to sawdust media did not affect the growth, whereas Ca sulfate addition suppressed the mycelial growth appreciably. The efficiencies of Ca accumulation in the fruiting were studied based on mycelial growth experiments on Ca-supplemented sawdust media. Supplementation with 0.1 to 5% Ca phosphate increased the Ca content in the fruiting body by 4.5-6.5 fold, to a level of $314.6{\pm}22.7$ to $449.7{\pm}29.3$.

  • The cultural characteristics and antibacterial activities of Cordyceps militaris and Paecilomyces tenuipes were compared. The mycelial growth was the highest on MCM (Mushroom Complete Medium) for C. militaris and on YMA (Yeast Malt Agar) for P. tenuipes. But the mycelial density on MMM (Mushroom Minimal Medium) was lower than other on media. The optimum mycelial growth was observed at $25^{\circ}C$. C. militaris was low mycelial growth when it was transferred over 5 times generation. The carbon source for the optimum mycelial growth was fructose of monosaccharide, maltose of disaccharide and dextrin of polysaccharide. The calcium nitrate of organonitrogen was found the best mycelial growth on C. militaris, while the sodium nitrate observed to be well for mycelial growth on P. tenuipes. The ammonium tartrate was observed to be the best among the inorganonitrogen used for mycelial growth. Antibacterial activities were found out just C. militaris against Bacillus cereus of Gram (+).

Effect of Water Potential on Mycelial Growth and Production of Sclerotia of Sclerotium cepivorum (Sclerotium cepivorum의 균사생장 및 균핵 생성에 대한 수분압의 영향)

  • The effect of water potential ($\psi_W$) and temperature on mycelial growth and sclerotial production of Sclerotium cepivorum was determined in potato dextrose agar(PDA) and potato dextrose broth (PDB) adjusted to different $\psi_W$ with NaCI, KCI, sucrose or polyethylene glycol (PEG) at 15, 20 and $25^{\circ}C$. the growth of mycellium was not significantly af. fected by $\psi_W$ values between -1,970 and -2,240J/Kg, but severely decreased lower than -2,240J/Kg. Dry weight was slightly increased at $\psi_W$values between -450 and -2,240 J/Kg. The reduction of dry weight wasslower than the reduction of mycelial growth as the $\psi_W$ decreased. The mycelial growth and dry weight were more severely influenced on PEG amended media than on other osmotica amended media. About 50% reduction of mycelial growth and dry weight was occurred about -1,000 and -2240 J/Kg, respectively. The production of sclerotion of sclerotial production occurred between -450 and -810 J/Kg. Sclerotium was not produced lower than -2,240 J/Kg. Mycelial growth and sclerotial production was better at $25^{\circ}C$ as the $\psi_W$ decreased than at $20^{\circ}C$ which is optimal temperaturein the undmended media. The influence of $\psi_W$ on mycelial growth and sclerotial production of S. cepivorum adjusted with NaCl, KCI sucrose or polyethylene glycol showed similar patterns.

Cross- and Double-Resistance of Benomyl-Resistant Botryosphaeria dothidea (Benomyl에 저항성인 사과 겹무늬썩음병균의 교차 및 이중저항성)

  • Mycelial growth resistant isolates of Botryosphaeria dothidea to benomyl showed 99~79% spore germination on the PSA media supplemented with 200~2,100 $\mu\textrm{g}$/ml of carbendazim and thiophanate-methyl to manifest the high cross-resistance in spore germination. Mycelial growth, 23~9 mm in colony diameter, also manifested the high cross-resistance of mycelial growth together with similarity of spore forming cross-resistance. Benomyl resistant isolates BR1, BR2 and BR3, grew 23~10 mm in colony diameter at 330~3,000 $\mu\textrm{g}$/ml of captafol, captan and oxine-copper showing the high double resistance of mycelial growth and spore formation with minor difference. However, within concentration range of the 3 fungicides tested, germinations of all the tested isolates were completely suppressed to show no double-resistance in the fungal spore germination.

  • According to the result of the observations on the effect of the reagents, $CuSO_4{\cdot}5H_2O$, $ZnSO_4{\cdot}7H_2O$ $MnSO_4{\cdot}4H_2O$, $HgCl_2$, upon the mycelial growth of psalliota campestris $HgCl_2$ showed the strongest effect on checking the mycelial growth $(,.008{\sim}0.01%)$ $CuSO_4{\cdot}5H_2O(0.030%)$, $ZnSO_4{\cdot}7H_2O(0.080%)$, came next and the $MnSO_4{\cdot}4H_2O(0.100%)$ was the weakest. Writer also found that showed the promoting effect of the mycelial growth in appropriate Concentration (at $0.015%\;CuSO_4{\cdot}5H_2O$, $0.030%ZnSO_4{\cdot}7H_2O$).
