Hypoglycemic Effects of Crude Extracts of Moutan Radicis Cortex (목단피 추출물의 혈당 강하 효과)

  • Hypoglycemie effect of Moutan Radicis Cortex (MRC) extract contained in Yukmijihuang-hwan was determined by investigating insulin-sensitizing and ${\alpha}-glucoamylase-suppressing$ actions. MRC was extracted with 70% ethanol, fractionated by XAD-4 column chromatography with mixture solvent of methanol and water, and utilized for hypoglycemic effect assay. Significant insulin sensitizing activities of MRC extracts were observed in 3T3-L1 adipocytes, giving MRC extracts with 1 ng/mL insulin reach glucose uptake level increased by 50 ng/mL of insulin alone. MRC methanol extracts of 20, 40, 60, and 80% suppressed ${\alpha}-glucoamylase$ activity in vitro. Peak serum glucose levels and area under curve were lower in Sprague Dawley male rats treated with MRC ethanol extract than those treated with cellulose in oral glucose tolerance test using 2 g dextrin/kg body weight. These data suggest MRC extracts contain effective insulin -sensitizing and ${\alpha}-glucoamylase-suppressing$ compounds for hypoglycemic activity.

Studies on the Anti-apoptotic Effect of the Mudanpi (목단피가 세포고사의 억제에 미치는 영향에 관한 연구)

  • Mudanpi (Cortex Moutan Radicis; the root cortex of Paeonia suffruticosa Andrews) is an important Chinese crude drug used in many oriental prescriptions. 1,2,3,4,6-Penta-O-galloyl-beta-D-glucose (PGG), a major component of this crude drug, has been shown to possess potent antioxidant, anti-mutagenic and anti-proliferative effects. In this study, I examined whether PGG could protect Neuro 2A cells, a kind of neuronal cell lines, from oxidative damage through the induction of HO-1 expression and HO activity. Exposure of Neuro 2A cells to PGG (10-50μM) resulted in a concentration- and time-dependent induction of HO-1 mRNA, and protein expressions and heme oxygenase activity. PGG protected the cells from hydrogen peroxide-induced cell death. The protective effect of PGG on hydrogen peroxide-induced cell death was abrogated by zinc protoporphyrin IX (ZnPP IX), a HO inhibitor. These results indicate that PGG is a potent inducer of HO-1 and HO-1 induction is responsible for the PGG-mediated cytoprotection against oxidative damage.

Single Oral Dose Toxicity Study of WK-38 in Rats (랫트에서 WK-38의 단회경구투여 독성에 관한 연구)

  • Acute toxicity of WK-38, a herbal preparation for the atherosclerosis, was examined using male and female Sprague-Dawley rats. WK-38 is composed of Rhei Rhizoma, Magonoliae Cortx, Moutan Cortex Radicis. Rats were treated with the WK-38 intragastrically at 0 mg/kg, 5 mg/kg, 50 mg/kg, 500 mg/kg or 2,000 mg/kg and observed for two weeks. No mortality was observed at the doses used. Abnormal clinical signs such as eye bleeding, nasal bleeding and hyperemia had been shown temporary after administration. All rats were appeared to be healthy and normal during the 2 week observation. Also there was no difference in net body weight gain, gross pathological findings, and urine analysis among the groups rats treated with different doses of the WK-38.

Modification and Validation of Analytical Method for Oxypaeoniflorin and Paeoniflorin in Moutan Cortex Radicis Extract (목단피 추출물의 Oxypaeoniflorin 및 Paeoniflorin의 분석법 개선 및 검증)

  • The aim of this study was the validation of a modified analytical method for determination of oxypaeoniflorin and paeoniflorin in Moutan Cortex Radicis extract. For validation of the analytical method, we modified established analytical methods and validated improvement. For validation, the specificity, linearity, precision, accuracy, limit of detection (LOD), and limit of quantification of oxypaeoniflorin and paeoniflorin were measured by high performance liquid chromatography. The results show that the correlation coefficients of the calibration curve for oxypaeoniflorin and paeoniflorin were 1.0000 and 0.9998, respectively. The LOD for oxypaeoniflorin and paeoniflorin were $0.23{\mu}g/mL$ and $0.25{\mu}g/mL$, respectively. The inter-day and intra-day precision values of oxypaeoniflorin and paeoniflorin were 0.70~3.19% and 1.74~2.43%, and 0.32~0.92% and 0.62~2.28%, respectively. The inter-day and intra-day accuracies of oxypaeoniflorin and paeoniflorin were 98.33~102.11% and 97.72~118.12%, and 98.44~101.56% and 97.10~112.00%, respectively. Therefore, the analytical method was validated for the detection of oxypaeoniflorin and paeoniflorin in Moutan Cortex Radicis.

