Characteristics of Mulberry Cultivar "Sangberry"(Morus alba L.) for Fruit Production (오디 생산용 뽕품종 "상베리"의 특성)

  • We bred a mulberry cultivar named Sangberry, through local adaptability test, which is under registration as a new cultivar for fruit production. Local adaptability test had been carried out at four places(Suwon, Kongju, Jangseong and Sangju) for six years from 2005. This is tetraploidy variety belonging to(Morus alba L.) made by colchicine treatment on growing point of winter buds. Sangberry was high yielding cultivar in fruit productivity by 70% compared to control cultivar "Chungilppong(Morus alba L.)" for four years. Although Sangberry was a little lower in sugar content of mulberry fruits, it contains more bioactive materials like C3G, rutin and amino acids than Chungilppong. It is adaptable to every where except the places where cold damage and sclerotic disease happen frequently.

Interaction Between time of Nodal Explant Collection and Growth Regulators Determines the Efficiency of Morus alba Micropropagation

  • The hormonal requirement suiting micropropagation of Morus alba during any season throughout the year was studied. Sprouting frequency from axillary buds of M. alba was greatly influenced by the time of explant collection, the highest value was achieved when nodal explants were collected at the end of bud dormancy period (late in March) and cultured on Murashige and Skoog (MS) medium supplemented with low concentration (0.5 mg/L) of BAP, kinetin or IBA (85-68%). In addition, they showed higher axillary bud sprouting on growth-regulators-free medium (49%) than others collected in autumn or winter and cultured on medium supplemented with various growth regulators (47-48%). Regardless of that period, young explants with greenish buds collected in summer exhibiting high sprouting frequency (66%) on MS medium supplemented with 0.5 mg/L kinetin and 0.5 mg/L GA3. Shoot multiplication via adventitious bud formation was achieved when the nodal explants were cultured on MS medium supplemented with 2 mg/L BAP and 0.2 mg/L IBA. Further multiplication via nodal explants of in vitro grown shoots was obtained on MS medium supplemented with 0.5 mglL BAP and 0.5 mg/L GA3. While half strength MS medium supplemented with low concentration (0.5 mg/L) of IBA, IAA or 2,4-D stimulated adventitious root formation, IBA was the best. After transfer the plantlets to the soil, acclimatization for three weeks was essential prerequisite for survival in high frequency (92%). Peroxidase activity is related to break of bud dormancy where maximum enzyme activity was detected when the lateral buds were induced to commence growth under field condition (early in spring) or in vitro.

Hypoglycemic Effect and Hepatic Detoxification Activity of Extracts from Crataegus fructus and Morus alba L. in Alcohol-treated Rats

  • To consider potentially new sources which have hypoglycemic effect and accelerating alcohol detoxification, this study was designed to investigate the effect of Crataegus fructus and Morns alba L. in alcohol-treated rats. I compared the body weight, glucose, aspartate aminotransferase (AST), alanine aminotransferase (ALT), alcohol dehydrogenase (ADH), and aldehyde dehydrogenase (ALDH) of rats administered both alcohol and extract of experimental plants to rats treated with alcohol alone. Administration of extracts of C. fructus and M alba, respectively, resulted in a significant reduction in the blood glucose level and the activities of ADH of liver compared to the control rats, and administration of extract of M. alba showed significantly lower on bodyweight gain in the rats than in other treated rats. In contrast, the activities of ALDH of liver were increased. The activities of AST and ALT between the only alcohol-treated rats and the alcohol and experimental plants-treated rats were no significant difference. The results suggest that C. fructus and M alba have a hypoglycemic effect, and reduce liver damage by accelerating acetaldehyde metabolism in alcoholic rats, so the combined effect of C. fructus and M alba may be considered as an alternative remedy for hangovers, alcohol-induced overweight and alcohol-induced diabetes.

  • In our country there are five species of Morus genus including Morus alba L.. Also their varieties and hybrids are distributed so much. In sucession of previous report we collected control and marketing specimens of Mori Cortex Radicis, comparative experiments were pharmacognostically carried out to identify the control specimen by the differences of external and internal morphology. It was difficult to identify marketing specimens by external morphology, because they are similar in spite of conparating with control specimen which the origin is definite. In internal morphology, medullary ray is developed near the cambium to primary bark in control specimen A(Morus alba series) and C(M. Lhou series), but less developed in B(M. bombycis series). The difference of these three series was observed. The thickness of cork layer is almost the same($7{\sim}12$ layers) in A and C series, but B is thin layer and sample E(that on the market) is generally more thick and has a stick cork cell. The kinds of starch, Ca-oxalate and latex, cell centents were same, but it was easy to identify them by the differences of their distribution. The bast fibre of D(wild specimen) and E were light lignified, latex tube of A and C series was richer distributed than others. These results show that the origin of Mori Cortex Radicis on the market can be appreciated in four groups of Korean Morus genus which are M. alba, M. bambycis, M. Lhou series and the others.

