Structure and Bioactivity of Boholamide A from a Tidal Mudflat Actinomycete (갯벌 방선균 유래 Boholamide A의 구조 및 생리활성에 대한 연구)

  • LC/MS approach targeting secondary metabolites of bacterial strain resulted in the discovery of boholamide A (1), from the culture of marine actinomycete strain which was isolated from a tidal mudflat in Muan, Republic of Korea. Boholamide A (1), a cyclodepsipeptide with HDMN, APD, glycine, and valine was structurally determined by using 1D/2D NMR spectroscopy, mass spectrometry and UV spectroscopy. Boholamide A (1) showed the inhibitory activity against Bacillus subtilis, with IC50 value of 0.08 mM.

  • Recently, Intense competition is expected to bring about continuous change with the rapid global aviation industry. In particular, domestic passenger demand has been steadily increasing over the past five years, with domestic and international shipments growing at a CAGR of 8.4% and 10.0%, respectively. In addition to the economic value of the airport, social interest due to greenhouse gas emissions is also rising. In this study, we analyzed the efficiency of airport operation by using DEA model(Directional Technology Distance Function) considering undesirable output for 14 airports in Korea. As a result of analysis, all airports except Gimhae, Yangyang, Cheongju and Yeosu were analyzed as efficient airports. However, only Gimpo, Gimhae, Jeju, Muan, Daegu and Gwangju were analyzed as efficient airports considering the undesirable output.

A Study on the Habitat Use of Waterbirds and Grading Assessment of the Tidal Flat at Muan Bay in Jeollanamdo, Korea (전라남도 무안만에 도래하는 수조류의 서식지 이용 및 갯벌등급 평가)

  • This research conducted a survey on waterbirds visiting this area four times by season from February to October in 2007 to look into the habitat use of waterbirds, to do a value and grade testing of the tidal flat by dividing the foreshore on Muan Bay located in Jeollanam-do into four areas (Dongam, Guro, Bokryong and Wangsan tidal flats). The survey results revealed that there existed a total of 15,755 individuals of 54 species including 2 species of grebes, 7 species of herons, 7 species of dabbling ducks, 6 species of diving ducks, 20 species of waders, 3 species of gulls and 9 other species and this survey also observed 9,291 individuals of the wading birds as a dominant group on Muan Bay. In these classified groups, the gulls and waders were observed to mostly use Dongam tidal flat as their habitat, while the group using Guro tidal flat as their habitat was mostly grebes, dabbling and diving ducks. As a result of UPGMA clustering analysis in consideration of the species and number of individuals, there appear the close similarities between Dongam and Bokryong tidal flats and so do Guro and Wangsan tidal flats. Taking a look at the grading of tidal flats by setting up ecological indexes, such as diversity index, abundance index, and dominance index, etc. legally reserved species and maximum number of individuals as a standard, the rank for the value and importance degree of Bokryong tidal flat appeared higher than that of the other three tidal flats. Like this, the gradation of tidal flats according to waterbirds are judged to able to suggest objective data on the issue of proper judgment and designation of valuable tidal flat areas and its subsequent effective preservation and management.

Occurrence of Plant Parasitic Nematodes in Chrysanthemum and ITS and D3-28S rDNA Characterization of Pratylenchus spp. (국화재배지의 식물기생선충 분포조사 및 뿌리썩이선층의 ITS와 D3-28S rDNA 특성조사)

  • A survey was conducted to find out the major plant parasitic nematode in Chrysanthemum morifolium fields in Korea from May to June in 2005. A genus of Pratylenchus was determined as the most important plant parasitic nematode based on analysis of total 50 samples from 8 cities of chrysanthemum field. Pratylenchus showed 86% occurrence rate and average numbered 1,095 per 200cc soils and 1g root. Five Pratylenchus isolates, 'Muan', 'Masan', 'Tean', 'Gumi', 'Jeongup', were selected for the molecular identification of the species of Pratylenchus, and ITS and D3-28S ribosomal DNA were amplified by PCR. For the ITS, only 'Muan' isolate was differentiated by total 1 kb PCR amplification, which was 200 bp larger than all the other isolates. There was no size variation in amplified D3-28S rDNA and all isolate represented approximately 320 bp of PCR product. Sequence data of D3-28S rDNA were analysed by MegAlign program in DNASTAR software and phylogenetic tree was constructed. Sequence homology was 100% between 'Gumi' isolate and 'Tean' isolate and 'Jeongup' isolate was also close to these isolates by 99.7% sequence homology. 'Gumi', 'Tean' group and 'Jeongup' isolate were determined to be closely related to Pratylenchus vulnus by 96.7% and 96.3% similarity in respectively. D3 sequence of 'Masan' isolate was 100% identical to P. penetrans, and 'Muan' isolate showed 99.7% similarity to P. brachyurus. This result was congruent with the branch divergence pattern shown in phylogenetic tree.