Effect of Moutan Cortex Radicis on gene expression profile of differentiated PC12 rat cells oxidative-stressed with hydrogen peroxide (모단피의 PC12 cell 산화억제 효과 및 neuronal 유전자 발현 profile 분석에 대한 연구)

  • Yukmijihwang-tang has been widely used as an and-aging herbal medicine for hundred years in Asian countries. Numerous studies show that Yukmijihwangtang has anti-oxidative effect both in vivo and in vitro. It has been reported that Moutan Cortex Radicis extract (MCR) was the most effective herb in Yukmijihwang-tang on undifferentiated PC12 cells upon oxidative-stressed with hydrogen peroxide. The purpose of this study is to; 1) evaluate the recovery of neuronal damage by assessing the anti-oxidant effect of MCR on PC12 cells differentiated with nerve growth factor (NGF), 2) identify candidate genes responsible for anti-oxidative effect on differentiated PC12 cells by oligonucleotide chip microarray. PC12 cells, which were differentiated by treating with NGF, were treated without or with hydrogen peroxide in the presence or absence of various concentration of MCR. Cell survival was determined by using MTS assay. Measurement of intracellular reactive oxygen species (ROS) generation was determined using the H2DCFDA assay The viability of cells treated with MCR was significantly recovered from stressed PC12 cell. In addition, wide rage of concentrations of MCR shows dose-dependent inhibitory effect on ROS production in oxidative-stressed cells. Total RNAs of cells without treatment(Control group), only treated with H₂O₂ (stressed group) and treated with both H₂O₂ and of MCR (MCR group) were isolated, and cDNAs was synthesized using oligoT7(dT) primer. The fragmented cRNAs, synthesized from cDNAs, were applied to Affymetrix GeneChip Rat Neurobiology U34 Array. mRNA of Calcium/calmodulin-dependent protein kinase II delta subunit(CaMKII), neuron glucose transporter (GLUT3) and myelin/oligodendrocyte glycoprotein(MOG) were downregulated in Stressed group comparing to Control group. P2X2-5 receptor (P2X2R-5), P2X2-4 receptor (P2X2R-4), c-fos, 25 kDa synaptosomal attachment protein(SNAP-25a) and GLUT3 were downregulated, whereas A2 adenosine receptor (A2AR), cathechol-O-methyltransferase(COMT), glucose transporter 1 (GLUT1), EST223333, heme oxygenase (HO), VGF, UI-R-CO-ja-a-07-0-Ul.s1 and macrophage migration inhibitory factor (MIF) were upregulated in MCA group comparing to Control group. Expression of Putative potassium channel subunit protein (ACK4), P2X2A-5, P2X2A-4, Interferon-gamma inducing factor isoform alpha precursor (IL-18α), EST199031, P2XR, P2X2 purinoceptor isoform e (P2X2R-e), Precursor interleukin 18 (IL-18) were downregulated, whereas MOO, EST223333, GLUT-1, MIF, Neuronatin alpha, UI-R-C0-ja-a-07-0-Ul.s1, A2. adenosine receptor, COMT, neuron-specific enolase (NSE), HO, VGF, A rat novel protein which is expressed with nerve injury (E12625) were upregulated in MCR group comparing to Stressed group. The results suggest that decreased viability and AOS production of PC12 cell by H₂O₂ may be, at lease, mediated by impaired glucose transporter expression. It is implicated that the MCR treatment protect PC12 cell from oxidative stress via following mechanisms; improving glucose transport into the cell, enhancing expression of anti-oxidative genes and protecting from dopamine cytotoxicity by increment of COMT and MIF expression. The list of differentially expressed genes may implicate further insight on the action and mechanism behind the anti-oxidative effects of herbal extract Moutan Cortex Radicis.