  • Ethanol 75% extracts of Morus alba Linne(Ma) and Sophora flavescens AITON(Sf) were fractionated by chloroform($CHCl_{3}$), ethyl acetate(EtOAc), butanol(BuOH) and water. Each fractionate was added to tryptic soy broth varying in extract levels and 5 strains of Listeria monocytogenes(Lm) were inoculated in them for comparing growth inhibition at $32^{\circ}C$. The $CHCl_{3}$ and EtOAc fraction of Ma were superior in growth inhibition of 5 strains but only $CHCl_{3}$ fraction of Sf showed comparatively higher inhibition to Lm ATCC 19113. Lm ATCC 19111 and 19113 were totally inhibited by 500 ppm of EtOAc fraction and Lm ATCC 19112, 19114 and 15313 were also absolutely inhibited by 1000 ppm of $CHCl_{3}$ fraction of Ma 75% ethanol extract.

Analysis of Functional Constituents in Mulberry (Morus alba L.) Twigs by Different Cultivars, Producing Areas, and Heat Processings

  • Four functional constituents, oxyresveratrol 3'-O-${\beta}$-D-glucoside (ORTG), oxyresveratrol (ORT), t-resveratrol (RT), and moracin (MC) were isolated from the ethanolic extract of mulberry (Morus alba L.) twigs by a series of isolation procedures, including solvent fractionation, and silica-gel, ODS-A, and Sephadex LH-20 column chromatographies. Their chemical structures were identified by NMR and FABMS spectral analysis. Quantitative changes of four phytochemicals in mulberry twigs were determined by HPLC according to cultivar, producing area, and heat processing. ORTG was a major abundant compound in the mulberry twigs, and its levels ranged from 23.7 to 105.5 mg% in six different mulberry cultivars. Three other compounds were present in trace amounts (<1 mg/100 g) or were not detected. Among mulberry cultivars examined, "Yongcheon" showed the highest level of ORTG, whereas "Somok" had the least ORTG content. Levels of four phytochemicals in the mulberry twigs harvested in early September were higher than those harvested in early July. Levels of ORTG and ORT in the "Cheongil" mulberry twigs produced in the Uljin area were higher than those produced in other areas. Generally, levels of ORTG and ORT in mulberry twigs decreased with heat processing, such as steaming, and microwaving except roasting, whereas those of RT and MC did not considerably vary according to heat processing. These results suggest that the roasted mulberry twigs may be useful as potential sources of functional ingredients and foods.

  • With extracts from Cheongmoknosang, anti-inflammatory effect was examined in LPS-stimulated Raw 264.7 cells. LPS (10 ng/ml) treatment increased the production of inflammatory cytokines, $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ but the ethanol extracts from Cheongmoknosang slightly decreased the production of $TNF-{\alpha}$ and also reduced the expression of iNOS and the production of COX-2. It seems that anti-inflammatory effects of ethanol extracts from Cheongmoknosang is partly due to the inhibition of iNOS and COX-2 expression by inhibiting nuclear translocation of $NF-{\kappa}B$ and AP-l in Raw 264.7 cells.

  • Flower buds of the mulberry(Morus alba L., Morus bombycis Koidz.) were cultured under different conditions such as basal media, and various concentrations of plant growth substances. Effects of the culture conditions on growth of the buds and organ regeneration were investigated and the result obtained are as follows: Murashige and Skoog(M.S.) medium was more effective on budding and growth of female(Keomseolppong) and male(Kaeryangppong) flower buds isolated directly from branches, compared to Greshoff & Doy(G.D.) and Wolter & Skog(W.S.) media. The growth of the female buds was promoted at higher concetration of benzyl amino purine(BAP) i.e., 2.0ppm. The female and male buds cultured after cuuting for seven days showed better growth than those without cutting treatment. The females and the males bloomed to form healthy stigmas and anthers, respectively, when cultured on M.S. media containing high Kinetin with low concentration of indole acetic acid(IAA).

Detection of Suitable Sites for Effective Cultivation of Morus alba L. (뽕나무의 효과적 재배를 위한 적지 탐색)

  • This study was conducted to scientifically analyze on the trends of cultivation area and production, and to detect the suitable site for Morus alba cultivation in forest of Buan-gun for effective cultivation. During last 3 years, the cultivation area of M. alba has steadily been increased, and the mulberry production of Jeollabuk-do has been researched as the greatest in Korea. Especially Buan-gun has the largest cultivation area of M. alba. On the other hand, as the result of detecting the suitable site for M. alba cultivation by factors of altitude, aspect, slope, soil texture, and organism content using GIS, there was the 663 ha suitable site of the total of 16,608 ha. In this area, Byeonsan-myeon, Sangseo-myeon, and Jinseo-myeon were included with the largest area, therefore, it was considered that these were the favorable regions for the expansion of M. alba cultivation area. The result of this study would be expected to be used as the effective information when establishing the expansion plan for M. alba cultivation area focused on the forest of Buan-gun in the future.