  • This study evaluates the usefulness and capability of surface electrical resistivity technique for identifying the weak zones or subsurface cavities in karst area with limestone formation. Weak zones or cavities near surface can be potentially dangerous and several problems are associated with collapse of roads or buildings accompanied by subsidence phenomena. In this paper, both two and three dimensional resistivity investigation were conducted to investigate subsidence along a road in Yongweol-ri, Muan-gun, South Korea. The results of the resistivity survey using dipole-dipole array provide a clear view of the weathered regolith, the distribution of weak zones or cavities and bedrock. Several low resistivity areas were identified and subsequent drilling led to the discovery of several weak zone or clay-filled underground cavities. The drilling results show excellent correlation with the resistivity images. It is illustrated, the ability of electrical technique to produce high resolution images of subsurface, which are useful for subsidence assessment. Also the results of this study have demonstrated that two and three dimensional electrical resistivity surveys are useful for delineating the subsidence area. Based on resistivity imaging, the map of hazardous zone has been developed.

Genetic Diversity of Culturable Endophytic Fungi Isolated from Halophytes Naturally Growing in Muan Salt Marsh (무안 염습지에 자생하는 염생식물(halophyte)의 뿌리로부터 분리된 내생진균(endophytic fungi)의 유전학적 다양성)

  • Native halophytes, such as Suaeda maritima, Limonium tetragonum, S. japonica, Zoysia sinica, and Phragmites australis were collected from the Muan salt marsh. Ninety endophytic fungi were isolated from the roots of the collected halophytes. Molecular insights inferred by internal transcribed spacer containing ITS1, 5.8s, and the ITS2 region showed that all the fungal strains belong to ten orders, i.e., Capnodiales (4.44%), Cystofilobasidiales (1.11%), Dothideales (3.33%), Eurotiales (53.33%), Glomerellales (3.33%), Hypocreales (8.89%), Mucorales (1.11%), Pleosporales (15.56%), Sordariales (1.11%), and Trichosphaeriales (1.11%). The rest (6.67%) of all fungal isolates were not identified. Ninety fungal strains were confirmed at the genus level, containing Acremonium, Alternaria, Aspergillus, Aureobasidium, Cephalosporium, Chaetomium, Cladosporium, Colletotrichum, Cryptococcus, Didymella, Dothideomycete, Emericellopsis, Epicoccum, Eupenicillium, Fusarium, Gibberella, Gongronella, Macrophoma, Microsphaeropsis, Nigrospora, Paecilomyces, Paraconiothyrium, Penicillium, Phaeomyces, Phoma, Pleosporales, Purpureocillium, and Talaromyces. Of all the endophytic fungi identified from the various halophytes, Aspergillus and Penicillium of Eurotiales had the highest abundance.

Response of Flax Varieties to Fertilizer Levels (아마품종의 시비량 반응)

  • This study was conducted to find out the optimum fertilizer level for three flax varieties Wiera, Stoment-Goss and Taijungsun#l from the winter cropping on drained paddy field of new experimental field in Muan at 6 compositions of fertilizer levels. Maturation dates was delayed in dressing plots, that was delayed to 3 or 4 days in Wiera and Stoment -Goss, 1 or 2 days in Taijungsun#l and it was early in Wiera and Stoment-Goss, 1 or 2 days in Taijungsun#l and it was early in the non dressing plot. Stem length was longest, oil content was increased, stem and seed yield were heaviest at the compositions of fertilizer amounts with N-P$_2$O$\sub$5/-K$_2$O were 8-6-4 Kg/10a. Maturation dates, Stem length, Oil content, Stem yield and Seed yield showed significant difference between fertilizer level and showed positive correlation. Judging from the results reported above, in optimum fertilizer amounts for N-P$_2$O$\sub$5/-K$_2$O from the winter cropping on drained paddy field of new experimental field in Muan seemed to be 8-6-4 Kg /l0a