Effects of some herbal drugs on gingival fibroblast and periodontal ligament cellular activity (생약 제제가 세포활성도에 미치는 효과)

  • Healing of periodontal tissues require the migration and proliferation of gingival fibroblasts and periodontal ligament cells. There is many evidences that the some agents like cytokines and polypeptide growth factors are mediate these cellular events in wound healing. Recently someone is interested in herbal drugs on periodontal tissue healing processes. The purpose of this study was to examine the effects of 4 herbal drugs, Carthami Flis, Moutan Redias Cortex, Scirpi Rhisoma, Seed of Carthamus tinctorius L. on human gingival fibroblasts and periodontal ligament cells. Periodontal ligament cells and gingival fibroblasts were primarily cultured from extracted premolar with non-periodontal diseases. The powder from extracted. herbal drugs were prepared with distilled water. Cells were cultured with DMEM at $37^{\circ}C$, 5% $CO_2$, 100% humidity incubator, and treated with each herbal drugs with proper concentration for 1, 2, and 3 days. The cell activity was determined by ELISA reader using MTT assay. There was the most significant elevation in $10^{-3}g/ml$ of almost herbal drugs on cellular activities. The result of this study demonstrated that Carthami Flis, Moutan Radicis Cortex, Scirpi Rhisoma, Seed of Carthamus tinctorius L. appears to have beneficial effect on healing process after periodontal treatment.

Contents of Paeonol and Inorganic Components in Herbaceous Root-Bark of Paeonia moutan SIMS (목란근피(牧丹根皮)의 Paeonol 및 무기성분(無機成分) 함량(含量))

  • To get a fundamental data fur management of security, validity, Quality, of Paeonia suffruficosa roots as medicinal herb, the content of paeonol, ash, and inorganic components in Moutan Radicis Cortex produced in 8 regions of Korea was investigated from Oct. 1990 to May 1991. The results are summarized as follows: The average content of paeonol was 0.65%. The content of ash was 3.53%, acid-insoluble ash 0.77% in average, both of them were suitable to the standard value as Korean medical herbs. Total nitrogen content was 1.03%, phosphate 0.55%, potassium 0.82%, and calcium 1.50% in average. The content of MgO was 0.21ppm, 325.83ppm in Fe, 1.82ppm in Cr, 3.73ppm in Cu, 18.93ppm in Zn and Mn 21.27ppm, respectively. Especially Hg was not detected in the products of all regions. The average content of Cd was 0.25ppm, Pb 3.09ppm in Moutan Radicis Cortex, so they were estimated suitably to use as medicinal herb.

  • Objectives : Yukmi-Jihwang-Tang(YJT) has been used in Oriental Medicine as a drug for tonifying and nourishing yin. So, the purpose of this Study was to investigate effects of Yukmi-Jihwang-Tang extract(YJTE) on the T-lymphocytes and peritoneal macrophages in mice. Methods : YJTE consists of the following components; Rehmaniae Radix Preparata(熟地黃), Dioscoreae Rhizoma(山藥), Corni-Fructus(山茱萸), Hoelen alba(白茯), Moutan Cortex Radicis(牧丹皮), Alismatis Rhizoma(澤瀉). Result : The results of this Study were obtained as follow; The administration of YJTE did not affect T-lymphocytes apoptosis. YJTE decreased sub-population of TH and TC cells, and proliferation of T-lymphocytes too. But YJTE accelerated phagocytic activity and Nitric Oxide(NO) production from peritoneal macrophages in mice. Conclusions : These results suggest that the administeration of YJTE suppresses T-cell mediated immunity, but activates peritoneal macrophages.

Effects on Bacteria Growth of Grude Drugs in Korean Market (시판생약이 세균발육에 미치는 영향)

  • Effects on the bacteria gwoth of 96 kinds of crude drugs described in "Pen-Tsao-Kang-Mu" was investigated. Galla Rhois, Moutan Cortex Radicis, Aconiti Tuber, Euphorbiae Radix and PAeoniae Radix showed the antibacterial activity but Cnidii Rhizoma, Coicis Semen, Angelicae gigantis Radix, Asparagi Radix and Liriopes Tuber show the gwoth promoting action of all bacteria used in this experience. It can be seen that 10 kinds of crude drugs classified as poisonous plants inhibited the growth or had no effect by never promoted. 32 kinds of crude drugs as drugs acting on the gastrointestinal system showed antibacterial activity on E. coli $O_{55}$ and E. coli $O_{111}$. 18 kinds on inflammation showed antibacterial activity on Staphylococcus aureus and Sarcina lutea.

  • Keokhachukeu-tang is the one of Chinese traditional prescription used for the treatment of liver disease. This prescription consists of Carthami Flos (6g), Persicae Semen (9g), Pteropi Faeces (9g), Corydalis Tuber (9g), Moutan Radicis Cortex (6g), Paeoniae Radix rubra (9g), Angelicae gigantis Radix (9g), Cnidii Rhizoma (9g), Linderae Radix (12g), Cyperi Rhizoma (12g), Aurantii Fructus (9g), and Glycyrrhizae Radix (3g). Water extract of Keokhachukeu-tang showed a moderate hepatoprotective effect on tacrine-induced cytotoxicity in Hep G2 cells. (omitted)