Temporal and Spatial Fluctuations of Coastal Water Quality and Effect of Small Tide Embankment in the Muan Peninsula of Korea (무안반도 연안수질의 시ㆍ공간적 변동과 소규모 방조제의 영향)

  • In this study, we estimated the seasonal fluctuations of water quality and effect of small tide embankment in coastal water around the Muan Peninsula, which is located in the northern part of Mokpo city, and layer farming ground is spread around there. Some physical and chemical factors were analyzed to characterize water quality from Jan. to Oct. in 1994. The results were as follows: Dissolved oxygen was slightly under saturation in the almost areas of July, and in some bottom layer at ebb tide of October. Distribution of COD showed high values that over 2㎎/L in October and flood tide of April by the discharge of freshwater and resuspension of benthic sediment, which exceeded water quality criteria II. Maximum values of dissolved inorganic nitrogen ware appeared in surface layer during the flood tide of October, while minimum of that showed in surface layer in April. Concentration of dissolved inorganic phosphorus was higher at July than the others, which ranged from 0.24 to 2.08㎍-at/L. Mostly mean values of N/P ratio were lower than 16, it mean that nitrogen is more limiting nutrient than phosphorus for the growth of phytoplankton. The values of eutrophication index were in the range of 0.07~0.81. However, very high values due to increase of COD were estimated near the tide embankment and southern part in relation to tidal current in October. Water quality around tide embankment was suddenly changed worse within a short period after opening the water gate during the rainfall.

The Evaluation of the Water Quality in Coastal Boundary on Tidal flat (통계분석기법을 이용한 전남 갯벌 해역 수질특성)

  • To understand characteristics of the water quality on the coastal boundary on tidal flat, field observations between 2008 and 2009 were undertaken twice a month at five coastal areas (Muan bay, Tando bay, Hampyeong bay, Shinan Jido and Yeongkwang coastal areas). Yearly water temperature difference was large with the range between $1.3^{\circ}C$ and $31.1^{\circ}C$. Salinity was about 32 but was the lower less than 20 for the heavy rainfall season. DO was high in winter and low in summer according to the variation of water temperature. pH represented the variation similar to DO. Suspended solid was averagely high over 100 mg/l in Yeongkwang coastal area, especially. COD did not revealed large variation with the value of about 1 mg/l. DIN and DIP concentration were high when freshwater was highly input in summer. DIN concentration was low for winter and early spring but DIP concentration did not show the seasonal variation with the continuous increase from July 2009 to December 2009. Chlorophyll a appeared high for spring with approximately $10\;{\mu}g/l$ and was higher for summer in Yeongkwang coastal area than other sites. The results of principal component analysis conducted to compare the characteristics of water quality observed in study areas showed the distinguishable features as follows. The freshwater input fluctuation appeared as the first factor in Muan and Tando bays, and the change of water temperature was the first factor in Shinan Jido and Yeongkwang coastal areas. The influence mixed with the variation of freshwater outflow and the change of water temperature in Hampyeong bay was to be the first factor.

Physicochemical Characteristics of Native Korean Allium wakegi Araki according to Different Seed Bulb Origins (종구산지에 따른 한국산 쪽파(Allium wakegi Araki)의 이화학적 특성)

  • The aim of this study was to investigate the physicochemical characteristics of native Korean Allium wakegi Araki samples, which were grown from seven local seed bulbs(Yesan, Muan, Anmyon-do, Deokjeok-do, Jeju-do, Yecheon, and China) to produce high quality native Korean Allium wakegi Araki. For the proximate composition of samples, moisture contents were in the range of 90.69~92.43%. The crude protein content of the Jeju-do sample was highest compared to the other samples. However, there were no significant differences in total sugar contents between samples. The seed bulb origin did not affect the hardness of the stem part, but was high for the leaves of the Yesan sample compared to the other samples. The results for anti-oxidative activity were as follows: Yesan(2.30 mg/mL) > China(2.51 mg/mL) > Muan (2.56 mg/mL) > Yecheon(2.74 mg/mL) > Jeju-do(2.85 mg/mL) > Anmyon-do(2.87 mg/mL) > Deokjeok-do(3.18 mg/mL). In terms of mineral and amino acid contents, the Yesan sample showed the highest levels, respectively, compared with the other samples. Food values such as contents of total phenolics and pyruvic acid were highest in the Jeju-do sample. These results show that the physicochemical characteristics of Allium wakegi Araki were significantly different according to different seed bulb origins.
